Myeloid-specific SIRT1 Deletion Aggravates Hepatic Inflammation and Steatosis in High-fat Diet-fed Mice

  • Sirtuin 1 (SIRT1) is a mammalian $NAD^+$-dependent protein deacetylase that regulates cellular metabolism and inflammatory response. The organ-specific deletion of SIRT1 induces local inflammation and insulin resistance in dietary and genetic obesity. Macrophage-mediated inflammation contributes to insulin resistance and metabolic syndrome, however, the macrophage-specific SIRT1 function in the context of obesity is largely unknown. C57/BL6 wild type (WT) or myeloid-specific SIRT1 knockout (KO) mice were fed a high-fat diet (HFD) or normal diet (ND) for 12 weeks. Metabolic parameters and markers of hepatic steatosis and inflammation in liver were compared in WT and KO mice. SIRT1 deletion enhanced HFD-induced changes on body and liver weight gain, and increased glucose and insulin resistance. In liver, SIRT1 deletion increased the acetylation, and enhanced HFD-induced nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$), hepatic inflammation and macrophage infiltration. HFD-fed KO mice showed severe hepatic steatosis by activating lipogenic pathway through sterol regulatory element-binding protein 1 (SREBP-1), and hepatic fibrogenesis, as indicated by induction of connective tissue growth factor (CTGF), alpha-smooth muscle actin (${\alpha}$-SMA), and collagen secretion. Myeloid-specific deletion of SIRT1 stimulates obesity-induced inflammation and increases the risk of hepatic fibrosis. Targeted induction of macrophage SIRT1 may be a good therapy for alleviating inflammation-associated metabolic syndrome.

Pathological study on rabbit haemorrhagic disease in young rabbits

  • We investigated the pathological changes in young rabbits which were experimentally infected with rabbit hemorrhagic disease virus (RHDV). Experimental infection of RHDV was carried out in both thymectomized and non-thymectomized young immature rabbits and adult rabbits. None of young rabbits infected with RHDV died during the experiment. Histologically, single or focal hepatocellular degeneration and necrosis with mild lymphocyte infiltration were observed in the rabbits killed at 30 hours and 5 days PI. Lymphocyte infiltration was more severe at 5 days PI than at 30 hours PI. RHDV antigens were mainly detected in the degenerating hepatocytes adjacent to the infiltrated lymphocytes at 30 hours PI and 5 days PI. In electron microscopical observation, infiltrated lymphocytes in the lesions had large nuclei without cytoplasmic granules and interdigitated with adjacent hepatocytes. It is assumed that infiltrated lymphocytes in hepatic lesions in RHDV infected young rabbits are T-lymphocytes and originate from peripheral lymphoid organs or tissues rather than from thymus.

Dexamethasone 투여(投與)가 토끼 virus성(性) 간염(肝炎) 유기(誘起)에 미치는 영향(影響) (Effect of dexamethasone administration on the induction of viral hepatitis in native rabbit)

  • In order to know the effect of dexamethasone on the induction of the rabbit viral hepatitis, the pathological changes were observed in the native rabbits, 2 to 6 months old in age, that were injected by dexamethasone and liver emulsion of Angora rabbit naturally infected with viral hepatitis. The results were summarized as follows: The native rabbits injected by dexamethasone and liver emulsion were infected with viral hepatitis and died between 2 and 7 days after inoculation. Clinical signs and gross lesions were very similar to those of Angora rabbit naturally occurred, In microscopical findings, the hepatic lesions were characterized by peripheral necrosis of the lobules, and peripheral necrosis of the lobules with fatty changes of hepatic cells was occurred in a few cases. Perivascular lymphocytic infiltration in the central nervous system was observed in some cases, The lesions of the other organs were very similar to those of Angora rabbit naturally occurred. On the other hand, the native rabbits that were injected by only liver emulsion of Angora rabbit naturally infected with viral hepatitis were not infected with the disease except very few cases.

의약품의 병용투여에 관한 연구 (VII) (Studies on the Concurrent Administration of Medicines(VII))

  • These studies were attempted to investigate the preventive effect of Ganoderma lucidum extract administered concurrently with glutathione on the liver damage induced by carbon tetrachloride ($CCl_4$) in rats. S-GOT and S-GPT activities of all the pre-treatment groups were significantly decreased, as compared with those of the control intoxicated by $CCl_4$. The concurrent administrations of Ganoderma lucidum extract with glutathione (100+100mg/kg, 200+100mg/kg, and 400+100mg/kg, respectively) were more effective than the individual administrations. i.e., Ganaderma lucidtcm extract (100, 200 and 400mg/kg, respectively) and glutathione (100, 200 and 400mg/kg, respectively). On the determination of lipid-peroxidation in liver, the concurrent administrations of Ganoderma lucidum extract with glutathione (100+100mg/kg, and 200+100mg/kg, respectively) significantly reduced the liver TBA values. Although hepatic cellular necrosis and fatty acid deposit were remarkably increased by $CCl_4$ intoxication, the concurrent administration of Ganoderma lucidum extract with glutathione (200+100mg/kg) reduced the pathological changes of parenchymal cell necrosis and fatty changes around centrilobalar area of the control. These findings indicate that the concurrent administrations of Ganoderma lucidum extract with glutathione showed better improvements than the individual administrations of them in all pathological aspects, in particular, against hepatitis and hepatic necrosis due to the cellular necrosis and fatty infiltration.

