Red ginseng oil promotes hair growth and protects skin against UVC radiation

  • Background: A wide range of environmental factors, such as diseases, nutritional deficiencies, ageing, hormonal imbalances, stress, and ultraviolet (UV) radiation, may affect the structure and function of the skin that covers the entire surface of the human body. In this study, we investigated roles of red ginseng oil (RGO) in enhancing skin functions, including hair growth and skin protection, using mouse models. Methods: For hair growth experiment, shaved dorsal skins of C57BL/6 mice were topically applied with vehicle, RGO, RGO's major compounds, or minoxidil for consecutive 21 days and skin tissues were examined the hair growth promoting capacity. For skin protection experiment, SKH-1 hairless mice were topically applied with vehicle or RGO twice a day for three days prior to exposure to UVC radiation at 20 kJ/cm2. Skin tissues were collected to evaluate skin protective effects of RGO. Results: Topical application of RGO to C57BL/6 mice effectively promoted hair regeneration by inducing early telogen-to-anagen transition and significantly increasing the density and bulb diameter of hair follicles. Major compounds, including linoleic acids and β-sitosterol, contributed to RGO-promoted hair growth. Treatment with RGO as well as its major components upregulated expression of hair growth-related proteins. Furthermore, in SKH-1 hairless mice, RGO had a protective effect against UVC-induced skin damage by inhibiting inflammation and apoptosis, as well as inducing cytoprotective systems. Conclusion: These data suggest that RGO may be a potent agent for improving skin health and thereby preventing and/or treating hair loss and protecting skin against UV radiation.

효모성분추출물의 탈모치료 효과와 줄기세포활성화제 역할 (HAIR loss treatment effect and stem cell activator role of Yeast Constituent Extract)

  • The objective of this study is to find out the effect of yeast on hair loss treatment and the role of hair follicle stem cell activator, which is important in hair growth. The authors have recently produced a substance, which has no disgusting odor, does not precipitates and does not easily corrupt, to use instead of yeast acquired from raw rice wine(Makgeolli). The substance is named Yeast Constituent Extract(YCE). In this research, the Produced YCE was applied on the hair loss area of 10 Androgenic alopecia patients, twice every day for 6 months, in order to test the effect of hair loss treatment and the role of stem cell activator. As a result, all of the patients showed a significant growth of hair after 3 months of test, and showed much more growing, thickening and strengthening of hair after 6 months. As a result of measuring the number of hair strings in the same scalp region of the patients after 6 months, it is found that the density of hair has increased, indicating that the hair loss treatment was effective. Also the hair follicle stem cell was isolated from the patients and the contents of growth factors (IGF, VEGF, FGF, HGF) derived from hair follicle stem cell were measured with ELISA. As result, the amount is found to be about 10 times greater than before the test. The hair follicle stem cell contains many growth factors that affect growth of hair, so it takes a highly important role in hair loss treatment. The YCE that the authors have produced was found to be effective in increasing the contents of growth factors that are derived from hair follicle stem cell. Thus it can be inferred that the YCE plays a role as a stem cell activator that activates the hair follicle stem cells. In conclusion, the YCE is considered to be highly effective for hair loss treatment and to have a role as a stem cell activator.

Hair Zinc Level Analysis and Correlative Micronutrients in Children Presenting with Malnutrition and Poor Growth

