Identification of a Second Type of AHL-Lactonase from Rhodococcus sp. BH4, belonging to the α/β Hydrolase Superfamily

  • N-acyl-homoserine lactone (AHL)-mediated quorum sensing (QS) plays a major role in development of biofilms, which contribute to rise in infections and biofouling in water-related industries. Interference in QS, called quorum quenching (QQ), has recieved a lot of attention in recent years. Rhodococcus spp. are known to have prominent quorum quenching activity and in previous reports it was suggested that this genus possesses multiple QQ enzymes, but only one gene, qsdA, which encodes an AHL-lactonase belonging to phosphotriesterase family, has been identified. Therefore, we conducted a whole genome sequencing and analysis of Rhodococcus sp. BH4 isolated from a wastewater treatment plant. The sequencing revealed another gene encoding a QQ enzyme (named jydB) that exhibited a high AHL degrading activity. This QQ enzyme had a 46% amino acid sequence similarity with the AHL-lactonase (AidH) of Ochrobactrum sp. T63. HPLC analysis and AHL restoration experiments by acidification revealed that the jydB gene encodes an AHL-lactonase which shares the known characteristics of the α/β hydrolase family. Purified recombinant JydB demonstrated a high hydrolytic activity against various AHLs. Kinetic analysis of JydB revealed a high catalytic efficiency (kcat/KM) against C4-HSL and 3-oxo-C6 HSL, ranging from 1.88 x 106 to 1.45 x 106 M-1 s-1, with distinctly low KM values (0.16-0.24 mM). This study affirms that the AHL degrading activity and biofilm inhibition ability of Rhodococcus sp. BH4 may be due to the presence of multiple quorum quenching enzymes, including two types of AHL-lactonases, in addition to AHL-acylase and oxidoreductase, for which the genes have yet to be described.

사골농축액 시료 중에 함유된 아미노산 정량분석에 대한 오차 요인 및 측정불확도 추정 (Error factors and uncertainty measurement for determinations of amino acid in beef bone extract)

  • 본 연구는 액체크로마토그래피를 이용하여 사골농축액 시료 중에 23종의 아미노산 정량분석 시 발생할 수 있는 측정불확도를 평가하였다. 아미노산 분석에 관여하는 측정불확도 인자로 시료량, 최종 무게, 표준품 무게, 표준품 순도, 검정곡선, 회수율 및 재현성 등을 확인하였다. GUM(Guide to the expression of uncertainty in measurement)과 EURACHEM에 근거하여 측정불확도에 대한 수학적 계산 및 통계분석을 수행하였다. 사골농축액 시료 중 총 아미노산 함량은 36.18 g/100 g이었으며, 95% 신뢰수준(k, 2.05~2.36)에서 확장불확도는 3.81 g/100 g이었다. 측정불확도의 주요 요인은 분석시의 회수율과 반복성(25.2%) > 시료전처리(24.5%) > 검정곡선(24.0%) 순으로 나타났다. 따라서 아미노산 분석 시 불확도를 줄이기 위해서는 실험자들의 주의와 개인적인 숙력도의 향상이 요구된다.

Steroidal Saponin을 이용한 위유, 황정의 분류 및 함량 분석법 개발 (Content Analysis and Classification for Polygonati Odorati Rhizoma and Polygonati Rhizoma by Steroidal Saponin)

  • In present study, classification and quality control of Genus Polygonatum were developed using the isolated from Polygonati Odorati Rhizoma and Polygonati Rhizoma. 3 components were isolated from Butanol fractions of Polygonati Rhizoma, and 2 components were isolated from Hexane and Butanol fractions of Polygonati Odorati Rhizoma. All the components were obtained using silica gel and ODS column chromatography. The compounds were identified as adenosine, 14-hydroxylfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-D-galactopyranosyl-26-O-${\beta}$-D-glucopyranoside, 22-O-methyl-14-hydrocxyfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-Dgalactopyranosyl-26-O-${\beta}$-D-glucopyranoside, ${\beta}$-Sitosteryl-3-O-${\beta}$-D-D-glucopyranoside, 14-hydoxylfurost-5-ene-3-O-${\beta}$-Dglucopyranosyl-($1{\rightarrow}2$)-O-[${\beta}$-D-xylopyranosyl-($1{\rightarrow}3$)]-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-D-galactopyranoside through physicochemical data, spectroscopic methods ($^1H$-NMR, $^{13}C$-NMR, Mass) according references. The quality control of genus Polygonatum were conducted using HPLC quantitative analysis of 14-hydroxylfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-${\beta}$-D-glucopyranosyl-($1{\beta}4$)-O-${\beta}$-D-galactopyranosyl-26-O-${\beta}$-D-glucopyranoside, 14-hydoxylfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-[${\beta}$-D-xylopyranosyl-($1{\rightarrow}3$)]-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-D-galactopyranoside in 30 samples collected throughout Korea and China. This method provided a tool for standardization of mix or misusing the commercial Odorati Rhizoma and Polygonati Rhizoma. As a result, contained quantity of 14-hydroxylfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-D-galactopyranosyl-26-O-${\beta}$-D-glucopyranoside was measured $0.008{\pm}0.006%$ and 14-hydoxylfurost-5-ene-3-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}2$)-O-[${\beta}$-D-xylopyranosyl-(13)]-O-${\beta}$-D-glucopyranosyl-($1{\rightarrow}4$)-O-${\beta}$-Dgalactopyranoside was measured $0.026{\pm}0.012%$.

