The complement system is a part of the natural immune regulation mechanism against invading pathogens. Complement activation from three different pathways (classical, lectin, and alternative) leads to the formation of C5-convertase, an enzyme for cleavage of C5 into C5a and C5b, followed by C6, C7, C8, and C9 in membrane attack complex. The C9 is the last complement component of the terminal lytic pathway, which plays an important role in lysis of the target cells depending on its self-polymerization to form transmembrane channels. To address the association of C9 with traits related to disease resistance, the complete porcine C9 cDNA was comparatively sequenced to detect single nucleotide polymorphisms (SNPs) in pigs of the breeds Hampshire (HS), Duroc (DU), Berlin miniature pig (BMP), German Landrace (LR), Pietrain (PIE), and Muong Khuong (Vietnamese potbelly pig). Genotyping was performed in 417 $F_2$ animals of a resource population (DUMI: $DU{\times}BMP$) that were vaccinated with Mycoplasma hyopneumoniae, Aujeszky diseases virus and porcine respiratory and reproductive syndrome virus at 6, 14 and 16 weeks of age, respectively. Two SNPs were detected within the third exon. One of them has an amino acid substitution. The European porcine breeds (LR and PIE) show higher allele frequency of these SNPs than Vietnamese porcine breed (MK). Association of the substitution SNP with hemolytic complement activity indicated statistically significant differences between genotypes in the classical pathway but not in the alternative pathway. The interactions between eight time points of measurement of complement activity before and after vaccinations and genotypes were significantly different. The difference in hemolytic complement activity in the both pathways depends on genotype, kind of vaccine, age and the interaction to the other complement components. These results promote the porcine C9 (pC9) as a candidate gene to improve general animal health in the future.
In order to isolate substances that inhibit the hemolytic activity of human serum against eryth-rocytes, we have evaluated whole plants of the Orostachys japonicus species with regard to its anti-complement activity, and have identified its active principles following activity-guided isolation. A methanol extract of the O. japonicus, as well as its n-hexane soluble fraction, exhibited significant anti-complement activity on the complement system, which was expressed as total hemolytic activity. A bioassay-guided chromatographic separation of the constituents resulted in the isolation of three known compounds 1-3 from the active n-hexane fraction. The structure of these compounds were analyzed, and they were identified as hydroxyhopanone (1), $\beta-sitosteryl-3-O-\beta-D-glucopyranosyl-6'-O-palmitate$ (2), and $\beta-sitosteryl-3-O-\beta-D-glucopyranoside$ (3), respectively. Of these compounds, compound 2 exhibited potent anti-complement activity $(IC_{50}=1.0\pm0.1{\mu}M)$ on the classical pathway of the complement, as compared to tiliroside $(IC_{50}=76.5\pm1.1{\mu}M)$, which was used as a positive control. However, compounds 1 and 3 exhibited no activity in this system.

Seven known oleanolic acid glycosides (1-7) were isolated form the MeOH extract of Tiarella polyphylla. The structures were identified to be 3-O-($\beta$-glucopyranosyl) oleanolic acid (1), 3-O-[$\beta$-D-glucopyranosyl-(1 3)-$\beta$-D-glucopyranosyl] oleanolic acid (2), 3-O-D-[$\beta$-D-glucopyranosyl-(1 2)-$\beta$-D-glycopyranosyl] oleanolic acid (3), 3-O-[$\beta$-D-glucopyranosyl-(1 3)-$\beta$-D-glucopyranosyl] oleanolic acid 28-O-$\beta$D-glucopyranosyl ester (4), 3-O-[$\beta$-D-glucopyranosyl-(1 2)-$\beta$-D-glucopyranosyl] oleanolic acid 28-O-$\beta$-D-glucopyranosyl ester (5), 3-O-[a-L-rahmnopyranosyl-(1 3)-$\beta$-D-glucururonopyranosyl] oleanolic acid (6), and 3-O-[$\alpha$-L-rhamnopyranosyl-(1 3)-$\alpha$-D-glucuronopyranosyl] oleanolic acid 28-O-$\alpha$-D-glucopyranosyl ester (7) on the basis of physicochemical and spectral data. These triterpene glycosides were tested for the anti-complement activity and hemolytic activity. Bisdesmosidic saponins, 4, 5, and 7, showed anti-complement activity; in contrast, monodesmosidic saponins, 1-3, and 6, showed direct hemolytic activity. Methyl esterified monodesmosidic saponins showed anti-complement activity at a low concentration and hemolytic activity at a high concentration.
