Evidence of Multimeric Forms of HSP70 with Phosphorylation on Serine and Tyrosine Residues - Implications for Roles of HSP70 in Detection of GI Cancers

  • Background: Heat-shock protein70 (HSP70) are intracellular protein chaperones, with emerging evidence of their association with various diseases. We have previously reported significantly elevated plasma-HSP70 (pHSP70) in pancreatic cancer. Current methods of pHSP70 isolation are ELISA-based which lack specificity due to cross-reactivity by similarities in the amino-acid sequence in regions of the protein backbone resulting in overestimated HSP70 value. Materials and Methods: This study was undertaken to develop a methodology to capture all isoforms of pHSP70, while further defining their tyrosine and serine phosphorylation status. Results: The methodology included gel electrophoresis on centrifuged supernatant obtained from plasma incubated with HSP70 antibody-coupled beads. After blocking non-specific binding sites, blots were immunostained with monoclonal-antibody specific for human-HSP70, phosphoserine and phosphotyrosine. Conclusions: Our novel immunocapture approach has distinct advantages over the commercially available methods of pHSP70 quantification by allowing isolation of molecular aggregates of HSP70 with additional ability to precisely distinguish phosphorylation state of HSP70 molecules at serine and tyrosine residues.

  • In order to identify potential biomarkers of environmental monitoring, we evaluated heat shock genes expressions as effects of various environmental pollutants (nonylphenol, bisphenol-A, 17a­ethynyl estradiol, bis(2-ethylhexyl)phthalate, endosulfan, paraquat dichloride, chloropyriphos, fenitrothion, cadmium chloride, lead nitrate, potassium dichromate, benzo[a]pyrene and carbon tetrachloride) on larvae of aquatic midge Chironomus tentans (Diptera, Chironomidae). Heat shock protein 70 gene expression increased in most of chemicals treated larvae compared to control. The response was rapid and sensitive to low chemical concentrations but not stressor specific. In conjunction with stressor specific biomarkers, heat shock protein 70 gene expression in Chironomus might be developed for assessing exposure to environmental stressors in the fresh water ecosystem. Considering the potential of Chironomus larvae as biomonitoring species, heat shock gene expression has a considerable potential as a sensitive biomarker for environmental monitoring in Chironomus.

Effects of Venlafaxine and Dexamethasone Treatment on HSP70 Expression in Rat C6 Glioma Cells (흰쥐 C6 신경교종 세포에서 Venlafaxine 그리고 Dexamethasone 처리가 열충격 단백질 70의 발현에 미치는 영향)

  • Object:The intracellular action of the antidepressant, venlafaxine, was studied in C6-gliomas using heat shock protein 70(HSP70) immunocytochemistry and HSP70 Western blots because HSP70 is associated with stress and depression. Methods:To examine how the glucocorticoid affects the expression of HSP70 in nerve cells, the rat C6 glioma cell was treated with dexamethasone for 6 hours. In addition, venlafaxine was administered to the experimental groups of C6 glioma cells for 1, 6, 24, and 72 hours each, after which the expression of HSP70 was investigated. Finally, venlafaxine and dexamethasone were simultaneously administered to the experimental groups for 1, 6, 24, and 72 hours, followed by an investigation of the expression of HSP70. Results:The short term(1 hour) venlafaxine treatment significantly increased the level of HSP70 expression. The short term treatment of venlafaxine with dexamethasone also increased the level of HSP70 expression but this reduction was not statistically significant. The long term(72 hours) venlafaxine with dexamethasone treatment significantly reduced the level of HSP70 expression. The long term treatment of venlafaxine also reduced the level of HSP70 expression but this reduction was not statistically significant. Dexamethasone(10uM, 6hours) did not affect the level of HSP70 expression compared with controls. Conclusion:Venlafaxine increases the expression of HSP70 at short term treatment, but prolonged treatment with dexamethasone suppresses the expression of HSP70.