  • Purpose: Zinc deficiency can induce serious clinical problems in the gastrointestinal (GI) system and immune system and can affect growth and development. It is more severe in younger patients. Chronic zinc deficiency is reflected more precisely in hair than in serum. We studied hair zinc levels and other hair and serum micronutrients in chronic malnourished children to identify which micronutrients are affected or correlated with the other ones. Methods: Hair mineral analyses were performed in 56 children (age, 1-15 years) presenting with malnutrition, poor growth, poor appetite, anorexia, with/without other GI symptoms (diarrhea, abdominal pain, constipation) from August 2012 to March 2015. Biochemical studies for macronutrients and major micronutrients were also conducted. Results: Hair zinc deficiency was diagnosed in 88%, and serum zinc deficiency was diagnosed in 55% of the children. There was no statistical correlation between serum and tissue zinc level. Hair zinc levels were highly correlated with serum vitamin D (r=-0.479, p=0.001), which also showed correlation with hair levels of magnesium and calcium. (r=0.564, 0.339, p=0.001, 0.011). Hair calcium level was correlated with serum pre-albumin (r=0.423, p=0.001). These correlations may explain the phenomenon that the major clinical manifestation of zinc deficiency is poor body growth. Clinical symptoms were resolved in most children after zinc supplementation. Conclusion: Hair zinc and mineral analyses are useful as a therapeutic guide in the clinical investigation of children with malnutrition and poor growth.

In Vivo Hair Growth Promotion Effects of Ultra-High Molecular Weight Poly-γ-Glutamic Acid from Bacillus subtilis (Chungkookjang)

  • We investigated the effect of ultra-high molecular weight poly-γ-glutamic acid (UHMW γ-PGA) on hair loss in vitro and in vivo. 5-Alpha reductase is an enzyme that metabolizes the male hormone testosterone into dihydrotestosterone. By performing an in vitro experiment to analyze the inhibitory effects of UHMW γ-PGA on 5-alpha reductase activity, we determined that UHMW γ-PGA did in fact inhibit 5-alpha reductase activity, indicating the use of UHMW γ-PGA as a potential 5-alpha reductase inhibitor in the treatment of men with androgenetic alopecia. To evaluate the promotion of hair growth in vivo, we topically applied UHMW γ-PGA and minoxidil on the shaved dorsal skin of telogenic C57BL/6 mice for 4 weeks. At 4 weeks, the groups treated with UHMW γ-PGA showed hair growth on more than 50% of the shaved skin, whereas the control group showed less hair growth. To investigate the progression of hair follicles in the hair cycle, hematoxylin and eosin staining was performed. Histological observations revealed that the appearance of hair follicles was earlier in the UHMW γ-PGA-treated group than in the control group. The number of hair follicles on the relative area of shaved skin in the UHMW γ-PGA-treated group was higher than that observed on the shaved skin in the control group. These results indicate that UHMW γ-PGA can promote hair growth by effectively inducing the anagen phase in telogenic C57BL/6 mice.

한약재 함유 헤어토닉 및 식품이 C57BL/6 마우스의 탈모 모델에서 모발성장에 미치는 영향 (Effects of Hair Tonic and Food including Korean Medicinal Herbs on Hair Growth in an Alopecia Model of C57BL/6 Mice)

  • Objective : This study was carried out to investigate effect of korean medicinal herbs-included hair tonic (MHT) and food (MF) on hair growth in an alopecia model of C57BL/6 mice. Methods : Six-weeks old mice were given laboratory rodent chow diet for 1 week. Then, we used a depilatory for hair removal on mice. The next day, mice were randomized and separated in 3 groups of 6 mice; normal group (N, vehicle epidermal application+normal diet-treated group), positive control group (C, 3% minoxidil sulfate epidermal application+normal diet-treated group), and the treatment group (T, HT epidermal application+ MF diet-treated group). The hair regrowth was determined by photograph, which was taken at 7, 14 and 21 days. And scoring indices, hair density and hair thickness, were evaluated by Folliscope at same day. In addition, the hair regrowth was analyzed insulin-like growth factor (IGF-1) in the dorsal skin of mice. Results : As a result, not only external appearance but also hair density and hair thickness on dorsal skin were increased more in C, T groups than in N group at 21 days with mixed treatment. Therefore, distinct regrowth pattern is appeared by treatment of herbal tonic and natural food for 21 days. Also, the expressions of IGF-1 on dorsal skin were higher in C, T groups than in N group. Conclusion : These results suggested that this herbal hair tonic and natural food has hair growth promoting activities and may be useful for treatment of bald or alopecia.