국내유통농산물 중 잔류농약 모니터링 및 안전성 평가 (Monitoring of Pesticide Residues and Risk Assessment of Agricultural Products Consumed in South Korea)

  • 본 연구는 물리.화학적인 특성이 서로 다른 37종의 농약 성분을 GC/MS/MS로 농약 성분별 MRM에 대한 동시다성분 분석법을 개발하였으며, 이 분석법에서 제시한 MRM을 이용하여 농산물 중 잔류하고 있는 농약성분의 정성 및 정량이 동시에 가능하게 하였고, 이 분석법은 기존의 장비(GC, HPLC, GC/MS 등)를 이용한 검사법에 비해 1건 처리 시 소요되는 분석시간이 짧으며(1건 분석 시 소요시간 30분 이내) 검사결과의 정확성에서 우수하였다. 잔류농약분석에 일반적으로 사용되는 GC-ECD 및 GC-NPD를 사용하지 않고 정성 및 정량이 동시에 가능한 GC-MS/MS를 사용하여 MRM mode를 이용한 농약성분의 동시 분석 및 분석법에 따른 validation을 실시하였다. 그 결과 real sample인 콩과 레몬의 경우 모든 농약에서 70% 이상의 회수율을 보였으며, correlation coefficient($r^2$)도 대부분의 농약에서 0.99 이상의 좋은 직선성을 나타내었다. 또한 분석법을 근거로 국내유통 중인 농산물의 잔류실태를 파악하기 위해 모니터링을 실시하였으며 검출된 농약성분에 한해서는 섭취하는 농약에 대한 위해도를 평가하였다. 국내의 대형시장에 유통되는 농산물 약 1,000여건을 수거하여 확립된 분석법으로 잔류농약 모니터링을 실시하였다. 그 결과 23개의 농산물에서 잔류농약이 검출되었는데 주로 엽채류에서 잔류농약이 검출되었고 금귤류에서 주로 검출된 과실류는 2.2%의 검출율을 보였다. 이 중 MRL을 초과하는 수준의 농약이 검출된 것은 엽채류 중 하나인 무 잎에서 검출되어 0.1%의 부적합율을 나타냈다. 검출된 농약과 그 검출량을 토대로 농약의 1일 섭취허용량(acceptable daily intake, ADI)을 바탕으로 음식을 통해 매일 섭취하는 농약이 사람에게 미치는 위해도를 평가한 결과 ADI 대비 $0.0000{\sim}0.3462%$의 현저히 낮은 수준으로 나타났다.

Characterization of Streptomyces Species Causing Potato Scab in Korea: Distribution, Taxonomy, and Pathogenicity

