한국인 다소비 채소의 에탄올 추출물이 3T3-L1 지방세포와 HepG2 간장세포의 지질축적, MCP-1과 Adiponectin의 분비에 미치는 영향 (Effects of Ethanol Extracts from Commonly Consumed Vegetables in Korea on Differentiation and Secretion of MCP-1 and Adiponectin in 3T3-L1 Adipocytes and Lipid Accumulation in HepG2 Hepatocytes)

  • 본 연구는 한국인이 다소비하는 채소 중 국민건강영양조사 자료와 관련연구 문헌을 통하여 9가지를 선정하고, 3T3-L1 지방전구세포와 HepG2 간장세포를 이용하여 채소의 80% 에탄올 추출물이 비만과 대사증후군 관련 지표에 미치는 영향을 조사하여, 다음과 같은 결과를 얻었다. 1. 3T3-L1 지방전구세포와 HepG2 간장세포에서 세포생존율은 고사리 0.01~0.1 mg/mL 농도에서, 나머지 8가지 시료는 0.02~0.2 mg/mL의 농도에서 세포생존율에 영향을 미치지 않았다. 2. 분화된 3T3-L1 지방세포의 Oil-red O 결과, 고사리를 제외한 8가지 시료는 한 가지 이상 농도에서 대조군에 비하여 유의적으로 지방분화 억제 효과를 보였으며, 시료에 따라 지방분화율을 25~90% 정도 감소시켰다. 그 중에서 쑥갓(0.02 mg/mL, 68%; 0.2 mg/mL, 61%)과 쑥(0.02 mg/mL, 57%; 0.2 mg/mL 90%)의 에탄올 추출물은 50% 이상의 지방분화 저해율을 보였다. 3. 3T3-L1 지방전구세포의 지방분화를 효과적으로 감소시킨 다소비 채소 7가지에 대하여 염증성 사이토카인인 MCP-1 단백질 분비를 측정하였을 때, 애호박, 부추, 쑥갓과 쑥은 대조군에 비하여 유의적으로 감소하였다. 4. Adiponectin의 단백질 분비는 7가지 시료에서 모두 유의적으로 감소하였으며, 유전자 발현은 애호박, 부추, 쑥갓과 쑥의 에탄올 추출물에서 대조군에 비하여 유의적으로 감소하였으며, 지방분화 관련 유전자인 PPAR-${\gamma}$의 발현은 쑥, aP2의 발현은 부추와 쑥의 에탄올 추출물에서 대조군에 비하여 유의적으로 감소하였다. 5. 비만에 의한 NAFLD의 모델로, HepG2 간장세포에 oleic acid를 처리하여 지방 독성을 유발한 후 채소의 80% 에탄올 추출물을 처리하였을 때 시료의 농도에 따른 지질축적 저해율의 차이는 있었지만, 9가지 시료 모두에서 간장세포의 지질축적을 저해하였다. 그 중 부추(0.02 mg/mL, 26%; 0.2 mg/mL, 29%)와 고구마줄기(0.02 mg/mL, 25%; 0.2 mg/mL, 20%) 에탄올 추출물은 처리한 농도 모두에서 감소 효과가 있었으며, 저농도에서 고농도 수준의 감소 효과를 보였다. 6. 지방과 간장세포의 지질축적 억제, 지방세포에서 과분비된 MCP-1의 감소, 지방분화관련 유전자의 발현 감소 효과가 있는 시료로 애호박, 부추, 쑥갓과 쑥이 선정되었다.

HepG2 cell에서 한인진, 울금, 차전자 추출물(ACP)의 항산화 및 항이상지질혈증 효과 (Antioxidant and Anti-dyslipidemic Effect of Artemisiae iwayomogii Herba, Curcumae longae Radix, and Plantaginis Semen Complex Extract(ACP) on HepG2 Cells)

  • This study was performed to investigate the antioxidant and antidyslipidemic effects of Artemisiae iwayomogii Herba, Curcumae longae Radix and Plantaginis Semen complex extract(ACP) on HepG2 cells. We measured total polyphenols, total flavonoids, radical scavenging activity, and ABTS radical scavenging activity of ACP to evaluate its antioxidant activity. HepG2 cells were treated with ACP. Then, we evaluated ROS production; intracellular GSH content; GPx, GR, SOD, and catalase activities; free fatty acids and MDA levels; and mRNA expression levels of ACAT1 and HMG-CoA reductase. Results: ACP contains polyphenols and flavonoids and increased the DPPH and ABTS radical scavenging activities in HepG2 cells in a dose dependent manner. Also, ACP significantly reduced ROS production in HepG2 cells compared to the control group and significantly increased the GSH content, and elevated the enzyme activities of GPx, GR, and catalase in HepG2 cells compared to the control group. In addition, ACP reduced the mRNA expression of ACAT1 and HMG-CoA reductase in HepG2 cells compared to that in the control group. Conclusion: These results suggest that ACP has an antioxidant effect and may suppress the expression of dyslipidemia - associated genes and thus may be useful for the improvement of dyslipidemia.