  • The primary concern has been focused on the response and adaptation of mouse fibroblast tumor cells to heat-shock in the level of membrane surface proteins, using two labeling techniques, lactoperoxidase-catalyzed iodination and galactose oxidase-sodium borohydride. Cells arrested in $G_1$ phase exhibited the highest level of LETS protein and high molecular proteins than did cells passing through $G_1/S, S, G_2$ and M, and unsynchronized cells. Confluent cells were found to show an increase in 125K proteins and a decrease in 130K and 100K proteins selectively. The adaptation processes of tumor cells after heat-shock were observed. All the proteins above 80K were reduced immediately after heat-shock, whereas 70K protein increased markedly 24 hours after heat-shock. The 70K protein and high molecular proteins returned to normal level in 48 hours. The 70K protein was found to be trypsin-sensitive and was similarly labeled by galactose-oxidase as well as by lactoperoxidase. It was, therefore, concluded that 70K protein is glycoprotein located on the surface membrane and might be the HSP 70. Possible function of heat-shock protein on the surface membrane and the relation of this protein to differential heat-sensitivity of tumor cells are discussed.

  • The purpose of the study was for investigating the effect of therapeutic ultrasound irradiation on HSP70 expression in knee of degraded rat articular cartilage. Knee of ten Sprague-Dawley male rats were immobilized for 4 weeks and divided at random into the control and continuous ultrasound applicated group. The continuous ultrasound applicated group was irradiated with frequency 1MHz, intensity $1W/cm^2$ for 5 minutes. The control group was sham sonication. The immunoreactivity of HSP70 was increased in degraded articular cartilage after untrasound irradiation. These results suggest that therapeutic ultrasound can enhance HSP70 expression in degraded articular cartilage.

  • We examined the expression of the heat shock proteins (HSPs) and glucose-regulated proteins (GRPs) during phorbol 1 2-myristate 1 3-acetate (PMA)-induced megakaryocytic differentiation of human er"'throleukemia K562 cells. PMA-treated K562 cells showed a cell growth arrest and alteration in morphology and patterns of gpllIa and c-myc expression, characteristic of megakaryocytic differentiation. During the megakaryocytic differentiation, HSP9OA, HSP9OB, and HSP28 mRNA and protein levels markedly decreased, while GRP78/B and GRP94 mRNA levels were enhanced. On the other hand, HSP7OA and HSP7OB mRNA levels were reduced, but HSP7O protein levels were not changed by PMA treatment. These results suggest specific roles for the HSPs and GRPs in K562 cell proliferation and megakaryocic differentiation.tion.

The Cytotoxicity of Kahweol in HT-29 Human Colorectal Cancer Cells Is Mediated by Apoptosis and Suppression of Heat Shock Protein 70 Expression

  • Although coffee is known to have antioxidant, anti-inflammatory, and antitumor properties, there have been few reports about the effect and mechanism of coffee compounds in colorectal cancer. Heat shock proteins (HSPs) are molecular chaperones that prevent cell death. Their expression is significantly elevated in many tumors and is accompanied by increased cell proliferation, metastasis and poor response to chemotherapy. In this study, we investigated the cytotoxicity of four bioactive compounds in coffee, namely, caffeine, caffeic acid, chlorogenic acid, and kahweol, in HT-29 human colon adenocarcinoma cells. Only kahweol showed significant cytotoxicity. Specifically, kahweol increased the expression of caspase-3, a pro-apoptotic factor, and decreased the expression of anti-apoptotic factors, such as Bcl-2 and phosphorylated Akt. In addition, kahweol significantly attenuated the expression of HSP70. Inhibition of HSP70 activity with triptolide increased kahweol-induced cytotoxicity. In contrast, overexpression of HSP70 significantly reduced kahweol-induced cell death. Taken together, these results demonstrate that kahweol inhibits colorectal tumor cell growth by promoting apoptosis and suppressing HSP70 expression.

Effeets of Environmental pH on the Heat Shoek Proteins and Thermosensitivity of Mouse Fibroblasts and SCK Tumor Cells* (생쥐의 섬유아세포와 SCK 종양세포의 Heat Shock Protein과 열감수성에 미치는 pH의 영향)

