진교(秦?, Gentianae Macrophyllae Radix)의 품질표준화에 관한 연구 (Standardization of Gentianae Macrophyllae Radix)

  • The Radix of Gentiana macrophylla (Gentianaceae) has been used for dispelling wind-damp, relaxing the sinews, and clearing heat from deficiency. In order to evaluate its quality, the method for the quantitative determination of gentiopicroside as standard compound has been developed. Gentiopicroside was a major compound of Gentianae Macrophyllae Radix. It was analyzed by HPLC using 30% MeOH as a solvent system. The amount of gentiopicroside from Gentianae Macrophyllae Radix was in the range of $2.22{\pm}1.22%\;(n=18)$.

향신료의 약물대사효소 CYP3A4 저해효과 (Inhibitory Effect of a Drug Metabolizing Enzyme CYP3A4 on Spices)

  • For the determination of inhibiting cytochrome P450(CYP)3A4 activity, an improvement HPLC method was established by using a new internal standard and solvent system. Moreover, CYP3A4 amount for a optimum reaction of enzyme was determined by a comparative study with a variety concentration of enzyme. Using a established method, inhibitory effect of CYP3A4 that is drug metabolizing enzyme Investigated on EtOAc extracts of 5-class spices. As a result of experiment, EtOAc extract of white pepper (Piper nigrum L.) showed strong inhibitory activity. On a continuous experiment, the fraction 2, 4 and 5 of while pepper extract showed remarkable inhibitory activity. Pipeline, a main constituent of pepper was not included in these fraction. It is suggested that major compounds for the inhibitory activity of white pepper may be other ingredient that is not piperine.

Studies on Microbial Transformation of Meloxicam by Fungi

  • Screening-scale studies were performed with 26 fungal cultures for their ability to transform the anti-inflammatory drug meloxicam. Among the different fungi screened, a filamentous fungus, Cunninghamella blakesleeana NCIM 687, transformed meloxicam to three metabolites in significant quantities. The transformation of meloxicam was confirmed by high-performance liquid chromatography (HPLC). Based on the liquid chromatography-tandem mass spectrometry (LC-MS/MS) data, two metabolites were predicted to be 5-hydroxymethyl meloxicam and 5-carboxy meloxicam, the major mammalian metabolites reported previously. A new metabolite was produced, which is not detected in mammalian systems. Glucose medium, pH of 6.0, temperature of $27^{\circ}C$, 5-day incubation period, dimethylformamide as solvent, and glucose concentration of 2.0% were found to be suitable for maximum transformation of meloxicam when studied separately. It is concluded that C. blakesleeana can be employed for biotransformation of drugs for production of novel metabolites.

  • In this work, the quantitative estimation of fatty acids in sesame and perilla oil by high performance liquid chromatography (HPLC) was investigated. The analysis of fatty acids were separated by HPLC using a differential refractometer as a detector. With micro Bondapak $C_{18}FFAA$ column and acetonitril, chloroform and tetrahydrofuran mixture as a solvent. In the fatty acid compositions, sesame oil was composed mainly of linoleic and oleic acids 49.6 % and 34.7%. In perilla oil, the amounts of oleic, linoleic and linolenic acids were 13.6%, 14.5% and 63.8%, respectively.

목향(Saussureae Radix)으로부터 Costunolide의 분리 및 함량분석 (Isolation and Quantitative Determination of Costunolide from Saussurea Root)

  • Isolation and quantitative determination of costunolide from Saussurea lappa Clarke (Compositae) has been conducted by using HPLC method. Costunolide in an acetone extract from the crude drug was separated on a RP-18 column using a $MeOH-H_20$ mixture (65:35) as an eluent and the average content is about $1.32{\sim1.42%.$ The content of costunolide in dried extract was decreased by about 24% in seven days. However it showed a slight decrease in solution. It is highly recommended that quantitative determination of costunolide from Saussureae Radix should be conducted as early as possible after solvent extraction.

Monascus purpureus에서 화색식용색소의 분리 및 화학적 특성 (Isolation and Chemical Characterization of Yellow Food Pigments from Monascus Purpureus)

  • The isolation and chemical characterization of yellow food pigments from Monascus purpureus were studied according to the compositions of media. Monacus yellow pigments were isolated and purified by solvent fractionation, silicagel column chromatography, TLC and HPLC. The retention time of Monascus yellow pigments isolated by HPLC was respectively 5 min(I) and 9 min(II) as the yeast malt extract agar(YMA) media and was respectively 4.6 min(III), 5 min(I) 5.7 min(IV), 8.3 min(V), 9 min(II) and 10.7 min(Ⅵ) at the malt extract agar(MEA) media. The structure of monascin(I), ankaflavin(II), 6,11-dihydrorubropunctatin(III), 6,11-dihydromonascorubrin(V) and unknown compounds(IV,Ⅵ) was elucidated by EI-Mass, H and C NMR, UV-visible spectrometer. Therefore, it was suggested that 6,11-dihydrorubropunctatin(III) and 6,11-dihydromonascorubrin(V) are new intermediates of Monascus yellow pigments.

A Convenient HPLC/ELSD Method for the Quantitative Analysis of Betaine in Lycium chinense

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

  • The methanol extracts of AraIia elata showed antimicrobial activity against bacteria. The antimicrobial active principle was successively purified with solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, silica gel partition column chromatography and HPLC. The active substance was isolated by HPLC on $C_{18}$ column with an acetic acid-MeOH system. The active substance was identified as trans-4-hydroxycinnamic acid by MS, $^1H-NMR\;and\;^{13}C-NMR$.

Antioxidant Activities of Agrimonia pilosa Ledeb: In Vitro Comparative Activities of Its Different Fractions

  • The antioxidant potentials of the methanol extract and its various fractions from Agrimonia pilosa Ledeb were measured as 1,1-diphenyl-2-picrylhydrazyl (DPPH), and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities as well as the reducing power decreasing NO in vitro. The methanol extract of Agrimonia pilosa Ledeb showed significantly strong scavenging effects on free radicals of DPPH, ABTS, and NO. Hexane fraction (HF), ethyl acetate fraction (EF) and n-butyl alcohol fraction (BF) were prepared by solvent fractionation. By comparison of polyphenol contents among BF and HF, EF with high polyphenol contents showed the highest DPPH, ABTS, and NO scavenging effects with the value of 50.2%, 80.4%, and 65.7%, respectively, at the highest tested dose. HPLC analysis revealed the presence of various phenolic compounds in EF of Agrimonia pilosa Ledeb. These results suggest that EF could be considered as natural antioxidant sources and dietary nutritional supplements to prevent oxidation-related diseases.

옥수수 종실의 Brassinosteroid 활성물질 탐색 (Brassinosteroid Substances in Immature Zea mays Seeds)

  • 옥수수 종설에 존재하는 brassinosteroid 활생물질 을 탐색하기 위하여 MeOH로 추출하고, 이 추출물을 rice inclination test를 지표로 용매분획, silica g gel adsorption chromatography, Sephadex LH-20 c chromatography, charcoal adsorption chromato­g graphy, bondesil chromatography 등의 수법으로 b brassinosteroid 활생물질을 정제한 다음, silica gel 흡착 chromatography로 활생 성분을 분리하고 H HPLC에 의해 활성 본체를 구명한 결과, castas­t terone과 teasterone이 동정되었는데, 대부분 cas­t tasterone형태로 존재하였다. 옥수수 종실에 함유된 b brassinosteroid함량은 brassinolide로 환산하여 생 체중량 g당 3-8ng 수준이였다.
