꿀벌 (Apis mellifera)의 lymph 액으로부터 정제된 항균성 펩타이드인, Apidaecin Ⅰb는 18개의 아미노산 잔기로 구성된 염기성 non-helical 펩타이드이다. 본 연구에서는 우리나라 연안의 적조 (HABs, harmful algal blooms)를 일으키는 4종의 적조생물 (Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides 및 Gymnodinium catenatum)에 대한 Apidaecin Ⅰb의 살조효과를 조사하였다. Apidaecin Ⅰb의 4종의 적조생물에 대한 살조효과는 12.5$\mu{g}/mL$부터 50$\mu{g}/mL$에서 세포의 lysis 또는 ecdysis와 같은 형태로 현미경으로 관찰할 수 있었다. 또한 Apidaecin Ⅰb는 C. marina에 대해서 A. tamarense, C. polykrikorides 및 G. catenatum보다 더욱 강한 살조효과를 나타내었다. 이러한 HABs에 대한 Apidaecin Ⅰb의 살조효과 연구는 새로운 살조물질을 개발하기 위한 자료가 될 것으로 생각된다.
Quantifying the abundance of Chattonella species is necessary to effectively manage the threats from ichthyotoxic raphidophytes, which can cause large-scale mortality of aquacultured fish in temperate waters. The identification and cell counting of Chattonella species have been conducted primarily on living cells without fixation by light microscopy because routine fixatives do not retain their morphological features. Species belonging to the Chattonella marina complex, including C. marina and C. marina var. ovata, had high genetic similarities and the lack of clear morphological delimitations between the species. To estimate the abundance of C. marina complex in marine plankton samples, we developed a protocol based on the droplet digital polymerase chain reaction (ddPCR) assay, with C. marina complex-specific primers targeting the internal transcribed spacer (ITS) region of the rDNA. Cell abundance of the C. marina complex can be determined using the ITS copy number per cell, ranging from 25 ± 1 for C. marina to 112 ± 7 for C. marina var. ovata. There were no significant differences in ITS copies estimated by the ddPCR assay between environmental DNA samples from various localities spiked with the same number of cells of culture strains. This approach can be employed to improve the monitoring efficiency of various marine protists and to support the implementation of management for harmful algal blooms, which are difficult to analyze using microscopy alone.
전세계적으로 2000년대 이후로 남조류에 의한 녹조의 발생이 증가하고 있으며, 이와 관련된 환경 문제가 공중건강을 위협하고 인간 활동을 제한하고 있다. 1970년대 캐나다의 호수를 중심으로 수행된 다년간의 현장 연구를 통하여, 녹조발생의 핵심적인 영양 제한인자로 인이 제시되었고, 인 저감에 대한 수계 관리가 진행되었다. 그러나 2000년대 이후, 대형 담수수계에서 특히 Microcystis에 의한 녹조 현상에서는 인뿐만 아니라 질소가 남조류 녹조 발생에 미치는 영향이 부각되고 있다. 한국의 담수 수계에서도 이와 비슷한 남조류에 의한 위해성 녹조 번성 특징을 갖고 있으므로, 이러한 패러다임의 변화는 국내 담수수계의 영양염류 관리에서 중요한 의미를 갖는다. 최근 국제적인 관련 연구를 통하여, 위해성 남조류 번성을 막기 위해 제안된 방법은 다음과 같다. 1) 질소와 인을 함께 관리하는 전략, 2) 폐수의 수집 및 처리, 3) 호소 유입수의 사전처리, 4) 저니의 준설, 5) 체류시간의 단축, 6) 조류의 효율적 회수법, 7) 조류의 침강 및 응집 등이 제시되고 있다. 추가적으로 남조류의 생태학적 특성에 기반한 지속가능하고 통합적인 담수수계의 녹조 관리기법이 수립되어야 한다. 녹조를 유발하는 남조류는 척결되어야 할 생물체가 아니라, 담수 수계에 필수적인 미생물 구성원이기 때문이다.
