MMP-1 inhibitory compounds were isolated from 120 Korean traditional edible plants. UP- 1 activity significantly increased linearly with increasing UVB dose in normal human foreskin fibroblast HS68 cell, showing maximum activity at approximately 35 $mJ/cm^2$, whereas in HaCaT cell, normal human keratinocyte, no increase was observed. Maximum secretion of MMP-1 after UVB treatment occurred around 36-48 k after treatment. MMP-1 inhibitory compound isolated from cold-water fraction of Cataegus pinnatifida Bunge showed the mort potent activity. The MMP-1 inhibitory compound was deduced as a peptide based on the fact that pronase digestion decreased the activity whereas periodate oxidation did not. The most potent UP- 1-inhibitory protein, CP-2Va-2, showing an activity of 88.5% against MMP-1, was isolated through sequential column chromatography on DEAE-Toyopearl 650C, Butyl-Toyopearl 650M, and Bio-Gel P-30. Molecular weight of CP-2Va-2 determined through high performance liquid chromatography and SDS PACE was 19 and 20 kDa. respectively, signifying a monomeric structure.
Objectives : Draconis Resina (DR), the resin of Daemonorops draco Bl., is used to circulate the blood and to stop bleeding. It also has been used to generate flesh including ulceration. The present study investigated the effects of DR extract on collagen metabolism in human fibroblasts and tyrosinase activity in mushroom tyrosinase.Methods : The effect of DR extract on type I procollagen production (collagen type I synthesis) and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts cell line. Hs68 cells after ultraviolet B (UVB, 312 nm) irradiation was measured using the enzyme - linked immunosorbent assay (ELISA). The tyrosinase activity was also measured to find out the whitening effects in mushroom tyrosinase by ELISA method.Results : There was no cytotoxicity at DR extract at concentrations of 10 μg/ml, 30 μg/ml, and 100 μg/ml. DR extract significantly inhibited the increase of collagenase activity, whereas it did not show on the reduction of type I procollagen in UVB damaged Hs68 cells. DR extract did not reduce the L - DOPA oxidation. However, it significantly reduced the tyrosinase activity by DR extract at concentraions of 0.1 mg/ml, 1 mg/ml and 10 mg/ml.Conclusions : In conclusion, DR showed the anti-wrinkle and whitening effects via the inhibition of collagenase production and the tyrosinase activity. These results suggest that DR may have potential as an anti-aging ingredient in cosmetic herb markets.
Background: Ginseng has been used as a traditional medicine and functional cosmetic ingredients for many years. Recent studies have focused on the potential biological effects of the ginseng berry and its ingredients. (+)-Syringaresinol (SYR) is enriched in ginseng berry and its beneficial effects on the skin have been recently reported. However, little is known about the its effects on the wound healing process of skin. Methods: Here, we evaluated the skin wound healing effect of (+)-SYR using the human fibroblast Hs68 cell and ex vivo pig and human skin tissue model. Scratch wound test and hydrogen peroxide (HPO) induce chemical wound model were employed. Results: (+)-SYR promoted the migration and proliferation of Hs68 cells without significant cytotoxicity at the tested concentrations. Especially, in ex vivo pig and human skin tissue, HPO-induced chemical wound was recovered almost completely by (+)-SYR. In line with the finding in Hs68, the protein expression levels of TGF-β and PCNA, a proliferation marker were increased, demonstrating the beneficial effects of (+)-SYR on skin wound repair. Conclusion: Collectively, we demonstrated that (+)-SYR from ginseng berry, can enhance the wound healing effect by accelerating cell proliferation and skin regeneration, suggesting the potential utility of (+)-SYR for skin wound repair.

In this study, we would like to confirm the collagen recovery effect through cell experiments of the flavonoid apigenin and baicalein and propose results that can support the collagen recovery effect through comparative transcriptome analysis. As a result of the study, it was confirmed that apigenin and baicalein were effective in recovering type I collagen damaged by UV in the HS68 cell line, and that both substances resulted in changes in the expression of ECM-related genes. Not only did it show a common mechanism for regulating the ECM, but it also showed the results of changing different categories of genes, making it possible to predict that it would have various effects on cells.

Objectives : This study is to investigate the anti-aging and antioxidant effects of Naesohwangryeon-tang(NSHRT) extract in Hs68 fibroblasts. Methods : The fibroblast cell line Hs68 cells were used and MTT analysis was performed to measure the cell viability of Naesohwangryeon-tang, and then MMP-1 was induced with the cytokine TNF-𝛼 to confirm the anti-aging effect through real-time PCR and ELISA. In addition, ROS was induced with the TNF-𝛼 to confirm the anti-oxidant effect through DCF-DA. The signaling pathway according to the antioxidant effect was performed by western blot. Results : Naesohwangryeon-tang did not show cytotoxicity at all concentrations(100-800㎍/㎖) treated on Hs68 cells, and the inhibitory effect of MMP-1 was also confirmed. In addition, it was confirmed that the intracellular ROS increased by TNF-α was decreased by Naesohwangryeon-tang, and it was confirmed that the expression of NQO1 was increased in a concentration-dependent manner of Naesohwangryeon-tang. Conclusions : These results suggest that Naesohwangryeon-tang can be used as an anti-aging material by demonstrating the anti-aging and antioxidant effects of Naesohwangryeon-tang.

