Finite element analysis (FEA) combined with the concepts of linear elastic fracture mechanics (LEFM) provides a practical and convenient means to study the fracture and crack growth of materials. In this paper, a numerical modeling of crack propagation in the cement mantle of the reconstructed acetabulum is presented. This work is based on the implementation of the displacement extrapolation method (DEM) and the strain energy density (SED) theory in a finite element code. At each crack increment length, the kinking angle is evaluated as a function of stress intensity factors (SIFs). In this paper, we analyzed the mechanical behavior of cracks initiated in the cement mantle by evaluating the SIFs. The effect of the defect on the crack propagation path was highlighted.
This paper describes an experimental study of growing ductile crack-tip behaviors by means of computer image processing technique and finite element method. Here, the displacement field near a crack-tip is first measured by the computer image processing technique. Combined with the finite element technique, strain and stress fields are evaluated from the measured displacement. Then, crack-tip parameters such as J-integral are evaluated using the obtained displacement, strain and stress fields. The present technique Is applied to a growing crack problem of tensile CT specimen made of 304 stainless steel. The ductile crack growth behaviors accompanied by unloading are discussed in detail through the comparison between the experimental and the theoretical results.
The polycrystalline 3C-SiC thin films heteroepitaxially grown by LPCVD method using single precursor 1,3-disilabutane at $850^{\circ}C$. The crystallinity of the 3C-SiC thin film was analyzed by XRD and FT-IR. Residual strain was investigated by Raman scattering. The surface morphology was also observed by AFM and voids or dislocations between SiC and $SiO_2$ were measured by SEM. The grown poly 3C-SiC thin film is very good crystalline quality, surface like mirror, and low defect and strain. Therefore, the polycrystalline 3C-SiC is suitable for harsh environment MEMS applications.

Strain BH5 was isolated from naturally fermented Kimchi and identified as a bacteriocin producer, which has bactericidal activity against Micrococcus flavus ATCC 10240. Strain BH5 was identified tentatively as Lactococcus lactis by the API test and some characteristics. Lactococcus lactis BH5 showed a broad spectrum of activity against most of the non-pathogenic and pathogenic microorganisms tested by the modified deferred method. The activity of lacticin BH5, named tentatively as the bacteriocin produced by Lactococcus lactis BH5, was detected at the mid-log growth phase, reached its maximum during the early stationary phase, and decreased after the late stationary phase. Lacticin BH5 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms as tested by the spot-on-lawn method. Its antimicrobial activity on sensitive indicator cells was completely disappeared by protease XIV or ${\alpha}$-chymotrypsin. The inhibitory activities of lacticin BH5 were detected during treatments up to 100$^{\circ}C$ for 30 min. Lacticin BH5 was very stable over a pH range of 2.0 to 9.0 and was stable with all the organic solvents examined. The cell concentration and bacteriocin production in strain BH5 were maximum when grown at 30$^{\circ}C$ in a modified MRS medium supplemented with 0.5% tryptone, 1.0% yeast extract, and 0.5% beef extract as nitrogen sources. It demonstrated a typical bactericidal mode of inhibition against Micrococcus flavus ATCC 10240. Lacticin BH5 was purified through ammonium sulfate precipitation, ethanol precipitation, and CM-Sepharose column chromatography. The apparent molecular mass of lacticin BH5 was estimated to be in the region of 3.7 kDa, by the direct detection of bactericidal activity after SDS-PAGE. Mutant strain NO141 which was isolated by nitrosoguanidine mutagenesis produced about 4 fold more bacteriocin than the wild type.

Comparison of acoustic emission, ultrasonic testing and crack gauge in 3-point bending testing have been studied. As the results, COD is indirectly assumed by strain gauge rate and grid pitch width when crack gauge grid is out. Acoustic emission is qualitatively able to measure crack growth by total count but ultrasonic testing has a difficulty in measuring it because of echo height fluctuation according to the change and pressure of UT. probe.

