Soft Tissue Augmentation with Silk Composite Graft

  • Purpose: The objective of this study was to evaluate the interaction between 4-hexylresorcinol (4HR) and antibody as that affects the performance of a silk-4HR combination graft for soft tissue augmentation in an animal model. Methods: The silk graft materials consisted of four types: silk+10% tricalcium phosphate (TCP) (ST0), silk+10% TCP+1% 4HR (ST1), silk+10% TCP+3% 4HR (ST3), and silk+10% TCP+6% 4-HR (ST6). The antibody binding assay tested the 4HR effect and scanning electron microscopic (SEM) exam was done for silk grafts. The animal experiment used a subcutaneous pocket mouse model. The graft - SH0 or SH1 or SH3 or SH6 - was placed in a subcutaneous pocket. The animals were killed at one, two, and four weeks, postoperatively. The specimens were subjected to histological analysis and lysozyme assay. Results: Groups with 4HR applied showed lower antibody binding affinity to antigen compared to groups without 4HR. In the SEM examination, there was no significant difference among groups. Histological examinations revealed many foreign body giant cells in ST0 and ST1 group at four weeks postoperatively. Both ST3 and ST6 groups developed significantly lower levels of giant cell values compared to ST0 and ST1 groups (P < 0.001) at four weeks postoperatively. In the lysozyme assay, the ST1 and ST3 groups showed denser signals than the other groups. Conclusion: 4HR combined silk implants resulted in high levels of vascular and connective tissue regeneration.

Study on Inhibition of Microorganism Derived from Bovine Frozen Semen in In Vitro Culture of Bovine Follicular Oocytes (소 난포란의 체외배양에 있어서 소 동결정액유래 미생물 억제에 관한 연구)

  • This study was accomplished to illuminate factors of contamination of microbes in the culture medium and effect of antibiotics on prevention of contamination in the medium when bovine follicular oocyte was matured, fertilized and developed in vitro. 1. When washed or unwashed semen diluted with TCM 199 was incubated for 24∼72hr, contamination was come out. 2. When diluted semen with TCM 199 which has penicillin, streptomycin, gentamycin, kanamycin or nystatin was incubated for 24∼72hr, contamination was not come out only in kanamycin. 3. When imported semen which was diluted in TCM 199 with penicillin, streptomycin, gentamycin, kanamycin or nystatin was incubated for 24∼72hr, contamination was not come out in all treatments. 4. When semen which was diluted in BO, CZB, Ham's F10 or TCM 199 was incubated for 24∼72hr, kanamycin showed no contamination in all treatments, but gentamycin showed contamination in CZB, Ham's F10 and TCM 199. 5. When the semen diluted in BO was moved at 24hr after incubation into BO and incubated for 72hr, contamination was not come out, but when it was moved into the TCM 199 and incubated for 72hr, contamination was come out at 48 to 72hr of incubation. 6. When the semen diluted in BO, BO+BSA or BO+FBS containing gentamycin, kanamycin or nystatin was incubated for 24∼72hr, the diluted semen in BO or BO+BSA showed no contamination in all antibiotics but the diluted semen in BO+FBS showed no contamination only in kanamycin. 7.The Pseudomonas cepacia, Serratia liquefaciens, Klebsiella pneumaniae was respectively isolated in the semen of A, B, and C bull and the microbes are highly affected by amikacin, tobramycin and kanamycin. 8. When bovine folicular oocyte was in vitro matured, fertilized and developed in the simple medium with kanamycin, 26.6% was developed to over 32cell stage embryo.

Infiltration Rate of Some Upland Soils in Korea (우리나라 밭토양의 수분침투속도(水分浸透速度)에 관하여)

  • The infiltration rates of the upland soils on hill side slope were investigated in situ using rainulator of which rainfall intensity was 100mm/hr. The soil moisture profile after the water infiltration was compared with that calculated from Youngs' equation. The results obtained are as follows: 1. Time required for infiltration rate to reach constant during rainfall was 15 to 25 min. The infiltration rate measured after 30 min was considered to be final infiltration rate. 2. The final infiltration rates of clay soils were lower than 10mm/hr., loamy soils 10 to 20., coarse loamy soils 20 to 30, and sandy soils higher than 30mm/hr., respectively. 3. The saturated hydraulic conductivity of the surface soil of Samgag sandy loam was 0.47mm/min., Songjeong clay loam0.16 mm/min., and Jeonnam silty clay loam 0.14mn/min., respectively. 4. The soil moisture profile calculated from Young's equation was in close agreement with measured in situ.

