Histone Deacetylase Inhibitor Trichostatin A Enhances Antitumor Effects of Docetaxel or Erlotinib in A549 Cell Line

  • Background and Objective: Histone deacetylase (HDAC) inhibitors represent a promising class of potential anticancer agents for treatment of human malignancies. In this study, we investigated the effect of trichostatin A (TSA), one such HDAC inhibitor, in combination with docetaxel (TXT), a cytotoxic chemotherapy agent or erlotinib, a novel molecular target therapy drug, on lung cancer A549 cells. Methods: A549 cells were treated with TXT, erlotinib alone or in combination with TSA, respectively. Cell viability, apoptosis, and cell cycle distribution were evaluated using MTT (3- (4, 5-dimethylthiazol-2-yl) -2, 5-diphenyltetrazolium bromide) assay, Hochst33258 staining and flow cytometry. Moreover, immunofluorescent staining and Western blot analysis were employed to examine alterations of ${\alpha}$-tubulin, heat shock protein 90 (hsp90), epidermal growth factor receptor (EGFR), and caspase-3 in response to the different exogenous stimuli. Results: Compared with single-agent treatment, co-treatment of A549 cells with TSA/TXT or TSA/erlotinib synergistically inhibited cell proliferation, induced apoptosis, and caused cell cycle delay at the $G_2/M$ transition. Treatment with TSA/TXT or TSA/erlotinib led to a significant increase of cleaved caspase-3 expression, also resulting in elevated acetylation of ${\alpha}$-tubulin or hsp90 and decreased expression of EGFR, which was negatively associated with the level of acetylated hsp90. Conclusions: Synergistic anti-tumor effects are observed between TXT or erlotinib and TSA on lung cancer cells. Such combinations may provide a more effective strategy for treating human lung cancer.

Fabrication Process and Impact Characteristic Analysis of Metal Matrix Composite for Electronic Packaging Application (전자패키징용 금속복합재료의 제조공정 해석 및 충격특성평가)

  • This study developed fabrication process of $SiC_p/Al$ metal matrix composites as electronic packaging materials by squeeze casting method. The $SiC_p$ preform were fabricated in newly designed preform mold using about 0.8 % of inorganic binder(SiO$_2$) and 5 vol.% of $Al_2O_3$fiber. To infiltrate the molten metal into the preform, fabrication condition such as the temperature and the pressure were selected. Applying the fabrication conditions, heat transfer analysis were preformed using finite element method and thus analyzed the temperature distribution and cooling characteristic during the squeeze casting. For the fabricated composites, impact toughness and thermal expansion coefficient were measured. The metal matrix composites developed in this study have 0.2~0.3 J impact toughness, $8~10 ppm/^{\circ}C$ thermal expansion coefficient and $2.9~3.0g/cm^3$density which is appropriate properties for electronic packaging application.

Experimental Study of Internal Distribution of Temperature and Relative Humidity in Notebook LCD Module (노트북용 액정 모듈 내부의 온도 및 상대 습도 변화에 관한 실험적 연구)

  • As a part of basic research to improve screen quality, which deteriorates when strain occurs within optical sheets in LCD modules for laptop computers, a measurement system is needed to analyze the influence of heat and moisture, which are the main factors causing the strain. It is assumed that the existence of an air gap less of than 0.5 mm inside the LCD module causes humid air from outside to permeate through the module. To investigate this phenomenon, in this study a thin printed circuit board (PCB) of compact sensors that measure temperature and humidity is inserted into LCD modules, and thus, the changes in temperature and humidity can be analyzed in real time. The results can be used as basic data for simulation of the deformation behavior and structural design of LCD modules in the future.

  • Thermal problems that are directly related to the lifetime of an electronic device are becoming increasingly important owing to the miniaturization of electronic devices. To solve thermal problems, it is essential to study thermal stability through thermal diffusion and insulation. A honeycomb sandwich plate has anisotropic thermal conductivity. To analyze the thermal deformation and temperature distribution of a system that employs a honeycomb sandwich plate, the thermal and elastic properties need to be determined. In this study, the thermal and elastic properties of a honeycomb sandwich plate, such as thermal conductivity, coefficient of thermal expansion, elastic modulus, Poisson's ratio, and shear modulus, are predicted. The properties of a honeycomb sandwich plate vary according to the hexagon size, thickness, and material properties.

