Water temperature (WT) is a major environmental factor of metabolic rate in fish; it directly affects food intake, ammonia excretion, oxygen consumption (OC), growth, and survival. Thus, the purpose of this study was to investigate changes in the OC and the hematological response of olive flounder Paralichthys olivaceus because of WT changes. In Exp. I, WT was increased from 20℃ to 29℃ within 18 h and maintained at 29℃ for 96 h. Then, WT was decreased from 29℃ to 20℃ within 18 h and maintained at 20℃ for 24 h. In Exp. II, WT was decreased from 20℃ to 11℃ within 18 h and maintained at 11℃ for 96 h. Then, WT was increased from 11℃ to 20℃ within 18 h and maintained at 20℃ for 24 h. The Exp. III maintained that the Exp. I and II was consecutively. In Exp. I, the OC increased from 116.7 mg O2 kg-1 hr-1 to 317.5 mg O2 kg-1 hr-1 with increasing WT. In Exp. II, the OC decreased from 96.5 mg O2 kg-1 hr-1 to 71.3 mg O2 kg-1 hr-1 with decreasing WT. In Exp. III, OC tended to increase or decrease in inverse proportion to temperature. In Exp. I, cortisol, glucose, and aspartate aminotransferase (AST) values increased with increasing WT. In Exp. II, Cl-, osmolality, AST, and alanine aminotransferase (ALT) values significantly changed during the experimental period: glucose values increased, whereas cortisol values decreased with decreasing WT. Exp. III was shown to be a more stressful environment to olive flounder than Exp. I and Exp. II. The results of our study will be useful for evaluating current aquaculture procedures of olive flounder and developing techniques to minimize stress in aquaculture farms.
Magnesium (Mg) plays an essential role in physiological and metabolic reactions. Recently, there has been an increased interest in the role of Mg deficiency, particularly the relationship between serum Mg value and inflammatory response. This study was designed to determine the relationship between serum Mg deficiency with inflammatory response, electrolytes and hematological alteration over long-term periods. Sixteen male Sprague-Dawley rats were divided into two groups: control (n=8), and Mg deficiency group (MgD group, n=8). Chow and normal water (tap water) were regularly provided to the control group and Mg-depleted chow and third distilled water were regularly provided for 60 days to the MgD group. Body weights, Serum Mg, $K^+$, inorganic phosphorus (IP) and total iron binding capacity (TIBC) levels in the MgD group were lower than those of the control group (P<0.05). Granulocyte fraction and MCV, RDW and PDW levels were higher, whereas lymphocyte fraction, erythrocyte, hemoglobin and MCHC levels were lower in the MgD group than in the control group (P<0.05). MCP-1 and TNF-${\alpha}$ levels in the MgD group were greater than those of the control group (P<0.05). In conclusion, the results of the present study suggest that Mg deficiency over a long-term period had not altered total leukocyte concentration in the blood, but had detrimental effects, including disturbances of electrolytes balance, disturbance of iron indices, potential anemia and elevation of pro-inflammatory cytokine. However, further studies should be performed to determine the relationship between serum Mg deficiency and major organ damage or alteration.
In order to investigate the of effects of nalbuphine on immune system in mice, we examined the various immunological parameters. After single oral administration of nalbuphine (130, 260, 390 mg/kg, i.p.) to female ICR mite, the weights of bodies and organs (thymus, spleen, liver, kidney), and hematological parameters were examined on day 2, 4, 6, and 8. The increased rate of body weight, relative weight of organ, and hematological parameters in nalbuphine -treated groups, were not significantly changed when compared with control group. However, number of WBC was decreased by the treatment of nalbuphine. To assess the effects of nalbuphine on humoral immune responses, splenic IgM plaque forming cell (PFC) and serum IgM were assayed. When nalbuphine wat administered after immunization with SRBC, but not before immunization, splenic IgM PFC and ,serum IgM level against SRBC were significantly lowered in a dole -dependent manner. These results indicate that the suppressive effects of nalbuphine on primary humoral immune response may be dependent on the timing of its administration relative to the initial antigenic sensitization.
