차가버섯과 어성초 함유 발효 조성물이 인체 위암 AGS 및 대장암 HCT-15 세포 생육에 미치는 영향 (Effect of Fermented Compositions Containing Inonotus obliquus with Houttuynia cordata on Growth of Human AGS Gastric and HCT-15 Colon Cancer Cells)

  • 어성초 분말을 포함하는 차가버섯 발효 조성물의 물 추출물이 인체 위암 세포 AGS 및 대장암 세포 HCT-15, 그리고 정상세포 NIH3T3 fibroblast의 세포 생육에 미치는 영향을 검토하였다. 세포독성 실험은 세포수 count와 MTT 방법으로 측정하였다. 어성초 분말을 포함하는 차가버섯 발효 조성물의 제조는 차가버섯과 어성초 분말을 혼합하고, 여기에 대두 발효 미생물원을 다시 혼합시켜 습도 $50{\sim}60%$,온도 $30{\sim}37^{\circ}C$에서 30일 정도 발효시킨 후 건조 시킨 분말에서 5% 물 추출물을 얻어 동결건조 시켜 실험에 제공하였다. MTT assay 방법에서 차가버섯 발효 조성물의 수용성 추출물 0.16, 0.4, 0.8, 1.6 및 4.0 mg/ml 첨가 농도에서 AGS 세포 생육은 13, 25, 40, 67 및78% 억제 되었으며, HCT-15세포 생육은 22, 40, 50, 69및 76%씩 각각 억제되었다. 그러나 동일한 실험조건에서 정상 세포수 NIH3T3은 86% 이상의 생존율을 나타내었다. 차가버섯 발효 조성물의 수용성 추출물은 인체 대장암 세포주 HCT-15와 위암 세포수 AGS에 대해 생육억제 작용이 강한 반면, 정상세포 NIH3T3에 대해서는 세포 독성을 거의 나타내지 않아 가장 바람직한 암예방 또는 항암식품 개발가능성을 제시하였다.

hARIP2 is a Putative Growth-promoting Factor Involved in Human Colon Tumorigenesis

  • Activin is a multifunctional growth and differentiation factor of the growth factor-beta (TGF-${\beta}$) superfamily, which inhibits the proliferation of colon cancer cells. It induces phosphorylation of intracellular signaling molecules (Smads) by interacting with its type I and type II receptors. Previous studies showed that human activin receptor-interacting protein 2 (hARIP2) can reduce activin signaling by interacting with activin type II receptors; however, the activity of hARIP2 in colon cancer has yet to be detailed. In vitro, overexpression of hARIP2 reduced activin-induced transcriptional activity and enhanced cell proliferation and colony formation in human colon cancer HCT8 cells and SW620 cells. Also, hARIP2 promoted colon cancer cell apoptosis, suggesting that a vital role in the initial stage of colon carcinogenesis. In vivo, immunohistochemistry revealed that hARIP2 was expressed more frequently and much more intensely in malignant colon tissues than in controls. These results indicate that hARIP2 is involved in human colon tumorigenesis and could be a predictive maker for colon carcinoma aggressiveness.

고려인삼 중 Petroleum Ether 추출물의 인체 암세포 증식억제효과 (Inhibitory Effect of Petroleum Ether Extract of Panaz Ginseng Root against Growth of Human Cancer Cells)

  • This study was attempted to screen the cytotoxic activity of petroleum ether ex- tract from panax ginseng root against human colon cancer cells. Two extracts of panax ginseng root, crude and partially purified, were used for this experiment. The crude extract was prepared by extraction with petroleum ether using Soxhlet aparatus for 12 to 15 hours from panax ginseng and the extract was partially purified by silicic acid column with mixture of petroleum ether: ethyl ether (70 : 30, v/v). Three species of human colon cancer cells, HRT-18, HCT-48 and HT-29, were maintained in DMEM (Dulbecco's modified Eagle medium), and the cells were cultured in DMEM containing serial concentration of the crude or partially purified fraction to observe the cytotoxic activity of the both extracts. The effects of incubation time and concentration of the both extracts in culture medium against the growth of the each cancer cell were determined. The results obtained are summarized as follows: 1. The doubling times of the HRT-18, HCT-48 and HT-29 cells were about 20, 24 and 22 hours, respectively. 2, The inhibitory effects of the crude extract on the growth of cancer cells were increased according to the rise of concentration of the extract and incubation time. 3. The inhibitory effect of partially purified fraction on the growth of HRT-18 cell was about 4 times stronger than that of the crude extract under same experimental condition. 4 The inhibitory effects of the crude and purified fraction on the growth of each cancer cell were shown difference by the kind of the cancer cell. In view of the above results, it could be said that the petroleum ether extract of panax ginseng root inhibited the division of the human colon cancer cell, in vitro.

