하이개부에 발생한 모기질종 (Pilomatricoma of the Subauricular Region: Report of Case)

  • Pilomatricoma or calcifying epithelioma is a benign tumor of hair follicle origin. Pilomatricoma presents a slowly enlarged and asymptomatic mass that located dermal or subdermal area. It showed mostly on face and upper limb and head and neck represents 50% of all case. And it developed almostly in the first 2 decade of life. Histologically, encapsulated mass composed of solid small basophilic cells and eosinophilic ghost cells. Surgical excision is the treatment choice and rarely recurs. The purpose of this article is to represent of pilomatricoma on subauricular region that treated with mass excision and local transpositional flap. Althrough the presurgical diagnosis of pilomatricoma may be difficult, clinicians must keep in mind the differential of head and neck masses that located subdermal layers.

The primary cilium as a multiple cellular signaling scaffold in development and disease

  • Primary cilia, single hair-like appendage on the surface of the most mammalian cells, were once considered to be vestigial cellular organelles for a past century because of their tiny structure and unknown function. Although they lack ancestral motility function of cilia or flagella, they share common ground with multiciliated motile cilia and flagella on internal structure such as microtubule based nine outer doublets nucleated from the base of mother centrioles called basal body. Making cilia, ciliogenesis, in cells depends on the cell cycle stage due to reuse of centrioles for cell division forming mitotic spindle pole (M phase) and assembling cilia from basal body (starting G1 phase and maintaining most of interphase). Ciliary assembly required two conflicting processes such as assembly and disassembly and balance between these two processes determines the length of cilia. Both process required highly conserved transport system to supply needed substance to grow tip of cilia and bring ciliary turnover product back to the base of cilia using motor protein, kinesin and dynein, and transport protein complex, IFT particles. Disruption of ciliary structure or function causes multiple human disorder called ciliopathies affecting disease of diverse ciliated tissues ranging from eye, kidney, respiratory tract and brain. Recent explosion of research on the primary cilia and their involvement on animal development and disease attracts scientific interest on how extensively the function of cilia related to specific cell physiology and signaling pathway. In this review, I introduce general features of primary cilia and recent progress in understanding of the ciliary length control and signaling pathways transduced through primary cilia in vertebrates.

인체 모유두세포의 증식에 미치는 갈근 에탄올추출물의 효과 (Effect of Puerariae Radix Ethanol Extract on the Proliferation of Human Dermal Papilla Cells)

  • In this study, we investigated the effect of Puerariae Radix ethanol extracts (EPR). The effect of the EPR on proliferation of human hair dermal papilla cells(HHDPCs) by MTT assay and observed Expression of mechanisms that regulate cell proliferation extracellular signal-regulated kinase(ERK) and Akt by western blot. The results showed EPR increased the proliferation of HHDPCs and up-regulation phosphorylation of ERK and Akt. ERK and Akt increased by EPR inhibited phosphorylation by PD98059 (ERK inhibitor) and LY294002 (Akt inhibitor), and cell proliferation was also inhibited. These results suggested EPR increases the proliferation of HHDPCs through phosphorylation of ERK and Akt, and therefore is a beneficial effect for the alopecia treatment.

  • To elucidate pathogenicity to hamster of encephalomyocarditis virus $K_3$ strain that was isolated in Korea from the swine with reproductive failures, adult male syrian hamsters were experimentally infected intraperitoneally with the virus at $10^{7.0}\;TCID_{50}/0.1ml$ and pathological and immunohistochemical studies were performed. The results obtained through the experiment were as follows. 1. Clinical signs such as depression, unkempt hair and bilateral parlysis of hind limbs were observed. 2. At necropsy, mild congestion was observed in the cerebrum, liver, kidney and lung, and atrophy was evident in testis. 3. In microscopic observation, degeneration and necrosis of the nervous cells and perivascular mononuclear cell infiltration were manifested in central nerve system, and various degrees of degeneration and necrosis of parenchymal cells were detected in pancreas, lacrimal gland, liver, kidney and testis. 4. In immunohistochemical observation, strong positive reactions were observed in degenerated parenchymal cell of testis, and weak positive reactions, in hepatocytes.

  • Intact mammalian hair and wool fibres are multi-compartmental composite materials consisting of a sulphur-rich outer protective cuticle layer surrounding elongated, highly keratinized, cortex cells. The cortex cells themselves are made up of crystalline, filamentous, low-sulphur a-helical keratin molecules embedded in a matrix of highly cross-linked, globular high-sulphur keratins. It is the structurally organised and highly disulphide cross-linked nature of these materials that provides them with their remarkable mechanical properties. However these mechanical properties are sensitive to environmental conditions such as water content, temperature and chemical treatment and the importance of their ultra-structural arrangements to overall mechanical properties in different environments is still not fully understood.(omitted)

  • The crude extract of Quintinia acutifolia Kirk inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (3 mm inhibition zone at $150\;{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (3 mm inhibition zone at $150\;{\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}$ $50,000\;{\mu}g/mL$ at $150\;{\mu}g/disc$). However, Candida albicans (ATCC 14053) did not observed the antimicrobial activity and the cytotoxic activity to BSC monkey kidney cells ($({\alpha})$ 5 mg/mL, $150\;{\mu}g/disc$).

