1H-NMR and HPLC analysis on the chiral discrimination of β-blockers using (S)-2-tert-butyl-2-methyl-1,3-benzodioxole-4-carboxylic acid

  • In the group of commonly prescribed β-blocker drugs, one of the enantiomers is generally relatively more active than the others. This study aims to develop a technique for the chiral analysis of select β-blockers based on proton nuclear magnetic resonance (1H-NMR) spectrometry. (S)-2-Tert-butyl-2-methyl-1,3-benzodioxole-4-carboxylic acid ((S)-TBMB) was synthesized and utilized as a chiral derivatizing agent. Pure β-blocker enantiomers were isolated from racemates by semi-preparative liquid chromatography prior to derivatization. The reaction time and concentration of (S)-TBMB were controlled to improve the derivatization procedure. No racemization was found during the analysis. High-performance liquid chromatography (HPLC) analysis was also performed for comparative purposes. High agreement between the NMR and HPLC methods was achieved in the determination of (R)-metoprolol in a standard solution of the (S) isomer.

HPLC를 이용한 식품의 ascorbic acid 함량의 분석과 조리에 의한 변화 (Analysis of ascorbic acid contents in raw, processed, and cooked foods by HPLC)

  • The ascorbic acid contents of 101 food items were analyzed by HPLC to provide database to estimate dietary intakes of ascorbic acid of Korean. Foods with high contents of ascorbic acid were green vegetables, citrus fruits, strawberry, kiwi, and fruit juices. This analysis data of ascorbic acid contents in some food items showed significant deviations compared with other Food Composition Table. Ascorbic acid content in soups were lower than those of raw foods by about 57%. The ascorbic acid contents in blanched or seasoned after blanching vegetables and boiled or steamed meals turned out to be decreased by about 52.3% and 47.5%, respectively, but the degrees were varied with the kind of foods as well as cooking methods. The ascorbic acid intakes from 18 most frequently consumed meals in Korea were determined to be about 1/2 of Food Composition Table according to this analysis data. The results showed the importance of accurate food database in assessing nutrient intake levels of population.

근적외부스펙트럼 측정법을 이용한 록소프로펜의 정량화 및 밸리데이션 (The Quantification and Validation of Loxoprofen using Near-infrared(NIR) Spectrum Method)

  • 본 논문에서는 록소프로펜 제제의 분석 시간을 단축하고 제품 출하 속도를 향상시키기 위하여 근적외부(NIR) 스펙트럼 측정법을 이용하여 기존에 사용하던 HPLC 방법과 상호 비교해 보고자 하였다. 제제학적으로 사용되는 다른 첨가제들과 함께 록소프로펜 혼합시료를 제조하고, NIR 분광광도계와 HPLC로 록소프로펜을 정량하고 평가하였다. 두 시험법의 밸리데이션 항목으로 특이성, 정확성 및 정밀성을 실시하였다. NIR 측정법이 검증되었고, 제약산업 분야의 제조공정 중 품질관리를 위한 적합한 결과를 얻었다. 결론적으로 록소프로펜을 분석하는데 있어서 NIR 측정법을 사용하면 기존의 HPLC 정량법을 NIR의 결과로 대치할 수 있을 것이다.

HPLC-ELSD법에 의한 길경의 platycodin D 정량분석 (Quantitative Analysis of Platycodin D from Platycodon grandiflorum by HPLC-ELSD)

  • 길경의 부탄올 분획으로부터 지표성분을 분리, 정제하기 위해 $SiO_2$ 컬럼크마토그래피를 실시한 결과 1종(화합물 1)의 단일 물질을 순수 분리하였으며 화합물 1의 구조는 $^1H$, $^{13}C$ NMR 등 분광학적 관측 결과 platycodin D로 확인되었다. Platycodin D의 HPLC 정량법을 검토한 결과, 검출기는 증기화광산란검출기(ELSD)를, 컬럼은 역상컬럼을 사용하여 길경 MeOH 추출액으로부터 효율적인 지표성분 정량이 가능하였다. 추출조건을 재검토한 결과 환류추출보다는 초음파 추출이 더 효율적이었으며 추출용매로는 80% MeOH이 다른 용매조건보다 추출효율이 우수하였다. 건조조건의 검토에서는 platycodin D의 함량이 양건이나 양건과 건조기 혼용건조 보다 건조기 단독 건조시 더 높았으며 특히 $60^{\circ}C$ 열풍건조에서 그함량이 0.083%로서 가장 높았다. 묽은 에탄올 엑스 함량도 건조기만을 이용한 열풍건조가 양건과 건조기 열풍건조를 혼용한 방법보다 4% 정도 더 높았다.

