Identification of differentially expressed genes in the developmental stages from olive flounder Paralichthys olivaceus using an annealing control primer system

  • We employed a new and improved differential display reverse transcription-polymerase chain reaction (DDRT-PCR) method, which involves annealing control primers (ACPs), to identify the genes that are specifically or prominently expressed in olive flounder (Paralichthys olivaceus) juveniles (35 days post-hatch; dph) compared to larval-stage (dph 21) flounder. Using 60 ACPs, we identified eight differentially expressed genes (DEGs) and basic local alignment search tool (BLAST) searches revealed eight known genes. Gene expression levels were confirmed by RT-PCR. Phosphoglucose isomerase (PGI) was highly expressed at 21 dph, while nephrosin, myosin light chain (MLC), myosin heavy chain (MHC), carboxypeptidase A, chymotrypsin B, fish-egg protein, and matrix protein were expressed at 35 dph. PGI, MLC, and MHC expression was further analyzed by RT-PCR. The differentially expressed genes identified in this study may provide insights into the molecular basis of development in olive flounder.

Susceptibility of Tea Red Spider Mite, Tetranychus kanzawai (Acari: Tetranychidae) to Cyenopyrafen (차응애의 cyenopyrafen에 대한 감수성)

  • The acaricidal activity of cyenopyrafen on the tea red spider mite, Tetranychus kanzawai was evaluated in laboratory and field studies. In the laboratory, cyenopyrafen was highly effective against egg, larva, protonymph and adult female of T. kanzawai. In surviving adult females of T. kanzawai exposed to cyenopyrafen, we observed significant reduction in reproduction and no egg hatch. Effectiveness of cyenopyrafen against adult female of T. kanzawai on bean leaves was lasted for 15 days at treatments of 125 and 83.3 ppm. In a field study, cyenopyrafen exhibited excellent activity to mixed developmental stages of T. kanzawai. Based on the results, cyenopyrafen could be used for the control of T. kanzawai and treatment of this acaricide at 83.3 ppm was recommended as the field rate.

Effects of Nanometer Sized Silver Materials on Biological Toxicity During Zebrafish Embryogenesis

  • Commercial nanometer sized silver is widely used for its antibacterial effect; however, nanoparticles may also have ecotoxicological effects after being discharged into water. Nanometer sized silver can flow into aquatic environments, where it can exert a variety of physiologically effects in living organisms, including fish. The present study aimed to investigate the effect of nanometer sized silver on the development of zebrafish embryos, analyze the properties of commercial nanometer sized silver and define the toxicity relationship between embryogenesis and hatched flies. The commercial nanometer sized silver was analyzed in the $Ag^+$ ion form. The hatch rate decreased in the nano-silver exposed groups (10 and 20 ppt); furthermore, the hatched flies had an abnormal notochord, weak heart beat, damaged eyes and curved tail. The expression of the Sel N1 gene decreased in the nano-silver exposed groups, and the catalase activities of the exposed groups increased relative to those in the control group. Therefore, the $Ag^+$ ions in commercial nanometer sized silver could accumulate in aquatic environments and seriously damage the development of zebrafish embryos.

  • Megaselia scalaris, also known as the scuttle fly, has a humpbacked morphology and was categorized as an insect undergoing complete metamorphosis. Megaselia scalaris was reported to be found in diverse environments. Recently, the scuttle fly has been a focus of studies by forensic scientists, geneticists, and developmental biologists. However, detailed research into scuttle fly development is still unclear. Therefore, the current study has been performed to elucidate the development of Megaselia scalaris. The scuttle fly undergoes four distinct stages of development: egg, larva, pupa and adult fly. After mating, the scuttle fly eggs hatch within one day and become larvae. The newly generated larvae go through 1, 2, and 3 instar stages within a period of 5~10 days. The instar stage 1 takes 1~2 days, instar stage 2 takes 1~2 days, and instar stage 3 takes 3~5 days. Depending on the environmental temperature, the pupae stage takes approximately 10~15 days to complete. The pupa stage can be divided into five sub-stages: 1~3 days, 4~6 days, 7~9 days, 10~12 days and 13~15 days. After molting, the newly formed adult scuttle fly can fly after one day. The morphological characteristics of the scuttle fly during each stage of development was detailed within this study.

Effect of Isocyanate Curing Agents on the Physical Properties and Flame Retardancy of Flame-Retardant PU Coatings (이소시아네이트 경화제 종류에 따른 PU 난연도료의 도막물성 및 난연효과)

  • PU type flame-retardant coatings (TBAO/L-75, TBAOL ; TBAO/N-100, TBAON) were prepared by blending bromine-containing modified polyester (TBAO) which was synthesized in our earlier work. with two kinds of isocyanate curing agents, Desmodur L-75 and Desmodur N-100. Physical properties of the prepared flame-retardant coatings were tested. TBAOL shows better hardness than TBAON, while TBAON shows better viscosity, accelerated weathering resistance, yellowness index and lightness index difference than TBAOL. There were no remarkable differences in fineness of grind, $60^{\circ}$ specular gloss, cross-hatch adhesion, and abrasion resistance of TBAOL and TBAON. There was no discernable difference in flame-retardancy between the two flame-retardant coatings, TBAOL and TBAON. When the content of tribromo acetic acid, which is flame-retarding component, was 30wt% the LOI value was in a range of $29{\sim}30%$, which indicates that the two coatings are good flame-retardant coatings.

