GDI 고압펌프의 유동특성에 관한 연구 (A Study of the Fluidic Characteristics of High-Pressure Fuel Pumps for GDI Engines)

  • 고압연료펌프는 GDI 엔진의 핵심 구성요소로써, 엔진출력 및 연료 효율을 향상시키기 위해서는 고압연료펌프의 유동특성을 연구하는 것이 필요하다. 본 연구에서는 유압해석툴인 AMEsim을 이용하여 고압연료펌프의 통합 모델을 생성하여 유동해석을 수행하였다. 하지만, AMEsim은 시스템 해석을 위한 1차원 모델이므로 복잡한 유동현상이 발생하는 부근에서의 해석 결과는 정확하지 않은 단점이 있으므로 본 연구에서는 전산해석프로그램인 Fluent를 이용하여 난류유동이 발생하는 체크밸브의 흡입부와 토출부에서 유량과 알짜힘을 계산하였다. 다양한 압력조건과 밸브 간극변화에 따른 CFD 해석 결과는 AMEsim모델에 대한 룩업테이블로 사용되어 AMEsim의 결과를 보완함으로써 고압연료펌프에 대한 성능 분석결과의 정확성을 향상시키는 결과를 얻을 수 있었다.

  • To determine which are the culturally specific factors of Korean bereavement, this chapter focuses on the view of death and the traditional mourning process which reflect Korean values and norms. The formation of the Korean view and understanding of death has been strongly influenced by three of its major traditional religions: Shamanism, Buddhism, and Confucianism (Park:1994: Hao:1999) and Christianity more recently. Each religion has a different view of death and the appropriate expression of mourning. Korea accommodates funeral customs and rules strictly as a cultural system and has retained these traditions over a long period; hence, some of the traditional funeral rituals still remain in modern Korean life, although some of the rites have been simplified. We have looked at the various ways in which grief and mourning is displayed and shared in a collective manner over a long period of time. This fits in well within the other Eastern cultures that are collectively organized, and contrary to the Eurocentric models do not hastily seek to detach the living from the dead and recognize that grief is a long process, and different individuals may take different amounts of time to recover from the grief. The view of death and bereavement in Korea has sprung from the roots of three Korean religions, together with the recent addition of Christianity, although they mainly result from the three earlier religions. The beliefs of these religions are still closely linked together in the rituals of Korean bereavement on both conscious and unconscious levels. The influence of these religions is evident in practice through the bereaved family's mourning reactions, funeral rites and customs and its views about death. Korea used to have a period of mourning for three years, following traditional mourning rites; then the chief mourner and the bereaved families could return to their normal life. In spite of this long mourning process for the bereaved family, once the funeral ceremony is finished, people expect the bereaved family not to express their grief in public; even the bereaved family does not like to talk about death. The process for bereaved people is related to mourning processes in terms of detachment from the deceased in order to start a new life. Relatives and the community recommend the performance of the kut ceremony for relieving the grief of the bereaved. When one family member dies in an unlucky way, the bereaved family may have some fear or other psychological reactions of grief such as pain, depression, insomnia and nightmares, hallucinations or other physical reactions. Unlucky deaths give the bereaved a very painful time and these types of reactions are often more serious than reactions to natural death. But through the kut ceremony, the bereaved family can start to make a new relationship with the deceased. The taboo of this type of death and death generally remains a crucial aspect of the isolation that bereaved people might face and the collective nature of mourning(even where it is still present) is unable to address this aspect of the privatization of grief.

$^{99m}Tc$ 표지 방사성의약품을 이용한 핵의학 영상검사를 받은 환자에서 방사선 적응반응의 개인간 차이에 대한 연구 (Individual Variation of Radiation Adaptive Responses in Patients Undergoing Imaging Studies Using $^{99m}Tc$ Labeled Radiopharmaceuticals)