Lgr5와 결합하는 신규 헵타펩타이드를 이용한 인체 모낭 세포의 활성과 모낭줄기세포 분화 유도 (Novel Heptapeptide Binds to the Lgr5 Induces Activation of Human Hair Follicle Cells and Differentiation of Human Hair Follicle Bulge Stem Cells)

  • 본 연구에서는 7 개의 아미노산으로 이루어진 헵타펩타이드의 Lgr5 binding에 따른 인체 모낭 구성 세포의 활성에 대한 영향을 확인하였다. 표면 플라즈몬 공명(surface plasmon resonance, SPR) 시스템을 이용하여 헵타펩타이드가 Lgr5에 결합하는 것을 확인하였다. 인체 모유두세포(human hair follicle dermal papilla cell, HHFDPC)에 헵타펩타이드를 처리한 결과, 농도 의존적인 세포 증식이 나타났으며 β-catenin의 세포 내핵 이동 및 하위 유전자인 LEF1, Cyclin-D1, c-Myc의 발현 증가가 관찰되었다. 그리고 세포 증식 기전 관련 인자인 Akt와 ERK의 인산화 수준이 증가되었으며, 성장인자인 hepatocyte growth factor (HGF), keratinocyte growth factor (KGF), vascular endothelial growth factor (VEGF) 발현이 유도되었다. 또한 인체 모모세포(human hair germinal matrix cell, HHGMC)의 분화 관련 전사 인자와 인체 외모근초세포(human hair outer root sheath cell, HHORSC)의 분화 표지 인자들도 헵타펩타이드 처리 시 높은 발현율을 보였다. 추가적으로 우리는 헵타펩타이드의 인체 모낭줄기세포(human hair follicle stem cell, HHFSC) 분화에 대한 영향을 조사하였다. 그 결과, HHFSC 표지인자들의 mRNA와 단백질 수준이 감소하였고 반면에 분화 표지인자들은 증가하였다. 상기의 결과들은 헵타펩타이드가 인체 모낭 구성 세포에서 Wnt/β-catenin 경로를 촉진시켜 증식 또는 분화를 유도할 수 있음을 보여준다. 이를 토대로 종합해 볼 때, 본 연구의 헵타펩타이드는 모발 성장을 유도하고 탈모 개선에 도움을 줄 수 있는 기능성 원료로 사용될 수 있을 것으로 보인다.

우슬의 에탄올 추출물이 모발 성장에 미치는 영향 (Effect of ethanol extract from Achyranthis Radix on hair growth)

  • Objective : As more and more people are interested in appearance in modern society, the increasing number of hair loss population can have an important impact on psychological and social problems such as depression and inappropriate interpersonal symptoms. Therefore, much research is being done on treatments for alopecia using herbal extracts with relatively few side effects. This study was investigated about the effect of Achyranthis Radix (AR) extract with ethanol solvent on hair growth. Methods : We determined the promoting efficacy of AR-ethanol extract compared with minoxidil (MNXD) on the growth of human hair dermal papilla cells (HDPCs). Cell viability was measured by MTT assay and cell proliferation was confirmed by cell cycle analysis from flow cytometry in HDPCs. Also, we monitored the safe concentration range through MTT assay. And protein expression of hair growth-related genes (insulin-like growth factor 1 (IGF-1), Wnt3a, Protein kinase B (Akt), Extracellular signal-regulated kinase (Erk)) was monitored by western blot. Results : On cell cycle analysis, the G2/M phase was higher than that of the DW group in AR ethanol extract group at 0.05 and 0.1 mg/㎖. All protein expression levels of HDPCs were increased in AR ethanol extract groups and the MNXD group, compared to the DW group, respectively. Conclusion : As mentioned above, AR extract increased cell proliferation and the protein expression of IGF-1, Wnt3a, Akt, Erk in HDPCs. These results suggest that AR ethanol extract has promoted hair growth and it might be potential hair growth supplement.