The occurrence of harmful algae on the coast of Korea has been a cause of damage to the aquaculture industry and deterioration of the coastal ecosystem environment. A method is required to predict their outbreak in real-time at the site. Therefore, this study attempted to develop a small hybrid optical sensor and real-time monitoring system based on LiDAR that can be used in the field and laboratory and can be applied to various platforms. FMS-L specifically suggested the amount of Chlorophyll a (Chl a) in the sample by measuring and analyzing the fluorescence emitted by the irradiating light. The accuracy of FMS-L was verified by measuring the concentrations of standard Chlorophyll a substances and Margalfidinium polykirkoids. In addition, the precision was verified by comparing the measurement results of FMS-L using commercial equipment Phyto-PAM-II. This equipment is compact and easy to move. Therefore, it can be easily applied to field surveys, allows short time measurements (10 s), and can be applied at a distance of 10 m from the measurement site.
One of the most threatening consequences of eutrophic freshwater reservoirs is algal blooming which typically occur after the long a mega drought or/and irregular rainfall under influence of climate change. The long-term experiences of chemical treatment are known as a most practical effort to reduce health concerns from human exposure of harmful cyanobacteria as well as to preserve ultimate freshwater resources. Even though these conventional chemical treatment methods do not completely solve the algal residue problem in water treatment plant or directly in the water bodies, they still have big advantages as fast and efficient removal process of cyanobacteria due to cheaper, easier to manage. This review summarizes their chemical treatment scenarios of the representative coagulants, pre-oxidants and algaecides composed to chemical compounds which immediately may help to manage severe cyanobacteria blooms in the summer seasons.

Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.

Exploring mixotrophy of dinoflagellate species is critical to understanding red-tide dynamics and dinoflagellate evolution. Some species in the dinoflagellate genus Karenia have caused harmful algal blooms. Among 10 Karenia species, the mixotrophic ability of only two species, Karenia mikimotoi and Karenia brevis, has been investigated. These species have been revealed to be mixotrophic; however, the mixotrophy of the other species should be explored. Moreover, although K. mikimotoi was previously known to be mixotrophic, only a few potential prey species have been tested. We explored the mixotrophic ability of Karenia bicuneiformis, Karenia papilionacea, and Karenia selliformis and the prey spectrum of K. mikimotoi by incubating them with 16 potential prey species, including a cyanobacterium, diatom, prymnesiophyte, prasinophyte, raphidophyte, cryptophytes, and dinoflagellates. Cells of K. bicuneiformis, K. papilionacea, and K. selliformis did not feed on any tested potential prey species, indicating a lack of mixotrophy. The present study newly discovered that K. mikimotoi was able to feed on the common cryptophyte Teleaulax amphioxeia. The phylogenetic tree based on the large subunit ribosomal DNA showed that the mixotrophic species K. mikimotoi and K. brevis belonged to the same clade, but K. bicuneiformis, K. papilionacea, and K. selliformis were divided into different clades. Therefore, the presence or lack of a mixotrophic ability in this genus may be partially related to genetic characterizations. The results of this study suggest that Karenia species are not all mixotrophic, varying from the results of previous studies.
