Genome-wide Drug-induced Haploinsufficiency Screening of Fission Yeast for Identification of Hydrazinocurcumin Targets

  • Hydrazinocurcumin (HC), a synthetic derivative of curcumin, has been reported to inhibit angiogenesis via unknown mechanisms. Understanding the molecular mechanisms of the drug's action is important for the development of improved compounds with better pharmacological properties. A genome-wide drug-induced haploinsufficiency screening of fission yeast gene deletion mutants has been applied to identify drug targets of HC. As a first step, the 50% inhibition concentration $(IC_{50})$ of HC was determined to be $2.2{\mu}M$. The initial screening of 4,158 mutants in 384-well plates using robotics was performed at concentrations of 2, 3, and $4{\mu}M$. A second screening was performed to detect sensitivity to HC on the plates. The first screening revealed 178 candidates, and the second screening resulted in 13 candidates, following the elimination of 165 false positives. Final filtering of the condition-dependent haploinsufficient genes gave eight target genes. Analysis of the specific targets of HC has shown that they are related to septum formation and the general transcription processes, which may be related to histone acetyltransferase. The target mutants showed 65% growth inhibition in response to HC compared with wild-type controls, as shown by liquid culture assay.

Combustion Control through the DME Injection Timing in the Hydrogen-DME Partially Premixed Compression Ignition Engine (DME 분사 시기 조절을 통한 수소-DME 부분 예혼합 압축착화 연소 제어)

  • Hydrogen-dimethy ether(DME) partially premixed compression ignition(PCCI) engine combustion was investigated in a single cylinder compression ignition engine. Hydrogen and DME were used as low carbon alternative fuels to reduce green house gases and pollutant. Hydrogen was injected at the intake manifold with an injection pressure of 0.5 MPa at fixed injection timing, $-210^{\circ}CA$ aTDC. DME was injected directly into the cylinder through the common-rail injection system at injection pressure of 30 MPa. DME inejction timing was varied to find the optimum PCCI combustion to reduce CO, HC and NOx emissions. When DME was injected early, CO and HC emissions were high while NOx emission was low. As the DME injection was retarded, the CO and HC emissions were decreased due to high combustion efficiency. NOx emissions were increased due to the high in-cylinder temperature. When DME were injected at $-30^{\circ}CA$ aTDC, reduction of HC, CO and NOx emissions was possible with high value of IMEP.

  • Effects of dietary nutrient content on the biological index and serum chemistry of olive flounder Paralichthys olivaceus achieving compensatory growth were investigated. Six treatments were prepared in triplicate. Fish were hand-fed with the control (C) diet twice daily for 8 weeks (8W-C) or fish were starved for 2 weeks and then hand-fed with the C, high protein (HP), high carbohydrate (HC), high lipid (HL), or intermediate protein, carbohydrate and lipid (IPCL) diets for 6 weeks, referred to as 6W-C, 6W-HP, 6W-HC, 6W-HL, and 6W-IPCL, respectively. Weight gain of fish in the 8W-C, 6W-HP, and 6W-IPCL treatments was higher than that of fish in the 6W-C treatment. Condition factor (CF) of the fish in the 6W-HP, 6W-HC and 6W-IPCL treatments was higher than that of fish in the 8W-C and 6W-C treatments. The hepatosomatic index (HSI) of fish in the 6W-HC, 6W-HL and 6W-IPCL treatments was higher than that of the fish in the 8W-C, 6W-C and 6W-HP treatments. Serum chemistry except triiodothyronine ($T_3$) was not significantly different among the treatments. In conclusion, CF and HSI of the fish could be indices reflecting compensatory growth, whereas $T_3$ seemed to play a partial role in achieving compensatory growth.