  • From 1996 to 1999, potato-growing areas in Korea were surveyed for identification and distribution of potato scab pathogens. Potato scab was widely distributed in the mass cultivation areas, especially in Jriu island, southern areas of Chonnam and Gyounggi provinces, and the alpine area of Gangwon province. Jeju island was the most affected area by this disease. A total of 55 Streptomyces strains were isolated from potato scab lesions, among which 40 strains were pathogenic on progeny tubers. Among the pathogenic strain, 21 strains were identified as previously described S. scabies, 7 Strains as S. turgidiscabies, and 5 Strains as S. acidiscabies, while 7 strains were observed as having distinct phenotypic properties. These strains were classified into six distinct clusters based on phenotypic characteristics and selected representative strains for each cluster. S. scabies (S33) had grey spores in a spiral chain. Mean-while, S. turgidiscabies (S27) had grey spores, S. acidiscabies (S71) had white spores, S. luridiscabiei (S63) had yellow-white spores, S. puniciscabiei (S77) had purple-red spores, and S. niveiscabiei (S78) had thin and compact white spores, all in a rectiflexuous chain. Pathogenicity was determined by the production of thaxtomin A and homologs of necl and ORFtnp genes. In TLC, representative strains S27, S71, S63, S77, and S78 produced a yellow band that co-migrated with the authentic thaxtomin A. However, thaxtomin A was not detected in chloroform extracts from oatmeal broth culture and Slice tuber tissue of S. luridiscabiei (S63) and S. puniciscabiei (S77) by HPLC analysis. In addition, no homologs of necl and ORFtnp genes in S. acidiscabies (S71), S. luridiscabiei (S63), S. puniciscabiei (S77), and S. niveiscabiei (S78) were detected by PCR and Southern hybridization analysis.

Optimization of Indole-3-Acetic production by phosphate solubilization bacteria isolated from waste mushroom bed of Agaricus bisporus

  • A total of 35 phosphate solubilizing bacterial strains were isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam and screened for the production of indole acetic acid (IAA). The best IAA producing strain was identified as Pantoea rodasii using 16S rRNA analysis. In addition to the IAA production, this strain could act as an efficient phosphate solubilizer (1100 ${\mu}g$ $ml^{-1}$ after 5 days of incubation) also. The selected strain was cultured under different conditions in order to assess the optimum conditions for maximum IAA production. The nutrient broth (NB) medium was recorded as the best medium, where the maximum IAA production (229 ${\mu}g$ $ml^{-1}$) was recorded at the start of stationary phase (12 hours after inoculation) of the bacteria growth. The performance of the strain was found to be maximum at the temperature of $30^{\circ}C$ followed by $25^{\circ}C$. IAA production was found to be increased with increasing tryptophan concentration (from 0.1 to 0.6%), however beyond this limit, a slight reduction in IAA production was observed. The strains' ability to produce IAA was further confirmed by extraction of crude IAA and subsequent TLC analysis. A specific spot from the extracted IAA preparation was found corresponding with the standard spot of IAA with same $R_f$ value. The results of HPLC analysis conducted in identifying and quantifying the IAA production more precisely, are in agreement with the results of the assessment done with colorimetric method. As revealed by the results of the pot experiment, the isolated strain could significantly enhance the growth (as measured by shoot and root growth) of mung bean plants compared to that of non-inoculated plants. Therefore it can be concluded that the present strain, Pantoea rodasii has great potential to be used as bio-inoculants.

주류의 오크라톡신 모니터링 (Monitoring of Ochratoxin in Alcoholic Beverages)

  • 본 연구를 통해 주류 중 와인, 맥주, 곡주(막걸리), 과실주 및 약주의 오크라톡신 A 오염 실태를 조사하였으며, 와인은 0.21 ng/mL, 맥주는 0.028 ng/mL, 곡주(막걸리)는 0.18 ng/mL, 과실주 및 약주는 0.18 ng/mL의 평균 검출량을 보였으며, EU에서 설정하고 있는 기준인 2.0 ng/mL보다 훨씬 낮은 수준이었다. 하지만 우리나라의 기후와 지역의 특성을 고려하여 고온 다습한 남쪽지역에서 생산되는 주류의 오크라톡신 A에 대한 오염실태 조사가 조금 더 이루어져야 할 것으로 생각된다. 또한 주류를 통한 오크라톡신 A의 섭취추정량 0.0008 ng/b.w.day는 EU에서 권고하는 오크라톡신 A 일일섭취한계량(TDI) 중 주류를 통해 섭취하는 오크라톡신 A의 TDI값의 4.5%인 0.2 ng/b.w.day에 비하여 훨씬 적은 양으로 우리나라는 주류에 의한 오크라톡신 A에 대하여 안전하다고 판단된다.

