  • To evaluate the effect of surface contaminated with Escherichia coli O157:H7 (E. coli O157:H7) on the microbiological safety of lettuce, this study was conducted to investigate the attachment, biofilm producing, survival, and cross-contamination of E. coli O157:H7 on stainless steel and polyvinyl chloride (PVC). The attachment rate of E. coli O157:H7 on PVC was 10 times higher than that on stainless steel after exposure 1 h in cell suspension. However, there was not a difference between two types of surface after exposure for 6 h and 24h. The biofilm producing of E. coli O157:H7 was TSB > 10% lettuce extracts > 1% lettuce extracts > phosphate buffer. When two kinds of materials were stored at various conditions ($20^{\circ}C$ and $30^{\circ}C$, relative humidity (RH) 43%, 69%, and 100%), the numbers of E. coli O157:H7 at $30^{\circ}C$, RH 43% or RH 69% were reduced by 5.0 log CFU/coupon within 12 h regardless of material type. Conversely, the survival of E. coli O157:H7 at RH 100% was lasted more than 5 days. In addition, the reduction rate of E. coli O157:H7 was decreased in the presence of organic matter. The transfer efficiency of E. coli O157:H7 from the contaminated surface to lettuce was dependent upon the water amount of the surface of lettuce. Especially, the transfer rate of E. coli O157:H7 was increased by 10 times in the presence of water on the lettuce surface. From this study, the retention of E. coli O157:H7 on produce contact surfaces increase the risk cross-contamination of this pathogen to produce. Thus, it is important that the surface in post harvest facility is properly washed and sanitized after working for prevention of cross-contamination from surface.

  • This study was performed to determine the optimal ratio for preparing an extract comprising the Ecklonia cava and Hizikia fusiformis complex as a therapeutic material for alleviating inflammatory respiratory diseases. First, to examine the optimal ratio for preparing the complex (SD-EH), Ecklonia cava and Hizikia fusiformis extracts were used; four extracts with different mixing ratios were prepared. The effects of the SD-EH extract on MUC5AC mRNA expression in PMA-treated NCI-H292 cells were analyzed; it was confirmed that the MUC5AC expression was significantly reduced after treatment with the SD-EHA-001 (E(100) : H(0)), SD-EHB-001 (E(90) : H(10)), SD-EHC-001 (E(80) : H(20)), and SD-EHD-001 (E(70) : H(30)) extracts. Western blotting was used to determine whether the SD-EH extract affects the expression levels of COX-2 and MMP-9 in PMA-treated A549 cells. The protein expression levels of COX-2 and MMP-9 were significantly lower (p < 0.001) in the cells treated with the SD-EHC-001 (E(80):H(20)), SD-EHD-001 (E(70) : H(30)), and SD-EHE-001 (E(60) : H(40) extracts than in the cells treated with PMA alone. The SD-EHC-001 (E(80) : H(20)) extract markedly downregulated the expression levels of MUC5AC, COX-2, and MMP-9. Therefore, the SE-EH extract may serve as a potential therapeutic agent for treating inflammatory respiratory diseases.

Let $${\cal{M}}_X=\{{\sum}{\limits_{i=1}^n}E_{i}Xf_{i}:n{\in}{\mathbb{N}},f_{i}{\in}{\cal{H}}\;and\;E_{i}{\in}{\cal{L}}\}$$ and $${\cal{M}}_Y=\{{\sum}{\limits_{i=1}^n}E_{i}Yf_{i}:n{\in}{\mathbb{N}},f_{i}{\in}{\cal{H}}\;and\;E_{i}{\in}{\cal{L}}\}.$$ Then the following are equivalent. (i) There is an operator A in $Alg{\cal{L}}$ such that AX = Y, Ag = 0 for all g in ${\overline{{\cal{M}}_X}}^{\bot},A^*A=AA^*$ and every E in ${\cal{L}}$ reduces A. • The properties of three esterases (E2, E6 and E11) which were previously purified from Pieris rapae L. were determined and physiological role of E6 was inferred using immunological methods. Based on inhibitor study, all of the purified esterases were found to be carboxylesterases (EC 3.1.1.1). The Km values for E2, E6 and E11 were determined to be 6.89 X 10-$^4$M. 3.19 $\times$ l0-$^4$M and 3.69 X 10-$^4$M, respectively. The molecular weights of E2, E6 and E11 were estimated to be 42 KD, 81 KD and 174 KD, respectively. The isoelectric points of E2, E6 and E11 were estimated to be pH 5.54, pH 5.89 and pH 6.50, respectively. The concentration of E6 during development was highest at the late 5th instar larval stage and that according to organs at the same stage was highest in midgut. These results suggest that E6 might be a hydrolase involved in the digestion of dietary lipids.