경기도 광릉에서 채집한 자실체로부터 분리한 Lentinus edodes IY105균을 배양하여 얻은 균사체를 알칼리로 추출하여 얻은 물질이 면역계와 sarcoma 180이 이식된 mouse에 미치는 영향을 검토하였다. Lentinus edodes IY105 배양균사체의 알칼리 추출물은 항보체활성과 alternative complement hemolytic 활성을 가지고 있었고, carbon clearance 활성을 가지고 있었고, carbon clearance 활성을 나타내는 것으로 보아 mouse의 망내계에 존재하는 macrophage를 활성화시켜 이 물질에 대한 탐식기능을 증가시킴을 알수 있었다. 또한, 이물질은 mouse의 용혈반 형성 세포수를 증가시키는 것으로 보아 항체생산 자극효과가 있는 것으로 추측되었으며, 종양세포인 sarcoma 180이 이식된 mouse의 암을 50 정도 억제하는 효과를 나타내었다. 알칼리 추출물의 성분을 분석한 결과, 당 17, 단백질 42로 구성된 단백질다당체가 주성분으로 단백다당체는 16종의 아미노산과 52종류의 당으로 구성되어 있었다.
본 연구는 담배가 면역계에 미치는 영향을 탐색하는 일환으로 한국산 각종 담배가 가토보체의 활성에 미치는 영향을 조사하였다. 담배를 veronal buffered saline으로 추출한 후 이를 가토보체와 0.1 : 1의 비율로 혼합한 후 $37^{\circ}C$에서 60분간 반응시켜 반응전후의 보체역가를 보체 용혈검사로 측정하였다. 담배 A 의 경우 60분간 반응후에는 보체역가가 55.92% 감소되었으며 담배 B 는 48.62%, 담배 C 는 42.62%, 담배 D 는 49.12%, 담배 E 는 42.26%, 담배 F 는 53.26%, 담배 G 는 71.96%, 담배 H 는 60.90%, 담배 I 는 61.14% 감소되었다. 한국산 담배 9종 모두가 보체역가를 적게는 42.26% 크게는 71.96% 감소시킨 것으로 보아 담배 모두가 정도의 차이는 있지만 보체를 활성화시켰다.
천연으로부터 보체 저해활성 물질을 선발할 목적으로 용혈성 보체측정법(hemolytic complement assay, $CH_{50}$)을 이용한 미량 탐색법을 확립하였다. 보체결합 반응은 microplate에서 수행하였으며, 표준 용혈(50% hemolysis)을 설정하기 위한 최적조건은 classical pathway (CP)의 경우 양 적혈구 농도를 $5.0{\times}10^8\;cells/ml$로 하였을때 hemolysin 희석농도 $1/75{\sim}1/100$ 및 보체 희석농도 $1/80{\sim}1/120$이었으며, alternative pathway (AP)의 경우는 토끼 적혈구 농도 $4.0{\times}10^8\;cells/ml$에서 보체 희석농도 l/5, EGTA 4mM 및 $Mg^{2+}\;4{\sim}8mM$ 이었다. 비수용성 화합물의 검정에는 dimethyl sulfoxide를 1% 수준이 되도록 시료의 농도를 조절하여 첨가하였다. 본 실험 조건에서 3종의 phenylpropanoid는 모두 농도에 비례하여 항보체 활성을 나타내었는데, 그중 rosmarinic acid의 항보체 활성은 CP의 경우 $0.063{\sim}0.5mM$에서 $5.4{\pm}3.6%{\sim}95.8{\pm}0.2%$, AP의 경우 $0.063{\sim}1\;mM$에서 $35.1{\pm}0.9%{\sim}95.6{\pm}1.1%$으로 측정되었다.
