Development of a Bottle-Free Multipurpose Incubator for Generating Various Bacterial Culture Conditions

  • The purpose of this study was to develop a multipurpose incubator, without the gas cylinders (bottles) which are required for $H_2$ and $CO_2$ supplementation. In our bottle-free multipurpose incubator, the $H_2$ and $CO_2$ were generated by chemical reactions induced within the chamber. The reaction between sodium borohydride and acetic acid at a molar ratio of 1:1 was used to generate $H_2$, according to the following formula: $4NaBH_4+2CH_3COOH+7H_2O{\rightarrow} 2CH_3COONa+Na_2B_4O_7+16H_2$, whereas the other reaction, citric acid and sodium bicarbonate at a 1:1 molar ratio, was used to generate $CO_2$, according to the following formula: $C_6H_8O_7+3NaHCO_3{\rightarrow}Na_3(C_6H_5O_7)+3H_2O+3CO_2$. Five species of obligate anaerobic bacteria, one strain of capnophilic bacterium, and one strain of microaerophilic bacterium were successfully cultured in the presence of their respective suitable conditions, all of which were successfully generated by our bottle-free multipurpose incubator. We conclude that, due to its greater safety, versatility, and significantly lower operating costs, this bottle-free multipurpose incubator can be used for the production of fastidious bacterial cultures, and constitutes a favorable step above existing anaerobic incubators.

  • Let R be a ring with left identity e and suitably-restricted additive torsion, and Z(R) its center. Let H : $R{\times}R{\times}R{\rightarrow}R$ be a symmetric 3-additive mapping, and let h be the trace of H. In this paper we show that (i) if for each $x{\in}R$, $$n=<<\cdots,\;x>,\;\cdots,x>{\in}Z(R)$$ with $n\geq1$ fixed, then h is commuting on R. Moreover, h is of the form $$h(x)=\lambda_0x^3+\lambda_1(x)x^2+\lambda_2(x)x+\lambda_3(x)\;for\;all\;x{\in}R$$, where $\lambda_0\;{\in}\;Z(R)$, $\lambda_1\;:\;R{\rightarrow}R$ is an additive commuting mapping, $\lambda_2\;:\;R{\rightarrow}R$ is the commuting trace of a bi-additive mapping and the mapping $\lambda_3\;:\;R{\rightarrow}Z(R)$ is the trace of a symmetric 3-additive mapping; (ii) for each $x{\in}R$, either $n=0\;or\;<n,\;x^m>=0$ with $n\geq0,\;m\geq1$ fixed, then h = 0 on R, where denotes the product yx+xy and Z(R) is the center of R. We also present the conditions which implies commutativity in rings with identity as motivated by the above result.

Synthesis of Azelastine.HCl from 4-Chlorophenyl Acetic Acid (4-염화페닐 아세트산을 이용한 염산 아젤라스틴의 합성)

  • A kind of Antihistamines, Azelastine HCl which known as modern H1-blockers, was synthesized by four step process using phthalic anhydride, 4-chlorophenylacetic acid, hydrazine 2HCl. The first step was the reaction of removing carboxyl group and hydroxyl group and the second step was saponification of 3-(4-chlorobenzylidene)phthalide. The third step was the nucleophilic addition reactions of primary amines and the fourth step was addition reaction of N-methyl-1-aza-bicyclo[3,2,0]heptane to 4-(4-chlorobenzyl)-1-(2H)phthalazinone. As a result, product was analyzed by FT-IR and $^1H$-NMR and could be obtained with a yield of 80%.