하계 우리나라 연안내만에서 빈번하게 발생하는 유해 적조생물 제어는 수산피해를 최소화하기 위한 중요한 국가적 현안문제이다. 본 연구에서는 Thiazolidinedione 유도체 물질인 GreenTD 농도 구배별로 유해 미세조류 4종(Chattonella marina, Heterosigma akashiwo, Cochlodinium polykriokides, Heterocapsa circularisquama)과 무해 미세조류 3종 (Chaetoceros simplex, Skeletonema sp., Tetraselmis sp.)에 대해 생물 고밀도 실험군과 저밀도 실험군에서 살조물질 농도별 살조효율과 선택성을 조사하였다. 유해종에 속하는 침편모조류 C. marina와 H. akashiwo는 각각 GreenTD 0.5와 $0.2{\mu}gL^{-1}$ 농도에서 단시간에 확실한 효과를 보였으며, 14일 동안의 관찰에서도 재성장을 보이지 않았다. 적조생물 C. polykrikoides은 GreenTD $0.2{\mu}gL^{-1}$ 이상의 농도에서 광합성활성이 현저하게 떨어졌고, 살조효율 역시 80% 이상으로 나타났다. 특히, GreenTD $0.2{\mu}gL^{-1}$에서도 C. polykrikoides가 재성장하지 않은 것으로 보아, 본 물질은 C. polykrikoides에 대한 살조효과가 우수할 것으로 판단된다. H. circularisquama는 고밀도 실험군에서 GreenTD $0.5{\mu}gL^{-1}$, 저밀도 실험군에서는 GreenTD $0.2{\mu}gL^{-1}$ 농도에서부터 일정하게 영향을 받는 것으로 파악되었다. 규조류 C. simplex와 Skeletonema sp.에 대해서는 생물농도가 고밀도일 때 GreenTD $0.2{\mu}gL^{-1}$에서는 크게 영향을 받지 않았으며, 초기 일정한 영향을 받은 후 시간 경과와 더불어 재성장이 이루어졌다. 특히 녹조류 Tetraselmis sp.는 최고농도인 GreenTD $1.0{\mu}gL^{-1}$에서도 일정하게 높은 값을 유지하였다. GreenTD 농도와 생물밀도에 따른 차이가 뚜렷하게 나타났으나, 전반적으로 살조물질의 효과는 침편모조류>와편모조류>규조류>녹조류 순으로 나타났다. 결과적으로 GreenTD 물질은 유해종에는 높은 살조능력이 있고, 무해종에는 일시적으로 광합성활성에 영향을 주지만, 시간의 경과에 따라 회복되는 것을 알 수 있다. 따라서, 적조생물 C. polykrikoides 제어하기 위해서는 고밀도 실험군에서 80.8%의 살조효과를 보인 GreenTD $0.2{\mu}gL^{-1}$의 농도가 적절할 것으로 판단되며, 현장 적용시 일시적인 희석등을 고려하여 적정농도보다 높게 살포하면 일정하게 높은 살조효율을 가질 것이며, 이는 경제성을 고려하여 충분한 경쟁력이 있는 물질로 기대된다.
최근 해수담수화 기술에 관한 관심이 증가하면서 해수 전처리 기술의 중요성이 날로 증가하고 있다. 해수담수화 기술은 미래 수처리 핵심기술로 자리매김하고 있고 이를 위한 전처리 기술의 올바른 선택과 운영은 향후 해수담수화 기술의 효율향상과 공정 최적화를 위해 중요하게 고려되어야 할 것이다. 해수담수화 전처리 기술의 목적은 주로 해수에 존재하는 입자성 물질, 콜로이드성 물질, 유기물질, 무기물질 그리고 미생물 오염물질 등의 처리를 통해 후단 담수화 기술의 효율성을 향상시키기 위함이나 전처리 기술 대상 처리물질의 범위는 매우 다양하여 맞춤형 전처리 기술의 적절한 적용이 필요하다. 해수담수화에서 올바른 전처리 기술의 적용은 후단 담수화 시설의 높은 처리효율 및 문제점을 최소화시킴과 동시에 해수의 큰 수질변동과 기후적인 그리고 지역적인 영향 등에 즉각적으로 대처할 수 있으므로 전처리 기술의 운영전략은 미래 해수담수화 기술의 성공여부를 결정짓기 위해 매우 중요하게 다루어져야 한다. 또한 최근에 많은 관심을 가지고 있는 해수 미세조류의 번성은 담수화 전처리 기술의 선정에 있어서 잠재적인 장애가 되고 있어 이에 대한 올바른 이해도 반드시 필요하다. 본 총설에서는 해수담수화 전처리 기술에 관한 그동안의 연구동향을 분석하고 해수담수화 전처리 기술의 선택 및 운전 최적화 달성을 위한 향후 도전과제들을 제시하고자 한다.
Real-time PCR을 적조출현주의보 발령에 활용하였다. Cochlodinium polykrikoides 적조예보를 위해 $10cells\;mL^{-1}$ 이상에서 적조출현주의보를 발령하였는데 현미경 검경은 저밀도에서 정확도가 낮기 때문에 이를 보완하기 위해 C. polykrikoides 특이적 real-time PCR을 사용하였다. 6월 말경에 C. polykrikoides이 통영, 남해 해역에 저밀도로 광범위하게 출현하였고 2015년 8월 2일 적조출현주의보를 발령하였다.