  • $^{99m}Tc$ 표지 방사성의약품을 이용한 핵의학 영상검사를 받은 환자들에서 방사선 적응반응이 개인간에 어느 정도 차이가 있는지 알아보고자 하였다. $^{99m}Tc-DTPA$ 신장 신티그라피를 시행한 환자 23명, $^{99m}Tc-MDP$ 골 신티그라피를 시행한 환자 18명 및 $^{99}mTc-TF$ 심근관류 신티그라피를 시행한 환자 21명, 총 62명의 환자를 대상으로 방사성의약품 주사 전, 그리고 주사후 4시간째 채혈하여 림프구를 채취 배양하였다. 고선량 방사선조사는 배양 46시간째 $^{137}Cs$ 조사기를 사용하여 2 Gy의 감마선을 조사하였다. 불안정 염색체의 발생빈도를 Ydr값으로 표현하였고, 방사선 적응반응 지수(k)는 저선량 전처치후 Ydr값을 전처치 전 Ydr값으로 나누어 k값이 1 미만일 때 방사선 적응반음이 유도되는 것으로 간주하였다. 대상환자 62명 중 61명에서 방사선 적응반응이 유도되었고 $^{99m}Tc-DTPA,\;^{99m}Tc-MDP$ 및 Tc-99m-TF 환자군에서 k값의 변동계수 (CV)는 각각 35%, 34%, 21%였다. k값은 성별, 나이 및 병명과 상관관계가 없었다. 핵의학검사를 시행한 환자들에서 방사성 의약품에 의해 유도된 방사선 적응반응은 다양한 개인차가 있었으며, 성별, 나이 또는 병명과 관계없는 다른 원인에 의한 것으로 판단되었다.

Facile Synthesis of In2S3 Modified Ag3PO4 Nanocomposites with Improved Photoelectrochemical Properties and Stabilities

  • In this work, Ag3PO4/In2S3 nanocomposites with low loading of In2S3 (5-15 wt %) are fabricated by two step chemical precipitation approach. The microstructure, composition and improved photoelectrochemical properties of the as-prepared composites are studied by X-ray diffraction pattern (XRD), field emission scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), photocurrent density, EIS and amperometric i-t curve analysis. It is found that most of In2S3 nanoparticles are deposited on the surfaces of Ag3PO4. The as-prepared Ag3PO4/In2S3 composite (10 wt%) is selected and investigated by SEM and TEM, which exhibits special morphology consisting of lager size substrate (Ag3PO4), particles and some nanosheets (In2S3). The introduction of In2S3 is effective at improving the charge separation and transfer efficiency of Ag3PO4/In2S3, resulting in an enhancement of photoelectric behavior. The origin of the enhanced photoelectrochemical activity of the In2S3-modified Ag3PO4 may be due to the improved charge separation, photocurrent stability and oriented electrons transport pathways in environment and energy applications.

The Uneven Distribution of Mating Type Genes in Natural and Cultivated Truffle Orchards Contributes to the Fructification of Tuber indicum

  • The aim of this study was to investigate the pattern of distribution of mating type (MAT) genes of Tuber indicum in ectomycorhizosphere soils from natural T. indicum-producing areas and cultivated truffle orchards and ascocarp samples from different regions. Quantitative real-time PCR and multiplex PCR were used to weight the copy numbers of MAT1-1-1 and MAT1-2-1 in natural truffle soils and cultivated orchard soils. The effect of limestone on the pattern of truffle MAT genes and the correlation between soil properties and the proportion of MAT genes were also assessed. These results indicated that an uneven and nonrandom distribution of MAT genes was common in truffle-producing areas, cultivated truffle orchards, and ascocarps gleba. The competition between the two mating type genes and the expansion of unbalanced distribution was found to be closely related to truffle fructification. Limestone treatments failed to alter the proportion of the two mating type genes in the soil. The content of available phosphorus in soil was significantly correlated with the value of MAT1-1-1/MAT1-2-1 in cultivated and natural ectomycorhizosphere soils. The application of real-time quantitative PCR can provide reference for monitoring the dynamic changes of mating type genes in soil. This study investigates the distributional pattern of T. indicum MAT genes in the ectomycorhizosphere soil and ascocarp gleba from different regions, which may provide a foundation for the cultivation of T. indicum.