Fine needle aspiration biopsy에 의한 소의 지방간진단(脂肪肝診斷) (Fine needle aspiration biopsy for the diagnosis of fatty liver in cattle)

  • 소의 지방간(脂肪肝)의 이환율(罹患率)을 조사(調査)하고 fine needle aspiration biopsy에 의한 지방간(脂肪肝)의 세포학적진단(細胞學的診斷)을 시도(試圖)하였다. 육안적(肉眼的) 또는 부유법(浮遊法)에 의한 간장지방함량(肝臟脂肪含量)에 기초(基礎)한 소의 지방간(脂肪肝)의 발생율(發生率)은 한우(韓牛)암소에서 0.30%, 유우(乳牛)암소에서 4.70%, 유우(乳牛)숫소에서 0.15%였다. 지방간(脂肪肝)은 육안적(肉眼的) 소견(所見)으로 간장(肝臟)이 종대(腫大)되고 변연(邊緣)이 둔하며 가볍고 창백(蒼白)에서 주황색(朱黃色) 색조(色調)를 보이나 간장(肝臟)의 색조(色調)와 지방간(脂肪肝)의 중증도간(重症度間)에 환상(桓常) 상관관계(相關關係)가 인정(認定)되지는 않았다. 대부분의 정상간장(正常肝臟)과 중도(重度)의 지방간(脂肪肝)을 제외(除外)하고는 간장(肝臟)의 지방침윤(脂肪浸潤)은 중심정맥주위(中心靜脈周圍)에서는 거대지방구(巨大脂肪球)와 변연부(邊緣部)에서는 소지방구(小脂肪球) 그리고 맥관부(脈管部)에 지방침윤(脂肪浸潤)의 소견(所見)을 보였다. 부유법(浮遊法)에 의한 간장지방함량(肝臟脂肪含量)과 비교(比較)한 세포학상(細胞學上)의 민감도(敏感度), 특이도(特異度), 정확도(正確度)는 정상(正常)에서 94.4%, 95.2%, 94.9%, 경도(輕度)에서 64.3%, 100%, 87.2%, 중등도(中等度)에서 100%, 83.39%, 82.2%, 그리고 중도(重度)에서 모두 100%였다. 세포학적(細胞學的) 소견(所見)은 조직학적(組織學的) 소견(所見)과 잘 일치(一致)하였다. Fine needle aspiraton biopsy에 의한 합병증(合倂症)은 임상적(臨床的)으로 인지(認知)되지 않았다. 결론적(結論的)으로 needle aspiration biopsy에 의한 세포학적(細胞學的) 진단(診斷)은 소의 지방간(脂肪肝) 진단(診斷)에 있어서 조직학적(組織學的) 기법(技法)과 비교(比較)했을 때 단순(單純)하고 신속(迅速)하며 안전(安全)하며 경제적(經濟的)인 방법(方法)으로 사료(思料)된다.

한국재래산양에서 발생한 리스테리아병 (A Case of Listeriosis Occurred in the Korean Native Goat)

  • A survey was performed on the outbreak of listeriosis in a Korean native goat in Animal Breeding Station of Gyeongsang National University, Chinju, Korea. Clinical signs in the affected goat were fever, dullness, inappetence, nasal discharge, slobbering, dysphagia, circling, incoordination, recumbency, unilateral facial paralysis, torticollis, dyspnea, spasmodic paddling movement of the limbs and death at 6 days after onset. No significant gross lesions were seen. The histopathological lesions were consisted of perivascular edema and cuffs by round cells and microabscess with infiltration of neutrophils, lymphocytes and macrophages in the cerebrum. Gram-positive organism was observed in the microabscess lesions. Listeria monocytogenes was isolated exclusively from the brain tissue by cultural examination and the lesions induced by experimental infection with the isolate were characterized by severe conjunctivitis in rabbit and hepatic necrotic foci in mouse. This seems to be the first report of listeriosis in Korean native goat.

Histologic Alterations in the Liver of Black Bengal Goats Infected with Fasciola gigantica

  • A total of 77 Black Bengal goats (67 females and 10 males) of 3.5 to 5.0 years old between 18 and 20 kg liveweights were used in this study. The whole liver with gall-bladder from respective carcass was collected at slaughter following fecal and postmortem examinations. Tissue sections of all lobes of each liver were prepared and stained with hematoxylin and eosin following a standard procedure. Results confirmed the presence of flukes surrounded by fibrous capsule in the liver. All the animals were suffering from chronic Fasciola gigantica infections. The blood vessels in most of the liver tissue were thickened due to proliferation of fibrous tissue around them. Focal infiltration of lymphocytes in the lobules, patches of focal accumulation of neutrophils and eosinophils were found in all the liver tissue. Proliferations of new bile ductules in the hepatic trinity around the bile ducts that were clogged by the flukes were also observed in most tissue sections.

