Inhibitory Effect of Shimotsu-to, a Traditional Chinese Herbal Prescription, on Acute Inflammation in Rats and Guinea Pigs

  • We examined the effect of topical application of Shimotsu-to, a traditional Chinese herbal prescription, on carrageenin-induced edema in rats and ultraviolet radiation-induced erythema in guinea pigs. Shimotsu-to (5% in water) markedly suppressed an acute edema of rat hindpaw induced by 1% carrageenin, and was more effective than any other single crude drug componcent of Shimotsu-to, Topical treatment with this prescription also inhibited ultraviolet erythema on the back skin of guinea pigs (a human sunbrun model). These results suggest the therapeutic effect on acute inflammation by topical application of Shimotsu-to.

  • Effect of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on the early- (EAR) and late-phase asthmatic responses (LAR) of guinea pigs were studied in vivo. Guinea pigs were sensitized by injection of 100 mg of ovalbumin (OA). Twenty-one days after sensitization, animals were challenged with exposure to aerosolized 1 % OA for five minutes in double-chambered plethysmograph box with jet nubulizer. Immediately and twenty-four hours after challenge, EAR and LAR ashmatic responses were determined the tidal volume (TV), respiration rate (RR) and specific airway resistance (sRaw), and then animals anethetized and taken the bronchoalveolar lavage fluid (BALF) by lavage the lung with HEPES buffer through cannulation into trachea. A Study on Antigenicity of G009, a Polysaccharide Isolated from Gandoderma lucidum IY009 in Mice and Guinea pigs (영지의 단백다당체 G009의 마우스와 기니픽에 있어서의 항원성에 관한 연구)

  • In the present study, the antigenic potential of G009, a polysaccharide isolated from Ganoderma lucidum IY009, was determined in BALB/C mice and Hartley guinea pigs. Antigenicity tests, including passive cutaneous anaphylaxis (PCA), active systemic anaphylaxis (ASA) and indirect hemagglutination test (IHA) were performed according to the established guidelines of National Institute of Safety Research. The results were as follows: 1. Mice showed no production of antibodies against G009 sensitized with an adjuvant, aluminum hydroxide gel (alum), when judged by the heterologous PCA test in rats. Meanwhile, antibodies against ovalbumin (OVA) sensitized with alum were clearly detected. 2. In the studies with guinea pigs, both the sensitization of G009 alone and of G009 with complete Freund's adjutant (CFA) did not produce positive reactions in homologous PCA. In the case of ASA, however, G009 alone and G009 with CFA produced positive reactions. 3. No G009 specific reaction was observed in an IHA assay using sera isolated from G009 sensitized mice. These findings suggest that G009 have no antigenicity potential in mice but may have weak antigenicity in guinea pjgs.

Effect of Crude Ginseng Saponin on Clinical Pathological Parameters of the Female Adult Guinea Pigs Exposed to 2,3,7,8-Tetrachlorodibenzo-p-dioxin

  • This study was carried out to investigate the effect of crude ginseng saponin (CGS) on clinical pathological parameters in adult female guinea pigs exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). A total of 80 guinea pigs (800$\pm$20 g) were divided into 8 groups: group 1 (normal control group) was given vehicle (com oil containing small amount of acetone and DMSO) and saline; group 2 (single TCDD-treated) received TCDD (1 $\mu\textrm{g}$/kg, i.p.) and saline (i.p.); groups 3 and 4 were administered CGS at daily i.p. doses of 10 and 20 mg/kg for 4 weeks, respectively; groups 5 and 6 were administered CGS (10 and 20 mg/kg, respectively) for 5 weeks starting 1 week before TCDD-exposure; groups 7 and 8 were administered CGS (10 and 20 mg/kg, respectively) for 3 weeks from 1 week after TCDD-exposure. CGS was prepared by Diaion HP-20 adsorption chromatography. Body weight of G2 was significantly decreased from the 2nd week after TCDD-exposure (p<0.01). Body weights of the CGS-treated groups were also decreased by TCDD-exposure, but the weight loss was greatly retarded compared with that of G2. Increase in blood glucose, amylase, lipase, total cholesterol. triglyceride, AST and LDL-cholisterol levels by TCDD exposure was significantly attenuated by the CGS-treatment (p<0.05). From these results, we found that saponin the main active ingredient of ginseng, played a protective role against TCDD-induced toxicity in not only male but female guinea pigs.

Immunoreactivity of PCNA in the Cerebellum of Developing Guinea Pig

  • The investigation of the embryonic development of the cerebellum has a long history. The postnatal normal development of the cerebellum in rodents and other animals became a popular topic for morphological investigations nearly a century ago. However, surprisingly, only a few studies are available regarding the prenatal normal development of the rodent cerebellum, especially in guinea pigs. Cell proliferation is essential for the development of the nervous system. The assessment of cell proliferation can be achieved by using various methods. In this study, we investigated the cell proliferation of the cerebellar cortex in guinea pigs at different stages of pregnancy and in postnatal life. Fetuses were obtained by cesarean section at 50 or 60 days of gestation (dg). Immunohistochemistry was performed with proliferating cell nuclear antigen (PCNA) antibody in the cerebellum. Strong PCNA immunoreactivity was observed in the external granular layer (EGL), which is a neurogenic zone in the cerebellum. The proportion of PCNA-IR cells was greater at 1 week than at 60 dg in lobule I, but not lobule VIII. After 50 dg, the width of the EGL continued to decline until 1 week, due to the maturation of the EGL cells. These results demonstrate the pattern of PCNA immunoreactivity in the developing cerebellum of guinea pigs. This serves as a guideline to study abnormal cerebellum development.

