In vivo Visualization of Human White Matter Tract by Diffusion Tensor Imaging Fiber Tractography (DTI-FT)

  • Purpose: To evaluate the white matter fiber connectivity of normal human using diffusion tensor MRI. Method: Normal young healthy volunteers (2 women and 1 man) and 3 brain tumor patients participated in this study. All studies were performed using a 1.5T Philips Gyroscan Intern system. Diffusion weighted imaging was performed using single-shot echo planar imaging, with navigator echo phase correction and SENSE. Diffusion weighting was performed along six independent axes, using diffusion weighting of b=800s/$\textrm{mm}^2$. 128matrix, 23cm FOV, 2.5mm slice thickness were used for Imaging parameters. Data were processed on a Window-2000 PC equipped with IDL and PRIDE (Philips Medical System). Corticospinal tract was traced from mid-pons level via posterior limb of internal capsule. Corpus callosum, cerebellar peduncles and frontal fibers were traced by fiber tractography.

에토석시미드의 HPLC 분석법 및 한국인에서의 약동학적 특징 (HPLC Determination and Pharmacokinetic Profile of Ethosuximide in Korean Subjects)

  • Ethosuximide is an oral anticonvulsantic agent used in the first choice anti-absence seizure drug. The purpose of this study was to assess the pharmacokinetic profile of the ethosuximide in healthy Korean volunteers and to develop the efficient assay method of ethosuximide in human plasma. The pharmacokinetics of ethosuximide administered orally was evaluated after a dose of 500 mg. Ethosuximide was assayed from plasma by a specific HPLC method reading absorbance at 195 nm. AUC was 1222$\pm$160 $\mu\textrm{g}$/$m\ell$$.$hr, $C_{max}$ 14.2l$\pm$1.74 $\mu\textrm{g}$/$m\ell$, $T_{max}$ 1.06$\pm$0.62 hr and half-life 77.83$\pm$12.46 hr. The half-life in Korean was longer than, in Caucasian (53∼56 hr).).).

  • 건강한 지원자를 대상으로 전기압력밥솥으로 조리한 수원464 백미와 일품종 백미를 섭취시켜 혈당의 반응을 비교한 결과식후 60분 후에서 수원464 백미섭취군의 혈당이 일품종 백미섭취군에 비해 유의성있게 더 감소하였다(p<0.01). 수원464의 혈당치가 일부 구간에서 현저히 낮은 이유는 수원464의 난소화성전분함량이 일반쌀에 비하여 높기 때문이라고 해석된다.

Biological Monitoring of Human Exposure to Volatile Halogenated Hydrocarbons Using Urinalysis with Capillary GC-ECD

  • For the risk assessment of human exposure to volatile halogenated hydrocarbons, a dynamic purge trap/on-column cryofocusing method using capillary gas chromatograph-$^{63}Ni$ electron capture detector and thermal desorption unit was applied to analyze the free forms, metabolites of 1, 1, 2-trichloroethylene and 1, 1, 2, 2-tetrachloroethylene. The urine sample was diluted with distilled water, hydrolyzed and sealed. Then the inert gas was infused to purge out free 1, 1, 2-trichloroethylene, free 1, 1, 2, 2-tetrachloroethylene and urichloroethanol. These compounds were trapped to $Tenax^R$ / GC-gas trap device throughout clean up tube. Being undertectable to gas chromatograph directly, trichloroacetic acid was methyl esterificated and trapped in the manner above mentioned. The optimal incubation time to get best recovery of methyl ester was 4 hours at $60^circ$C. The concentrations of free volatile halogenated hydrocarbons and their metabolites in urine were obtained of free volatile halogenated hydrocarbons and their metabolites in urine were obtained from 5 healthy volunteers. This analytical method is expected to make the biological monitoring more precise and convenient.

Immunomodulatory Response Induced by Ginseng

  • There has been continuing interest in the development of synthetic and natural compounds that modify the immune response particularly for the treatment of AIDS and cancer. During the past fifty years, numerous scientific studies have been published on ginseng (Foster and Chongxi, 1992). Modern human studies have investigated preventive effect of ginseng on several kinds of cancer (Yun et al, 1993,Yun, 1995,Yun and Choi, 1998), its long term immunological effect on HIV patients (Sankang, 1989, Cho et al, 1997), its effect on cell mediated immune functions in healthy volunteers (Scaglione et al, 1990). Similarly non clinical studies on animal model system have studied the chemopreventive action of ginseng on cancer (Kumar, 1993,98) and immunological properties of ginseng (Kim et al, 1990, Tomoda et al, 1993, Yun et al, 1993, Mizuno et al, 1994,Lee et al, 1997, Park et al, 2001,Yoshikawa et al, 2001, Wang et al, 2001). The precise mechanism of action of ginseng, however, not clearly understood. Considering its wide-ranging therapeutic effects, this study is being undertaken to elucidate the general mode of action of ginseng, especially to test our hypothesis that its biological action may be mediated by the immune system.

  • Lately, soybeans have received considerable public attention for their potential roles in the prevention of the chronic diseases. Epidemiologic study showed that Asian countries have significant health benefits because of the high contents of the isoflavones in their traditional diets(soybean-rich diet). This study was carried out to determine pharmacokinetic parameters of isoflavone in Korean woman. Pharmacokinetic study of three soy products(isogen, soymilk, and fermented soybean) with different ration of aglycone/glucoside in 26 healthy female volunteers(20-30 years of age) was performed. A Preliminary Study on the Effectiveness of Far-Infrared Emitting Ceramic Mattresses in Improving Sleep Quality

  • Objectives : Far-infrared (FIR) lights have been investigated for sleep quality intervention. We sought to measure the advantageous effects of FIR in sleep using polysomnographic data as the objective outcomes. Methods : The ten healthy volunteers were enrolled in a single-center, prospective, patient-blind, single-arm trial. Individuals slept on a sham mattress and a FIR emitting mattress with polysomnography for one night each. Results : Sleep efficiency showed an increasing trend but was not statistically significant. PSQI-K significantly decreased (p=0.013). The latency to REM of the baseline was shorter than that of the intervention (p=0.008). Though there was no statistical significance, Stage N1 and N2 were shortened, and Stage N3 was prolonged after the intervention compared to the baseline. Conclusions : The FIR-emitting mattresses improved sleep quality on self-reported insomnia. We suggested the candidate for the markers altered by the FIR therapy, such as the normalization of REM latency and increased N3 sleep.