Preparation Nanosized TPA-Silicalite-1 with Different Silica Sources and Promoters (다양한 실리카 원과 결정화 촉진제를 이용한 나노크기의 TPA-Silicalite-1 제조)

  • In this study, nanosized TPA-silicalite-1 was synthesized with a suitable molar composition of TPAOH: $SiO_2$: $H_2O$ for the development of zeolite ceramic membranes to utilize as gas separation. As silica sources, TEOS, LUDOX AS-40 and CAB-O-SIL were used with the starting material of TPAOH. $NaH_2PO_4$, and a variety of acids and bases were used as promoters after TPAOH, $SiO_2$, $H_2O$ gel synthesis. To decrease synthesis time, a two step temperature change method was applied to the synthesis of TPA-silicalite-1 at a low temperature. TPA-silicalite-1 synthesized was analyzed with XRD, SEM, BET and TGA. As a result, TPA-silicalite-1 powders with a particle size of 100 nm and a specific surface area of $416m^2/g$ were obtained as optimum synthesis conditions when the two stage temperature change method was used with $NaH_2PO_4$ as promoter.

  • This study was aimed to decrease AOX(Adsorbed Organic Halide Compounds) by applying bleaching methods of A(HNO$_3$ + NaNO$_2$) and $D^{(H/L)}$(dual pH chlorine dioxide bleaching) to conventional OCEDED bleaching stages. And so we investigate the effects of NSA treatment and the influence of production rate of ion species in chlorine dioxide and pulp mixtures according to various pH as well as dual pH on pulp bleaching. Finally the effects of AOC_DED^{(H/L)}ED^{(H/L)}$ bleaching stage were investigated by measuring AOX, brightness, kappa number and viscosity of pulps. A stage was treated by using 4% $HNO_3$ and 0.05~2.0% $NaNO_2$ on pulp and $D^{(H/L)}$ stage was like that pulp mixed with chlorine dioxide solution was adjusted at pH 7.0 and reacted in pulp cosistency 3%, S~20min, $70^{\circ}C$, and then successively for 160~175min at pH 4.0. It was found that suitable $NaNO_2$ addition rate was at 0.6% where the brigtness of pulp was most higher and pulp viscosity was not decreased much. Chlorate ($CIO_3$) was decreased according to pH increase but chlorite($CIO_2$) was highly decreased according to pH increase. And chloride was slightly increased with pH increase. The applying of A and D^{(H/L)}$ bleaching stages to some various multiple bleaching stages ($AOC_DED^{(H/L)}/EopD^{(H/L)} etc.$) had good results that brightness was increased more 2~3% ISO than conventional method(OCEDEopD), but viscosity was dropped by only 1~2cps.

  • Let $f_{\beta}=h_{\beta}+\bar{g}_{\beta}$ and $F_a=H_a+\bar{G}_a$ be harmonic mappings obtained by shearing of analytic mappings $h_{\beta}+g_{\beta}=1/(2isin{\beta})log\((1+ze^{i{\beta}})/(1+ze^{-i{\beta}})\)$, 0 < ${\beta}$ < ${\pi}$ and $H_a+G_a=z/(1-z)$, respectively. Kumar et al. [7] conjectured that if ${\omega}(z)=e^{i{\theta}}z^n({\theta}{\in}\mathbb{R},n{\in}\mathbb{N})$ and ${\omega}_a(z)=(a-z)/(1-az)$, $a{\in}(-1,1)$ are dilatations of $f_{\beta}$ and $F_a$, respectively, then $F_a\tilde{\ast}f_{\beta}{\in}S^0_H$ and is convex in the direction of the real axis, provided $a{\in}[(n-2)/(n+2),1)$. They claimed to have verified the result for n = 1, 2, 3 and 4 only. In the present paper, we settle the above conjecture, in the affirmative, for ${\beta}={\pi}/2$ and for all $n{\in}\mathbb{N}$.