In this paper, the fatigue behaviour of typical axisymmetric forward extrusion die is investigated and extrusion process is analyzed by the rigid-plastic finite element method and elasto-plastic finite element method. To approach the crack problem involving crack initiation and propagation in extrusion die, LEFM(Linear Elastic Fracture Mechanics) is introduced and singular element which models stress.strain singularity in the crack tip vincity has been used to obtain an accurate stress intensityu factor values and other results. Form the displacement around the crack tip the stress intensity factor and the effective stress intensity factor at the beginning of the die inlet radius has been calculated. Applying proper fatigue crack propagation criterion such as Paris/Erdogan fatigue law to this data the angle and direction of fatigue crack growth has been simulated and these are compared with some experimental results. Using the computed crack growth rate, fatigue life of the extrusion die has been evaluated.
A total of 28 traditional drinks derived from 23 plant species in 19 families were tested for their in vitro growth-inhibiting effects against Bifidobacterium adolescents, Bifidobacterium bifidum, Lactobacillus acidophilus, Lactobacillus casei, Clostridium perfringens, and Escherichia coil using a paper disc agar diffusion method under anaerobic condition. The responses varied with bacterial strain, plant species and tissue sampled. In a test with C. perfringens at 5 and 10 mg/disc, potent growth inhibition was produced from the extracts of Eucommia ulmoides stems, Pinus densiflora leaves and shoots, Thea sinensis leaves (green and oolong teas) and Zingiber officinale roots. All materials tested did not adversely affect the growth of bifidobacteria, lactobacilli, and E. coli. These results may be an indication of at least one of the pharmacological activities of these plant-derived drinks.

Effects of supplementation with ruminal epithelial cells on fiber-degrading activity and cell growth of Ruminococcus albus (R. albus, strain 7) was tested using a basal substrate of rice straw and formulated concentrate. Cultures of R. albus alone and R. albus with rumen protozoa were grown at $39^{\circ}C$ for 48 h with an 8.4% crude protein (CP) substrate, 33% of the CP supplemented with either ruminal epithelial cells or defatted soybean meal. The ruminal epithelial cells had lower amounts of rumen soluble and degradable protein fractions as compared to defatted soybean meal, as determined by an enzymatic method, and the same was found with amino acid composition of protein hydrolysates. Ruminal epithelial cells were directly utilized by the R. albus, and resulted in greater growth of cell-wall free bacteria compared to defatted soybean meal. The effect of epithelial cells on bacterial growth was enhanced by the presence of rumen protozoa. In consistency with cultures of R. albus and R. albus with rumen protozoa, fermentative parameters such as dry matter degradability and total volatile fatty acid did not differ between supplementation with ruminal epithelial cells or defatted soybean meal.
The protein-bound innerpolysaccharides (IPS) produced by suspended mycelial cultures of Inonotus obliquus have promising potentials as an effective antidiabetic as well as an immunostimulating agents. To enhance IPS production, intensive strain improvement process should be carried out using large amount of UV-mutated protoplasts. During the whole strain-screening process, the stage of solid growth-culture was found to be the most time-requiring step, thus preventing rapid screening of high-yielding producers. In order to reduce the cell growth period in the solid growth-stage, therefore, solid growth-medium was optimized using the statistical methods such as (i) Plackett-Burman and fractional factorial designs (FFD) for selecting positive medium components, and (ii) steepest ascent (SAM) and response surface (RSM) methods for determining optimum concentrations of the selected components. By adopting the medium composition recommended by the SAM experiment, significantly higher growth rate was obtained in the solid growth-cultures, as represented by about 41% larger diameter of the cell growth circle and higher mycelial density. Sequential optimization process performed using the RSM experiments finally recommended the medium composition as follows: glucose 25.61g/L, brown rice 12.53 g/L, soytone peptone 12.53 g/L, $MgSO_4$ 5.53 g/L, and agar 20 g/L. It should be noted that this composition was almost similar to the medium combinations determined by the SAM experiment, demonstrating that the SAM was very helpful in finding out the final optimum concentrations. Through the use of this optimized medium, the period for the solid growth-culture could be successfully reduced to about 8 days from the previous 15~20 days, thus enabling large and mass screening of high producers in a relatively short period.
BACKGROUND: This study was performed for selecting yeast mutants with a high tolerance for targeted metals, and determining whether yeasts strains tolerant to multiple heavy metals could be induced by sequential adaptations. METHODS AND RESULTS: A mutant yeast strain tolerant to the heavy metals cadmium (Cd), copper (Cu), nickel (Ni), and zinc (Zn) was selected by sequential elevated exposures to each metal with intermittent mutant isolation steps. A Cd-tolerant mutant was isolated by growing yeast cells in media containing $CdCl_2$ concentrations that were gradually increased to 1 mM. Then the Cd-tolerant mutant was gradually exposed to increasing levels of $CuCl_2$ in growth media until a concentration of 7 mM was reached, thus generating a strain tolerant to both Cd and Cu. In the subsequent steps, this mutant was exposed to $NiCl_2$ (up to 8 mM), and a resultant isolate was further exposed to $ZnCl_2$ (up to 60 mM), allowing the derivation of a yeast mutant that was simultaneously tolerant to Cd, Cu, Ni, and Zn. CONCLUSION: This method of inducing tolerance to multiple targeted heavy metals in yeast will be useful in the bioremediation of heavy metals.