Antigenicity of EPO (Erythropoietin) in Guinea Pigs, Mice and Rats (기니픽, 마우스 그리고 랫드에서 EPO (Erythropoietin)의 항원성에 관한 고찰)

  • The antigenicity of EPO (erythropoietin) was investigated in guinea pig, mice and rats. Antigenicity tests-active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA) of this materials were performed according to the established Regulation of Korean National Institute of Safety Research (1996, 4, 16). The results were followed: 1. After sensitizaion with EPO emulsified with complete Freund's adjuvant (CFA), guinea pigs didn't show any anaphylatic shock symptom in the ASA test 2. After sensitization with antisera of EPO sensitized mice, blue spots were observed on the hypodermis of back of rats in the PCA test, but diameter of each spot was smaller than 5 mm. From the results of this investigation, the antigenicity of EPO was negative under the present experimental condition.

The Mechanism of Thermoregulatory Action of Capsaicin Is Different from That of Its Antinociceptive Effect in Guinea Pig

  • In the present study, we investigated the mechanisms of antinociceptive effect and thermoregulatory action of capsaicin in guinea pigs. The administration of capsaicin (5 mg/kg, s.c.) caused a significant decrease in frequency of eye wiping, an indicative of nociceptive threshold. This antinociceptive effect of calsaicin was abolished by co-administration of capsazepine (30 mg/kg, s.c.) with capsaicin, suggesting the involvement of a vanilloid receptor in the antinociceptive action of capsaicin. The administration of capsaicin (1 mg/kg, s.c.) produced a significant decrease in body temperature of guinea pigs. The maximum decrease in body temperature by 2 degrees was shown 1 hour after the treatment, and this decrease was not reversed by coadministration of capsazepine. In conclusion, it is suggested that the mechanism of action of capsaicin-induced thermoregulation involves different pathways from that of capsaicin-induced antinociception.

  • Antigenicity tests-ASA(Active Systemic Anaphylaxis), PSA(Passive Systemic Anaphylaxis), PCA(Passive Cutaneous Anaphylaxis)- of Bovine Somatotrophine(BST, Lucky Ltd.) were performed according to the established regulations of National Institude of Safety Research. The results were as follows: 1. No specific clinical signs related anaphylaxis were observed. Therefore, it was considered that BST has no antigenicity in guinea pigs and rabbits. 2. No blue spots were observed on the back of guinea pig in the peA test; BST-related IgE was not produced.

Capsaicinoids-induced Neurotoxic Desensitization in Guinea Pig: Antinociception and Loss of Substance P-like Immunoreactivity from Peripheral Sensory Nerve Endings in Bronchi

  • Antinociceptive and desensitizing effects of systemically administered capsaicinoids (capsaicin and KR25018) were investigated in guinea pig. Nociceptive sensitivity to chemical stimulus was examined to test sensory function, and the content of substance P-like immunorractivity (SP-LI) in bronchi was determined as a peripheral marker of capsaicin-sensitive primary afferent neurons. Guinea pigs were pretreated s.c. with several doses of capsaicin (1,2.5,5, 10 mg/kg) or KR25018 (1, 2.5, 5, 10 mg/kg) one week prior to the experiments. Frequency of eye wiping was significantly decreased by capsaicin and KR25018 in a pretreatment dosedependent manner. In capsaicin- or KR25018-pretreated guinea pigs, there was a significant loss of SP-LI in bronchial tissue extracts. In summary, a newly synthesized capsaicin analogue H725018 exhibited antinociceptive effect against chemical stimulus in guinea pig, with comparable potency to capsaicin. This desensitizing activity of capsaicin or KR25018 might be related to the loss of SP-LI in peripheral afferent nerves.

Studies on Protective Efficacy of Escherichia coli Vaccines (송아지 대장균 백신개발에 관한 연구)

  • The oil emulsion and alhydrogel vaccines were prepared from a strain of enterotoxigenic Escherichia coli isolated from calves with diarrhea and their protective efficacy and immunogenicity were tested in Guinea-pigs. Enterotoxigenic Escherichia coli, isolated from calves with diarrhea, has K99 and F4l antigen as 46.2% and 50.9% with 48 and 53 strains respectively out of 104 strains. The protective efficacy of the gel and oil vaccines were 60% and 80% respectively. Agglutinin titers to sera of Guinea-pigs vaccinated with experimental gel and oil vaccines peaked at 5 and 6 weeks after vaccination.