  • This study aimed to inhibit Escherichia coli (E. coli) O157:H7 artificially contaminated in fresh meat using bacteriophage. Among 14 bacteriophages, the highly lytic bacteriophage BPECO19 strain was selected to inhibit E. coli O157:H7 in artificially contaminated meat samples. Bacteriophage BPECO19 significantly reduced E. coli O157:H7 bacterial load in vitro in a multiplicity of infection (MOI)-dependent manner. E. coli O157:H7 was completely inhibited only in 10 min in vitro by the treatment of 10,000 MOI BPECO19. The treatment of BPECO19 at 100,000 MOI completely reduced 5 Log CFU/cm2 E. coli O157:H7 bacterial load in beef and pork at 4 and 8h, respectively. In chicken meat, a 4.65 log reduction of E. coli O157:H7 was observed at 4 h by 100,000 MOI. The treatment of single bacteriophage BPECO19 was an effective method to control E. coli O157:H7 in meat samples.

  • Recently, pre-engineering building (P.E.B.) systematic frames are increasingly being used in steel factory buildings, but almost of the related techniques are dependent on the engineering program (e.g, MBS, LTI), which is usually imported from other countries. These are designed under the AISC-ASD because at present there is no Korean design code for P.E.B. frames. Also, there are few studies onbehaviour and we need to develop the element techniques by using H-shaped components. In particular, there is a tendency towards overestimated design because column-rafter connections have been designed with extended end plate type joint, which is treated asrigid joint,so structural examinations are needed. Therefore, this study represents a basic step in ascertaining the application of P.E.B. systematic frames by using H-shaped column-rafter connectionwith flush type end plate. Its structural performance is compared with that of existing extended type joint using a structural performance test. The structural behaviour of specimen was understood qualitatively and the possibility of application (e.g, design aid charts) of semi-connection (flush type) with H-shaped column-rafter was determined.

  • Low pathogenic avian influenza (LPAI; H9N2) and Newcastle disease (ND) are economically important poultry diseases in Korea. In this study, we investigated pathological features and virus distribution in the lymphoid tissues of chicks experimentally infected with H9N2 and/or ND virus. Six-weeks-old SPF chickens were divided into 4 groups, Control (C), H9N2 (E1), NDV (E2), and H9N2+NDV (E3). E1 group was challenged with 0.1 ml A/Kr/Ck/01310/01 (H9N2) $10^{5.6}$ $EID_{50}$ intranasally, E2 group was challenged with 0.5 ml KJW (NDV) $10^{5.0}{\sim}10^{6.0}$ $ELD_{50}$ intramuscularly, and E3 group was challenged with H9N2, followed 7 days later by NDV. In histopathological examination, E1 group showed depletion and necrosis in bursa of Fabricius, thymus, cecal tonsil, and spleen, whereas E2 and E3 groups were noted severe lymphocyte depletion and necrosis with destruction of lymphoid organs structures. In TUNEL assay, apoptotic bodies were detected in lymphoid organs of all experimental groups, which was most severe in E3 group. H9N2 and ND viruses were predominantly detected in cecal tonsil of E1, E2, and E3 groups by PCR and immunohistochemistry (ICH). In conclusion, co-infection of H9N2 with NDV caused severe pathologic lesions and apoptosis in lymphoid tissues compared to single infections.

  • Seasonal occurrence of the egg parasitoid of bugs, Ooencyrtus nezarae (Hymenoptera: Encyrtidae), was monitored at soybean fields and university campus using traps baited with aggregation pheromone of bean bug, Riptortus clavatus, in Jinju, Gyeongnam province, Korea. The female O. nezarae captured at university campus in 2003 and 2004 was 317.5 and 103.4 times as many as males, respectively, and it was 12.6 times at soybean field in 2004. The female began to occur from late August, showing its peak around September 20 at soybean field in 2004. At university campus in 2003 and 2004, the female began to occur from mid or late August, and showed its peak early or mid September. At the two monitoring sites the catches of female sharply declined after October The E2HZ3H only showed attractiveness to O. nezarae, among three components of the aggregation pheromone of bean bug, (E)-2-hexenyl (Z)-3-hexenoate (E2HZ3H), (E)-2-hexenyl (E)-2-hexenoate (E2HE2H), and myristyl isobutyrate (MI). A mixture in a ratio of 20:20:10 or 16.7:16.7:16.7 of E2HZ3H:E2HE2H:MI attracted significantly more females than the mixture of 7:36:7 ratio did. This higher attractiveness of the former two blends may be attributed to the higher amounts of E2HZ3H in the blends.
