We investigated the induction of resistance to Haemaphysalis longicornis infestation in rabbits that had been immunized with recombinant H. longicornis P27/30 protein. The success of immunological control methods is dependent upon the use of potential key antigens as tick vaccine candidates. Previously, we cloned a gene encoding 27 kDa and 30 kDa proteins (P27/30) of H. longicornis, and identified P27/30 as a troponin I-like protein. In this study, rabbits that were immunized with recombinant P27/30 expressed in Escherichia coli showed the statistically significant longer feeding duration for larval and adult ticks (P<0.05), low engorgement rates in larval ticks (64.4%), and an apparent reduction in egg weights, which suggest that H. longicornis P27/30 protein is a potential candidate antigen for a tick vaccine. These results demonstrated that the recombinant P27/30 protein might be a useful vaccine candidate antigen for biological control of H. longicornis.

The study examined the seasonal pattern of larvae, nymph, and adult life stages for Haemaphysalis longicornis and the rate of infection with severe fever from the thrombocytopenia syndrome virus in ticks collected from 12 sections (Jichukdong), 14 sections (Uldaeri), and 18 sections (Howondong) in Bukhansan dullegil for April-October 2019. Haemaphysalis longicornis ticks have been considered the main vector for severe fever with thrombocytopenia syndrome (SFTS). Haemaphysalis flava and Ixodes nipponensis were collected using the dragging and flagging method. The ratios of Haemaphysalis longicornis of the collected ticks were 91% (Jichukdong), 94% (Uldaeri), and 98% (Howondong). Monthly distributional studies of Haemaphysalis longicornis based on the developmental stage showed that the adults peaked in September while nymphs were collected more frequently from April through June. The larvae peaked in September and October. SFTS virus detection was performed using 2 × OneStep RT-PCR and nested PCR. On the other hand, no SFTS virus-specific gene was detected in 1,158 ticks of Haemaphysalis longicornis. This result provides estimates of the population densities for the life stages of Haemaphysalis longicornis and the associated disease risk in Bukhansan dullegil, where many people have visited since opening in 2010.
Chemotherapeutic treatment is still the foundation of tick control programs. This study investigated the acaricidal efficacy of cypermethrin alone and in combination with chlorpyrifos against Haemaphysalis (H.) longicornis. Unfed larval ticks were exposed to 0.1, 1.0, and 10 mg/mL cypermethrin for 60 min, after which the acaricidal efficacy was examined based on tick mortality. All compounds showed similar suppression curves, with the best control being achieved by cypermethrin and chlorpyrifos (1 : 1 ratio) at 10 mg/mL. Effective cypermethrin concentrations for tick control were two to seven times higher than the recommended doses, indicating resistance by H. longicornis.
The objective of this study was to evaluate the efficacy of an imidacloprid 10% and flumethrin 4.5% polymer matrix collar against the developmental stages of Haemaphysalis longicornis infesting dogs using the hair from treated dogs in a semi-in-vitro assay set. When incubated with 0.5 g of the hair collected from the dogs installed with the drug-embedded collar after 10 days, average death rate of the larval, nymphal, and adult H. longicornis was 21.5%, 77.9%, and 100% at 30 min, 1 hr, and 2 hr, respectively. This study showed the larval stages as well as the nymphal and adult stages of H. longicornis ticks are killed upon contact with the hair from dogs treated with the collar within 2 hr.
Larvae, nymphs, and adult stages of 3 species of ixodid ticks were collected by tick drag methods in Seoul during June-October 2013, and their infection status with severe fever with thrombocytopenia syndrome (SFTS) virus was examined using RT-PCR. During the period, 732 Haemaphysalis longicornis, 62 Haemaphysalis flava, and 2 Ixodes nipponensis specimens were collected. Among the specimens of H. longicornis, the number of female adults, male adults, nymphs, and larvae were 53, 11, 240, and 446, respectively. Ticks were grouped into 63 pools according to the collection site, species, and developmental stage, and assayed for SFTS virus. None of the pools of ticks were found to be positive for SFTS virus gene.
A chitinase cDNA named CHT1 was cloned from the hard tick, Haemaphysalis longicornis, and the enzymatic properties of its recombinant proteins were characterized. The CHT1 cDNA encodes 930 amino-acid (aa) residues including a 22 aa putative signal peptide, with the calculated molecular mass of the putative mature protein 104 kDa. The E coli-expressed rCHT1 exhibited weak chitinolytic activity against $4MU-(GlcNAc)_3$. The rCHT1 protein with higher activity was obtained using recombinant Autographa californica multiple nuclear polyhedrosis virus (AcMNPV), which expresses rCHT1 under polyhedrin promoter. These findings suggest that the rCHT1 expressed in baculovirus-mediated Sf 9 cells has a high activity than E coli-expressed rCHT1.
Tick saliva is critically important for continuous attachment to the host, blood feeding for days, and transmission of tick-borne pathogens. To characterize the patterns of inflammatory cytokine gene expression during its attachment and blood sucking time, peripheral blood samples of rabbits infested with Haemaphysalis longicornis ticks were collected at different intervals. Blood histamine concentration was evaluated as well as gene encoding IFN-${\gamma}$, TNF-${\alpha}$, IL-2, IL-6, IL-4, and IL-10 were compared with non-infested rabbits. Blood histamine concentration of tick-infested rabbits during fast feeding time was significantly higher than that of non-infested rabbits. In both nymph and adult tick infested rabbits, expression of TNF-${\alpha}$ and IFN-${\gamma}$ genes were decreased significantly (P<0.05), while expression of IL-4, IL-6, and IL-10 were increased 1.3 to 7 folds in adult infested rabbits with the exception of IL-6 that was significantly (P<0.05) decreased in nymph infested rabbits. IL-2 was not expressed in either nymph or adult infestation. H. longicornis saliva is capable of modulate host responses through a complex correlation with histamine and Th1, Th2 mediated cytokines that suppress the inflammatory responses directed toward inflammatory mediators introduced into the host during tick feeding.