Elimination capacities of toluene and ammonia in the bio-filter system depending on type of media (담체 종류에 따른 바이오필터의 톨루엔과 암모니아 분해능 평가)

  • Contribution of immobilized media with bacteria to the odor removal was evaluated in a lab scale bio-filter compared to that with sponge or ceramic media without the immobilized bacteria. Candida tropicalis for volatile organic compounds and ammonium oxidizing bacteria (AOB) for inorganic compounds were used as seeds in lab-scale bio-reactors. Three different type of media in the bio-reactors that immobilized bioreactor (IBR), sponge bioreactor (SBR), and ceramic bioreactor (CBR) were examined, respectively. An empty bed contact time (EBCT) of the bio-filters was fixed as 60 seconds, and the inlet concentration of toluene was changed from 20 ppm to 200 ppm to observe the removal efficiency depending on the concentrations. As a result, the maximum elimination capacities of IBR, SBR, and CBR were 166 $g/m^3/hr$, 138 $g/m^3/hr$, and 138 $g/m^3/hr$, respectively. In addition, toluene as an organic compound and ammonia as an inorganic compound were applied together with different inlet concentrations varied from 80 ppm to 250 ppm of toluene and from 2.5 ppm to 40 ppm of ammonia. The toluene maximum elimination capacities in IBR, SBR, and CBR were 97.4 $g/m^3/hr$, 59.5 $g/m^3/hr$, and 81.9 $g/m^3/hr$, respectively. The ammonia maximum elimination capacities were reached as 7.2 $g/m^3/hr$ in IBR, 6.6 $g/m^3/hr$ in SBR, and 7.0 $g/m^3/hr$ in CBR.

The Oxidative Effects of Benzo[a]pyrene in Rat Hepatocyte Primary Culture (랫드 간세포 일차배양에서 Benzo[a]pyrene의 산화 효과)

  • The objectives of present study were to investigate the effects of benzo[a]pyrene(BaP) on cytotoxicity, lipid peroxidation and antioxidant enzymes in rat hepatocyte primary culture. Primary cultures of rat hepatocytes were incubated for 24 hr, 48 hr or 72 hr in the presence of various concentrations (0, 10, 20, 30, 50 or 100 $\mu.$ M) of BaP. Cytotoxicity and cell viability were determined by measuring glutamic oxaloacetic transaminase(GOT) activity, lactate dehydrogenase(LDH) activity and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MIT) value. Lipid peroxidation was evaluated using thiobarbituric acid reactive substances(TBARS) assay. Effects on antioxidant system were determined by measuring glutathione peroxidase(GPx) activity, glutathione reductase(GR) activity and glutathione concentration. Activities of GOT and LDH, MTT value as well as TBARS concentration were not affected by up to 100 $\muM$ of BaP for 24 hr incubation. However, BaP at the concentration of 50 $\muM$ for 48 hr incubation or at the concentration of 30 $\muM$ for 72 hr incubation began to increase LDH activity and TBARS concentration but decrease MTT value, representing that BaP caused cytotoxicity and decreased cell viability in dose- and time-dependent manners. GPx activity began to be decreased by BaP at the concentration of 50 $\muM$ for 72 hr incubation. Whereas, GR activity began to be decreased by BaP at the concentration of 20 $\muM$ for 72 hr incubation. Glutathione concentration began to be decreased by BaP at the concentration of 20 $\muM$ for 72 hr incubation and was further reduced to 90% by 100 $\muM$ of BaP. These results demonstrate that BaP caused cytoctoxicity and decreased cell viability by increasing lipid peroxidation and decreasing glutathione concentration as well as activities of GPx and GR.

Effects of PNF Technique on Delayed Onset Muscle Soreness After Eccentric Exercise

  • This study examined the effects of hold-relax with agonist contraction (HR-AC) on the symptoms of delayed onset muscle soreness (DOMS) induced by intensive eccentric exercise of the non-dominant biceps brachii. Ten men (mean age=26.7 yrs, mean height=172.1 cm, mean weight=66.2 kg) and ten women (mean age=27.4 yrs, mean height=165.9 cm, mean weight=60.7 kg) who had not participated in a regular exercise program for the upper extremities in the previous six months were randomly assigned to one of two experimental groups: the HR-AC group, or the control group. We measured joint range of motion (ROM), maximal voluntary isometric contraction (MVIC), and muscle soreness before eccentric exercise, and 24, 48, and 72 hours after eccentric exercise. The subjects in the HR-AC group received the HR-AC technique in the non-dominant biceps brachii. The HR-AC technique was applied 24 and 48 hours after eccentric exercise. There was no significant difference between the HR-AC and the control group. However, the HR-AC group, compared to the control group, had a significant difference between the time points of the various parameters. Increased ROM (p<.05), decreased muscle soreness (p<.05), and reduced MVIC (p<.05) were found in the HR-AC group after 72 hours. Decreased ROM (p<.05) and MVIC (p<.05), and increased muscle soreness (p<.05) were observed in the control group. These findings suggest that the HR-AC technique effectively reduces muscle soreness and increases ROM 72 hours after eccentric exercise.