지골피, 동충하초, 가시오가피 복합추출물이 고포도당 조건에서 배양한 HepG2 세포의 당대사 관련 효소 활성에 미치는 영향 (Effects of Mixed Extract from Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus on Glucose-Regulating Enzymes of HepG2 in Hyperglycemic Conditions)

  • 본 연구는 지골피, 동충하초, 가시오가피의 복합추출물을 고포도당 조건(4.5 g/L)에서 배양한 HepG2 세포를 이용하여 당대사 관련 주요 효소인 glucokinase, acetyl CoA carboxylase의 활성을 RT-PCR과 western blotting 방법을 이용하여 측정하였다. 복합추출물을 HepG2 세포에 24시간 처리한 후 GK mRNA와 protein의 발현 양을 측정한 결과 각각 $168{\pm}0.04%$, $182.4{\pm}0.03%$로 증가하였으며, ACC 효소의 활성은 고포도당으로 배양하였을 때 ACC mRNA 및 protein 발현 양은 각각 $127.3{\pm}0.02%$, $126.7{\pm}0.24%$로 증가 하였다. 결론적으로 지골피, 동충하초, 가시오가피의 복합추출물이 GK 및 ACC mRNA 발현을 증가시킴으로써 항당뇨 효과를 나타냄을 관찰하였다.

Impact of Co-transfection with Livin and Survivin shRNA Expression Vectors on Biological Behavior of HepG2 Cells

  • Objective: To construct short hairpin RNA (shRNA) eukaryotic expression vectors targeting Livin and Survivin genes, and to explore the impact of co-transfection of Livin and Survivin shRNA expression vectors on the biological behavior of HepG2 cells. Methods: shRNA eukaryotic expression vectors pSD11-Livin and pSD11-Survivin were designed and constructed then transfected into HepG2 cells separately or in combination. mRNA and protein expression in transfected cells was assessed by quantitative fluorescence PCR and Western blotting, respectively. Cell proliferation was measured by MTT assay and cell apoptosis by TUNEL assay. Results: The Livin and Survivin shRNA eukaryotic expression vectors were successfully constructed and transfected into HepG2 cells. The relative mRNA expression levels of Livin and Survivin in HepG2 cells co-transfected with pSD11-Livin and pSD11-Survivin were $0.12{\pm}0.02$ and $0.33{\pm}0.13$, respectively, which was significantly lower than levels in cells transfected with either pSD11-Livin or pSD11-Survivin (P<0.05). The relative protein expression levels of Livin and Survivin in the co-transfected cells were also significantly decreased compared to single-transfection (P<0.05). The inhibition rate of cell growth in the co-transfection group was higher than that in the single-transfection groups at 48 h, 60 h, or 72 h after transfection (P<0.01). The apoptotic rate increased to the greatest extent in the co-transfection group relative to any other group (P<0.05). Conclusions: Co-transfection with pSD11-Livin and pSD11-Survivin was more efficient than transfection with either vector alone in reducing the mRNA and protein expression of Livin and Survivin genes in HepG2 cells. Co-transfection also inhibited the proliferation of transfected cells more than the other groups, and induced cellular apoptosis more effectively.

Does physiotherapy after rotator cuff repair require supervision by a physical therapist?: a meta-analysis

  • Background: A supervised physiotherapy program (SPP) is a standard regimen after surgical rotator cuff repair (RCR); however, the effect of a home-based exercise program (HEP), as an alternative, on postoperative functional recovery remains unclear. Therefore, the purpose of this meta-analysis was to compare the functional effects of SPP and HEP after RCR. Methods: We searched electronic databases including Central, Medline, and Embase in April 2022. The primary outcomes included the Constant score, American Shoulder and Elbow Surgeons score, University of California Los Angeles shoulder score, and pain score. Secondary outcomes included range of motion, muscle strength, retear rate, and patient satisfaction rate. A meta-analysis using random-effects models was performed on the pooled results to determine the significance. Results: The initial database search yielded 848 records, five of which met our criteria. Variables at 3 months after surgery were successfully analyzed, including the Constant score (mean difference, -8.51 points; 95% confidence interval [CI], -32.72 to 15.69; P=0.49) and pain score (mean difference, 0.02 cm; 95% CI, -2.29 to 2.33; P=0.99). There were no significant differences between the SPP and HEP. Other variables were not analyzed owing to the lack of data. Conclusions: Our data showed no significant differences between SSP and HEP with regard to the Constant and pain scores at 3 months after RCR. These results suggest that HEP may be an alternative regimen after RCR. Level of evidence: I.