Proteomic tools allow large-scale, high-throughput analyses for the detection, identification, and functional investigation of proteome. For detection of antigens from Haemaphysalis longicornis, 1-dimensional electrophoresis (1-DE) quantitative immunoblotting technique combined with 2-dimensional electrophoresis (2-DE) immunoblotting was used for whole body proteins from unfed and partially fed female ticks. Reactivity bands and 2-DE immunoblotting were performed following 2-DE electrophoresis to identify protein spots. The proteome of the partially fed female had a larger number of lower molecular weight proteins than that of the unfed female tick. The total number of detected spots was 818 for unfed and 670 for partially fed female ticks. The 2-DE immunoblotting identified 10 antigenic spots from unfed females and 8 antigenic spots from partially fed females. Matrix Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry (MALDI-TOF) of relevant spots identified calreticulin, putative secreted WC salivary protein, and a conserved hypothetical protein from the National Center for Biotechnology Information and Swiss Prot protein sequence databases. These findings indicate that most of the whole body components of these ticks are non-immunogenic. The data reported here will provide guidance in the identification of antigenic proteins to prevent infestation and diseases transmitted by H. longicornis.

This study investigated the seasonal distribution of ticks in Boeun, Chungbuk, South Korea, from 2016 to 2020. Over the five-year period, ticks were collected annually from four different sites. A total of 17,704 ticks belonging to three tick species (Haemaphysalis longicornis, Haemaphysalis flava, and Ixodes nipponensis) were collected. H. longicornis was the dominant species across all four sites with the highest density of 68.40% of the total collected specimens, followed by H. flava (3.53%) and I. nipponensis (0.06%). The larvae of unidentified species were also collected: 11.81 T.I. (28.01%). The H. longicornis population peaked during the spring season (May-June), whereas the larval population peaked during August and September. H. longicornis was collected the most from four sites (coniferous forest, broad-leaf forest, mountain path, and copse), with the exception of the larvae, which was collected the most in grassland and grave. H. flava was collected from all sites, but in a small proportion. The SFTS virus was not found in any of the 828 pools of ticks during 2016-2020. Based on the results of this study, the continuous surveillance of the tick population is recommended to mitigate the spread of diseases by these vectors.

This study was attempted to develop a rearing method of the tick(Haemaphysalis longicornis) at the laboratory in winter. The rearing conditions for ticks in winter were summarized as follows; Even in the winter, under controlled Incubator on 25~3$0^{\circ}C$ and 90~95%, temperature and humidity, respectively, it is possible to rear the ticks normally as on summer. in the usual laboratory room temperature and humidity, 20~$25^{\circ}C$ and 51 ~77%. In the ticks collected in summer, the life span of the ticks, from hatching to death, was 91~129(112.8$\pm$15.2) days in the lagoratory, and the number of the oviposited eggs from a tick were about 1,680~2,460 and the hatching rate of the oviposited eggs was about 95(92~98)%. The life span of the ticks which were reared in the laboratory in winter was 89-138(112.2$\pm$21.1) days, and the number of the oviposited eggs from the tick were about 1,382~2,674 and the hatching rate of them was about 89.5(87~92)%. In the rearing of the tick at the laboratory, the dogs, rabbits and mice were able to use the hosts for the tick. The proper temperature to feed the ticks on the cattle in the cold season was obtained by $Hokalong^{\circledR}$ which were attached on the out side of sac which covered bovine ear where attached ticks.