고련피 추출물의 항암활성 (Anti-cancer Activities of Extract from the Bark of Melia azedarach L. var. japonica Makino)

  • In the present study, the anti-cancer activity of 80% ethanol extracts from 120 kinds of medicinal herbs and native plants were investigated. Among them, the barks of Melia azedarach L. var. japonica Makino showed the highest cytotoxicity in HCT-15 human colon cancer cell. With this result, we carried out hollow fiber (HF) assay and anti-metastasis study to confirm the anti-cancer effects of M. azedarach var. japonica. In MTT assay, M. azedarach var. japonica.inhibited the proliferation of HCT-15 cells in dose-dependent manner. HF assay was carried out using A549 human adenocarcinoma cell, HCT-15 and SK-Hep1 human liver cancer cell via intraperitoneal (IP) and subcutaneous (SC) site. As a results, SK-Hep1 implanted in IP site showed the highest cytotoxicity. The result from metastatic model using B16/BL6 mouse corresponded to that of HF assay. These results suggest that the ethanol extract from M. azedarach var. japonica. might have a potent anti-cancer activity and advanced study is needed for the development of novel natural anti-cancer drug.

Inhibitory effects of calcium against intestinal cancer in human colon cancer cells and $Apc^{Min/+}$ mice

  • The aim of the study was to investigate the inhibitory effects of calcium against intestinal cancer in vitro and in vivo. We first investigated the effects of calcium treatment in HCT116 and HT29 human colon cancer cells. At the concentration range of 0.8-2.4 mM, calcium significantly inhibited cell growth (by 9-29%), attachment (by 12-26%), invasion (by 15-31%), and migration (by 19-61%). An immunofluorescence microscope analysis showed that the treatment with calcium (1.6 mM) for 24 h increased plasma membrane ${\beta}$-catenin but decreased nuclear ${\beta}$-catenin levels in HT29 cells. We then investigated the effect of dietary calcium on intestinal tumorigenesis in $Apc^{Min/+}$ mice. Mice received dietary treatment starting at 6 weeks of age for the consecutive 8 weeks. The basal control diet contained high-fat (20% mixed lipids by weight) and low-calcium (1.4 mg/g diet) to mimic the average Western diet, while the treatment diet contained an enriched level of calcium (5.2 mg calcium/g diet). The dietary calcium treatment decreased the total number of small intestinal tumors (by 31.4%; P < 0.05). The largest decrease was in tumors which were ${\geq}$ 2 mm in diameter, showing a 75.6% inhibition in the small intestinal tumor multiplicity (P < 0.001). Immunohistochemical analysis showed significantly reduced nuclear staining of ${\beta}$-catenin (expressed as nuclear positivity), but increased plasma membrane staining of ${\beta}$-catenin, in the adenomas from the calcium-treated groups in comparison to those from the control group (P < 0.001). These results demonstrate intestinal cancer inhibitory effects of calcium both in human colon cancer cells and $Apc^{Min/+}$ mice. The decreased ${\beta}$-catenin nuclear localization caused by the calcium treatment may contribute to the inhibitory action.

Calcium Signaling of Dioleoyl Phosphatidic Acid via Endogenous LPA Receptors: A Study Using HCT116 and HT29 Human Colon Cancer Cell Lines

  • In the present study, we have tested the effect of dioleoyl phosphatidic acid (PA) on intracellular $Ca_{2+}$ concentration ($[Ca^{2+}]_{i}$) in two human colon cancer cell lines (HCT116 and HT29). PA and lysophosphatidic acid (LPA), a bioactive lysolipid, increased $[Ca^{2+}]_{i}$ in both HCT116 and HT29 cell lines. Increases of $[Ca^{2+}]_{i}$ by PA and LPA were more robust in HCT116 cells than in HT29 cells. A specific inhibitor of phospholipase C (U73122), however, was not inhibitory to the cell responses. Pertussis toxin, a specific inhibitor of $G_{i/o}$ type G proteins, however, had an inhibitory effect on the responses except for an LPA-induced one in HT29 cells. Ruthenium red, an inhibitor of the ryanodine receptor, was not inhibitory on the responses, however, 2-APB, a specific inhibitor of inositol 1,4,5-trisphosphate receptor, completely inhibited both lipid-induced $Ca^{2+}$ increases in both cell types. Furthermore, by using Ki16425 and VPC32183, two structurally dissimilar specific antagonists for $LPA_{1}/LPA_{3}$ receptors, an involvement of endogenous LPA receptors in the $Ca^{2+}$ responses was observed. Ki16425 completely inhibited the responses but the susceptibility to VPC32183 was different to PA and LPA in the two cell types. Expression levels of five LPA receptors in the HCT116 and HT29 cells were also assessed. Our data support the notion that PA could increase $[Ca^{2+}]_{i}$ in human colon cancer cells, probably via endogenous LPA receptors, G proteins and $IP_{3}$ receptors, thereby suggesting a role of PA as an intercellular lipid mediator.

