Antitoxic Effect of Korean Red Ginseng Extract on Serum Protein of Mouse Treated with Methyl Mercury (생쥐의 혈청 단백질에 미치는 메틸수은 독성에 대한 홍삼 추출물의 해독 효과)

  • In order to investigate the antiotoxic effect of red ginseng extract on serum protein of mouse treated with methyl mercury playing a role as toxic contaminant in ecosystem, variations of the serum protein contents, electrophoretic patterns, and blood components were studied. Mice were divided into 3 groups: Control, group I treated only with methyl mercury, and group II treated together with methyl mercury and red ginseng extract. The total serum protein content of the control group was 5.8g/dl and those of groups I and II were slightly decreased as compared with the control. The control group showed 11 serum protein fractions and groups I and II showed 10 fractions except prealbumin. The amounts of albumin, ${\alpha}_1-$, ${\alpha}_2-$ globulin fractions were decreased and those $\beta$-, $\gamma$-globulin fractions were increased in groups I and II. The amount of each serum protein fraction in group II showed approximately the same level as the control. The hematocrit value and the number of white blood cells of groups I and II were increased, whereas the number of red blood cells showed the decrease as compared with the control.

  • Authors investigated the hematological examinations of 993 persons at rural region among the 1,7 41 of residents in-the part of Dang Jin (M:268, F:247) and Young Joo (M:270, F:208) in the Korea which was indicated as follows. 1. Average value and standard deviation of hematological items represents as follows: i) Hemoglobin Men Dang Jin-13.99$\pm$0.52. There are higher values of hemoglobin, hernatocrit, M.C.H.C. and WBC in men and women at Young Joo than Dang Jin.

  • We investigated liver protection effect of the Hovenia dulcis Thunb extraction which has treated with carbon tetrachloride ($CCl_4$, 1.0 ML/kg) in SD male rat (20 weeks). We observed the amount of leukocyte, erythrocyte, hemoblobin and hematocrit are increased and the number of plaque is decreased for treatment of $CCl_4$, However, after treatment of Hovenia dulcis Thunb extraction (15 mL/kg), we have same result that of control group which has treated with $CCl_4$ The blood biological data showed that the value of AST (Aspartate aminotransferase), ALT(Alanine aminotransferase) and LDH (Lactate dehydronase) are increased significantly for the negative control group compared to that of control group. For experimental group, respectively, In addition, it was confirmed that the recovery effect of a mysterious death of an interstitial cell of the experimental group is to be compared to that of the negative control group for a sample of pathological tissue.

The effects of n-hexane and tetrachlorodiphenyl on chicken (n-hexane 과 tetrachlorodiphenyl 이 닭에 미치는 영향)

  • The effects of n-hexane and tetrachlorodiphenyl on the growth rate, the blood picture and histopathological change induced were observed in chicken. A total of 350 chicken were fed various daily dosages of n-hexane and tetrachlorodiphenyl for the experimental period of 60 days. The results obtained were as follows. 1. The growth rate of chicken in the group fed 100 ppm of tetrachlorodiphenyl was reduced (p<0.05). However no significant results were observed in the other dosage groups. 2. The reduction of the growth rate was found only in the liver and the kidney but not found in the other orgns. 3. No significant results were found in the number of erythrocytes and hematocrit value. However, the number of leukcocytes of chicken in the group fed 100 ppm of tetrachlorodiphenyl was significantly low. 4. In both n-hexane and tetrachlorodiphenyl groups, the number of lymphocytes was found to decrease but the number of basophiles and eosinophiles were found to increase (p<0.05). 5. All the chicken fed 300 ppm of tetrachlorodiphenyl died between 7th and 9th days. 6. Fatty change of hepatic cells and cloudy swelling of epithelia of renal tubules were found in the group fed 0.5 ml of n-hexane. However, in the group fed more the 100 ppm, fatty change of hepatic cells was followed by necrosis and comparatively severe cloudy swelling was found in epithelia of renal tubules.

