Historical Study of Beef Cooking -IV. boiled beef(熟肉) and sliced of boiled beef(片肉)- (우육조리법(牛肉調理法)의 역사적(歷史的) 고찰(考察) -IV. 숙육(熟肉)과 편육(片肉)-)

  • The purpose of this study was to survey the various kinds of cooked beef products focusing on Sukyuk (boiled beef) and Pyunyuk (boiled beef slice) recorded on the historical literatures written from 1670 to 1945. Sukyuk and Pyunyuk were recorded 45 times in the references and could be classified into 11 groups based on major ingredients such as fresh meat, tough meat, rotten meat, tail, head, lung, cup of breast, testicles, pancreas, spleen and tung. Twenty two cooking methods were described on the records. Sukyuk and Pyunyuk based on fresh meat were described the most frequently. Pyunyuk based on head was described late in 18th centuries, but its recipe could not be found in any records and that of internal organs and tung were presented late in 18th centuries and early in 19th centuries, respectively. The major ingredients of Sukyuk and Pyunyuk were lean meat, tail, head, lung, cup of breast, testicles, pancreas, spleen and tongue Mulberry seed, fragment of roof tile and other sub-ingredients were used for softening or deodorizing the off flavor of the products.

Spontaneous Release of Bacteriophage Particles by Lactobacillus rhamnosus Pen

  • The identification of bacteriophage proteins on the surface of Lactobacillus rhamnosus Pen was performed by LC-MS/MS analysis. Among the identified proteins, we found a phage-derived major tail protein, two major head proteins, a portal protein, and a host specificity protein. Electron microscopy of a cell surface extract revealed the presence of phage particles in the analyzed samples. The partial sequence of genes encoding the major tail protein for all tested L. rhamnosus strains was determined with specific primers designed in this study. Next, RT-PCR analysis allowed detection of the expression of the major tail protein gene in L. rhamnosus strain Pen at all stages of bacterial growth. The transcription of genes encoding the major tail protein was also proved for other L. rhamnosus strains used in this study. The present work demonstrates the spontanous release of prophage-encoded particles by a commercial probiotic L. rhamnosus strain, which did not significantly affect the bacterial growth of the analyzed strain.

Studies on the Morphology and Stainability of Chicken Spermatozoa (닭 정자(精子)의 형태(形態)와 염색성(染色性) 조사(調査))

  • The present studies were designed to investigate the morphology and stainability of the chicken spermatozoa. Semen samples were collected by abdominal massage from 10 cocks of Arbor, Acres strain (egg breed) and 10 cocks of white Cornish strain (meat breed). The semen samples were diluted with Sarker's solution and were washed. Some of the semen smear slides were stained with seven differential stain methods and was compared with one another by light microscope. In addition to the staining already compared, the length of heads, middle pieces and tails of 400 spermatozoa of two chicken breed was measured with micrometer. The results obtained from these, studies were as follows: 1. Eosin stain appeared to give good results than hematoxylin, pre-treated protease and eosin or hematoxylin stain, pre-treated protease and hematoxylin-eosin stain, carbol-fuchsin, stain and Giemsa 9 technique in differential staining of spermatozoal three portions and pre-treated protease and eosin stain appeared as good staining methods for middle piece of spermatozoa. 2. The average length of chicken spermatozoa was $90.4{\pm}4.0{\mu}m$, and the average length of the head, middle piece and tail of spermatozoa was $13.0{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $73.6{\pm}3.8{\mu}m$ lesoectively. 3. The average length of spermatozoa of Arbor Acres strain was $89.2{\pm}5.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $12.9{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $72.5{\pm}4.7{\mu}m$ respectively. The average length of spermatozoa of with Cornish was $91.6{\pm}3.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $13.1{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $74.7{\pm}2.8{\mu}m$ respectively.