Effects of Garcinia cambogia Extract Feeding on Body Weight and Lipid Profiles in Rats Fed a High-carbohydrate or High-fat Diet

  • This study was designed to evaluate the effects of high-carbohydrate (HC) or high-fat (HF) diet with hydroxycitric acid (HCA)-containing Garcinia cambogia in rats. Seventy male Sprague-Dawley rats were randomly divided into 7 groups and raised with experimental diets containing different HCA levels (0, 1.6, and 3.2%) and calorie sources (carbohydrate, fat) for 8 weeks. Energy intake was significantly reduced in rats fed a HC diet with HCA compared to the HC diet without HCA. Body weight gain was significantly reduced in HCA groups. In the diet groups, plasma total lipid and triglyceride (TG) levels of the HCA groups were significantly lower than those of the group without HCA. There were no significant differences in energy intake and plasma lipid profile in HF groups. These results suggest that HC diet with HCA was more effective in the reduction of energy intake, body weight gain, and plasma lipid contents than those of HF diet with HCA.

Prediction of Damage Extents due to In-Compartment Explosions in Naval Ships (내부 폭발에 의한 함정의 손상 예측)

  • In order to reasonably predict damage extents of naval ships under in-compartment explosion (INCEX) loads, two conditions should be fulfilled in terms of accurate INCEX load generation and fracture estimation. This paper seeks to predict damage extents of various naval ships by applying the CONWEP model to generate INCEX loads, combined with the Hosford-Coulomb (HC) and localized necking (LN) fracture model. This study selected a naval ship with a 2,000-ton displacement, using associated specifications collected from references. The CONWEP model that is embedded in a commercial finite element analysis software ABAQUS/Explicit was used for INCEX load generation. The combined HC-LN model was used to simulate fracture initiation and propagation. The permanent failures with some structural fractures occurred where at the locations closest to the explosion source points in case of the near field explosions, while, some significant fractures were observed in way of the interfaces between bulkheads and curtain plates under far field explosion. A large thickness difference would lead to those interface failures. It is expected that the findings of this study enhances the vulnerability design of naval ships, enabling more accurate predictions of damage extents under INCEX loads.

Characteristics and Action Pattern of Alkaline Protease from Streptomyces gviseus HC-1141 (Streptomyces griseus HC-1141이 생성하는 Alkaline Protease의 특성 및 작용양상)

  • An alkaline protease producing microorganism was isolated from soil and identified as Streptomyces griseus HC-1141. The optimum pH and temperature for the purified enzyme activity were 8.0 and $60^{\circ}C$, respectively. The enzyme was relatively stable in the pH range of 7.0-9.0 and at the temperature below $60^{\circ}C$. The activity of purified enzyme was inhibited by $Hg^{2+}$, $Cu^{2+}$, $Zn^{2+}$, $Ba^{2+}$ and $Fe^{2+}$, whereas activated by $Mn^{2+}$ and $Ca^{2+}$. $\varepsilon$-Amino caproic acid, 2,4-dinitrophenol and iodine did not show inhibitory effect on the activity of alkaline protease, but p-chloromercuribenzoic acid, ethylendiaminetetraacetic acid showed inhibitory effect on the enzyme activity. These result suggested that the protease was metalloenzyme, and require a reactive SH group for the activity. The reaction of this enzyme follows typical Michaelis-Menten kinetics with the $K_m$ value of $2.229{\times}10^{-4}$M and the $V_{max}$ of $46.08 {\mu}$g/min for casein. The activation energy for the alkaline protease calculated by Arrhenius equation was 3.643 kcal/mol. This enzyme hydrolyzed casein more rapidly than the hemoglobin and egg albumin.

Effect of Dietary Structural to Nonstructural Carbohydrate Ratio on Rumen Degradability and Digestibility of Fiber Fractions of Wheat Straw in Sheep

