AtHAP3b Plays a Crucial Role in the Regulation of Flowering Time in Arabidopsis during Osmotic Stress

  • The HAP complex has been found in many eukaryotic organisms. HAP recognizes the CCAAT box present in the promoters of 30% of all eukaryotic genes. The HAP complex consists of three subunits - HAP2, HAP3 and HAP5. In this paper, we report the biological function of the AtHAP3b gene that encodes one of the HAP3 subunits in Arabidopsis. Compared with wild-type plants, hap3b-1 and hap3b-2 mutants exhibited a delayed flowering time under long-day photoperiod conditions. Moreover, the transcription levels of FT were substantially lower in the mutants than in the wild-type plants. These results imply that AtHAP3b may function in the control of flowering time by regulating the expression of FT in Arabidopsis. In a subsequent study, AtHAP3b was found to be induced by osmotic stress. Under osmotic stress conditions, the hap3b- 1 and hap3b-2 mutants flowered considerably later than the wild-type plants. These results suggest that the AtHAP3b gene plays more important roles in the control of flowering under osmotic stress in Arabidopsis.

  • Single nucleotide polymorphisms (SNPs) are the most abundant form of human genetic variation and are a resource for mapping complex genetic traits. A genome is covered by millions of these markers, and researchers are able to compare which SNPs predominate in people who have a certain disease. The International HapMap Project, launched in October, 2002, motivated us to start the Korean HapMap Project in order to support Korean HapMap infrastructure development and to accelerate the finding of genes that affect health, disease, and individual responses to medications and environmental factors. A Korean SNP and haplotype database system was developed through the Korean HapMap Project to provide Korean researchers with useful data-mining information about disease-associated biomarkers for studies on complex diseases, such as diabetes, cancer, and stroke. Also, we have developed a series of software programs for association studies as well as the comparison and analysis of Korean HapMap data with other populations, such as European, Chinese, Japanese, and African populations. The developed software includes HapMapSNPAnalyzer, SNPflank, HWE Test, FESD, D2GSNP, SNP@Domain, KMSD, KFOD, KFRG, and SNP@WEB. We developed a disease-related SNP retrieval system, in which OMIM, GeneCards, and MeSH information were integrated and analyzed for medical research scientists. The kHapMap Browser system that we developed and integrated provides haplotype retrieval and comparative study tools of human ethnicities for comprehensive disease association studies (http://www.khapmap.org). It is expected that researchers may be able to retrieve useful information from the kHapMap Browser to find useful biomarkers and genes in complex disease association studies and use these biomarkers and genes to study and develop new drugs for personalized medicine.

Morphology and Mechanical Properties through Hydroxyapatite Powder Surface Composite (Hydroxyapatite의 파우더 표면 복합화를 통한 형태 및 기계적 성질에 관한 연구)

  • In this study, new hydroxyapatite powder surface composites were investigated for protective effects against ultraviolet rays. Hydroxyapatite (HAp) is biocompatible and does not cause nebula phenomenon on skin. We investigated the surface modification of hydroxyapatite to improve UV block and skin usage. Dimethicone, lauroyl lysine, triethoxycaprylylsilane and silica were used as coating agents for the surface modification of HAp. To prepare the composite complex of the modified surface, the dimethicone, lauroyl lysine and triethoxycaprylylsilane were prepared by a dry process, and silica by a hydrothermal synthesis method. The HAp-silica was chosen as the best composite powder when measuring its sun protection levels. We investigated the characteristics of the surface of HAp-silica by SEM, particle size analyzer and energy dispersive spectrometry (EDS). Additionally, the stability in the formulation, UV block effect, and safety in BB creams were investigated. In conclusion, HAp-silica prepared by the modification of HAp complex surface improved the skin usage and UV block effect by enhancing the white cloudy phenomenon. These results indicate that HAp-silica may be used for UV block cosmetics.

Monitoring conservation effects on a Chinese indigenous chicken breed using major histocompatibility complex B-G gene and DNA Barcodes