Comparative Experiments to Assess the Effects of Accumulator Nitrogen Injection on Passive Core Cooling During Small Break LOCA

  • The accumulator is a passive safety injection device for emergency core cooling systems. As an important safety feature for providing a high-speed injection flow to the core by compressed nitrogen gas pressure during a loss-of-coolant accident (LOCA), the accumulator injects its precharged nitrogen into the system after its coolant has been emptied. Attention has been drawn to the possible negative effects caused by such a nitrogen injection in passive safety nuclear power plants. Although some experimental work on the nitrogen injection has been done, there have been no comparative tests in which the effects on the system responses and the core safety have been clearly assessed. In this study, a new thermal hydraulic integral test facility-the advanced core-cooling mechanism experiment (ACME)-was designed and constructed to support the CAP1400 safety review. The ACME test facility was used to study the nitrogen injection effects on the system responses to the small break loss-of-coolant accident LOCA (SBLOCA) transient. Two comparison test groups-a 2-inch cold leg break and a double-ended direct-vessel-injection (DEDVI) line break-were conducted. Each group consists of a nitrogen injection test and a nitrogen isolation comparison test with the same break conditions. To assess the nitrogen injection effects, the experimental data that are representative of the system responses and the core safety were compared and analyzed. The results of the comparison show that the effects of nitrogen injection on system responses and core safety are significantly different between the 2-inch and DEDVI breaks. The mechanisms of the different effects on the transient were also investigated. The amount of nitrogen injected, along with its heat absorption, was likewise evaluated in order to assess its effect on the system depressurization process. The results of the comparison and analyses in this study are important for recognizing and understanding the potential negative effects on the passive core cooling performance caused by nitrogen injection during the SBLOCA transient.

Converting Panax ginseng DNA and chemical fingerprints into two-dimensional barcode

  • Background: In this study, we investigated how to convert the Panax ginseng DNA sequence code and chemical fingerprints into a two-dimensional code. In order to improve the compression efficiency, GATC2Bytes and digital merger compression algorithms are proposed. Methods: HPLC chemical fingerprint data of 10 groups of P. ginseng from Northeast China and the internal transcribed spacer 2 (ITS2) sequence code as the DNA sequence code were ready for conversion. In order to convert such data into a two-dimensional code, the following six steps were performed: First, the chemical fingerprint characteristic data sets were obtained through the inflection filtering algorithm. Second, precompression processing of such data sets is undertaken. Third, precompression processing was undertaken with the P. ginseng DNA (ITS2) sequence codes. Fourth, the precompressed chemical fingerprint data and the DNA (ITS2) sequence code were combined in accordance with the set data format. Such combined data can be compressed by Zlib, an open source data compression algorithm. Finally, the compressed data generated a two-dimensional code called a quick response code (QR code). Results: Through the abovementioned converting process, it can be found that the number of bytes needed for storing P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can be greatly reduced. After GTCA2Bytes algorithm processing, the ITS2 compression rate reaches 75% and the chemical fingerprint compression rate exceeds 99.65% via filtration and digital merger compression algorithm processing. Therefore, the overall compression ratio even exceeds 99.36%. The capacity of the formed QR code is around 0.5k, which can easily and successfully be read and identified by any smartphone. Conclusion: P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can form a QR code after data processing, and therefore the QR code can be a perfect carrier of the authenticity and quality of P. ginseng information. This study provides a theoretical basis for the development of a quality traceability system of traditional Chinese medicine based on a two-dimensional code.

Vimentin Is Involved in Peptidylarginine Deiminase 2-Induced Apoptosis of Activated Jurkat Cells

  • Peptidylarginine deiminase type 2 (PADI2) deiminates (or citrullinates) arginine residues in protein to citrulline residues in a $Ca^{2+}$-dependent manner, and is found in lymphocytes and macrophages. Vimentin is an intermediate filament protein and a well-known substrate of PADI2. Citrullinated vimentin is found in ionomycin-induced macrophage apoptosis. Citrullinated vimentin is the target of anti-Sa antibodies, which are specific to rheumatoid arthritis, and play a critical role in the pathogenesis of the disease. To investigate the role of PADI2 in apoptosis, we generated a Jurkat cell line that overexpressed the PADI2 transgene from a tetracycline-inducible promoter, and used a combination of 12-O-tetradecanoylphorbol-13-acetate and ionomycin to activate Jurkat cells. We found that PADI2 overexpression reduced the cell viability of activated Jurkat cells in1a dose- and time-dependent manner. The PADI2-overexpressed and -activated Jurkat cells presented typical manifestations of apoptosis, and exhibited greater levels of citrullinated proteins, including citrullinated vimentin. Vimentin overexpression rescued a portion of the cells from apoptosis. In conclusion, PADI2 overexpression induces apoptosis in activated Jurkat cells. Vimentin is involved in PADI2-induced apoptosis. Moreover, PADI2-overexpressed Jurkat cells secreted greater levels of vimentin after activation, and expressed more vimentin on their cell surfaces when undergoing apoptosis. Through artificially highlighting PADI2 and vimentin, we demonstrated that PADI2 and vimentin participate in the apoptotic mechanisms of activated T lymphocytes. The secretion and surface expression of vimentin are possible ways of autoantigen presentation to the immune system.