사람 분변에서 분리한 Enterococcusfaecalis가 생성하는 BileSaltHydrolase의 특징 (Cloning and Characterization of a Bile Salt Hydrolase from Enterococcus faecalis Strain Isolated from Healthy Elderly Volunteers)

  • 담즙산 분해효소(Bile salt hydrolase, EC 3.5.1.24) 활성은 담즙산의 카르복실기와 결합되어 있는 아미노기(glycine or taurine)와의 amide 결합을 끊는 작용을 하며, 이 효소 활성은 사람이나 동물의 장내 미생물들에서 널리 분포하고 있다. 노인 분변으로부터 분리한 여러 균주의 Enterococcus faecalis중에서 BSH activity가 가장 높은 CU30-2를 선발하였다. BSH 유전자를 pET22b expression vector에 클로닝하여 Escherichia coli BL21(DE3) Gold를 이용하여 단백질을 발현하였다. 6x His-tag이 있는 BSH 효소를 $Ni^+$-NTA agarose column을 이용하여 분리 정제하였고, 6가지의 다른 담즙산염을 이용하여 기질 특이성을 비교하였다. E. faecalis CU30-2의 BSH 효소는 glycine이 결합된 담즙산염에 대한 효소활성이 taurine이 결합된 것에 대한 활성보다 약 50배 정도 높게 나타났다. 이 효소의 최적 pH와 온도는 각각 7.0과 40$^{\circ}C$로 확인되었다.

테놀민 정(아테놀올 50 mg)에 대한 신일아테놀올 정의 생물학적 동등성 (Bioequivalence of Sinil Atenolol Tablets to Tenormin Tablets (Atenolol 50 mg))

  • This study was conducted to compare the bioavailability of a generic product of Sinil Atenolol Tablets (Sinil Pharmaceutical Co., Ltd., Korea) with the innovator product, $Tenormin^{\circledR}$ Tablets in 20 healthy Korean volunteers. The volunteers received a single 50 mg dose of each atenolol formulation according to a randomized, two-way crossover design. Plasma samples were obtained over a 24-hour interval, and atenolol concentrations were determined by HPLC with a fluorescence detector. From the plasma atenolol concentration vs time curves, the following parameters were compared: area under the plasma concentration-time curve (AUC), peak plasma concentration $(C_{max})$, time to reach peak plasma concentration $(T_{max})$, and terminal first order elimination half-life $(t_{1/2})$. No statistically significant difference was obtained between the $T_{max}$ values, and the logarithmic transformed AUC and $C_{max}$ values of the two products. The 90% confidence for the ratio of the logarithmically transformed AUC and $C_{max}$ values of Sinil Atenolol Tablets over those of $Tenormin^{\circledR}$ Tablets were calculated to be between 0.99 and 1.07, and 1.04 and 1.16, respectively; both were within the bioequivalence limit of 0.80-1.25. The mean of $T_{max}$ in $Tenormin^{\circledR}$ Tablet group was 3.68 hour, and that in Sinil Atenolol Tablet group was 3.65 hour. The values of $t_{1/2}$ between the two products were found comparable, and the mean $t_{1/2}$ values of $Tenormin^{\circledR}$ Tablets and Sinil Atenolol Tablets were 5.9 and 6.0 hour, respectively. Based on these results, it was concluded that Sinil Atenolol Tablets were comparable to $Tenormin^{\circledR}$ Tablets in both the rate and extent of absorption, indicating that Sinil Atenolol Tablets were bioequivalent to the reference product, $Tenormin^{\circledR}$ Tablets

잡기동작 수행 시 입모양에 따른 뇌활성화 분석 (Analysis of Brain Activation due to Mouth Shape during Grip Movement)

  • Purpose : This study was performed to understand the relationship between hand and mouth shapes using functional magnetic resonance imaging(fMRI). Methods : Two healthy volunteers without any previous history of physical or neurological illness were recruited. fMRI was done that volunteers was 6 repeated of natural mouth, close mouth and open mouth while power grip and pinch grip movement. Results : Cerebral cortex activation was not well observed for the natural mouth during the power grip exercise. For the closed mouth, the temporal lobe, Broca's area, the prefrontal area related to thinking and judgment, the supplementary motor area, the auditory area and Wernicke's area were activated. For the open mouth, cortical activation was also observed in the temporal lobe, Wernicke's area, the prefrontal area related to thinking and the orbital frontal area related to visual sense. During the pinch grip exercise, cortical activation was observed for the natural mouth in the primary sensory area, Wernicke's area, the primary and supplementary motor area, and the prefrontal area. For the closed mouth, cortical activation was observed in the temporal lobe, Wernicke's area, the prefrontal area related to thinking, the secondary visual area, the primary sensory area and the supplementary motor area. In the case of the open mouth, cortical activation was observed in a few parts in the temporal lobe as well as Wernicke's area, the prefrontal area related to thinking, and other areas related to visual sense such as the primary visual area, the secondary visual area and the visual association area. Conclusion : Brain was more activation for close mouth and open mouth more than natural mouth movement.