생약 중 Fenpyroximate의 분석법 연구 (Analytical Methods of Fenpyroximate in Herbal Medicines)

  • Fenpyroximate는 생약 재배시 살응애제로 사용되는 피라졸계 살충제이다. 본 실험은 HPLC-PDA와 HPLC-MS/MS를 이용하여 생약 중 fenpyroximate의 잔류분석법 확립을 위해 수행하였다. 생약(감초와 홍화) 중 잔류하는 fenpyroximate를 아세톤으로 추출하고, 추출액을 다시 포화식염수와 dichloromethane을 이용하여 액-액 분배한 후 아미노프로필 카트리지와 florisil로 정제한 것을 HPLC-PDA와 HPLC-MS/MS로 정량하였다. 생약별로 fenpyroximate를 2 또는 3 수준으로 첨가하여 수행한 회수율 범위는 72.0~106.4%이었으며 이때의 변이계수는 0.2~4.4였다. 따라서 본 분석법은 생약 중의 fenpyroximate 잔류 분석법으로서의 충분한 재현성과 감도를 나타내었다.

PMFs Analysis of Krachaidum Products by HPLC and GC

  • Polymethoxyflavones (PMFs) are a group of polymethoxylated bioactive flavones with diverse biological activities, including anticancer, anti-inflammatory, antibacterial, and antiviral activities. PMFs are found from various plants such as orange, tangerine, and krachaidum. To establish the simple quantitative analytical methods for PMFs, chromatographic analysis was applied to the selected krachaidum foods because krachaidum contains diverse PMFs compared to other PMF-containing foods. Krachaidum is the rhizome of Kaempferia parviflora, and many commercial krachaidum products, such as tea, juice and wine, are commercially available and consumed as health functional foods in Asian countries. Apart from the claimed health promoting benefits, reliable quality assurance and legal guideline for the registration of these products are not available yet. Twelve PMFs were analyzed from the commercial krachaidum foods by GC-FID and HPLC-DAD. No single chromatographic method could not analyze 12 PMFs simultaneously. HPLC-DAD method was found more sensitive to detect PMFs. Based on our analysis data, we proposed 5,7-dimethoxyflaone and 5,7,4'-trimethoxyflavone as index components for the food products.

Quality Evaluation of Modified Bo-Yang-Hwan-O-Tang by Capillary Electrophoresis and High-performance Liquid Chromatography

  • High-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were used to identify five active components in the modified herbal decoction Bo-Yang-Hwan-O-Tang (mBHT), i.e., amygdalin, decursin, paeoniflorin, salvianolic acid B, and calycosin-7-O-${\beta}$-D-glycoside. These components were identified by comparing their retention times and mass spectra with those of reference compounds. The conditions of both analytical methods were optimized and validated. Sufficient separation of target analytes was achieved using a buffer consisting of 40 mM sodium borate and 60 mM sodium dodecylsulfate (SDS) containing 10% methanol (pH 9.5) at 250 nm for CE analysis and gradient elution with a water-methanol mobile phase and ultraviolet (UV) photodiode array detector (DAD) at 250 nm for HPLC analysis. The mBHT components were determined within 65 min by HPLC and 16 min by CE. All calibration curves showed high linearity (R > 0.999) within the ranges tested. Intra-day and inter-day precision were less than 1.6% and 1.8% for HPLC and 2.5% and 4.8% for CE, respectively. The accuracy of the methods ranged from 98.8% to 102.3% for HPLC and from 95.9% to 108.2% for CE.

HPLC/DAD를 이용한 6종(種) 우슬(牛膝)의 분류기준 연구;우슬(牛膝)(쇠무릎, Achyranthes japonica $N_{AKAI}$)로부터 20-hydroxyecdysone 분리.동정 및 산지별 우슬의 HPLC 패턴 비교 (A Study on Discriminative Criteria of 6 Kinds of Achyranthis Radix Using HPLC/DAD;Isolation and Identification of 20-hydroxyecdysone from Aclryranthes japonica $N-{AKAI}$ and Comparison of Patterns of Achyranthis Radix from Different Locations by HPLC)

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.

HPLC에 의한 멸치액젓의 Carbonyl 화합물 분석 (Separation and Quantitative Determination of Carbonyl Compounds in the Filtrated Myulchichuts by HPLC)

  • Qualitative and quantitative analysis for carbonyl compounds contained in the five kinds of filtrated myulchichuts have been made using HPLC. Carbonyl compounds were separated by converting them into 2,4-dinitrophenylhydrazine derivatives which, in turn extracting with hexane. The extract was dehydrated, filtered and examined on HPLC with $C_{18}column$. The quantitative determination of carbonyl species was achieved using standard calibration technique. Highly reproduceable results were obtained by this methods and the amounts of 2,4-dinitrophenylhydrazine derivatives were highly correlative to peak heights. As the results, we find that the seven kinds of carbonyl compounds are contained in each of myulchichuts examined in this study. Five of which are identified as pyruvic acid, formaldehyde, acetaldehyde, acetone and butanone. But the amounts of carbonyl compounds vary widely with the kinds of the filtrated myulchichuts.

The Effective Preparation of Flavonoids from Scutellaria baicalensis GEORGI by Diaion HP-20 Resin

  • Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.