Inhibition of SMP30 Gene Expression Influences the Biological Characteristics of Human Hep G2 Cells

  • Senescence marker protein 30 (SMP30), a hepatocellular carcinoma (HCe) associated antigen had been identified by our research group. To study its mechanisms of regulation and associations with the occurrence and development of HCe, we inhibited expression by RNAi technique, and observed effects on the biological characteristics of Hep G2 cells. In cell viability assays, cell growth in the experimental group (with siRNA transfection) was elevated. In Transwell invasion assays, compared with blank and control groups, numbers of invading cells in the experimental group were significantly increased, whereas in apoptosis assays, the percentage apoptosis demonstrated no differences, but after UV irradiation, that in the experimental group was higher than the other two groups. In a word, SMP30 can inhibit the proliferation and invasion of human hepatoma cells and thus can be regarded as a cancer suppressive factor.

Selective in vitro cytotoxicity of Hypericum hookerianum towards cancer cell lines

  • The methanol extracts of the aerial parts, leaves and stem of Hypericum hookerianum were tested for in vitro cytotoxicity on selected normal and cancer cell lines and anti tumor activity using DLA cells. Cell viability and morphological changes were assessed. Among the three extracts tested, the stem extract of Hypericum hookerianum showed potent cytotoxicity against HEp-2 and RD cell lines. The $CTC_{50}$(concentration required to reduce viability by 50%) of this extract was found to be $2.02\;{\mu}g/ml$ for RD cell line, $10.25\;{\mu}g/ml$ for HEp-2 cell line and $100.06\;{\mu}g/ml$ for Vero cell line. In the clonogenic assay, no colony formation was observed up to a concentration of $100\;{\mu}g/ml$. In the short term cytotoxicity studies using DLA cells, 50% viability was observed in the concentration range of $50-100\;{\mu}g/ml$ for aerial parts, $100-200\;{\mu}g/ml$ for stem and more than $200\;{\mu}g/ml$ for leaf extracts of Hypericum hookerianum. In the long-term activity using HEp-2 cell line, no colony formation was observed over a concentration of 200 mg/ml for the stem extract. Hypericum hookerianum stem extract was fractionated into petroleum ether, chloroform, ethyl acetate and methanol soluble fractions. The petroleum ether and chloroform soluble fractions showed higher cytotoxic activity against HEp-2 cell line when compared to the other two fractions. The methanol stem extract of Hypericum hookerianum has the potential for further investigation in animal models to determine its anti-tumor activity and to identify its active principles.

Alaternin and Emodin with Hydroxyl Radical inhibitory and/or Scavenging Activities and Hepatoprotective Activity on Tacrine-Induced Cytotoxicity in HepG2 Cells

  • The antioxidative and hepatoprotective potentials of two anthraquinones, alaternin (2-hydroxy-emodin) and emodin, to scavenge and/or inhibit hydroxyl radicals generated by the Fenton reaction and to protect tacrine-induced cytotoxicity in human liver derived HepG2 cells were evaluated, respectively. The inhibitory activity on hydroxyl radical generated in a cell-free chemical system (FeSO$_4$/$H_2O$$_2$) was investigated by a fluorescence spectrophotometer using a highly fluorescent probe, 2$^1$,7$^1$-dichlorofluorescein. The hydroxyl radical scavenging activity was determined by electron spin resonance spectroscopy using 5,5-dimethy-1-pyrroline-N-oxide as hydroxyl radicals trapping agents. Tacrine-induced HepG2 cell toxicity was determined by a 3-[4,5-dimethylthiazole-2yl]-2,5-diphenyltertrazolium bromide assay. Although the scavenging activity of alaternin on hydroxyl radical was similar to that of emodin in dose-dependent pat-terns, the inhibitory activity exhibited by the former on hydroxyl radical generation was stron-ger than that of the latter, with $IC_{50}$/ values of 3.05$\pm$0.26 $\mu$M and 13.29$\pm$3.20 $\mu$M, respectively. In addition, the two anthraquinones, alaternin and emodin showed their hepatoprotective activ-ities on tacrine-induced cytotoxicity, and the EC$_{50}$ values were 4.02 11M and 2.37 $\mu$M, respec-tively. Silymarin, an antihepatotoxic agent used as a positive control exhibited the EC$_{50}$ value of 2.00 $\mu$M. These results demonstrated that both alaternin and emodin had the simultaneous antioxidant and hepatoprotective activities.ies.