  • Objective: We report monitoring conservation effect for a Chinese indigenous chicken (Langshan) breed using major histocompatibility complex (MHC) and DNA barcords. Methods: The full length of MHC B-G gene and mitochondrial cytochrome oxidase I (COI) gene in generations 0, 5, 10, 15, 16, and 17 was measured using re-sequencing and sequencing procedures, respectively. Results: There were 292 single nucleotide polymorphisms of MHC B-G gene identified in six generations. Heterozygosity (He) and polymorphic information content (PIC) of MHC B-G gene in generations 10, 15, 16, and 17 remained stable. He and PIC of MHC B-G gene were different in six generations, with G10, G15, G16, G17 >G5>G0 (p<0.05). For the COI gene, there were five haplotypes in generations 0, 5, 10, 15, 16, and 17. Where Hap2 and Hap4 were the shared haplotypes, 164 individuals shared Hap2 haplotypes, while Hap1 and Hap3 were the shared haplotypes in generations 0 and 5 and Hap5 was a shared haplotype in generations 10, 15, 16, and 17. The sequence of COI gene in 6 generations was tested by Tajima's and D value, and the results were not significant, which were consistent with neutral mutation. There were no differences in generations 10, 15, 16, and 17for measured phenotypic traits. In other generations, for annual egg production, with G5, G10, G15, G16, G17>G0 (p<0.05). For age at the first egg and age at sexual maturity, with G10, G15, G16, G17>G5>G0 (p<0.05). Conclusion: Combined with the results of COI gene DNA barcodes, MHC B-G gene, and phenotypic traits we can see that genetic diversity remained stable from generations 10 to 17 and the equimultiple random matching pedigrees conservation population conservation effect of Langshan chicken was effective as measured by these criteria.

  • As the COVID-19 pandemic is prolonged, non-contact work has increased, as well as the demand for automation of simple and repetitive questions and tasks with success of using them. Therefore, companies are attempting to expand the area of automated business and apply various technologies such as AI to complex and various business processes of E2E to provide automation of all business. However, the extension to Intelligent Process Automation (IPA) is still in its beginning stage so that it is difficult to find practical use cases and related solutions. In this aspect, it is safe to say that there is insufficient evidence for companies which have various and complex enterprise processes to make a decision about the adoption. In this study, to solve this problem, a Hyper Automation Platform (HAP) that consists of RPA, Chatbot, and AI technology was proposed. Moreover, an implementation method that can bring intelligent process automation using HAP, and practical use-cases were provided so that it makes it possible to review the implementation of the HAP objectively and comprehensively. This study is meaningful and valuable to check the feasibility of the Hyper Automation concept and to actively utilize HAP.

Herbal Medicine Treatment of Refractory Epilepsy in Tuberous Sclerosis Complex : A Case Report

  • Infants with tuberous sclerosis complex (TSC) have a higher chance of experiencing seizures before the age of 1 year; in particular, they commonly accompany infantile spasms. In cases where infantile spasms resulting from TSC are drug-resistant, more severe neuro-developmental and cognitive impairments occur. This particular case dealt with an infant with TSC who continued to experience partial seizures and infantile spasms despite using two different kinds of antiepileptic drugs (AEDs). His spasms ceased on the seventh day of taking modified Yukmijihwang-tang (YMJ), at which point he stopped the use of all AEDs. He became seizure-free after a month of the treatment and modified hypsarrythmia was found to have been resolved in the electroencephalogram test. Until now, the infant has been taking YMJ for 16 months and is maintaining the seizure-free state without side effects. Moreover, his developmental status is continually improving, with a significant progress in language and cognitive-adaptive abilities. Such results suggest that YMJ can serve as an alternative treatment option for refractory epilepsy.

Determination of Hydroxyapatite Precipitation Condition from the $Ca-PO_4-H_2O$ System ($Ca-PO_4-H_2O$계로부터 수산화아파타이트의 침전조건 결정)

  • The formation and dissolution of hydroxides, carbonates and hydroxyapatite (HAp), which depend on the pH of solution, are important factor for the preparation of homogeneous and fine HAp, $Ca_{10-x}(HPO_4)_x(PO_4)_{6-x}(OH)_{2-x}(x=0)$, ceramic powder from the $Ca-PO_4-H_2O$ system. Since the solubility of each complex ion is a linear function of pH, the solubility diagram can be obtained by plotting the logarithmic molar concentrations calculated from the values of the equilibrium constants and solubility products for hydroxides, carbonates, and hydroxyapatite. The optimum pH condition for the formation of single phase $Ca_{10-x}(HPO_4)_x(PO_4)_{6-x}(OH)_{2-x}(x=0)$ powder in $Ca-PO_4-H_2O$ system at $25^{\circ}C$ was estimated as $10.5{\pm}0.5$ through the theoretical consideration. The HAp powder dried at $80^{\circ}C$ showed a fine agglomerated particles with a size of 75 nm. The HAp powder calcined at $1,000^{\circ}C$ consisted of nearly homogeneous particles with a size of 450 nm. Even though the dried HAp particles consisted of agglomeration, mechanical properties were superior due to fine microstructure after sintering.