Acute and chronic ultraviolet (UV) irradiation triggers severe skin photoaging processes, which directly disrupt the normal three-dimensional integrity of skin. UV light stimulates the expression of matrix metalloproteinases (MMPs) which degrade constituents of extracellular matrix (ECM) proteins. These MMPs reduce collagen synthesis and decrease skin elasticity and integrity, resulting in wrinkle formation. In this study, we identified Rosa multiflora hip extract (RME) as an effective anti-photoaging ingredient. First, cell proliferation activity of RME was verified using Hs68 human dermal fibroblast cell line. RME downregulated MMPs expression through the inhibition of activator protein (AP)-1. In addition, type I and IV collagen expressions were increased with RME treatment and UVB-induced inflammatory responses were also reduced after RME treatment. In conclusion, R. multiflora hip extract may effectively improve UVB-induced skin aging and wrinkle formation which may provide as an anti-aging, anti-wrinkle, and anti-inflammation ingredient in cosmetic industry.
CHSE(Chinook Salmon Embryo)-214 fish cell lines was cultured in Eagle's minimal medium (MEM) supplemented with 10% fetal bovine serum and 2mM-glutamin. Optimum growth temperature of CHSE-214 cell line was $20^{\circ}C$. Infectious pancreatic necrosis virus(IPNV) was successfuly multiplied and showed the cytopathic effect in CHSE-214 cell line. Infection symptom of IPNV was observed with inverted phase contrast microscopy. At 6h-12hrs post-infection, the cells infected with IPNV were similer to normal cells. At 18-24hrs post-infection, the cells were somewhat round form and a little swollen form than normal cells. At 30hs post-infection, the cells were becoming more abnormal cells. At 48-68 post-infection, the infected cells were lysed and showed the severe cytopathic effect.

DPPH radical-generating system was used to evaluate the antioxidant properties of the Rosaceae. The inhibitory effects of ethanolic extracts from Rosaceae plants were investigated on melanin biosynthesis which is closely related to hyperpigmentation. Of the Rosaceae extracts, Prunus sargentii, Rubus coreanus, Chaenomeles sinensis, Photinia glabra and Pyrus pyrifolia showed a potent inhibition of tyrosinase, the enzyme which converts 3-(3,4-dihydroxyphenyl) alanine (dopa) to dopachrome in the melanin biosynthetic process. Furthermore, MMT assay was used to check the cytotoxicity of extracts on the human foreskin fibroblast cell line, Hs68. Among the Rosaceae, bark of Prunus sargentii, bark wood of Photinia glabra and all parts or Chaenomeles sinensis showed more than $50\%$ inhibition of mushroom tyrosinase activity at 100 ug/mL and more than $80\%$ of strong DPPH radical-scavenging activity at 10 ug/mL. In audition to, they had no cytotoxic activity on Hs68. These results suggest that these extracts might be except a controler in pigmentation.

Acerola is an excellent ingredient because of its high natural vitamin C content, but it is difficult to stabilize and has hardly been studied as a cosmetic material. Therefore, this study developed a mixed liposome preparation for stabilizing acerola extract. As a safety test, the skin irritation test was evaluated by BCOP assay and HET-CAM assay. We evaluated the inhibition of tyrosinase activity, the whitening effect of melanin production, and the wrinkle effect of prochloragentype-I C-peptide production, and confirmed the possibility of functional cosmetics. In addition, a cream of liposomes containing acerola extract mixture was developed to evaluate the clinical studies of skin wrinkles and whitening. BCOP assay, HET-CAM assay and human skin primary irritation test results of liposomes containing acerola extract mixture showed no irritation and were safe from skin and eye. The result of tyrosinase activity by 75.8% at 1,000 ㎍/mL. As a result of the melanogenesis inhibition test, liposome with acerola extract showed the melanin content by 46.2% at 1,000 ㎍/mL that does not effect the viability of the B16F10 cell line. The result of collagen production test using ELISA kit, liposomes containing acerola extract mixture showed collagen synthesis ability by 152.1% at 1,000 ㎍/mL that does not affect the viability of the HS68 cell line. But it did not showed any inhibition of collagenase (MMP-1) activity at all concentrations in the MMP-1 activity inhibition test in the HS68 cell line. We performed clinical studies for the whitening and skin-wrinkle activity of cream containing acerola extract mixes liposome, was showed that the melanin contents and wrinkle was statistically significant reduction. These results suggest that liposomes containing acerola extract mixture have safe natural material, and skin wrinkle, whitening effects allowing their application in cosmetics as a natural product.
In this study, to develop a new anti-aging agent, we will examine the antioxidant activity and the inhibitory effects of the mosidae for the MMP-1 activities in UVB-irradiated HS68 human foreskin fibroblast cell lines and human keratinocytes (Ha- CaT cell line). And the consomme soup being prepared with mosidae according to different levels (3, 6, 9, 12%) and then quality characteristics will be determined by sensory evaluations and color values. It is also observed that the mosidae has the inhibition of UVB-induced MMP-1 activities. The consomme soup prepared with mosidae is found to reduce the level of $IC_{50}$ in SOD-like activities and hydroxy radical scavenging activities, respectively. And consomme soup prepared with mosidae (CS6) is the best by sensory evaluations. In conclusion, these results suggest that the consomme soup prepared with mosidae can be further developed as new anti-aging food.