양춘사 추출물이 HepG2 세포에서 콜레스테롤 합성에 미치는 효과 (Effects of Amomum Villosum Extracts on Cholesterol Synthesis in HepG2 Cells)

  • Dried fruits of Amomum villosum Lour. have been used an korean medicine to treat digestive diseases for a long time. It has been reported that Amomum villosum extracts(AVE) have effects for diabetes and steatosis in experimental models. But we did not find the report about the cholesterol synthesis inhibition effects of AVE. The objective of this study is to clarify the inhibitory effect of AVE against oleic acid and glucose-induced hypercholesterolemia in HepG2 cells. The results show that AVE had a significant inhibitory effect against oleic acid and glucose-induced cholesterol accumulation. Those effects seem to be caused by inhibition of AVE on oleic acid and glucose-induced decrease of HMG CoA reductase, which is the rate-limiting enzyme for cholesterol biosynthesis in liver. It is believed that the results of this study can provide basic data for the drug and functional food development of hypercholesterolemia treatments.

양극산화 임플란트 표면에 적용된 헤파린과 골형성단백질(rhBMP-2)이 치조골 증대에 미치는 효과: 방사선학적 평가 (Effect of immobilization of the recombinant human bone morphogenetic protein 2 (rhBMP-2) on anodized implants coated with heparin for improving alveolar ridge augmentation in beagle dogs: Radiographic observations)

  • 연구 목적: 본 연구는 골형성단백질의 서방출을 위해 헤파린과 골형성단백질 (rhBMP-2)을 화학적으로 고정시킨 양극산화 티타늄 임플란트가 골 결손부에서 임플란트 주변의 수직적 골증대에 미치는 효과를 방사선학적으로 평가하고자 시행되었다. 연구 재료 및 방법: Pure-Titanium을 사용하여 길이 7.0 mm, 직경 3.5 mm의 실험용 임플란트 18개를 제작하였다. 모든 임플란트를 양극산화처리 하였고, 플랫폼 하방 2.5 mm에 식립 기준선을 표시하였다. rhBMP-2가 코팅되지 않은 임플란트 집단을 대조군으로, dip and dry 방법으로 rhBMP-2를 물리적 흡착시킨 집단을 BMP군, 3,4-dihydroxyphenylalanine(DOPA)-heparin을 이식하고 rhBMP-2를 화학적으로 고정시킨 집단을 Hep-BMP군으로 설정하였다. 각군별6개씩의 임플란트를 3마리의 비글견 양측 하악에 한쪽에 3개씩 총 18개를 치조정 상방으로 2.5 mm 노출시켜 식립하였다. 식립 직후와 4주, 8주에 식립부위의 방사선학적 검사가 시행되었고, 각 시기별, 각 군별 임플란트의 근원심 변연골의 수직적 재생량에 대한 평균값과 표준편차를 얻었다. Kruskal-Wallis test와 Mann-Whitney U test를 이용하여 4주, 8주에서 대조군과 실험군들의 차이를 비교 분석하고, 유의 수준5%에서 통계적으로 검정하였다. 결과:방사선학적 관찰 결과 임플란트 근원심 변연골 재생량(평균값 ${\pm}$ 표준편차)은 4주에 대조군은 $0.09{\pm}0.22mm$, BMP군은 $1.02{\pm}0.72mm$, Hep-BMP군은 $1.29{\pm}0.51mm$ 였으며, 8주에서는 각각 $0.11{\pm}1.26mm$, $1.11{\pm}0.58mm$, $1.59{\pm}0.79mm$였다. 두 실험군 모두 4주와 8주에서 대조군과 비교 시 유의한 수직적 골증대를 나타냈으나(P<.05), Hep-BMP군과BMP군의 비교에서는 유의한 변연골 재생량 차이를 보이지 않았다(P>.05). 결론: 골형성단백질을 물리적으로 흡착시키거나 서방출 위해 헤파린을 이용하여 화학적으로 고정시킨 양극산화 임플란트 표면은 모두 골 결손부에서 임플란트 주변골의 수직적 증대에 효과적이었다. 그러나 방사선학적 관찰의 한계 내에서 골형성단백질의 이 두가지 적용방법간에는 수직적 골증대량에 유의한 차이가 없었다.
