석유화학공장에서의 한국형 위험기반검사에 의한 화학설비의 안정성 평가 (Analysis of Safety of the Chemical Facilities by Korea Risk Based-Inspection in the Petrochemical Plant)

  • As a way of improving the safety of the chemical facilities, the risk based-inspection(RBI) was executed for the facilities of the applied petrochemical plant using KS-RBI Ver. 3.0 program developed based on the API-581 based resource document(BRD). From an evaluation result of KS-RBI program, we could find the evaluation of the process safety management(PSM) for the applied plant, risk of the applied process, risk of static facilities and pipes, and the demage mechanism of the facilities. Also, we could suggest a proper inspection plan(frequency and method of inspection) using the calculated risk and the status of the facilities. Therefore, the applied plant could be achieved a reduced inspection cost by an extension of inspection frequency, improved productivity, improved reliability of the facilities, and a computerized history management.

Comparison of Bacterial Community of Healthy and Erwinia amylovora Infected Apples

  • Fire blight disease, caused by Erwinia amylovora, could damage rosaceous plants such as apples, pears, and raspberries. In this study, we designed to understand how E. amylovora affected other bacterial communities on apple rhizosphere; twig and fruit endosphere; and leaf, and fruit episphere. Limited studies on the understanding of the microbial community of apples and changes the community structure by occurrence of the fire blight disease were conducted. As result of these experiments, the infected trees had low species richness and operational taxonomic unit diversity when compared to healthy trees. Rhizospheric bacterial communities were stable regardless of infection. But the communities in endosphere and episphere were significanlty affected by E. amylovora infection. We also found that several metabolic pathways differ significantly between infected and healthy trees. In particular, we observed differences in sugar metabolites. The finding provides that sucrose metabolites are important for colonization of E. amylovora in host tissue. Our results provide fundamental information on the microbial community structures between E. amylovora infected and uninfected trees, which will contribute to developing novel control strategies for the fire blight disease.

Comparisons of inorganic amounts in paddy fields, rice straw and seed with varying severity of brown spot caused by Cochliobolus miyabeanus

  • In order to elucidate influence of nutritional status on rice brown spot caused by Cochliobolus miyabeanus, rice cultivation soils and rice straws were collected from paddy fields where ice brown spot occurred severely, moderately, a little and none respectively. Rice plant materials were analyzed to measure inorganic nutrients in rice straws and rice seeds. Analysis of chemical properties of rice paddy soil showed that EC and contents of available phosphate, cation and silicic acid in soil with severe infections were lower than those in healthy soil. This result suggests that amount and holding capacity of nutrient contents in soils collected from paddy field with infection of C. miyabeanus are relatively low compared to those in soils collected from healthy paddy field. Analysis of inorganic nutrients in rice straws showed that amount of macronutrient elements such as silicic acids, available phosphate and total nitrogen, and micronutrients such as copper, iron and zinc in rice straws from paddy field with infection were lower than those in healthy soil. Especially amount of iron and silicic acid were very low in rice straws from paddy field soils with infection Amount of inorganic nutrients such as iron and zinc in rice seeds was the same trend as those of rice straws. These results showed that one of major factors affecting rice brown spot was amount of nutrient contents in soil and rice straw.

Changes in the Composition and Microbial Community of the Pepper Rhizosphere in Field with Bacterial Wilt Disease

  • Bacterial wilt caused by Ralstonia solanacearum is considered one of the most harmful diseases of pepper plants. Recently, research on plant disease control through the rhizosphere microbiome has been actively conducted. In this study, the relationship with disease occurrence between the neighboring plant confirmed by analyzing the physicochemical properties of the rhizosphere soil and changes in the microbial community. The results confirmed that the microbial community changes significantly depending on the organic matters, P2O5, and clay in the soil. Despite significant differences in microbial communities according to soil composition, Actinobacteriota at the phylum level was higher in healthy plant rhizosphere (mean of relative abundance, D: 8.05 ± 1.13; H: 10.06 ± 1.59). These results suggest that Actinobacteriota may be associated with bacterial wilt disease. In this study, we present basic information for constructing of healthy soil in the future by presenting the major microbial groups that can suppress bacterial wilt.

