Analysis of flavonoid contents in the fruits of Acanthopanax species (A. chiisanensis, A. divaricatus, A. koreanum, A. senticosus, and A. sessiliflorus) was conducted by high performance liquid chromatography. A Discovery$^{(R)}$ C18 ($4.6{\times}250$ mm, 5 ${\mu}m$) column was used with a gradient mobile phase of water and acetonitrile (90 : 10 to 60 : 40 for 60 min) and UV detection was conducted at 350 nm. The contents of rutin, hyperin, quercetin, afzelin, and kaempferol were 0.063~0.540, 0.494~7.480, 0.584~0.704, 0.388~0.567, 0.190~0.471 mg/g, respectively, in the fruits of Acanthopanax species. Total content of flavonoids in the fruits of Acanthopanax species was highest in those of A. chiisanensis. Furthermore, hyperin was the most abundant compound in the fruits of Acanthopanax species. Consequently, our results demonstrate that the fruits of Acanthopanax species containing flavonoids have promising potential as a new income source of agriculture and industry in medicinal natural products, health supplements, and beverages.
This paper describes the quantitative determination of flavonoids, tannins and ellagic acid in the leaves from wild and cultivated variations of Rubus L. species (Rosaceae): raspberry (2 wild and 13 cultivars) and blackberry (3 wild and 3 cultivars). The content of flavonoids was analyzed using spectrophotometric (the Christ-M llers method) and HPLC analysis after acid hydrolysis. The content of tannins was determined by the weight method, with hide powder, described by German Pharmacopoeia 10 (DAB 10). Ellagic acid content was examined using the HPLC method after acid hydrolysis. Flavonoid content, determined using the Christ-Muller's method was higher for the blackberry leaves than for the raspberry leaves and varied between 0.46% and 1.05%. Quercetin and kaempferol were predominant in all samples analyzed using the HPLC method. The highest flavonoid content was found in the leaves of R. nessensis (1.06%); with results in all of the examined samples varying between 0.27% and 1.06%. The concentration of ellagic acid in all species was determined after acid hydrolysis and ranged from 2.06% to 6.89%. The leaves of raspberries are characterized by greater amounts of tannins (varying between 2.62% and 6.87%) than the leaves of other species. The results from this study indicate that the analyzed species are a rich source of flavonoids, ellagic acid and tannins, which may be used for the quality assessment of Rubus L. species leaves.

Acidic hydrolysis conditions prior to HPLC analysis of plant flavonoids was investigated by response surface methodology (RSM), using leaves of Ginkgo biloba L. Ten hydrolysis conditions using 0.5-2.5 M HCl and 0.5-2.5 hr hydrolysis time were chosen to form a central composite rotatable design (CCRD), and optimization by RSM was achieved by measuring myricetin, quercetin, and kaempferol levels by HPLC. Optimum hydrolysis condition was 1.5 M HCl for 1.5 hr. The levels of flavonoids obtained under the condition were in good agreement with predicted maximum values, with yields of more than 95%. These optimum hydrolysis condition was applied to analysis of flavonoid content in the leaves of Camellia sinensis, Ficus carica, and Sageretia theezans.
The leaves of Diospyros kaki Thunb. were used to treat heart disease and hypotension in traditionally East Asia. The purpose of this study is to simultaneously quantitiative analyze the content of flavonoids in leaves of D. kaki. The isolated flavonoids from the leaves of D. kaki, and the structure of the isolated flavonoids were analyzed based on 1H and 13C NMR spectrum compared to literature data. Simultaneous quantitative analysis of the isolated flavonoids was validated using high performance liquid chromatography (HPLC). Results showed that the calibration curves of the flavonoid compounds were confirmed that they have a large linearity with a correlation coefficient (R2) of 0.99. In the intra-day and inter-day analysis, accuracy and precision of five compounds were measured that accuracy was 94.39 to 114.47% and precision was less than 3.00%. Content analysis showed hyperoside (1.30 ± 0.09%), astragalin (0.81 ± 0.06%), trifoline (1.58 ± 0.07%), quercetin (0.13 ± 0.02%) and kaempferol (1.33 ± 0.25%).
For the utilization of chestnut inner bark as forest biomass, the diethyl ether solubles of hot water extract from chestnut inner bark was analyzed by HPLC. Each peak was identified by comparing with retention time of standard regents and their purity from obtained UV spectrum by RI detector. Identified 6 compounds were gallic acid, 3,5-dihydroxybenzoic acid, 2,4,6-trihydroxybenzoic acid and protocatecualdehyde as phenolic acids and aldehyde, and catechin and epicatechin as flavonoids.
Three flavonoid compounds - quercetin, luteolin, and kaempferol - were analyzed from two commercially available concentrated pomegranate extracts produced in Turkey and Italy, respectively. The samples were freeze-dried and hydrolyzed by 0.4 M hydrochloric acid in 50% ethanol at $80^{\circ}C$. HPLC (high-performance liquid chromatography) with DAD (diode array detection) at a wavelength of 260 nm was used for the detection of the three flavonoids. The detection limits of the three compounds were in hundreds of picograms and the signal-to-noise ratio ranged from 4 to 5: quercetin: >308 pg, s/n=4.0; luteolin: >262 pg, s/n=4.5; kaempferol: >688 pg, s/n=5.0. Quercetin, but not luteolin and kaempferol, was detected in the both pomegranate extracts. The concentrations of quercetin were $49.7\;{\mu}g/g$ and $27.7\;{\mu}g/g$ in the two pomegranate extracts made in Turkey and Italy, respectively.