Classification and Characterization of Bacteriophages of Lectobacillus casei (Lactobacillus casei Bacperiophage의 분류 및 특성에 관한 연구)

  • Phages of Lactobaciilus casei (PLC) isolated from plant drainage were classified and characterized. The results are as follows : 1. On the basis of host range pattern, phages could be divided into 2 groups (PLC-B and PLC-C). PLC-B group phages could be further divided into 5 sub-groups $(B_1, \;B_2, \;B_3, \;B_4, \;and\;B_5)$. Although PLC-C group phages had the same host range, they could be also divided into 2 sub-groups $(C_1\;and\;C_2)$ by morphlogical type. 2. It was $B_3$ group phages that represented a major proportion (44.4%) of phages tested. However, $B_1$ group phages were shown to have the widest host range. 3. Electron micrographs revealed that the phages fell into three different morphological types. $(B_1, \;B_2, \;and\;B_3)$ group phages hd a hexagonal head (52nm in diameter) and a sheathless noncontractile (245 nm in length). $B_4\;and\;C_2$ group phages had a hexagonal head (56 nm) and a short flexible tail (169nm) having no sheath. $B_5\;and\;C_1$ group phages were shown to have a hexagonal head (81 nm) and a contractile tail (140 nm) having a sheath, a base plate and tail fibers. 4. The inactivation of the phages by antisera indicated that serological relationships correlated completely with morphological types. 5. $B_1, \;C_1\;and\;C_2$ group phages produced a large (1, 2 mm in diameter) plaque with a clear ring. The morphology of plaques of $B_3\;and\;B_5$ group phages was the same as those produced by the above, but the average plaque sizes for $B_3\;and\;B_5$ were 0.8 mm abd 0.5 mm, respectively. $B_2\;and\;B_4$ group phages produced a small (0.5 mm) turbid plaque with an irregular edge. 6. The latent period and the average burst size of $B_1\;and\;B_3$ group phages were 90 min and 100, respectively. These phages reuqired calcium ions for their miltiplication. 7. $B_3$ group phages could not be absrobed to R-variant $KC_1$. 8. The order of resistance of phages to heat was $B_2\;>\;B_1, B_4\;and\;B_5\;>\;B_3\;and\;C_2, \;B_5$ group phages were more stable than $B_3$ in various pH values. $C_2$ group phages were more sensitive to UV irradiation than $B_1\;and\;B_3$ group phages. 9. Strains YIT9018 and IAM 1043 were induced by mitomycin C treatment. Phage particles detected in the lysates had a hexagonal head (38 and 49 nm, respectively), but no tail. Any sensitive indicator strain could not be isolated in spite of repaeated trials.

Effect of Seminal Sections of Tuber and Staking Methods on Growth and Yield of Chinese Yam (Dioscorea batatas Decne) (종근부위(種根部位) 및 지주형(支柱形)이 마의 생육(生育)과 수량(收量)에 미치는 영향(影響))

  • This study was conducted to determine the effect of seminal sections of tuber and staking methods on growth and yield of a chinese yam (Dioscorea batatas Decne). Tuber was divided into top head, head, median and tail which were used as seminal sections of tuber. The emergence of shoots was delayed in median and tail sections by 15 days as compared with top head and head sections. Sections from head and tail exhibited yield increase as high as 16 and 15%, respectively, compared to top head sections. Although flowering and bubil setting occurred almost at the same time in all staking methods, fresh weight of stem and leaves was higher in ${\wedge}-$, I-type stake supporting net than in the conventional stake. In the cultivar, 'Dan-ma', tuber yield didn't show a meaningful increase statistically by all staking methods. But ${\wedge}-$ and I-type stake supporting net increased the yield of the cultivar, 'Jang-ma', as high as 11 and 8%, respectively, compared to conventional stake. Also I-type stake was ideal supportor of net, considering the cost of management.