오레가노 추출물의 모발성장 촉진효과 (Effect of Origanum vulgare Extracts on Hair Regeneration)

  • This study was carried out to investigate the effect of Origanum vulgare extracts on cell proliferation of human hair dermal papilla cell (HHDPC) using sulforhodamine B (SRB) assay, antioxidant activity by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method, expression of insulin-like growth factor-1 (IGF-1) by analyzing reverse transcriptase polymerase chain reaction (RT-PCR) and hair growth in a shaving animal model of C57BL/6 mice topically applying with an amount of 0.1 mL once a day for 3 weeks. The mice were divided into 4 groups including normal group (saline, N), negative control group (dimethyl sulfoxide, NC), positive control group (5 mg/mL minoxidil, PC), and experimental group (Origanum vulgare extracts, OV). Treatment of OV didn't show cytotoxicity in HHDPC up to 10 ${\mu}g/mL$ and exhibited antioxidant activity with $IC_{50}$ of 31.0 ${\mu}g/mL$. IGF-1 expression in the skin was significantly (p<0.05) increased in the PC and OV compared to the N or NC. PC and OV also showed a prominently promoted hair regrowth compared to the N or NC in hair growth observation. The hair regrowth of OV was significantly higher than that of PC (p<0.05). Therefore, these results indicate that O. vulgare extracts effectively stimulated hair growth in an animal model.

나라수 힐링액 샴푸와 토닉의 모발 유수분과 양모 효능 및 안전성 연구 -단일기관, 단일군 전후 비교 예비 인체 적용 시험- (The Efficacy and Safety Human Study of Narasoo Healing Shampoo and Hair Tonic for Scalp's Lipids & Moisture and Hair Growth -One Center, one group pre-post comparison Pilot Human study-)

  • Objectives : This study was carried out to examine the change of scalp and hair condition and to evaluate the efficacy of hair growth, subject's satisfaction and safety of the Narasoo healing shampoo and Hair tonic Methods : We offered 25 subjects Narasoo healing shampoo and Hair tonic and observed the changes among the starting point, 3rd week and 6th week. Study methods include the measurement of water and sebum content of the scalp, photo of scalp and hair, thermographic images and questionnaires. Results : It is estimated that Narasoo healing shampoo and Hair tonic have the positive effects on increase of water contents hair gloss, removal of excessive hair keratin, facilitation of hair growth, and decrease of sebum contents. Conclusions : Several meaningful effects of Narasoo healing shampoo and Hair tonic are observed in this study. Additional research is needed forward.

측백추출물에 따른 발모 효능에 대한 연구 (Hair Growth Promoting Effect of Thuja orientalis Ethanol Extracts on Hair Loss-induced DBA1J Mice)

  • Thuja orientalis extract (TOS) has been used on the hemostatic roborant, sedatives and stressless in oriental countries. We investigate the inhibitory effects on the productions of pro-inflammatory cytokines such as interleukin-1beta (IL-1β), IL-6, tumour necrosis factor alpha (TNF-α), and nitric oxide (NO) and the hair promotiont of TOS in the macrophage celline RAW264.7 or hair loss-induced DBA1J mice. T08 (50-500 ㎍/㎖) per se had no cytotoxic effect non-stimulated cells, but this extracts concentration-dependently reduced the release of NO, (IL-1β), IL-6, and TNF-α in the stimulate RAW264.7 cells with lipopolysaccharide (LPS). Moreover, when applied TOS (500 ㎍/㎖) to DBA 1J/mice, hair growth was promoted remarkably. These data suggest that T08 promotes hair growth on hair loss induced model mice and these properties may contribute to the anti- hair loss activity of the Thuja orientalis.