  • The oral admistration of deer horn extract was proved to cause a considerable amelioration to the impairment of hepatic oxidative phosphorylation and to the decrease of ATP content in liver tissue induced by cholesterol ingestion. Deer horn extract was shown to accelerate considerably incorporation of $acetate-1-C^{14}$ into cholesterol in liver tissue and it tends to restore a decrease of GOT activity of liver tissue, caused by cholesterol administration. Histological examination showed that deer horn extract had a remakable preventive effect aginst fatty infiltration of organgs such as liver, heart, spleen, and adrenal gland of cholesterol given rabbits.

여주 추출물이 급성 납 중독된 흰쥐의 간과 콩팥에 미치는 효과 (Effects of Momordica Charantia L. Water Extracts on the Rat Liver and Kidney with Acute Toxicated by Lead)

  • 본 연구는 급성 납 독성에 대한 여주 추출물의 효과를 알아보기 위하여 시도되었다. 250 g 내외의 흰쥐를 대상으로 대조군, 납(1,000 ppm) 투여군, 납 투여 후 필리핀산과 일본산 여주 추출물을 5%와 10%로 희석하여 식수로 공급한 후 6주 후에 간과 콩팥 손상 억제 효과를 알아보기 위하여 생화학적 및 조직학적 실험을 실시하였다. 혈액중 AST, ALT와 BUN의 수치는 여주 추출물 투여군이 납 단독투여군보다 감소되었다. 간 조직의 광학현미경적 관찰에서 납 단독투여군은 간세포의 괴사와 염증세포의 침윤이 관찰되었으나 여주 추출물 투여군에서는 중심정맥 주위에 약간의 단핵세포의 조직 침윤만이 관찰되었다. 콩팥 조직의 광학현미경적 관찰에서 납 단독투여군은 콩팥토리 모세혈관의 위축과 괴사 현상이 관찰되었고 콩팥세관 주위 간질에 염증세포의 침윤이 관찰되었으나 여주 추출물 투여군에서는 일부 콩팥세관 주위에서 염증 세포의 침윤만이 관찰되었다. 이상의 연구 결과로 보아 여주가 납으로 유발된 간과 콩팥 손상을 보호하는 효과가 있는 것으로 사료되며 여주에 대한 다양한 연구가 필요할 것으로 사료된다.

실험적 간섬유화에서 Fat-storing Cell의 미세구조의 변화에 대한 연구 (Ultrastructural Changes of Fat-storing Cells in Experimental Hepatic Fibrosis)

  • 간 섬유화에서 fat-storing cell의 변화를 미세 구조학적 측면에서 관찰하고자, 사염화탄소를 mineral oil에 녹여 1 : 1로 회석한 후 Kg당 0.5 ml의 용량을 주 2회씩 12주간 흰쥐에 복강 주입하였다. 실험기간 동안 1주 간격으로 도살하고, 적출된 간조직을 광학현미경과 연역조직화학적 검사 및 전자현미경으로 관찰하여 다음과 같은 결과를 얻었다. 광학현미경적 소견상 소엽 중심주의 염증 반응 및 세포 침윤 현상은 사염화탄소 투여 후 1주부터 나타나서 4주 째 가장 심하였으며, 이시기에 desmin염색 반응 양성세포의 수도 증가되었다. 괴사 및 섬유화는 2주부터 나타나기 시작하여 6주부터 결절이 형성되기 시작하였고 괴사성 병변은 8주까지 지속되었다. 그러나 세포 침윤 정도는 감소되면서 desmin에 대한 반응성도 낮아졌고, 경변성 변화는 10주 이후부터 나타났다. 전자현미경적 소견상 fat-storing cell의 숫적 증가는 1주부터 관찰되었으며 지방적이 감소하고 rER이 팽창된 이행세포는 2주째 출현하여 4주에 가장 현저히 증가되었다. 섬유아세포는 3주부터 나타나기 시작하였는데, rER이 확장되었으며 지방적은 없거나 1-2개의 소적이 세포돌기에서 관찰되었다. 섬유화가 진행됨에 따라 이행세포 및 fat-storing cell은 감소하고 섬유아세포는 증가된 상태를 유지하였다. 이상의 결과로 보아 fat-storing cell은 간섬유화 과정에서 활성화되고 증식하여 형태학적으로 이행세포 및 섬유아세포로 전환함으로써, 정지상태에 있는 섬유아세포의 전구체로서 간섬유화에 중요한 역할을 하는 것으로 사료된다.