  • Microbes is becoming increasingly forensic possibility as a consequence of advances in massive parallel sequencing (MPS) and bioinformatics. Human DNA typing is the best identifier, but it is not always possible to extract a full DNA profile namely its degradation and low copy number, and it may have limitations for identical twins. To overcome these unsatisfactory limitations, forensic potential for bacteria found in evidence could be used to differentiate individuals. Prokaryotic cells have a cell wall that better protects the bacterial nucleoid compared to the cell membrane of eukaryotic cells. Humans have an extremely diverse microbiome that may prove useful in determining human identity and may even be possible to link the microbes to the person responsible for them. Microbial composition within the human microbiome varies across individuals. Therefore, MPS of human microbiome could be used to identify biological samples from the different individuals, specifically for twins and other cases where standard DNA typing doses not provide satisfactory results due to degradation of human DNA. Microbial forensics is a new discipline combining forensic science and microbiology, which can not to replace current STR analysis methods used for human identification but to be complementary. Among the fields of microbial forensics, this paper will briefly describe information on the current status of microbiome research such as metagenomic code, salivary microbiome, pubic hair microbiome, microbes as indicators of body fluids, soils microbes as forensic indicator, and review microbial forensics as the feasibility of microbiome-based human identification.

A Scanning Electron Microscopic Study on the Glochidial Encystment of a Freshwater Clam, Anodonta arcaeformis on the Host Fish, Carassius auratus

  • A scanning electron microscopic study on the glochidium and glochidial encystment of Anodonta arcaeformis on the host fish Carassius auratus was conducted. The shape of the glochidium was apparently subtriangular and its average size was $270\;{\mu}m\;\times\;260\;{\mu}m\;\times\;145\;{\mu}m$. The glochidial shell valves were of the same size, kept together by a ligament that is 50.4 ${\mu}m$ in length and 5.5 ${\mu}m$ in width. Each of the glochidial shell valve had a long hook studded with many spines on the superior face. A large area of at the apex of the valve surrounding the base of the hook was provided with numerous small spines which became progressively smaller toward the periphery of the area. The glochidial shell valve consisted of two layers. The mantle cells line the glochidial shell valves and some of hair cells were observed. A larval thread was 2.3 ${\mu}m$ in diameter. In the artificial infection of the glochidia to one of the natural hosts, Carassius auratus, it took about three to four hours to encyst the glochidia with epithelial cells of the fish fins. The encystment method was the cell migration from the neighboring epithelial cells.

SK-MEL-2 세포에서 청호(靑蒿) 추출물의 Melanin 생성, Tyrosinase 활성과 생성, MMP-1 및 MMP-9 생성 억제 효과 (Inhibitory Effect of Artemisiae Annuae Herba Extracts on Melanin Synthesis, Tyrosinase Activities and Production Levels of Tyrosinase, MMP-1 and MMP-9 in SK-MEL-2 Cells)

  • Objectives: Artemisiae Annuae Herba is the dried aerial part of Artemisia annua L. (AAL). In Oriental medicine, Artemisiae Annuae Herba (AAH) is traditionally used to treat fever. AAH clears summerheat or damp-Heat, clears deficiency fevers, cools the blood and stops bleeding, stops malarial disorders and relieves heat, clears liver heat and brightens the eyes. Recently, there were many studies about effects of AAH on anti-oxidative, anti-inflammatory, anti-cancer, hair growth and plasma lipid composition. So, we expected AAH has an availability that can effect on skin whitening and elasticity. Methods: The present study was designed to investigate the effects of AAH on skin whitening and elasticity in SK-MEL-2 cells. In this experiment, the effects of AAH on proliferation rates, melanin synthesis, tyrosinase activities and production levels of tyrosinase, MMP-1 and MMP-9 in vitro were examined. Results: AAH did not affect viability of SK-MEL-2 cells and inhibited melanin synthesis induced by ${\alpha}$-Melanocyte-stimulating hormone (${\alpha}$-MSH) significantly. In addition, AAH also inhibited tyrosinase activity and lowered tyrosinase level in SK-MEL-2 cells. Finally, AAH inhibited productions of Matrix metalloproteinase-1 (MMP-1) and Matrix metalloproteinase-9 (MMP-9). Conclusions: These data suggest that AAH can be used to treat patients with skin diseases such as freckled face and also used as skin whitening agent.

Attenuation of ROS Generation by KCNE1 Genes in Cisplatin-treated Auditory Cells

  • Potassium is essential for the proper functioning of the ears. The inner ear's endolymph differs from all other extracellular fluids (in its positive potential) and in the ionic compositions in the various parts of the endolymphatic space. Ion concentration of the endolymph is 150 mM of potassium, which is comparable to the concentrations in other organs. Cisplatin (cis-diamminedichloroplatinum II: CDDP) is one of the most effective anticancer drugs, widely used against various tumors. However, its clinical use is limited by the onset of severe side effects, including ototoxicity and nephrotoxicity. For ototoxicity, a number of evidences in cytotoxic mechanism of cisplatin, including perturbation of redox status, increase in lipid peroxydation, and formation of DNA adduct, have been suggested. Therefore, in this study, the author investigated the relationship between the potassium ions on cisplatin-induced cytotoxicity in HEI-OC1 cells associated with reactive oxygen species (ROS). KCNE1 gene expression by the concentration of intracellular potassium appeared in the plasma membrane and increased the concentration of intracellular potassium. Cisplatin decreased the viability of HEI-OC1 cells, but the KCNE1 gene increased. Also, the KCNE1 gene significantly suppressed generation of intracellular ROS by cisplatin. Western blot analysis showed that the KCNE1 gene increased phase II detoxification enzymes markers such as superoxide dismutase 1 (SOD1), superoxide dismutase (SOD2), NAD(P)H:quinine oxidoreductases (NQO1), which were associated with the scavenger of ROS. These results suggest that the KCNE1 gene for intracellular potassium concentration ultimately prevents ROS generation from cisplatin and further contributes to protect auditory sensory hair cells from ROS produced by cisplatin.