A Study on Characteristics of Lincomycin Degradation by Optimized TiO2/HAP/Ge Composite using Mixture Analysis (혼합물분석을 통해 최적화된 TiO2/HAP/Ge 촉매를 이용한 Lincomycin 제거특성 연구)

  • In this study, it was found that determined the photocatalytic degradation of antibiotics (lincomycin, LM) with various catalyst composite of titanium dioxide ($TiO_2$), hydroxyapatite (HAP) and germanium (Ge) under UV-A irradiation. At first, various type of complex catalysts were investigated to compare the enhanced photocatalytic potential. It was observed that in order to obtain the removal efficiencies were $TiO_2/HAP/Ge$ > $TiO_2/Ge$ > $TiO_2/HAP$. The composition of $TiO_2/HAP/Ge$ using a statistical approach based on mixture analysis design, one of response surface method was investigated. The independent variables of $TiO_2$ ($X_1$), HAP ($X_2$) and Ge ($X_3$) which consisted of 6 condition in each variables was set up to determine the effects on LM ($Y_1$) and TOC ($Y_2$) degradation. Regression analysis on analysis of variance (ANOVA) showed significant p-value (p < 0.05) and high coefficients for determination value ($R^2$ of $Y_1=99.28%$ and $R^2$ of $Y_2=98.91%$). Contour plot and response curve showed that the effects of $TiO_2/HAP/Ge$ composition for LM degradation under UV-A irradiation. And the estimated optimal composition for TOC removal ($Y_2$) were $X_1=0.6913$, $X_2=0.2313$ and $X_3=0.0756$ by coded value. By comparison with actual applications, the experimental results were found to be in good agreement with the model's predictions, with mean results for LM and TOC removal of 99.2% and 49.3%, respectively.

Semantic Modeling for SNPs Associated with Ethnic Disparities in HapMap Samples

  • Single-nucleotide polymorphisms (SNPs) have been emerging out of the efforts to research human diseases and ethnic disparities. A semantic network is needed for in-depth understanding of the impacts of SNPs, because phenotypes are modulated by complex networks, including biochemical and physiological pathways. We identified ethnicity-specific SNPs by eliminating overlapped SNPs from HapMap samples, and the ethnicity-specific SNPs were mapped to the UCSC RefGene lists. Ethnicity-specific genes were identified as follows: 22 genes in the USA (CEU) individuals, 25 genes in the Japanese (JPT) individuals, and 332 genes in the African (YRI) individuals. To analyze the biologically functional implications for ethnicity-specific SNPs, we focused on constructing a semantic network model. Entities for the network represented by "Gene," "Pathway," "Disease," "Chemical," "Drug," "ClinicalTrials," "SNP," and relationships between entity-entity were obtained through curation. Our semantic modeling for ethnicity-specific SNPs showed interesting results in the three categories, including three diseases ("AIDS-associated nephropathy," "Hypertension," and "Pelvic infection"), one drug ("Methylphenidate"), and five pathways ("Hemostasis," "Systemic lupus erythematosus," "Prostate cancer," "Hepatitis C virus," and "Rheumatoid arthritis"). We found ethnicity-specific genes using the semantic modeling, and the majority of our findings was consistent with the previous studies - that an understanding of genetic variability explained ethnicity-specific disparities.

Heterogeneous Catalysts for Hydrogen Generation Based on Ru-Incorporated Hydroxyapatite

  • Hydrolysis of sodium borohydride provides a safe and clean approach to hydrogen generation. Having the proper catalytic support for controlling this reaction is therefore a valuable technology. Here we demonstrate the capability of hydroxyapatite as a novel catalytic support material for hydrogen generation. Aside from being inexpensive and durable, we reveal that Ru ion exchange on the HAP surface provides a highly active support for sodium borohydride hydrolysis, exemplifying a high total turnover number of nearly 24,000 mol $H_2$/ mol Ru. Moreover, we observe that the RuHAP support exhibits a high catalytic lifetime of approximately one month upon repeated exposure to $NaBH_4$ solutions. In addition to examining surface area effects, we also identified the role of complex surface morphology in enhancing hydrolysis by the catalytic transition metal covered surface. Particularly, we found that a polycrystalline RuHAP catalytic support exhibits shorter induction times for the initial bubble formation as well as increased hydrogen generation rates as compared to a single crystal supports. The independent factor of a complex surface morphology is believed to provide enhanced sites for gas release during the initial stages of the reaction. By demonstrating the ability to shorten induction time and enhance catalytic activity through changes in surface morphology and Ru content, we find it feasible to further explore this catalyst support in the construction of a practical hydrogen generator.