식물성 식품재료를 대상으로 보체 용혈분석법$(TCH_{50})$ 을 이용하여 보체 활성화능(항보체 활성)에 대한 검색을 실시하였다. 총 38종의 식용 식물추출물 중 5종의 시료에서 대조구에 비해 60% 이상의 $TCH_{50}$ 감소를 일으키는 비교적 높은 활성을 나타냈으며 그 활성의 순서는 $1000{\mu}g/ml$ 시료 농도에서 생강>토란대>냉이>은행잎>달래이었다. 한편 가장 높은 활성을 보였던 생강에서 조제된 ZR-1의 경우 pronase 소화 후에는 활성의 변화가 없는 반면에 과요오드산 산화에 의해서는 급격한 활성의 감소를 나타냄으로써 ZR-1의 단백질 부위가 아닌 다당 부위가 활성에 기여함을 알 수 있었다. 또한 ZR-1의 항보체 활성은 $Ca^{2+}$ 이온 부재시 부분적인 감소현상을 보였으며, ZR-1을 정상인의 혈청과 반응 후 anti-humanC3를 이용하여 2차원 면역 전기영동을 행한 결과 C3의 분해산물을 관찰할 수 있었다. 또한 이들 획분은 $ACH_{50}$의 저해를 일으켰다. 동 결과로부터 ZR-1의 보체 활성화 양식은 classical pathway 뿐만 아니라 alternative pathway도 경유함을 알 수 있었다.
홑파래로부터 황산기를 함유한 다당체를 크로마토그래프로 분리정제하여 rhamnan sulfate가 항보체 활성화에 미치는 영향을 조사하였다. 항보체 활성능력은 F-4-3 획분을 비교군으로 Heparin H-180, Dextran과 비교해 결과 비교군보다 높았고, C4a와 C3a의 C convertase의 형성과 기능을 F-4-3 획분이 억제하였다. 이러한 보체 활성화 양식은 classical pathway 및 alternative pathway로도 경유함을 알 수 있었다.
김치 및 발효유 제품으로부터 분리한 유산균과 공시균주 12종을 대상으로 보체 용혈 분석법을 이용하여 면역계에서 중요한 역할을 담당하고 있는 보체계 활성화(항보체 활성, $TCH_{50}$) 정도를 측정한 결과, 김치로 부터 분리한 Lactobacillus plantarum이 타 유산균 종에 비해 높은 활성을 나타내었다. 이들 균주로 부터 조제된 세포벽 획분의 경우 세포질 획분보다도 높은 활성을 보였으며 각 획분의 활성은 농도 의존적 경향을 나타내었다. L. plantarum의 세포질 획분과 세포벽 획분의 경우 pronase 소화 후에는 활성의 변화가 없는 반면, 과요오드산 처리에 의해서는 급격한 활성의 감소를 나타내는데 이들 결과로부터 L. plantarum의 세포질과 세포벽 획분에 의한 보체계 활성화가 주로 다당 영역에 기인함을 알수 있었다. 한편 anti-human C3를 이용한 2차원 면역전기영동에 의해, $Ca^{++}$ 이온을 제거한 상태에서도 세포질과 세포벽 획분에 의한 C3 활성화 산물을 동정할 수 있었다. 또한 L. plantarum의 세포벽 획분에 의한 항보체 활성은 동일 조건에서 활성을 유지한 반면, 세포질 획분에 의한 활성화 정도는 동일 조건에서 상당히 감소하였다. 이상의 결과로부터 L. plantarum 세포벽 획분의 보체계 활성화 양식은 주로 alternative pathway의 활성화에 의한 것이며, 세포질 획분에 의한 활성화는 classical pathway와 alternative pathway 양 경로를 경유함을 알 수 있었다.
한약재를 대상으로 용철 보체 분석법$(TCH_{50})$을 이용, 항보체 활성물질에 대한 전반적인 검색을 실시하였다. 총 55종의 시료의 열수 추출물을 대상으로 동 활성을 측정한 결과, 대조구에 비해 70%이상의 $TCH_{50}(total\;complement\;hemolysis\;of\;50%)$의 감소를 일으키는 비교적 강력한 항보체 활성을 소유한 울금, 대복피, 조각자, 화살나무, 가시오갈피, 인동, 부자, 계피 등 9종의 시료를 선택할 수 있었다. 이들은 pronase 소화 후 활성을 유지한 반면, $NaIO_4$ 산화에 의해 급격한 활성의 감소를 나타냄으로써, 그 활성의 본체가 다당에 기인함을 알 수 있었다. 또한 가장 높은 활성을 보였던 대복피(Areca catchu) 다당의 경우 $Ca^{++}$ 이온 부재시 부분적으로 활성의 감소를 보였으며, 정상인의 혈청과 반응시 C3의 분해산물을 면역 전기 영동법에 의해 확인할 수 있었는 바, 동 사실로부터 대복피의 보체 활성화 기구는 classical pathway뿐만 아니라 alternative pathway도 경유함을 알 수 있었다.