  • Objectives : Literatures related to Bianque are studied to discover the path of development and the impact of Bianque school's pulse diagnosis system. Methods : Texts regarding Bianque were searched in history books such as Shiji and Zhanguoce, and medical texts such as the medical books of Mawangdui Han Tomb, Huangdineijng, Maijing, and Qianjinyifang to understand how the Bianque school's pulse diagnosis system was developed. Results : 1. Bianque school's pulse diagnosis system was used to inspect the distribution pattern of blood vessels and discover the location of the disease including the palpatation realm such as only hard or only fall. 2. The system of inspection was created when the diagnosis method that uses the color of the pulse by using the color of blood vessels was added to the diagnostic method of pulse condition. 3. Adding the concept of pulse to the visual information that derives from pulse condition becomes pulsation. This is a diagnostic method that falls under the realm of palpation, and it was used to discover the location of disease. 4. The qi of pulse is motor that induces pulse, and this concept is used in order to understand how normal and abnormal pulsations appear, and to treat the circulation disorder of qi and blood. 5. Cubit skin examination is a method that comprehensively take into account the upper arm skin's cold and heat, slippery and roughness, and relax and tension state. This method was used together with other diagnostic methods. As described above, it seems that the diagnostic method with blood vessels used by Bianque school seems to have developed from Bianque's special inspection ability to the stage where it uses palpation, and then to the stage of cubit skin examination which uses both palpation and inspection.

An Experimental Assessment of the Effects of Residual Stresses on Fracture Behavior of the Plate (압축잔류응력이 판의 파괴 특성에 미치는 영향에 관한 실험적 평가)

  • The quantitative assessment of the effect of the residual stress on fracture behavior was executed by some experiment and numerical analysis. First of all, artificial residual stresses were imposed on CT(Compact Tension) specimens by local heating using gas torch, and an appropriate distribution of residual stresses was obtained by thermal elastic-plastic FE analysis. To certify the result of the FE analysis, an experimental measurement was performed in accordance with ASTM standard. Fracture toughness test was executed on the several types of specimens. The first type was the specimen without residual stresses, and the others had different peak value of compressive residual stress at crack front via controlling the heat flux. All the test results were presented on th J resistance(JR) curves and discussed to verify the effect of compressive residual stresses on fracture behavior.

Required Pressurant Mass for Cryogenic Propellant Tank with Pressurant Temperature Variation (가압가스 온도에 따른 극저온 추진제탱크 가압가스 요구량)

  • The prediction of the required pressurant mass for maintaining the pressure of propellant tanks during propellant feeding is an important issue in designing pressurization system. The temperature of pressurant fed into propellant tank is the critical factor in the required pressurant mass and is one of the most crucial design parameters in the development of pressurization system including designing the weight of pressurant tanks and the size of heat exchanger. Hence a series of propellant drainage tests by pressurizing propellant stored in a cryogenic propellant tank have been performed with measuring the temperature distribution inside ullage and the required pressurant mass according to the temperature condition of pressurant. Results shows that the required pressurant mass decreases as the temperature of pressurant increases. However, the rate of the actual pressurant mass to the ideal required pressurant mass increases.

Study on the tensile restraint crack characteristics in underwater welds of marine steel plates (선용 강판 수중용접부의 인장 구속 균열 특성에 관한 연구)