  • 양서류를 이용한 내분비계장애물질의 환경독성시험기법을 확립하기 위해 Xenopus laevis의 변태 과정에 thyroid계 물질을 처리하여 이들의 효과에 의한 내분비계 장애물질의 판정 기법을 확립하고자 하였다. 올챙이의 발달 단계에 의한 내분비계 장애물질에 대한 민감도를 고려하여, embryo 단계에서 tail resorption 과정의 전단계인 stage 11로부터 30일간(방법1), stage 49로부터 28일간(방법2)와 tail resorption 과정 및 이후 stage 57로부터 17 내지 21일간(방법3)으로 구분하여 시험물질을 노출시켰으며 대조군과 비교하여, 시험물질에 노출된 올챙이의 기형적 변태 정도는 developmental stage 변화 속도, head-tail length 차이, 및 body weight 변화 등을 endpoints로 하였다.

The Effect of Antisperm Antibodies Detected by Immunobead Binding Assay on Fertilization and Cleavage of Human Oocytes In Vitro (Immunobead 검사로 검출된 항정자 항체가 인간 난자의 체외 수정 및 분할에 미치는 영향)

  • The effect of antisperm antibodies (ASA) on the human in vitro fertilization (lVF) process was evaluated by analyzing the IVF data between October and December 1988 at Seoul National University Hospital prospectively. The immunobead test (IBT) was used to identify Ig G, Ig A, and Ig M in the serum, semen, and follicular fluid from 93 couples undergoing in vitro fertilization-embryo transfer (lVF-ET ) . The fertilization rate in couples with ASA to sperm head of at least one isotype in female serum (n= 10) was significantly less than that in couples without ASA to sperm head (n=83; 28.5% versus 45.3% , p=0.028). The presence of ASA to sperm head in follicular fluid (n=8) also reduced fertilization rate from 45.3% to 24.4% (p=O.0l3). However, ASA binding to sperm head in male serum and semen did not predict fertilization. Similarly, ASA binding to sperm tail and tail-tip did not reduced the oocyte fertilization rate significantly in any of the fluids tested. The zygote cleavage rate was not reduced in the presence of ASA. These results suggest that the presence of ASA to sperm head in female serum and follicular fluid is associated with reduced fertilization in IVF-ET. Another observation is that the oocyte that do fertilize in the presence of antisperm antibodies can subsequently proceed with normal cleavage. The results of this investigation therefore suggest that the IBT is a useful test forscreening of women participat.ing IVF-ET program.

  • 3-Methoxy-4-cyano-2,9-dioxabicyclo[4.3.0]non-3-e ne (1) was prepared by (4 + 2) cycloaddition reaction of methyl ${\alpha}$-cyanoacrylate with 2,3-dihydrofuran. Compound 1 was ring-open polymerized by cationic catalyst such as boron trifluoride etherate to obtain alternating head-to-head (H-H) copolymer (2) of methyl $\alpha$ -cyanoacrylate and 2,3-dihydrofuran. For comparison, head-to-tail (H-T) copolymer (3) was also prepared by free radical copolymerization of the corresponding monomers. The H-H copolymer exhibited minor differences in its $^1H$-NMR and IR spectra, but in the $^{13}C$-NMR spectra significant differences were observed between the H-H and H-T copolymers. All of the H-H and H-T copolymers were soluble in common solvents and the inherent viscosities were in the range 0.2-0.3 dl/g.

  • Cropping of head and tail ends of rod in wire Rolling Process is required to aviod roll damage, and prevent cobbles. In order to reduce the crop loss, the new crop control system for rotary shear of Wire rolling Process has been developed. Performance shows the developed system cut precisely within setting length. As a result, it is expected to increase the yield ratio of products about 0.2 percent and stabilize the operantional condition.

  • In this paper, the resonant characteristic of a Tonpilz transducer with a fixed tail mass has been researched and the feasibility to utilize the transducer in that resonant frequency has been analyzed. The additional resonance is occurred by adhering an elastic tube to the tail mass of a conventional Tonpilz transducer and fixing the other end of the tube. The characteristic of this low resonant mode is analyzed by means of the finite element method, then it is shown that this mode is affected by the variation of the stiffness of the tube as well as the mass of the head mass and tail mass. Based on the analysis results, the Tonpilz transducer is designed to meet specific performance conditions.