Overexpression of Rcan1-1L Inhibits Hypoxia-Induced Cell Apoptosis through Induction of Mitophagy

  • Mitophagy, a cellular process that selectively targets dysfunctional mitochondria for degradation, is currently a hot topic in research into the pathogenesis and treatment of many human diseases. Considering that hypoxia causes mitochondrial dysfunction, which results in cell death, we speculated that selective activation of mitophagy might promote cell survival under hypoxic conditions. In the present study, we introduced the Regulator of calcineurin 1-1L (Rcan1-1L) to initiate the mitophagy pathway and aimed to evaluate the effect of Rcan1-1L-induced mitophagy on cell survival under hypoxic conditions. Recombinant adenovirus vectors carrying Rcan1-1L were transfected into human umbilical vein endothelial cells and human adult cardiac myocytes. Using the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide MTT assay and Trypan blue exclusion assay, Rcan1-1L overexpression was found to markedly reverse cell growth inhibition induced by hypoxia. Additionally, Rcan1-1L overexpression inhibited cell apoptosis under hypoxic conditions, as detected by annexin V-fluorescein isothiocyanate (FITC)/propidium iodide (PI) apoptosis assay. Meanwhile, the mitochondria-mediated cell apoptotic pathway was inhibited by Rcan1-1L. In contrast, knockdown of Rcan1-1L accelerated hypoxia-induced cell apoptosis. Moreover, Rcan1-1L overexpression significantly reduced mitochondrial mass, decreased depolarized mitochondria, and downregulated ATP and reactive oxygen species production. We further delineated that the loss of mitochondrial mass was due to the activation of mitophagy induced by Rcan1-1L. Rcan1-1L overexpression activated autophagy flux and promoted translocation of the specific mitophagy receptor Parkin into mitochondria from the cytosol, whereas inhibition of autophagy flux resulted in the accumulation of Parkin-loaded mitochondria. Finally, we demonstrated that mitochondrial 1permeability transition pore opening was significantly increased by Rcan1-1L overexpression, which suggested that Rcan1-1L might evoke mitophagy through regulating mitochondrial permeability transition pores. Taken together, we provide evidence that Rcan1-1L overexpression induces mitophagy, which in turn contributes to cell survival under hypoxic conditions, revealing for the first time that Rcan1-1L-induced mitophagy may be used for cardioprotection.

Helicobacter pylori Chaperone-Like Protein CagT Plays an Essential Role in the Translocation of CagA into Host Cells

  • Most of the Helicobacter pylori strains containing the cag pathogenicity island (PAI) have been associated with more severe gastric disease in infected humans. The cag PAI is composed of 27 proteins, and some of the components are required for CagA translocation into host cells as well as induction of proinflammatory cytokines, such as interleukin-8 (IL-8); however, the exact function of most of the components remains unknown or poorly characterized. In this study, we demonstrated that CagT (HP0532), which is an essential structural component of the cag PAI apparatus, plays an important role in the translocation of CagA into host epithelial cells. In addition to being located on the bacterial surface, CagT is also partially localized in the inner membrane, where it acts as a chaperone-like protein and promotes CagA translocation. However, CagT secretion was not detected by immunoprecipitation analysis of cell culture supernatants. Meanwhile, CagT was related to the introduction of IL-8 of the host cell. These results suggest that CagT is expressed on both the inner and outer bacterial membranes, where it serves as a unique type IV secretion system component that is involved in CagA secretion and cag PAI apparatus assembly.