한국산 마늘로 부터 분리한 Alliin과 에탄올 추출물의 In Vitro계 생리 활성 (Physiological Activity of Alliin and Ethanol Extract from Korean Garlic (Allium Sativum, L.))

  • 본 연구는 여러가지 유해세균과 인체장암세포에 대한 다양한 농도에서의 마늘 추출물의 효과를 고양하고자 계획하였다. 미생물에 대한 항균효과는 그 최소 발육저지농도가 alliin의 경우에는 $5,000{\sim}20,000\;ppm$, 그리고 ethanol extract의 경우에는 $1,250{\sim}10,000\;ppm$이었으며, 또한 이들 성분의 암세포증식억제는 첨가농도가 증가함에 따라 점차 강하게 억제시킴을 알 수 있었다. 그리고 HCT-15의 형태적 관찰에서도 위축됨을 알 수 있었으며, 위의 결과들로 부터 항균과 암세포증식억제에 있어서의 활성물질은 열에 안정하고 에탄올에 의해 추출되는 성분임을 알 수 있었다.

Chemotherapeutic Candidate Inducing Immunological Death of Human Tumor Cell Lines

  • The immunological death induction by EY-6 on the human tumor cell lines was screened. Human colon carcinoma (HCT15, HCT116), gastric carcinoma (MKN74, SNU668), and myeloma (KMS20, KMS26, KMS34) cells were died by EY-6 treatment with dose-dependent manner. CRT expression, a typical marker for the immunological death, was increased on the EY-6-treated colorectal and gastric cancer cells. Interestingly, the effects on the myeloma cell lines were complicated showing cell line dependent differential modulation. Cytokine secretion from the EY-6 treated tumor cells were dose and cell-dependent. IFN-${\gamma}$ and IL-12 secretion was increased in the treated cells (200% to over 1000% of non-treated control), except HCT116, SNU668 and KMS26 cells which their secretion was declined by EY-6. Data suggest the potential of EY-6 as a new type of immuno-chemotherapeutics inducing tumor-specific cell death. Further studies are planned to confirm the efficacy of EY-6 including in vivo study.

Cytotoxicity of Two Novel Cisplatin Analogues, $(CPA)_2Pt[DOLYM]$ and (DACH)Pt[DOLYM], to Human Cancer Cells In Vitro

  • Despite the impressive antitumor activity of cisplatin, two major limitations of the drug, that is severe side effects and drug-resistance of cancer cells, make its use difficult of r cancer therapy. These limitations have resulted in a greate deal of effort having been expended into structural modifications of cisplatin. In this study, we tested two novel cisplatin analogues, (CPA)2 Pt[DOLYM] (COMP-I) and (DACH)Pt[DOLYM] (CoMP-II), for the mode of cytotoxic action against human tumor cells comparing with cisplatin and carboplatin in vitro. These two novel analogues had considerable cytotoxic activities against five kinds of human solid tumor cells, and especially COMP-II was more effective on HCT15 colon cancer cells than other compounds. In addition, COMP-II had cytostatic activity at low concentrations (10~0.3${\mu}g/ml$), but other compounds revealed little effect on tumor growth at the low concentration.

소금의 보돌연변이 및 암세포성장억제 효과 (Effects of Different Kinds of Salt in the Comutagenicity and Growth of Cancer Cells)

  • 국내산 정제염, 천일염과 유럽산 천일염 등의 SEM에서의 형태, 무기질 성분, 보돌연변이와 암세포 in vitro 항암 기능성을 측정하였다. 정제염은 SEM을 이용한 관찰 시 구조가 규칙적이었고 천일염은 불규칙한 표면 구조를 가지고 있었다. 천일염의 크기도 차이가 있었고 프랑스 천일염과 한국천일염I은 다른 천일염보다 크기가 작았다. 무기질 함량은 프랑스 천일염과 한국 천일염I이 Ca, K, Mg 등의 함량이 다른 소금보다 더 많았다. pH도 프랑스 천일염과 한국 천일염I이 가장 높았다. 한국산 두 가지의 천일염은 무기질 함량에 따라 물리적 특성의 차이를 보였고 한국 천일염I은 프랑스 천일염과 비슷한 무기질 함량을 가지고 있었다. 소금은 보돌연변이 효과가 있었지만 특히 무기질 함량이 높았던 프랑스 천일염과 한국산 천일염I은 다른 소금보다 낮은 보돌연변이 효과를 나타내었다. HCT-116과 AGS 암세포에 대한 in vitro 항암 효과에서도 보돌연변이 효과와 비슷한 경향을 나타내었고 많은 무기질을 함유한 프랑스 천일염과 한국산 천일염I의 암세포 성장 억제효과와 암세포의 apoptosis 유도 효과가 가장 높았다. 이상의 결과로부터 소금의 기능성은 무기질 함량이 많은 천일염이 정제염보다 좋으며 천일염 중에도 Ca, K 등의 무기질 함량이 높을수록 기능성도 증가하는 것으로 나타났다.
