Abalones are gastropod mollusks belonging to the genus Haliotis. Pacific abalones are regarded as a very important marine gastropod mollusk in Korea, Japan, China, and also in food industries around the world. In Korea, 6 species of abalone have been reported to occur along the coasts: Haliotis discus hannai, Haliotis discus discus, Haliotis madaka, Haliotis gigantea, Haliotis diversicolor supertexta, and Haliotis diversicolor diversicolor. This study was performed to discriminate the genetic variances by the partial sequences of the mitochondrial cytochrome c oxidase subunit I (COI) genes and random amplified polymorphic DNA (RAPD) analysis against four species of Pacific abalone (H. discus hannai, H. discus, H. madaka, H. gigantea). COI gene is reasonably well conserved and has been sequenced in various invertebrate taxa. The RAPD analysis technique is a relatively simple and low cost method that allows differentiation of taxa without the need to know their genomes. In this study, we investigated the genetic diversity, phylogenetic relationships within each species. The COI and RAPD analysis were able to distinguish between H. gigantea and the other three species. However, these analysis methods were inadequate to distinguish between H. discus and H. madaka. These results are believed to be able to provide a basis data for future hybrid breeding research by defining the genetically closely related four species of abalone, which is to develop new hybrid abalone for export using hybrid breeding.
This study was conducted to examine the effects of alterations in water temperature (WT) on biochemical and immunological factors in the hemolymph of the abalones, Haliotis gigantea and H. discus discus. The abalone were exposed to various WT; 18, 20, 22, 24, 26 and $28^{\circ}C$ for 96 hours. In biochemical factors, total-protein (TP), glucose, magnesium (Mg), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not significant changes in hemolymph of H. gigantea and H. discus discus. But calcium was significantly increased by high WT (${\geq}24^{\circ}C$). In immunological factor, The phenoloxidase (PO) activity was decreased in hemolymph of H. gigantea and H. discus discus exposed to high temperature (${\geq}22^{\circ}C$) compared to the control (P < 0.05). Whereas alkaline phosphatase (ALP) was not significantly changed. These results suggested that high temperature adversely affects the immunity of H. gigantea and H. discus discus.

This study was conducted to investigate the effects of elevated water temperature (WT) on biochemical and immunological factors in the hemolymph of the abalones, Haliotis discus hannai and H. discus discus. The abalone were exposed to various WT; 20, 22, 24, 26 and $28^{\circ}C$ for 4 days. In the control and $20^{\circ}C$, total-protein (TP), glucose and calcium (Ca) in hemolymph of H. discus discus were higher than the values in H. discus hannai. The values of magnesium (Mg), alkaline phosphatase (ALP) and lysozyme in H. discus hannai were similar to the H. discus discus in the control. There were no significant alterations in TP, glucose and Mg levels of hemolymph in H. discus hannai and H. discus discus by WT increases. The values of Ca, ALP and lysozyme were increased in H. discus hannai exposed to the high temperature (26 and $28^{\circ}C$) compared to control, while the values in H. discus discus were not significant difference between the WT groups. The phenoloxidase (PO) activity was increased in hemolymph of H. discus hannai exposed to high temperature (${\geq}24^{\circ}C$) compared to the control (P < 0.05). These physiological and immunological parameters were significantly changed in H. discus hannai. However, these parameters in H. discus discus were barely altered at the high WT (P < 0.05). These results suggested that H. discus hannai is considered to be more sensitive than H. discus discus at the high WT.
Genetic parameters for growth-related traits were estimated in 9-month old of two Korean abalone subspecies, Haliotis discus hannai and H. discus discus, using multiple traits of animal model. The data were collected from the records of 3,504 individuals produced from 16 sires and 17 dams in H. discus hannai and 821 individuals produced from 3 sires and 4 dams in H. discus discus, which was evaluated at the Bukjeju branch, NFRDI, from May 20, 2004 to February 14, 2005. The heritability estimates obtained from restricted maximum likelihood (REML) method range from 0.29 to 0.31 for three growth traits (shell length, shell width and body weight) in H. discus hannai and from 0.22 to 0.28 in H. discus discus, respectively. The heritabilities for shell shape and condition factor were lower than others of growth traits such as ranging from 0.03 to 0.24 in H. discus hannai and from 0.06 to 0.11 in H. discus discus, respectively. Genetic and phenotypic correlations were >0.91 between shell parameters and weight in two abalone subspecies, respectively, indicating that breeding for weight gains could be successfully achieved by selecting for shell length.
Here, we report the physiological changes and growth in disk abalone, Haliotis discus discus, in relation to dietary supplementation with citrus pomace (CP) 6%, Ecklonia cava residue (ECR) 6%, and CP + ECR (3% + 3%). The composition and nutrient content, survival rate and growth rate were measured 0, 4, 8 and 12 weeks after feeding the supplemented diets of CP and/or ECR. Moreover, the experiment of low salinity stress (25psu) for environmental resistance was examined for a period of 48 hours after feeding the supplemented diets for 12 weeks. The activities of superoxide dismutase (SOD), catalase (CAT), lysozymes, respiratory burst, and phenoloxidase were measured. The moisture content and crude protein condition of the body were increased with the addition of ECR only (P<0.05). We observed higher levels of survival in the experimental group compared with the control group. Moreover, the growth disk abalone that were fed a diet containing ECR was higher compared with the control group. However, the growth of abalone fed a diet containing CP was similar to the control group. With a rearing condition of low salinity stress, survival rate and lysozyme activity were increased in the ECR group compared with the control group. Dietary ECR reduced the level of CAT activity to approximately 30% of the control, however the level of CAT activity in the ECR group was similar to the start level of the previous stress. These results suggest that dietary ECR gives rise to an enhanced immunity in disk abalone, as a result of the decrease in CAT and lysozyme activity in particular. Accordingly, the growth and survival rate were increased by feeding an ECR-supplemented diet in the rearing of disk abalone, Haliotis discus discus.