The purpose of this investigation was to study the hematological response of Saudi Arabian Baladi fowl to protein rearing regimens. Males and females were subjected to the following 4 protein rearing regimens: conventional, C; reverse protein, RP; 2 single-stage low protein, $SS_1$ and $SS_2$ using 15% and 12% CP diets, respectively. Regimen effect was highly significant ($$p{\leq_-}.01$$) on BW, PCY, TP and U-Ac and significant ($$p{\leq_-}.05$$) on TL. Serum chol levels were not affected by regimen. In general $SS_{2}$ birds showed the lowest values for all parameters studied, except for PCV. However, the differences were not significant in each case. Age and sex effects were highly significant ($$p{\leq_-}.01$$) for all parameters, however, the regimen X sex interaction was not significant except for PCV. Regimen X age interaction, on the other hand, was highly significant ($$p{\leq_-}.01$$) only for BW, TP and U-Ac concentrations. The data may suggest that low levels of protein in the rearing regimen is an important factor influencing levels of the blood parameters studied. The data also indicate a lack of clear relationship between hen-day egg production and the blood parameters studied.
The effects of feeding frequency on the growth, food consumption, body composition, and hematological response of the Korean rockfish Sebastes schlegelii were investigated for 77 days at ambient water temperatures (17.2-24.5℃) in a sea cage in Tongyeong, Korea. Three replicate groups of fish were hand-fed to satiation with a commercial diet in one of four different feeding frequency trials (one meal every 2 days, and one, two and three meals per day). At the end of the experiment, the mean weight gain, specific growth rate, and daily feed intake of fish fed one and two meals per day were significantly higher than those of fish fed one meal every 2 days or three meals per day. The feed efficiency of the fish fed three meals per day was significantly lower than that of the fish in the other groups. The glucose concentration of fish fed one meal every 2 days was significantly higher than that of the other groups. We conclude that the optimum feeding frequency for improving the growth of Korean rockfish weighing 100-200 g reared in sea cages is one meal per day under our experimental conditions.
Objective: To investigate the efficacy and safety of voriconazole in treating Chinese patients with hematological malignancies and invasive aspergillosis. Methods: From March 2007 to April 2012, patients with diagnoses confirmed by CT, GM test and/or PCR assays, were recruited into this study. Aspergillosis of all patients were treated with voriconazole 6 mg/kg intravenous infusion (iv) every 12 h for 1 day, followed by 4 mg/kg IV every 12 h for 10-15 days; Then, switch to oral administration that was 200mg every 12h for 4-12 weeks. Efficacy and safety were evaluated according to Practice Guideline of Infectious Diseases Society of America. Results: The overall response rate of 38 patients after voriconazole treatment was 81.6%. The median time to pyretolysis was 4.5 days. Treatment related side effects were mild and found in only 15.8% of cases. No treatment related deaths occurred. Conclusions: Voriconazole can considered to be a safe and effective front-line therapy to treat patients with hematological malignancies and invasive aspergillosis. Alternatively it could be used as a remedial treatment when other antifungal therapies are ineffective.
The acute toxicity of Paecilomyces sinclairii was tested in beagle dogs. We daily examined clinical signs, body weights, and hematological/biochemical examinations for 14 days after administration of Paecilomyces sinclairii with different dose levels (0, 0.4, 2 and 10 g/kg). There were no clinical signs and no significant changes in hematological and biochemical analysis. These results showed that Paecilomyces sinclairii did not induce any remarkable acute toxic response and the $LD_{50}$ was greater than 10 g/kg in beagle dogs.
The experiment was conducted for 210 days to determine the effect of feeding, and starvation, and exposure to sodium nitrite ($NaNO_2$) on the survival, physiological changes, hematological parameter, and stress response of Far Eastern catfish, Silurus asotus. The survival of the starved group was lower than that of the fed group during the experiment. Starvation resulted in retardation of growth, which provides an example of fish that failed to continue to grow and remain in a good condition. Blood analyses (cortisol and glucose) showed significant differences of stress response between the fed and starved groups exposed to $NaNO_2$ at the conclusion of the experiment (p<0.05). These results suggest that all nutritional parameters used for starvation and feeding with $NaNO_2$ stress in this experiment appear to be a useful index of nutritional status in Far Eastern catfish.