대두종자의 자반병 감염과 Peroxidase 활성도변화 (The Change of Peroxidase Activity in Soybean Seed Followed by Infection with Cercospora kikuchii)

  • Cercospora kikuchii에 의해서 발병된 대두종자의 부패균에 대한 획득저항성기작을 구명코자 본 연구를 실시하였다. 1. 대두의 건전종자, 자연열개종자, 인위적 상처종자 및 자반병 감염종자재의 전단백질 함량에는 차이가 없었으나 peroxidase활성도는 감염종자에서 월등히 높았다. 2. 대두의 건전종자와 자반병 감염종자의 종피 및 자엽내의 단백질함량에는 큰 차이가 없었으며 peroxidase활성도는 자엽에서는 차이 가 없었으나 종피에서 건전한 종피보다 자반병에 감염된 종자가 2.5배 정도 높은 활성도를 보였다. 3. Polyphenol oxidase는 대두의 건전종자의 자반병 감염종자 모두 활성도가 너무 낮아 측정할 수 없었다. 4. 전기영동에 의한 peroxidase isozyme pattern은 건전종자와 감염종자간에 차이가 없었다. 5. 자반병에 대한 저항성기작은 자반병균에 의하여 대주종피 자체내에서 생화학적 방어기작이 더욱 활성화 하였으며 peroxidase활성도의 증가는 병원균에 대한 반응으로 사료된다.

Comparative Analyses of Tomato yellow leaf curl virus C4 Protein-Interacting Host Proteins in Healthy and Infected Tomato Tissues

  • Tomato yellow leaf curl virus (TYLCV), a member of the genus Begomovirus, is one of the most important viruses of cultivated tomatoes worldwide, mainly causing yellowing and curling of leaves with stunting in plants. TYLCV causes severe problems in sub-tropical and tropical countries, as well as in Korea. However, the mechanism of TYLCV infection remains unclear, although the function of each viral component has been identified. TYLCV C4 codes for a small protein involved in various cellular functions, including symptom determination, gene silencing, viral movement, and induction of the plant defense response. In this study, through yeast-two hybrid screenings, we identified TYLCV C4-interacting host proteins from both healthy and symptom-exhibiting tomato tissues, to determine the role of TYLCV C4 proteins in the infection processes. Comparative analyses of 28 proteins from healthy tissues and 36 from infected tissues showing interactions with TYLCV C4 indicated that TYLCV C4 mainly interacts with host proteins involved in translation, ubiquitination, and plant defense, and most interacting proteins differed between the two tissues but belong to similar molecular functional categories. Four proteins-two ribosomal proteins, S-adenosyl-L-homocysteine hydrolase, and 14-3-3 family protein-were detected in both tissues. Furthermore, the identified proteins in symptom-exhibiting tissues showed greater involvement in plant defenses. Some are key regulators, such as receptor-like kinases and pathogenesis-related proteins, of plant defenses. Thus, TYLCV C4 may contribute to the suppression of host defense during TYLCV infection and be involved in ubiquitination for viral infection.

흰가루병에 성체식물저항성을 지닌 봄보리의 감염잎에서 아미노산함량의 변화 (Changes in Amino Acid Content in Infected Leaves of Spring Barley Plants Resistant to Powdery Mildew at Adult- Plant Stage)

  • 흰가루병에 대하여 감수성인 봄보리품종 Peruvian과 성체식물 저상성품종 Asse의 건전잎과 감염잎에 함유되어 있는 에탄올가용성 아미노산을 분석하였다. 접종1일 후에는 이들 두 품종의 제1엽내의 아미노산 함량은 건전잎과 비슷한 수준이었다. 포자형성 동안 Asse보다 Peruvian에서 아미노산의 증가에 뚜렷하였다. 감염된 제1엽, 제5엽에서 아미노산 함량은 엽당 병반수가 많을수록 증가하였고, 감수성 품종 Peruvian의 감염잎이 Asse보다 아미노산 함량이 더 높았다.