현재 우리나라에 서식하는 전복은 말전복(Haliotis gigantea), 까막전복(Haliotis discus), 참전복(Haliotis discus hannai), 시볼트전복(Haliotis sieboldii), 오분자기(Haliotis diversicolor superfexta), 둥근전복(Haliotis diversicolor diversicolor) 등 6종이 있고, 이 중 참전복은 가장 맛이 좋아 주로 날 것으로 또는 건제품 및 죽의 형태로 소비되는 고가의 중요한 연안 수산물이다. 특히, 전복은 식품학적 측면으로 볼 때, 오래 전부터 우리나라, 중국 및 일본 등지에서 상당히 귀하게 여기는 기능성 수산물이어서 시장 가치가 매우 높다고 할 수 있다. Changes in 13 free amino acids (FAA) of Haliotis discus discus exposed to various salinities were investigated using a high performance liquid chromatography (HPLC). Taurine, glycine and alanine are three major contributors to the total FAA in the gill tissues of H. discus discus. Concentration of taurine was 114 imol/g dry tissue weights accounting for 76.64% of total FAA in H. discus discus. Levels of most FAAs in H. discus discus exposed to low and high salinities for 24 h decreased dramatically. Taurine concentration was slightly increased in the samples exposed to 20 psu and 25 psu for 48 h, and greatly increased after 120 h exposure. After 48 hrs exposure to 20 psu, 25 psu, 30 psu and 40 psu, methionine in H. discus discus was not detected; no methionine was detected in the sample exposed to 20 psu, 25 psu, 30 psu and 40 psu after 120 h of exposure. Taurine:glycine ratio increased depending upon hyper-hypoosmotic condition as well as period of osmotic stress. These data indicate that taurine, glycine and methionine play important role in regulating osmotic stress in H. discus discus. This study suggesting that FAA analysis is a useful tool to diagnose osmotic stress to H. discus discus.
Serratia marcescens characterized by the ability to produce red pigments inhabits various ecological niches. A strain Serratia marcescens PYU was isolated from abalone (Haliotis discus) collected at the West Sea in Korea. The isolated strain was gram-negative, motile, rods like coccus, oxidase-negative, and catalase-positive; and formed red pigment. S. marcescens PYU was grown in the presence of 0~10% (w/v) NaCl, at pH 4~9, and at $10{\sim}40^{\circ}C$. The strain PYU produced red pigment, and the extracted pigment showed antibacterial activity against Streptococcus iniae and Lactococcus garviae which has been known as an important fish pathogens. Further studies are underway to elucidate the direct relationship between the red pigment and antibacterial activity.
The present study investigated the feces production rate and fecal nitrogen production ratio of abalone, Haliotis discus hannai fed seaweed, Undaria pinnatifida. The abalone, with shell lengths of 3 cm, 5 cm, and 7 cm, were tested at temperatures of $12^{\circ}C$, $16^{\circ}C$, and $20^{\circ}C$ in a semi-recirculating aquaculture system. Under the given experimental conditions, the weight specific feces production rate (FPw) of the abalone increased with the decrease of shell length and the increase of temperature. The relationship of the FPw was. $FPw=-3.092+206.573/T - 1916.8/T^2+0.141L -0.037L^2(r^2=0.9412)$. In addition the abalone produced 17.8-22.5% of fecal material against ingested seaweed, Undaria pinnatifida, and $18.4-22.4\%$ of nitrogen ingested total nitrogen.

Abalones are gastropod mollusks belonging to the genus Haliotis. Fishery products are continuously declining worldwide while abalone production from aquatic farms has considerably increased. Although abalones are regarded as very important marine resources and considered to have big potential in sea-food farming industry around world, the slow growth rate of Pacific abalones is considered to be one of the most serious problems. For the genetic improvements in cultured abalone, advances in various breeding techniques for abalone have been reached through the introduction of selection, crossbreeding, hybridization, and polyploidy in several commercially important abalone species. Six species of abalone have been reported to be distributed along the coasts of Korea: Haliotis discus hannai, Haliotis discus discus, Haliotis madaka, Haliotis gigantea, Haliotis diversicolor diversicolor, and Haliotis diversicolor supertexta. The hybridization between these abalones may be one of the advanced technologies, and the preliminary experiments of interspecific hybrids between abalone species distributed in northern pacific areas including Korea, China, and Japan have been conducted. In this study, we reviewed the phylogenetic relationship of northern pacific abalone species which have the potential traits for aquaculture in Korea and their identifications. We also examined the development of molecular markers and some other aspects of the genetic approaches for successful development of hybrids.