Structural characteristics of a red ginseng acidic polysaccharide rhamnogalacturonan I with immunostimulating activity from red ginseng

  • Background: Many researchers reported that the various immune activities of red ginseng are due to acid polysaccharides. But, the exact structural characteristics of the acidic polysaccharide in red ginseng have not been fully elucidated. Therefore, we isolated the acidic polysaccharide from red ginseng and characterized the structural property of the active moiety of this polysaccharide, which contributes to the immunostimulatory activity of red ginseng. Methods: A polysaccharide (RGP-AP-I) was purified from red ginseng via size-exclusion chromatography using Sephadex G-100. Immunostimulatary activity of RGP-AP-I was investigated via anti-complementory and macrophage stimulatory activity. The structure of RGP-AP-I was characterized by HPLC, sugar composition, β-glucosyl Yariv reagent and methylation analysis. Results: Peritoneal macrophages stimulated using RGP-AP-I significantly augmented the production of various cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factor (TNF)-α. The primary structure of RGP-AP-I was elucidated by assessing its sugar composition and methylation analysis. RGP-AP-I is a 96 kDa acidic polysaccharide, and comprises nine different monosaccharides, which mainly include sugars such as rhamnose (Rha, 9.5%), galacturonic acid (GalA, 18.4%), galactose (Gal, 30.4%), and arabinose (Ara, 35.0%). RGP-AP-I exhibited an considerable reaction with the β-glucosyl Yariv reagent, revealing the presence of arabino-β-3,6-galactan. Methylation analysis indicated that RGP-AP-I comprises 21 different glycosyl linkages, such as 3-, 4-, 6- and 3,6-linked Galp; 5-linked Araf; 2,4-linked Rhap; and 4-linked GalAp, which are characteristics of rhamnogalacturonan I (RG-I). Conclusion: we assumed that the immunostimulatory activity of RGP-AP-I may be due to the RG-I structure, which comprises a main chain with a repeating linkage unit, [→2)-Rhap-(1→4)-GalAp-(1→] and three groups of side chains such as (1→5)-linked arabinan, (1→4)-linked galactan, and arabino-β-3,6-galactan, which branch at the C(O)4 positions of Rha residues in the main chain of RGP-AP-I.

독도 번행초에서 분리된 내생균류의 배양적 특성과 Aspergillus tubingensis YH103의 gibberellin A7의 생산 (Gibberellin A7 production by Aspergillus tubingensis YH103 and cultural characteristics of endophytic fungi isolated from Tetragonia tetragonoides in Dokdo islands)

  • 독도에 자생하는 번행초의 뿌리로부터 순수 분리하여 형태적으로 상이한 17개의 내생균류를 선별하였다. 또한 분리된 균류들에 대하여 각각의 염농도와 pH 농도 구배에 따라 생장 시험을 확인하였다. 내생균류에 대해 각각 난장이벼의 유묘에 식물생장활성시험을 진행하였고, 그 결과 YH103 균주가 가장 높은 활성을 나타내었다. 계통분석은 Maximum likelihood 방법을 활용하여 결합된 ITS영역, beta-tubulin 및 calmodulin 유전자 염기서열을 분석하여 분리된 균주의 유연관계를 분석하였다. YH103 균주의 배양여과액을 HPLC와 GC/MS SIM을 이용하여 분석한 결과 식물호르몬인 지베렐린 $GA_4$, $GA_7$, $GA_8$$GA_{19}$가 확인되었다. 최종적으로 YH103 균주의 형태학적 관찰 및 결합된 유전자 염기서열의 분자적 분석을 통해 GA를 생산하는 새로운 Aspergillus tubingensis로 동정되었다.

아마인에 함유된 시안배당체의 분석과 저감화 (Analysis and Decrease of Cyanogenic Glucosides in Flaxseed)

  • 아마인의 linustatin과 neolinustatin은 가수분해되어 acetone, hydrogen cyanide(HCN) 그리고 sugar를 생성하는 diglucoside 시안배당체로써 가수분해물인 HCN의 독성으로 인해 식품의 안전성에 문제가 되고 있다. $80\%$ methanol 추출과 LC/MSn에 의한 정성분석과 RI-LC에 의한 아마인에서 함유된 시안배당체 함량을 조사한 결과, linustatin과 neolinustatin이 각각 평균 206.5 mg/100 g과 174.2 mg/100 g 함유되어 있었다. 또한 autohydrolysis 후 증류액으로부터 IC 분석 에 의 해 HCN 137.38 mg/kg이 가수분해되어 유리됨을 알 수 있었다. 시안배당체의 저감화를 위하여 가열처리를 수행한 결과, 아마인은 $200^{\circ}C$에서 2시간 이상 가열시 $85\%$ 이상의 linustatin과 neolinustatin 그리고 HCN의 제거효과를 보였다. 특히 $200^{\circ}C$에서 30분 가열시 $98\%$의 HCN이 제거됨을 관찰할 수 있었다.