Optimization of the Processing Conditions for Heated Garlic Juice by Response Surface Methodology

  • This study was designed to determine the optimum conditions of heating temperature, heating time and dilution rates for producing heated garlic juice by using central composite design of response surface methodology. Garlic 3Department of Biosystems Engineering, Chungbuk National University, Cheongju 361-763, Korea was heated using a high temperature and pressure treatment apparatus. Total soluble solid contents ranged from 4.4 at $130^{\circ}C$ 3 hr and 6 fold dilution to 5.89 at $115^{\circ}C$ for 2 hr and 8 fold dilution. The highest total acidity was 0.55% at $120^{\circ}C$ for 3 hr and 2 fold dilution. The pH ranged from 4.01 at $130^{\circ}C$, 3 hr and 6 fold dilution to 5.85 at $120^{\circ}C$, 1 hr and 6 fold dilution. From the results of statistical analysis on the sensory evaluation the predicted optimum processing conditions for best color, taste, flavor and overall acceptance were $119.41^{\circ}C$, 3.11 hr, 5.85 fold dilution, $118.23^{\circ}C$, 3.57 hr, 3.15 fold dilution, $120.54^{\circ}C$, 3.47 hr, 5.01 fold dilution and $119.54^{\circ}C$, 3.18 hr, 5.66 fold dilution, respectively. The application of response surface methodology for preparing heated garlic juice processing showed a good correlation with high significance.

Effects of Red Ginseng Intake on Muscle Injury Due to Eccentric Exercise

  • This study examined the effects of Korean red ginseng (RG) on muscle injury induced by eccentric exercise. Eighteen college male students were randomly assigned to either the RG group or the placebo group (P). The RG group ingested RG extract at 20 g/day (mixed with 200 mL of water) for 7 days prior to performing a downhill treadmill exercise and for 3 days after performing the downhill treadmill exercise, while the P group ingested 200 mL of water containing Agastachis Herba for 7 days prior to and 3 days following a downhill treadmill exercise. All subjects performed 2 bouts of a downhill treadmill exercise (6 km, -14 degree slope, 12 km/hr speed) with a 5 minute resting interval between bouts. Blood samples were drawn immediately before and after exercise, and at 1 hr, 24 hr, 48 hr, and 72 hr. Two-way repeated ANOVA documented that creatine kinase (CK) levels in the RG group were significantly reduced at 48 hr after exercise, as compared with P group, and remained constant until 72 hr after exercise. Lactate levels in the RG group were also significantly lower at 24 hr and 72 hr after exercise as compared to the P group. We conclude that supplementation of RGreduces CK levels and may prevent muscle injury induced by eccentric exercise.

Effects of Trichostatin A on In Vitro Development of Porcine Parthenogenetic and Nuclear Transfer Embryos

  • Developmental potential of cloned embryos is related closely to epigenetic modification of somatic cell genome. The present study was to investigate the effects of applying histone deacetylation inhibitor, trichostatin A (TSA) to activated porcine embryos on subsequent development of porcine parthenogenetic and nuclear transfer embryos. Electrically activated oocytes were treated with 5 nM TSA for different exposure times (0, 1, 2 and 4 hr) and then the activated embryos were cultured for 7 days. The reconstructed embryos were treated with different concentrations of 0, 5, 10 and 25 nM TSA for 1 hr. Also 5 nM TSA was tested with different exposure times of 0, 0.5, 1, 2 and 4 hr. And fetal fibroblast cells were treated with 50 nM TSA for 1, 2 or 4 hr and with 5 nM TSA for 1 hr. Cumulus-free oocytes were enucleated and reconstructed by TSA-treated donor cells and electrically fused and cultured for 6 days. In parthenogenetic activation experiments, 5 nM TSA treatment for 1 hr significantly improved the percentage of blastocyst developmental rates than the other groups. Total cell number of blastocysts in 1 hr group was significantly higher than other groups or control. Similarly, blastocyst developmental rates of porcine NT embryos following 5 nM TSA treatment for 1 hr were highest. And the reconstructed embryos from donor cells treated by 50 nM TSA for 1 hr improved the percentage of blastocyst developmental rates than the control group. In conclusion, TSA treatment could improve the subsequent blastocyst development of porcine parthenogenetic and nuclear transfer embryos.

  • E-Learning, a technology-based learning process is the obtaining of knowledge from the basis of IT technology. The development of e-Learning requires corporate educational staff as well as HR staff, who manage e-learning, to drastically change their roles. The main reason for this is that the properly adapted and highly experienced education and training abilities of HR staff could accelerate the adoption and usage of e-Learning but they need to clearly select and define an appropriate curriculum from among the extensive mass-customized content with an e-learning basis. Here the research has analyzed the influence of the introduction and usage of e-Learning in hotels on HR staff and their roles from the viewpoint of hermeneutic phenomenology.