Biological Control of Apple Ring Rot on Fruit by Bacillus amyloliquefaciens 9001

  • Apple ring rot disease, caused by Botryosphaeria dothidea (Moug. ex. Fr) Ces. et de Not., is one of the most important diseases on apple fruits. In this study, strain 9001 isolated from healthy apple fruits from an infested orchard was evaluated for its biocontrol activity against apple ring rot in vitro and in vivo. Strain 9001 showed obvious antagonistic activity to B. dothidea YL-1 when plated on potato dextrose agar. Soaking healthy apples in the bacterial suspensions of strain 9001 prior to artificial inoculation of fungal pathogen resulted in a dramatic decrease in disease incidence when compared to the control. Moreover, either field application in the growth season or postharvest treatment of apples from infected orchards with bacterial suspensions of strain 9001 resulted in significantly reduced disease incidence within the storage period for 4 months at room temperature. Based on the phylogenetic analysis of 16S rRNA and the gyrA gene, strain 9001 was identified as Bacillus amyloliquefaciens. These results indicated that B. amyloliquefaciens 9001 could be a promising agent in biocontrol of apple ring rot on fruit, which might help to minimize the yield loss of apple fruit during the long postharvest period.

배나무잎 이상반점증상에 관한 연구 - 3. 병원의 접목전염 - (Studies on the Pear Abnormal Leaf Spot Disease - 3. Graft Transmissibility of the Causal Agent -)

  • Nature of graft transmissibility of pear abnormal leaf spot disease was examined by various grafting methods in the greenhouse and field. When the diseased and symptomless twigs were collected in winter and grafted in the next spring to the seed-originated healthy root stock, the abnormal leaf spot was developed only in the case of the diseased twigs. Double grafting on a seed-originated healthy root stock, where the diseased and the symptomless twigs were used as 1st and 2nd scions, respectively, developed abnormal leaf spot lesions without exception on the 2nd scions. Tongue-graft with the diseased and the symptomless trees also incited abnormal leaf spots on the both trees. Abnormal leaf spots of were also developed on HN-39, an indicator pear tree, used as a 2nd scion in a double graft test, where the diseased twig and a seed-originated healthy tree were used as the 1st scion and the root stock, respectively. When the diseased twig was top-grafted to the healthy root stock, lesion development of abnormal spot was limited to the grafted twig itself in the 1st year, but expanded to the main branches in the 2nd year, and spread over the whole tree in the 3rd year. This result indicates that the causal agent of abnormal leaf spot disease is transmitted by graft.

First Report of Diaporthe actinidiae, the Causal Organism of Stem-end Rot of Kiwifruit in Korea

  • Post-harvest diseases of kiwifruit caused severe damages on the fruits during storage, transportation, marketing and consumption. Phomopsis sp. was reported to be one of the major causal organisms of post-harvest fruit rots of kiwifruit. Symptoms of stem-end rot caused by Phomopsis sp. appeared at the stem-end area of the fruit as it ripened. The brown pubescent skin at the area became soft and lighter in color than the adjacent firm healthy tissues. A watery exudate and white mycelial mats were frequently visible at the stem-end area forming a water-drop stain down the sides on the dry brown healthy skin. When the skin was peeled back, the affected flesh tissue was usually watersoaked, disorganized, soft and lighter green than the healthy tissue. Phomopsis sp. was consistently isolated from the diseased fruits, and its pathogenicity was confirmed by an artificial inoculation test on healthy fruit of kiwifruits. The mycological characteristics of the telemorph state of the fungus produced on potato-dextrose agar were in accordance with those of Diaporthe actinidiae. This is the first report on the occurrence of a telemorph state of D. actinidiae as the causal organism of stem-end rot of kiwifruit in Korea.