  • 생쥐의 섬 유아세포(MEP)와 종양세포(SCK)를 이용하여 정상세포와 종양세포 사이에 열 감수성의 차이가 있는지의 여부 및 환경의 pH가 이 세포들의 열감수성과 heat shock protein(HSP) 합성에 미치는 영향을 생존곡선과 HSP합성 kinetics등을 써서 검토하였다. MEF와 SCK 세포를 정상 pH(7.4) 또는 산성 pH(6.7)에서 42"C에서 2시간 온열처리 후 3일간에 걸쳐 생존을을 비교해 븐 결과, ME선와 SCK세포 사이에 생득적 열강수성의 차이는 없었고 산성 P광에서는 세포의 종류에 관계없이 열감수성 이 증감되었다. 온열처리의 결과 유도되는 내일성이 conditioning Leat의 크기와 어떤 관계가 있는지를 보기 위해서 45"C에서 5분 또는 20분을 주어본 결과 체은 conditioning heat를 주었을 때 내일성이 신속히 그리고 높은 수준으로 발생하였고, 이러한 열 감수성의 kinetics는 HSP의 합성 kinetics와 잘 일치하였다. 단백질, 특히 HSP 합성에 미치는 PH의 영 향을 알아보기 위해서 46"C에서 6분간의 heat shock를 주어 본 바 전반적인 단백질 및 major HSP의 합성양상에는 별로 차이를 보이지 않았다. 그러나 SCK 세포에 43"C에서 30분의 온열처리를 주고 새로 합성되는 HSPSP의 kinetics를 검토해 본 결과 정상 P반에서는 0-5시간에 합성이 일어나나 산성 PH에서는 3-9시간에 합성이 일어나서 몇시간의 합성지연이 관찰되었다. 아울러 HSP68, HSPTC, HSP87을 Peptidemapping하여 본 결과 HSP68과 HSP70 은 유사한 peptide fragment pattern을 보여 amino acid sequence는 유사하고 기능도 같을 것으로 추론되었으나 HSP87은 전혀 다른 pattern을 보였다. 전혀 다른 pattern을 보였다.

Expression of HSP90, HSP70 mRNA and Change of Plasma Cortisol and Glucose During Water Temperature Rising in Freshwater Adapted Black Porgy, Acanthopagrus schlegeli (담수 사육 감성돔, Acanthopagrus schlegeli의 수온 상승에 따른 HSP90, HSP70 mRNA의 발현 및 혈장 cortisol과 glucose 변화)

  • The objective of the present study was to investigate the expression of heat shock protein 90 (HSP90) and 70 (HSP70) mRNA as cellular stress responses, the levels of plasma cortisol with glucose as neuro-endocrine stress responses during water temperature rising in freshwater adapted black porgy, Acanthopagrus schlegeli. A cDNA fragment of 891 (HSP90) and 465 (HSP70) bp was cloned from black porgy testis by Reverse transcription-polymerase chain reaction (RT-PCR) with primers designed from the conserved regions of other teleost. The PCR product of HSP90 showed very high homology to red seabream (99%), rainbow trout (95%), Atlantic salmon (94%), zebrafish (94%) HSP90, HSP70 of black porgy was also highly similar to those of rainbow trout (96%), silver seabream (95%), zebrafish (95%) HSP70. Water temperature rising ($20{\sim}30^{\circ}C$) induced elevation of HSP90 mRNA in black porgy gonad, liver, brain, intestine and kidney, whereas it resulted in an induction of the HSP70 mRNA expression in gonad only. Plasma cortisol levels increased significantly at $30^{\circ}C$ in the fish compared to those at $20^{\circ}C$. Glucose levels of the fish showed a tendency of co-increase with cortisol during water temperature rising. These results suggest that increased HSP90 mRNA in liver with plasma cortisol following heat shock may be related to increasing glucose for homeostasis in this species.

Constitutive Expression of Small Heat Shock Protein Increases Thermotolerance in Transgenic Plant (저 분자량 Heat Shock Protein의 항상적 발현에 의한 형질전판 식물체의 고온내성 증가)

  • To investigate the function of chloroplast small HSP, transgenic tobacco plants (Nicotiana tabacum L. cv. Samsun) that constitutively overexpress the chloroplast small HSP (NtHSP21) from N. tabacum cv. Petit Havana SR1 were generated. Five homozygous lines of transformants showing different constitutive expression levels of the NtHSP21 were selected. To determine whether constitutive overexpression of NtHSP21 protein affects thermotolerance, wild-type and transformants were grown in Petri dishes, heat-stressed at 52$^{\circ}C$ for 45 min, and then incubated in normal growth condition. When heat-stressed wild-type plantlets were incubated at $25^{\circ}C$, leaf color gradually became white and all trio plantlets finally died within a week. As for the transformants, however, more than 70% of them remained green and survived under the conditions in which all the wild-type plants were dying. It was also found that the levels of NtHSP21 were correlated with the degree of thermotolerance. These results suggest that the NtHSP21 protein in transformants is responsible for the increase in thermotolerance.