Reference intervals of hematological and histopathological index at three different temperatures in Korean rockfish Sebastes schlegelii (조피볼락의 수온별 혈액학적 및 병리조직학적 정상치 탐색)

  • In this study, clinically healthy Korean rockfish were provided by a farm and then raised in a lab for 14 days at three different temperatures (10℃, 15℃, and 20℃) to establish hematological, blood biochemical, and histopathological reference intervals against normal fish. Hematocrit, MCV, MCHC, total protein, BUN, and GPT values in the blood showed significant differences among temperature groups. As the water temperature increased, neutrophil, thrombocyte, and lymphocyte counts also rose, while the monocyte value peaked at 15℃. The histopathological score revealed significant variations in the gills, stomach, and inflammation indices by temperature group; the gills and inflammation indices peaked at 20℃, whereas the stomach index peaked at 15℃. It is expected that information on these normal values will serve as a fundamental collection of data for further studies related to laboratory-based experiments.

Effect of Feeding on Sunflower Seed Oil Cake on Growth and Value of Erythrocyte in Albino Rats (해바라기 종자유박(種子油粕)의 급여(給與)가 흰쥐의 발육(發育)과 적혈구상(赤血球像)에 미치는 영향(影響))

  • Despite the extensive study of protein effect to man and domestic animals, protein effect of sunflower seed oil cake to the growth and value of erythrocyte in rats is still remained to be investigated. This study is to investigate the effect of protein of sunflower seed oil cake to the rat growth and value of erythrocyte as well as the liver weight, heart, kidneys and spleen of rats. Sixty to seventy-days-old, male Sprague-Dawley rats weighing 50-60g, were used in this experiment and 36 rats allotted to 4 groups: standard, 30% cake, 20% cake and low protein. The standard group was fed for 7 weeks on the ration supplemented with 17% casein, 30% and 20% cake groups with sunflower seed oil cake by 30% and 20%, respectively, and low protein group without any supplementation. To observe value of erythrocyte, blood was taken from heart. Value of erythrocyte, hemoglobin and hematocrit value were determined by usual method and oxygen consumption by the method of Watts and Gourley. The result of this experiment suggests that it is recommendable to supplement deficient amino acids in animal.

Changes of Cerebral Metabolism and the Related Factors during Cardiac Surgery

  • The effect of cardiopulmonary bypass (CPB) on cerebral physiology during heart surgery remains incompletely understood. This study was carried out to investigate changes of cerebral metabolism and the association between the changes and clinical factors during heart surgery. Seventy adult patients (n=70) scheduled for elective cardiac surgery were participated in the present study. Middle cerebral artery blood flow velocity (V$_{MCA}$), cerebral arteriovenous oxygen content difference (C(a-v)O$_2$), cerebral oxygen extraction (COE), and modified cerebral metabolic rate for oxygen (MCMRO$_2$) were measured during six phases of the operation; Pre-CPB, CPB-10 min, Rewarm-1 (nasopharyngeal temperature 34$^{\circ}C$), Rewarm-2 (nasopharyngeal temperature 37$^{\circ}C$), CPB-off, and Post-OP (at skin closure after CPB-off). Each relationship of age, arterial blood gas parameters, or other variables to V$_{MCA}2$, C(a-v)O$_2$, COE, or MCMRO$_2$ was evaluated. V$_{MCA}$ increased (P<0.0001) whereas C(a-v)O$_2$ decreased (P<0.01) throughout the five phases of the operation compared to Pre-CPB value (control). COE diminished at CPB-10, Rewarm-1, and CPB-off (P<0.05) while MCMRO$_2$ reduced at CPB-10 and Rewarm-1 (P<0.05) compared to Pre-CPB value. Positive correlation was found between age and cerebral metabolic parameters (V$_{MCA}$, C(a-v)O$_2$, COE, or MCMRO$_2$) during CPB (range r=0.24 to 0.38, p<0.05). Four cerebral metabolic parameters had partially negative or positive correlation with arterial blood gas parameters and other variables (arterial blood pH, $O_2$ tension, $O_2$ content, $CO_2$ tension, blood pressure, blood flow, temperature, or hematocrit) during the operation. In conclusion, CPB led to marked alterations of cerebral metabolism and age, pH, and $CO_2$ tension profoundly influenced the changes during cardiac surgery.