  • This study was a part of FSA study which was initiated by IMO and was applied to hatchway watertight integrity of bulk carriers. Hazards which were involved in high risk level were identified as follows. Ship Operation out of Design Criteria(Hatch Coaming Damage) and Poor Maintenance & Inspection(Securing Arrangement Damage). The potential risk was calculated by risk analysis and risk control option was made to reduce potential risk. The potential risk was about U$60,000/ship-year and could be reduced to about U$30,000/ship-year by applying RCO 1(Advanced system directly related to Hatchway Security). In addition, effectiveness of RCOs was shown by cost benefit assessment.

Effects of Dietary Ascorbic Acid on Performance, Carcass Composition and Bone Characteristics of Turkeys during High Summer Temperature

  • Heat stress is major welfare problem in the poultry industry. Huge economic losses occur every year because of mortality and decreased production due to high environmental temperatures. This experiment was conducted to determine the effects of level of inclusion of ascorbic acid supplementation to the diet on performance, carcass composition and bone characteristics of male turkeys during high summer temperature. A total of 120 day-old turkey chicks were wing banded at hatch and randomly distributed into 3 treatment groups. Three diets were formulated to provide a similar nutrient profile with the exception of using three graded levels of ascorbic acid, namely 0, 150 and 300 mg/kg of the diet. The experimental diets were used from 0 to 18 weeks of age. Body weights, feed intake and feed conversion ratio, slaughter weight, carcass yield, portions, composition and thigh and breast pigmentation of the turkeys were not affected by ascorbic acid (p>0.05). Also, shank and tibia bone characteristics were not affected by ascorbic acid (p>0.05).

  • Nowadays, as the development paeiod of new products becomes even shorter, the importance of Rapid Prototyping Technology(RPT) has been rapidly increased. The major application of RPT is an early verification of product designs and quick production of prototypes for testing. Moreover, RPT is applied not only as a second tooling process such as mold making and investment casting but also as a creating some physical structure in medical field. Despite the remarkable progress of RPT, it is required to improve various problems resulting from application such as production time, accuracy and materials. This paper presents a laser scan path generation for accuracy of stereolithographicparts The methodology of laser scan path generation is discussed based on the stereolithography, The procedure of this research is as follows : 1) Input laser scanning conditions such as a laser beam diameter and a laser scanning interval, 2) Reconstruct original contours without self intersecting offset, 3) Calculate offset about reconstructed contours, 4) Calculate intersection points between horizontal or vertical lines and offset contours for internal hatch, 5) Decide laser shutter on/off points. The algorithm developed and programmed by C language is verified as an efficient method after testing a number of STL files of mechanical parts.

  • Effect of fluctuating temperatures on the development of the immature beet armyworm, Spodoptera exigua (H bner), was analyzed. At constant rearing temperature regimes, the estimated developmental threshold temperatures were varied among stages and instars, but had an average 13$^{\circ}C$ from egg hatch to adult emergence. Based on the 13$^{\circ}C$ threshold temperature, we set up three different rearing temperature regimes having the same day-degrees. Two fluctuating temperature regimes changed significantly the developmenta1 period expected by the constant rearing temperature regime. Under the same thermophase temperature (25$^{\circ}C$), the thermocycling regime with the higher cryophase temperature (10$^{\circ}C$) decelerated the developmental rate probably by lowering temperature limit thor development, but that with the lower cryophase temperature (5$^{\circ}C$) gave a negative developmental effect.

A Study on the Preparation of Thermoplastic Powder Coating Material and Its Flame Retardancy (열가소성 분말 코팅소재 제조 및 난연특성 연구)

  • The purpose of this study is application to flame retardant powder coating(FRPC) material consisting of ammonium polyphosphate(APP) and magnesium hydroxide($Mg(OH)_2$) as a halogen free flame retardant into thermoplastic resin(LDPE-g-MAH). For improvement of adhesion, LDPE-g-MAH was synthesized from low density polyethylene(LDPE) and maleic anhydride(MAH). The mechanical properties as melt flow index, pencil hardness, cross-hatch adhesion and impact resistance of FRPC were measured. Also, the limited oxygen index(LOI) values were measured 17.3vol%, 31.1vol% and 33.7vol% for LDPE-g-MAH, FRPC-3(APP 15wt%, $Mg(OH)_2$ 15wt%) and FRPC-5(APP 30 wt%), respectively. The thermo gravimetry/differential thermal analysis(TG/DTA) of FPRC-3 was observed endothermic peak at $340^{\circ}C$ and $450^{\circ}C$, it was confirmed predominant thermal stability though the wide temperature range by APP and $Mg(OH)_2$. It was showed V-0 grade for FRPC-3 and FRPC-4(APP 20wt%, $Mg(OH)_2$ 10wt%) that a char formation and drip suppressing effect, and combustion time reduced by UL94(vertical burning test). It was confirmed that flame retardancy was improved with the synergy effect because of char formation by APP and $Mg(OH)_2$.