  • Generally the factors affected largely by the cold cracking sensitivity of the weld are the quantity of the diffusible hydrogen, the brittleness and hardness of the bond area and the tensile restraint stress. These factors have relation each other, and if we can reduce one of these factors, it becomes instrumental to the root cracks prevention of weld. This study deals with the gravity type-underwater-welding of KR Grade A-3 marine steel plate using E4303 welding electrode in order to compare wet-underwater-welding with in-air- welding, resulting in obtaining the tensile restraint characteristics, the hardness distribution, the quantity of diffusible hydrogen and the macro- and micro-crack properties in both underwater and in-air welds. The main results obtained are as follows: 1) The quantity of diffusible hydrogen measured for 48 hours is about 18cc/100g-weld-metal for the in-air-weld of one pass and about 48cc/100g-weld-metal for the underwater-weld of one pass which is about 3 times penetration of diffusible hydrogen compairing with the case of the in-air-weld. However, it was experimentally confirmed that, by the multi-pass welding of 2 to 5 passes, the diffusible hydrogen in the underwater weld metal can be reduced as much as 27 to 49%. 2) The hardness of the weld metal indicates the highest value in the heat affected zones of underwater weld for more rapid cooling rate, resulting in the higher sensitivity of cold cracking. So, it is desirable to soften the higher hardness in the HAZ by tempering effect such as the multi-pass welding in the underwater welding. 3) At the bond vicinity of the underwater weld HAZ, micro cracks were found as resulted by both more rapid cooling rate and more diffusible hydrogen and also by the stress corrosion cracking under the tensile restraint stress in the underwater. But this could be prevented by the tempering effect of the following weld bead such as the multi-pass welding.

Changes of the Flame Temperature and OH Radical in the Unsteady Extinction Process (비정상 소화 과정에서의 화염 온도 및 OH 라디칼의 변화)

  • A flame extinction phenomenon is a typical unsteady process in combustion. Flame extinction is characterized by various physical phenomena, such as convection, diffusion, and the production of heat and mass. Flame extinction can be achieved by either increasing the strain rate or curvature, by diluting an inert gas or inhibitor, or by increasing the thermal or radiant energy loss. Though the extinction is an inherently transient process, steady and quasi-steady approaches have been used as useful tools for understanding the flame extinction phenomenon. Recently, unsteady characteristics of flames have been studied by many researchers, and various attempts have been made to understand unsteady flame behavior, by using various extinction processes. Representative parameters for describing flame, such as flame temperature, important species related to reactions, and chemi-luminescence of the flame have been used as criterions of flame extinction. In these works, verification of each parameter and establishing the proper criterions of the extinction has been very important. In this study, a time-dependent flame temperature and an OH radical concentration were measured using optical methods, and the instantaneous change of the flame luminosity was also measured using a high-speed ICCD (HICCD) camera. We compare the unsteady extinction points obtained by three different methods, and we discuss transient characteristics of maximum flame temperature and OH radical distribution near the extinction limit.

Synthesis and Characterization of NiAl2O4 Inorganic Pigment Nanoparticles by a Reverse Micelle Processing (역-마이셀 공정에 의한 NiAl2O4 무기안료 나노 분말의 합성 및 특성)

  • $NiAl_2O_4$ nanoparticle was synthesized by a reverse micelle processing for inorganic pigment. $Ni(NO_3)_2{\cdot}6H_2O$ and $Al(NO_3)_3{\cdot}9H_2O$ were used for the precursor in order to synthesize $NiAl_2O_4$ nanoparticles. The aqueous solution, which consisted of a mixing molar ratio of Ni/Al, was 1:2 and heat treated at $800{\sim}1100^{\circ}C$ for 2h. The average size and distribution of synthesized $NiAl_2O_4$ powders are in the range of 10-20 nm and narrow, respectively. The average size of the synthesized $NiAl_2O_4$ powders increased with an increasing water-to-surfactant molar ratio and heating temperature. The crystallinity of synthesized $NiAl_2O_4$ powder increased with an increasing heating temperature. The synthesized $NiAl_2O_4$ powders were characterized by X-ray diffraction analysis(XRD), a field emission scanning electron microscopy(FE-SEM), and a color spectrophotometer. The properties of synthesized powders were affected as a function such as a molar ratio and heating temperature. Results indicate that synthesis using a reverse miclle processing is a favorable process to obtain $NiAl_2O_4$ spinels at low temperatures. The procedure performed suggests that this new synthesis route for producing these oxides has the advantage of being fast and simple. Colorimetric coordinates indicate that the pigments obtained exhibit blue colors.
