We investigated the induction of resistance to Haemaphysalis longicornis infestation in rabbits that had been immunized with recombinant H. longicornis P27/30 protein. The success of immunological control methods is dependent upon the use of potential key antigens as tick vaccine candidates. Previously, we cloned a gene encoding 27 kDa and 30 kDa proteins (P27/30) of H. longicornis, and identified P27/30 as a troponin I-like protein. In this study, rabbits that were immunized with recombinant P27/30 expressed in Escherichia coli showed the statistically significant longer feeding duration for larval and adult ticks (P<0.05), low engorgement rates in larval ticks (64.4%), and an apparent reduction in egg weights, which suggest that H. longicornis P27/30 protein is a potential candidate antigen for a tick vaccine. These results demonstrated that the recombinant P27/30 protein might be a useful vaccine candidate antigen for biological control of H. longicornis.
To determine effectively the chitinolytic activity of rCHT1 from the hard tick H. longicornis expressed in baculovirus-mediated Spodoptera frugtperda (Sf) 9 cells, a simple and sensitive assay system was established in solid phase using agarose gel containing ethylene glycol chitin as substrate. The various factors affecting the efficacy of the assay were also investigated. The effects of various temperature, dosages of proteins, pH of media and time courses of reaction were examined to verify the sensitivity of assay for chitinolytic activity of rCHT1 protein. It was found that the optimal reactive conditions were $37^{\circ}C$ of temperature, 12 to 15 hours of reactive times, $0.1{\mu}g$ of protein concentration and pH 5 to 7 of media. Using the assay system designed, the functional activities of H. longicornis rCHT1l protein could be evaluated simply and sensitively.
Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease characterized by fever and thrombocytopenia. Haemaphysalis longicornis ticks comprise the major population of ticks in the environment and have been considered as the main vector for SFTS virus (SFTSV). Here we investigated the distribution of ticks carrying SFTSV collected from the environment using the dragging or sweeping methods during April~October 2015 in Jeollanam-do, Korea. Sampling was taken from Songjeong, Omi, Bangkwang, Sandong areas in Jiri walking trails. Among the total 3,869 ticks collected, 3,823 ticks (98.8%) were H. longicornis, 41 (1.1%) were Amblyomma testudinarium, and 5 (0.1%) were Ixodes nipponensis. Classification results by regional groups of H. longicornis indicated that 1,613 ticks were collected in Sandong, 1,190 ticks in Omi, 603 ticks in Bangkwang, and 417 ticks in Songjeong. In monthly distributional studies of H. longicornis based on the developmental stages, nymph (325 ticks) was collected from May to October, 94% of larvae from April to June, and 94% of adult from June to August. These results showed the different dominant stage of ticks according to seasons. However, no SFTSV-specific gene was detected in 3,823 ticks of H. longicornis, 41 of A. testudinarium and 5 of I. nipponensis.
Chemotherapeutic treatment is still the foundation of tick control programs. This study investigated the acaricidal efficacy of cypermethrin alone and in combination with chlorpyrifos against Haemaphysalis (H.) longicornis. Unfed larval ticks were exposed to 0.1, 1.0, and 10 mg/mL cypermethrin for 60 min, after which the acaricidal efficacy was examined based on tick mortality. All compounds showed similar suppression curves, with the best control being achieved by cypermethrin and chlorpyrifos (1 : 1 ratio) at 10 mg/mL. Effective cypermethrin concentrations for tick control were two to seven times higher than the recommended doses, indicating resistance by H. longicornis.
The objective of this study was to evaluate the efficacy of an imidacloprid 10% and flumethrin 4.5% polymer matrix collar against the developmental stages of Haemaphysalis longicornis infesting dogs using the hair from treated dogs in a semi-in-vitro assay set. When incubated with 0.5 g of the hair collected from the dogs installed with the drug-embedded collar after 10 days, average death rate of the larval, nymphal, and adult H. longicornis was 21.5%, 77.9%, and 100% at 30 min, 1 hr, and 2 hr, respectively. This study showed the larval stages as well as the nymphal and adult stages of H. longicornis ticks are killed upon contact with the hair from dogs treated with the collar within 2 hr.

Ticks, blood-sucking ectoparasites, spread diseases to humans and animals. Haemaphysalis longicornis is a significant vector for tick-borne diseases in medical and veterinary contexts. Identifying protective antigens in H. longicornis for an anti-tick vaccine is a key tick control strategy. Enolase, a multifunctional protein, significantly converts D-2-phosphoglycerate and phosphoenolpyruvate in glycolysis and gluconeogenesis in cell cytoplasm. This study cloned a complete open reading frame (ORF) of enolase from the H. longicornis tick and characterized its transcriptional and silencing effect. We amplified the full-length cDNA of the enolase gene using rapid amplification of cDNA ends. The complete cDNA, with an ORF of 1,297 nucleotides, encoded a 432-amino acid polypeptide. Enolase of the Jeju strain H. longicornis exhibited the highest sequence similarity with H. flava (98%), followed by Dermacentor silvarum (82%). The enolase motifs identified included N-terminal and C-terminal regions, magnesium binding sites, and several phosphorylation sites. Reverse transcription-polymerase chain reaction (RT-PCR) analysis indicated that enolase mRNA transcripts were expressed across all developmental stages of ticks and organs such as salivary gland and midgut. RT-PCR showed higher transcript levels in syn-ganglia, suggesting that synganglion nerves influence enolase's role in tick salivary glands. We injected enolase double-stranded RNA into adult unfed female ticks, after which they were subsequently fed with normal unfed males until they spontaneously dropped off. RNA interference significantly (P<0.05) reduced feeding and reproduction, along with abnormalities in eggs (no embryos) and hatching. These findings suggest enolase is a promising target for future tick control strategies.