Inductoin of Radioresistance by Overexpression of Glutathione S-Transferase K1 (hGSTK1) in MCF-7 Cells (MCF-7 세포주에서 Glutathione S-Transferase K1 (hGSTK1) 과발현에 의한 방사선 내성의 유도)

  • Purpose : This study was conducted to assess the effects of x-irradiation on the expression of the novel glutathione S-transferase K1 gene. Materials and methods : Human glutathione S-transferase K1 (hGSTK1) DNA was purified and ligated to a pcDNA3.1/Myc-His(+) vector for the overexpression of hGSTK1 gene. MCF-7 cells were transfected with or without the recombinant hGSTK1 gene, and irradiated with 6 MV x-ray. After incubation of 14 days, cell survival was measured and compared. The expression of hGSTK1 and the effect of x-irradiation on hGSTK1 expression were also estimated in MCF-7 cells transfected with or without the hGSTK1 gene by RT-PCR. Results : Following 2 to 12 Gy of x-irradiation, the cell survivals were higher in the MCF-7 cells transfected with the hGSTK1 gene than in those without transfection. Despite the higher cell survival in the hGSTK1-transfected cells, RT-PCR for hGSTK1 mRNA revealed no significant differences according to radiation dose, fractionation, and time after irradiation. Conclusion : The MCF-7 cells transfected with the hGSTK1 gene showed higher cell survival than those without transfection of the gene. The hGSTK1 gene might be associated with the radiosensitivity of MCF-7 cell line and further analysis should be needed.

  • We find a graded Artinian level O-sequence of the form $H\;:\;h_0\;h_1\;\cdots\;h_{d-1}\;h_d\cdots$ $^{(d+1-1_)-st}h_d$ < $h_{d+s}$ not having the Weak-Lefschetz property. We also introduce several algorithms for construction of some examples of non-unimodal level O-sequences using a computer program called CoCoA.

Preparation and Stability of Silyl Adlayers on 2×1-Reconstructed and Modified Si(100) Surfaces (Si(100)-2×1 표면과 개질된 Si(100) 표면 상에서 실릴 (Silyl) 흡착충의 형성과 안정성)

  • Saturation-coverage silyl, $-SiH_3(a)$, overlayers were prepared from $Si_2H_6$ adsorption on three comparative surfaces: clean unmodified; D-precovered; and atomically roughened Si(100). Together with its precursor-mediated adsorption behavior, the surface reactivity of $Si_2H_6$ was found to be the highest on the unmodified Si(100)-$2{\times}1$ surface. This was correlated with its dissociative adsorption mechanism, in which both the $H_3Si-SiH_3$ bond scission and the dual surface $Si-SiH_3(a)$ bond formation require a surface dangling bond 'pair'. The unusually high thermal stability of $-SiH_3(a)$ on the unmodified surface was ascribed to a nearly close-packed $-SiH_3(a)$ coverage of ${\sim}0.9$ monolayer and the consequent lack of dangling bonds on the silyl-packed surface.

Optimal pH and Immersion Time for the Organic Acid-activating Treatment of Pyropia yezoensis and Ulva linza (방사무늬김(Pyropia yezoensis)과 잎파래(Ulva linza)의 유기산활성처리에 대한 최적 pH와 침지시간)

  • Herein, two experiments were performed to determine the appropriate pH range and immersion time for organic acid-activating treatment (OAT) in a Pyropia farm. The effects of pH (0.50, 0.75, 1.00, 1.25, 1.50, 1.75, and 2.00) on the cell mortality of Pyropia yezoensis and Ulva linza thalli were tested after 20 sec of immersion under OAT. In addition, the cell mortality of the two species was estimated under various combinations of immersion time (30, 60, and 120 sec) and pH (1.00, 1.50, and 2.00). Upon 20 sec of immersion under OAT conditions, the cell mortality of P. yezoensis did not differ at any pH but that of U. linza exceeded 90% at a pH range of 0.50-1.00. P. yezoensis showed little cell mortality upon 30 sec of immersion, but its mortality exceeded 55% upon 120 sec of immersion at a pH range of 1.00-1.50. U. linza showed 67.9-100% mortality at a pH of 1.50 and 100% mortality at a pH of 1.00, regardless of the immersion time (30-120 sec). These results indicate that for the effective removal of U. linza, the optimal pH range is 1.00-1.50 and the optimal immersion time is 20-30 sec.