  • The effect of different dietary structural carbohydrate (SC) to nonstructural carbohydrate (NSC) ratios on fiber degradation, digestion, flow, apparent digestibility and rumen fluid characteristics was studied with a design using 18 wethers fitted with permanent rumen and duodenum cannulae. All sheep were divided into six groups randomly, receiving six diets with varying SC to NSC ratios. All diets contained the same proportion of wheat straw and concentrate. The dietary SC to NSC ratios were adjusted by adding cornstarch to the concentrate supplements. The duodenal and fecal flows of dry matter (DM), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose (HC) and cellulose (CEL) were estimated using chromium-mordanted wheat straw as a flow marker. The degradation parameters of wheat straw DM, NDF, ADF, HC and CEL were determined by incubating the ground wheat straw in nylon bags in the rumen for different periods of time. There was no effect (p>0.05) of the different dietary SC to NSC ratios on rumen pH or $NH_3$-N, but acetate, propionate and butyrate concentrations were significantly affected (p<0.05 or p<0.01) by dietary SC to NSC ratios in the rumen fluid. When the dietary SC to NSC ratio was 2.86, the highest rumen degradability of wheat straw DM, NDF, ADF and CEL was found, but the highest apparent rumen digestibilities of DM, NDF, ADF, HC and CEL occurred at a 2.64 SC to NSC ratio. However, because of compensatory digestion in the hindgut, the apparent digestibilities of DM, NDF, ADF, HC and CEL were highest when the dietary SC to NSC ratio was 2.40. In conclusion, there is a optimal range of dietary SC to NSC ratios (between 2.86 and 2.40) that is beneficial to maximize wheat straw fiber degradation and apparent digestibility.

Biocontrol of gray mold of cucumber and tomato by epihpytic bacteria in field condition

  • Bacterial isolates, CC178, PTC25, HC39 and KY165 originally obtained from the leaves of cucumber or tomato were selected for biocontrol agents against gray mold of cucumber and tomato by in vivo cucumber seedling assay. Each suspension of the selected epiphytic bacteria were sprayed three times at seven-day interval from early stage of cucumber in a field. Incidence of gray mold on cucumber fruits treated with isolates CC178, PTC25, HC39 and KY165 was 15.3%, 18.2%, 23.6%, and 10.4%, respectively, whereas that of control was 38.0% after 7 days of final spray. On the other hand, treatment with the selected isolates, CC178, PTC25, HC39, and KY165 on tomato showed 2.2%, 1.3%, 2.9%, and 3.5% in the incidence of gray mold on leaves, whereas that of control was 9.3%. All selected isolates had strong antagonistic activity against Botrytis cinerea on dual culture plate assay.

  • 본 논문의 연구에서는 IEEE 802.11e HCCA에서 멀티미디어와 같은 트래픽에 대해 보다 안정적인 전송을 위하여 일정 시간 동안 채널을 사용할 수 있도록 HC (Hyper Coordinator)가 허가함에 있어서 IEEE 802.11 PHY (physical layer)의 모듈레이션 변화에 따른 동적인 대응이 필요함을 논하고, 문제 해결을 위한 방안을 제시하였다. IEEE 802.11e의 HC는 멀티미디어와 같은 트래픽의 전송을 위하여 BSS (Basic Service Set) 안에 있는 무선 단말기와 TSPEC (Traffic Specification)을 이용하여 CAP (Controlled Access Phase) 채널을 설정하고 관리한다. 그러나 HC가 TSPEC을 설정할 당시와는 달리 무선 단말기들은 이동하거나 물리적으로 채널 환경의 변화로 IEEE 802.11 PHY의 모듈레이터 변경으로 설정된 TXOP 듀레이션 (duration)이 적절하지 않게 된다. 이와 같은 문제점을 밝히고 해결 방안을 아래와 같이 제안한다. 실험을 위해서는 NS-2와 유사하지만 보다 세련된 시뮬레이터를 활용하였다.

  • From the cooking data, Japanese rice showed higher water uptake but lower expansion volume, pH and iodine blue value than those of Korean rice. Japanese rice had higher maximum viscosity, breakdown viscosity and pasting temperature but lower final viscosity and setback viscosity than those of Korean rice by RVA analysis. Japanese rice had higher LC (low compression) hardness, U stickiness and HC (high compression) stickiness, LC balance and HC balance, but had lower HC hardness and thickness in the tensipresser data. Also Japanese rice had higher stickiness and balance, and lower hardness from the texturometer analysis. Japanese rice showed higher a cooked taste score than that of the Satake cooked taste machine. The various mean values of Japanese rice after cooking showed better cooking characteristics than the Korean rice. These results might be caused because Japanese rice had a little lower amylose and protein content, but higher tat acidity content.