참진드기 3종류의 발육단계별 즉 유충, 자충, 성충에 대한 계절적 발생 유형을 2014년 4월에서 10월까지 조사하였다. 천끌기와 천젓기 방법으로 포천 영중면 금주리에서 작은소피참진드기 610개체, 개피참진드기 296개체 그리고 일본참진드기 15개체가 채집되었으며 같은 방법으로 철원 서면 자등리에서는 작은소피참진드기 254개체, 개피참진드기 345개체, 일본참진드기 15개체가 채집되었다. 작은소피참진드기의 성충은 6월에 가장 높은 채집 비율을 보였으며 자충은 4월에서 6월사이에 주로 채집되었다. 반대로 작은소피참진드기의 유충은 9월과 10월에만 채집되었다. 개피참진드기의 성충은 4월, 9월, 10월에 채집된 반면 자충은 4월부터 10월까지 채집되었다. 개피참진드기의 자충은 포천에서는 4월에 그리고 철원에서는 5월에 가장 많이 채집되었다. 개피참진드기의 유충은 작은소피참진드기의 유충과 같이 9월과 10월에만 채집되었다. 일본 참진드기의 성충과 자충은 주로 4월에 채집되었으나 유충은 채집되지 않았다. 천끌기과 천젓기 방법으로 채집된 특정한 진드기종의 발육단계별 밀도를 조사한 결과는 진드기의 계절별 밀도 평가와 진드기 매개 질병 위험 분석에 활용할 수 있다.
수증기증류 추출법에 의한 미국산 길초근(Valeriana officinalis)의 구성성분을 알아보기 위해 GC-MS 분석을 수행한 결과, 주요 성분이 patchouli alcohol (18.69%) 및 ${\beta}$-gurjunene (15.26%)인 것으로 나타났다. 그리고 길초근 정유의 살비활성 및 항균활성을 알아보기 위해 작은소피참진드기 및 긴털가루응애에 대하여 접촉독성법을 실시하였고, 식중독균 8 종에 대하여 agar disc diffusion법을 실시하였다. 그 결과, 긴털가루응애 성충에 대하여 $28.01{\mu}g/cm^2$의 $LD_{50}$값을 나타내었으며, 작은소피참진드기 유충 및 약충에 대하여 각각 178.26 및 $207.98{\mu}g/cm^2$의 $LD_{50}$ 값을 나타내었다. 또한 식중독균은 8종 균주에 대하여 항균활성을 나타내었으며, 특히 L.monocytogenes에 대하여 우수한 항균활성을 나타내었다. 따라서 본 연구를 통해 수증기증류 추출법에 의한 길초근 정유가 살비제 및 항생제로서의 가치가 있음을 확인하였다.
본 연구는 2016년부터 2020년까지 충북 보은지역의 참진드기류의 시기별 분포를 조사하였다. 참진드기류는 다양한 환경(매년 4지점)에서 채집하였고, 5년 동안 충북 보은지역에서 채집된 참진드기류는 2속 3종으로 작은소피참진드기(Haemaphysalis longicornis), 개피참진드기(Haemaphysalis flava), 일본참진드기(Ixodes nipponensis)로 총 17,704개체의 참진드기가 채집되었다. 작은소피참진드기는 가장 많은 28.83 T.I. (68.40%) 값을 보였고, 개피참진드기 1.49 T.I. (3.53%), 일본참진드기 0.02 T.I. (0.06%)순으로 채집되었고 종 동정이 되지 않는 유충은 11.81 T.I. (28.01%)로 조사되었다. 우점을 차지하는 작은소피참진드기는 봄철 (5-6월)에, 유충은 8월과 9월에 가장 많은 수가 채집되었다. 대부분의 채집지에서 작은소피참진드기가 가장 많이 채집되었고 무덤과 초지에서만 유충이 많이 채집되었다. 개피참진드기는 충북 보은지역에서 채집된 개체수는 적지만 모든 채집지에서 분포하는 것으로 나타났다. 5년간 채집된 3종의 참진드기류에 대하여 총 828 pools의 SFTS 감염률을 조사한 결과 모두 음성으로 병원체 감염이 확인되지 않았다.
충북 청주지역에서 채집된 작은소피참진드기(Haemaphysalis longicornis)에 대하여 다양한 식물체 메탄올 조추출물과 천궁유래 물질들을 이용하여 살비활성과 기피활성을 조사하였다. 총 24종의 식물추출물을 이용한 작은소피참진드기에 대한 살비활성을 조사한 결과, 천궁(Cnidium officinale)에서 93.3%의 살비율로 가장 높은 활성을 보였으며, 기피 효과의 경우 도라지, 미국자리공, 백목련, 어성초, 천궁, 칡잎 등에서 통계적으로 유의미한 차이를 보였다. 기피 및 살비활성이 가장 높은 천궁의 메탄올추출물을 용매 분획한 물질들과 천궁 유래물질인 butylidenephthalide의 활성을 비교한 결과, 헥산(hexane)층 처리 3일 후에 90%의 높은 살비효과를 보였으며 기피반응도 통계적으로 유의미한 차이를 보였다. 천궁유래물질인 butylidenephthalide은 처리 7일 후에 90%의 살비활성을 보였고 T-tube olfactometer 실험에서 모든 작은소피참진드기가 무처리구로 이동하여 기피활성이 있음을 알 수 있었다. 따라서 본 연구결과는 천궁 조추출물과 천궁유래물질인 butylidenephthalide를 이용하여 작은 소피참진드기의 방제 개발에 기초자료로 활용될 수 있을 것으로 생각된다.