Background: Dietary protein requirements are dependent on a variety of factors and water temperature is one of the most important abiotic factors affecting protein requirement of fish. This study was, therefore, conducted to investigate effects of water temperature on dietary protein requirement of fry Heteropneustes fossilis which has high demand in most of the Asian markets. Methods: Quadruplicate groups of 30 fish per treatment (2.97 ± 0.65 cm; 5.11 ± 0.34 g) were fed seven isoenergetic diets (17.9 kJ g-1 gross energy; 14.99 kJ g-1 digestible energy) containing dietary protein levels ranging from 28 to 52% at two water temperatures (18 and 26 ℃). Experimental diets were fed to apparent satiation as semi-moist cakes thrice daily at 17:00, 12:00, and 17:30 h for 12 weeks. For precise information, various growth parameters, protein deposition, hematological parameters, metabolic enzymes, and stress response were analyzed, and effects of water temperature on dietary protein requirement was recommended on the basis of response from above parameters. Results: Groups held at 26 ℃ attained best growth, feed conversion, and protein deposition at 44% dietary protein indicating that temperature affected dietary protein requirement for optimum growth of H. fossilis fry and protein requirement seems to be satisfied with 44% dietary protein. Interestingly, interactive effects of both dietary protein levels and temperature were not found (P > 0.05). Fish reared at 18 ℃ had comparatively higher values for aspartate and alanine transferases than those reared at 26 ℃ water temperature which exhibited normal physiological value for these enzymes indicating that body metabolism was normal at this temperature. Hematological parameters also followed same pattern. Furthermore, fish reared at 26 ℃ water temperature exhibited more resistant to thermal stress (P < 0.05). The 95% maximum plateau of protein deposition data using second-degree polynomial regression analyses exhibited dietary protein requirement of fry H. fossilis between 40.8 and 41.8% of diet at 26 ℃ water temperature. The recommended range of dietary protein level and protein/digestible energy ratio for fry H. fossilis is 40.8-41.8% and 27.21-27.88 mg protein kJ-1 digestible energy, respectively. Conclusions: Information developed is of high significance for optimizing growth potential by making better utilization of nutrient at 26 ℃ and, to develop effective management strategies for mass culture of this highly preferred fish species.
Ninety advanced Catla catla fingerlings (av. wt. 16 g) were randomly distributed in six treatment groups with three replicates each for an experimental period of 60 days to study the effect of dietary lipid source on growth, enzyme activities and immuno-hematological parameters. Six isoprotein (40.0-41.9%) and isocaloric (4,260 kcal $kg^{-1}$) semi-purified diets were prepared with varying levels of soybean oil (SBO) and cod liver oil (CLO) within a total of 8% lipid viz., $D_1$ (Control), $D_2$ (8% SBO), $D_3$ (6% SBO and 2% CLO), $D_4$ (4% SBO and 4% CLO), $D_5$ (2% SBO and 6% CLO) and $D_6$ (8% CLO). Highest SGR was noted in $D_5$ (0.73${\pm}$0.03) group, which was similar with $D_3$ (0.71${\pm}$0.02) and $D_4$ (0.69${\pm}$0.01) groups. Activity of intestinal lipase, hepatic glucose-6-phosphate dehydrogenase (G6PDH) and aspartate amino transferase (AST) of the lipid treatment groups were significantly higher (p<0.05) than the control group. The respiratory burst activity of the phagocytes (Nitroblue tetrazolium (NBT)) was highest in $D_2$ (1.95${\pm}$0.21) followed by $D_3$ (1.19${\pm}$0.15) group, which were significantly (p<0.05) higher than the other groups. Globulin level was significantly higher in $D_3$ (1.29${\pm}$0.08) than in the other groups expect $D_4$. Hemoglobin content and total erythrocyte count did not show any significant difference. From this study, it is concluded that a diet containing 6% soybean oil and 2% cod liver oil ($D_3$) yields higher growth and immune response in Catla catla fingerlings and would be cost effective.