Phytochemical constituents were isolated from Pinus koraiensis needles by open column chromatography. The structures isolated from the ethyl acetate fraction of P. koraiensis needles were elucidated as afzelin (1) and quercitrin (2) on the basis of spectroscopic analyses including $^1H$-NMR and MS. The amounts of afzelin (1) and quercitrin (2) in genus Pinus were determined by HPLC/UV analysis. Afzelin (1) and quercitrin (2) contents were highest in P. thunbergii needles. Because P. thunbergii needles contain high amounts of afzelin (1) and quercitrin (2), they have promising potential as new additives to natural medicinal products, health supplements, and beverages.

High-performance liquid chromatography was performed in order to analyze the changes in the flavonoid content (rutin, hyperin, afzelin, quercetin, and kaempferol) of Acanthopanax divaricatus and A. koreanum, in response to different cultivation methods (pinching height, planting time, and top dressing). The total flavonoid content of A. divaricatus and A. koreanum ranged from 0.201 to 0.690 mg/g with different pinching heights, 0.143 to 1.001 mg/g for different planting times, and 0.156 to 1.074 mg/g depending on the rate of fertilizer application. In both A. divaricatus and A. koreanum, the total flavonoid content in the upper section of the plant was greater than that in the lower section. These results demonstrate which cultivation methods maximize the flavonoid content of A. divaricatus and A. koreanum, and thus help to optimize flavonoid yields to improve production for nutraceutical, pharmaceutical, and cosmeceutical applications.
Content analysis of flavonoids (epicatechin, epicatechin gallate, and rutin) present in the leaves of Boehmeria nivea (originating from Geumsan-myeon, Biin-myeon, Hansan-myeon, and Baeksu-eup) and their commercial products (ramie tteok, ramie songpyeon, ramie bory-tteok, and ramie tea) was conducted by HPLC. The content of epicatechin, epicatechin gallate, and rutin was highest in the leaves of B. nivea from Geumsan-myeon (0.138 mg/g), Baeksu-eup (1.654 mg/g) and Geumsan-myeon (12.205 mg/g), respectively. With respect to commercial products, the content of epicatechin and epicatechin gallate was highest in ramie tea, with concentrations of 1.879 and 1.090 mg/g, respectively. Given these flavonoid concentrations, B. nivea leaf extracts have the potential to be used as additives in natural medicinal products, health supplements, and beverages.

The methanol extracts of flowers obtained from 67 plant species were screened for their inhibitory activity on aldose reductase (AR). Alnus japonica, Aster spathulifolius, Chionanthus retusus, Morus bombycis, Crysanthemum boreale, Persicaria tinctoria, Platycarya strobilacea, and Serratula coronata var. insularis exhibited potent aldose reductase inhibitory (ARI) activity. HPLC-UV analysis of quercetin, an AR inhibitory flavonoid, was performed on extracts showing strong ARI activity. Quercetin was detected in C. retusus, C. boreale, P. tinctoria, and S. coronata var. insularis at concentrations of 1.33, 1.56, 0.82, and $3.37mg\;g^{-1}$ extract, respectively, indicating that quercetin contributed to the ARI activity of these extracts. In the samples in which quercetin was absent, other compounds may be responsible for their potent ARI activity. These results serve as a basis for further studies regarding the bioactive components responsible for the inhibitory effects of various flower extracts on AR activity.