  • The double tracer study on erythrokinetics was carried out experimentally with radioactive iron ($^{59}Fe$) and chromium ($^{51}Cr$) in rabbits. The 0.1% canthalidin solution and 1% pot. perchlomate solution was given subcutaneously to 20 rabbits respectively. 3 and 6 days after injection, the blood chemistry, urine examination, ferrokinetics and apparent half survival time of RBC were ($^{51}Cr\;T\frac{1}{2}$)determined. Following were the results: 1) Red blood cell hematocrit and hemoglobin values were moderately reduced and B.U.N. and serum creatinine values were slight]y inercased in the canthalidin group, while B.U.N. and serum creatinine values were within normal limits in the pot. perchlomate group. Reticulocyte values were slight]y increased in the canthalidin group, while was normal range in the pot. perchlomate group. 2) Blood chemistry finding was not significant statistically in both experimental groups, but serum iron value was moderately reduced in both group. 3) Plasma volume was unchanged in both group, but red cell volume and whole blood volume were slightly reduced in both groups. 4) Results of ferrokinetics were as follows: i) The plasma iron disappearance rate was delayed in both groups. Plasma iron turnover rate, red cell iron utilization and red cell iron turnover rate were decreased in both groups, and then red cell iron turnover rate was more decreased than plasma iron turnover rate in both groups. Circulating red cell iron was slight]y increased in canthalidin group and red cell iron concentration was within normal range in both groups. ii) P.I.T.R.-R.C.I.T. value was moderately increased in the canthalidin group and slightly increased in the pot. perchlomate group. Reticulocyte index, red cell iron turnover index, plasma iron turnover index and effective erythropoiesis index were whole]y reduced in both groups. iii) The red cell life span was slightly shortened in the canthalidin group while was within normal range in pot. perchlomate group. The pathologic finding of renal biopsy of the canthalidin group shows a selective damage in glomerulus, while shows almost normal range or slight damage in tubules. And that of the pot. perchlomate group shows a selective damage in tubules with slight damage of glomerulus.

Evaluation of the health status of preschool children stratified based on the weight-length index (WLI)

  • This study was conducted to prepare basic materials and offer advice regarding dietary habits to prevent and cure childhood obesity by comparing and analyzing dietary habit, nutritional status, blood factors, and mineral contents of hair. All subjects were stratified by their weight-length index (WLI). According to the standard WLI values, 64.9% of children were within the normal value, 13.5% of children were underweight, and 21.6% of children were overweight and obese (WLI ${\geq}$ 110%). Overall, the score assessed dietary habit for all children was $21.32{\pm}2.55$ point (921 subjects), with 5.1% of children having excellent dietary habits and 3.1% having poor dietary habits. Additionally, 37.9% of underweight children, 37.6% of normal weight children, and 43.2% of overweight and obese children consumed higher amounts of protein than underweight children did (meat, fish, eggs, and soy products) (P < 0.05). Overweight and obese children consumed more fried foods than underweight or normal weight children (P < 0.05). Moreover, 38.0% of the children had hemoglobin levels of 12 g/dl, while 7.6% were anemic (11.1 g/dl). When a hematocrit level of 33% was taken as the standard, 11.0% of children were anemic. The plasma transferrin content was $263.76{\pm}54.52$ mg/dl in overweight and obese children. The mean values of Fe, Cu, Ca, Cr, Mn, Se, Na, K, Li, V, Co, and Mo were within the reference values, but the Zn concentrations of underweight, normal weight, and overweight and obese children were $67.97{\pm}28.51$ ppm, $70.09{\pm}30.81$ ppm, and $73.99{\pm}30.36$ ppm, respectively. The Zn concentration of overweight and obese children ($73.99{\pm}30.36$ ppm) was lower than that of the standard value (180~220 ppm). Therefore, a nutritional education program and new guidance for dietary pattern should be developed to reduce the number of underweight and overweight and obese children.

The Analysis of Ethanol Distribution Level in Biological Specimens (생체 시료중에서 Ethanol 농도 분포에 관한 연구)

  • Five kinds of packed column and two kinds of capillary column were used to get optimum condition for ethanol analysis by using fifteen different volatile, low molecular weight organic substances. Only two columns, Gaskuropack 54 and DB-1, showed good separation efficiency. In the adding salt-effect experiment 0.6N - perchloric acid, 1M - meta-phosphoric acid and saturated NaCl solution were used for alcohol concentration measurement of biological fluids and tissue specimens. Among adding salt experiment, adding saturated NaCl solution showed the most stable value of alcohol concentration. This fact might be due to the increased vaporization of alcohol in the saturated NaCl solution. In the time-course of blood alcohol concentration, the alcohol level was lineary decreased to the diameter of vessel containing specimens. This result was interpreted in view of ethanol level, weight, water content, and hematocrit value. The ethanol distribution levels were measured from samples of blood and tissue obtained from 25 postmortems cases investigated by NISI. This study showed that the distribution level was decreased in order of brain, blood, kidney, spleen, liver, and lung.
