비열 유전체장벽방전 플라즈마 발생기의 풍량에 따른 결핵균 성장억제 효능 (Effect of Non-thermal Dielectric Barrier Discharge Plasma by Air Volume against Mycobacterium Tuberculosis)

  • Objectives: The objective of this study was to evaluate the inhibitory effect of non-thermal dielectric barrier discharge (DBD) plasma by air volume against Mycobacterium tuberculosis (MTB). Methods: Plasma generators (TB-300, Shinyoung Airtec, Seongnam-si, Korea) were operated in a 2A type biosafety cabinet. The plasma generator was set to a wind flow rate of 14 ($80m^3/h$), 18 ($110m^3/h$), and 22 ($150m^3/h$), and exposure times were set to 0 hours, 3 hours, 6 hours, 9 hours, and 24 hours. Results: The inhibitory effects of plasma at air volume 14 with prolonged exposure time of three hours was 20%, 64% at six hours, 82.3% at nine hours, and 100% after 24 hours exposure. With air volume of 18, the inhibitory effects upon plasma exposure were 36% for three hours, and 100% from 24 hours. Greater air volume resulted in greater inhibition of tuberculosis bacterial growth. In particular, the maximum inhibitory effect (100%) was shown in air volume of 22 ($150m^3/h$) after three hours of plasma exposure. Conclusions: The results showed the correlating inhibitory effects of plasma on the growth of MTB in combination with increasing plasma exposure time and air volume.

느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과 (Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells.)

  • 인체 암세포계(MG-63 골육암 세포, HT-29 인체 결장암세포, K-562 백혈암세포)를 이용하여 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액에 의한 암세포 성장에 미치는 효과를 검토하였다. 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액은 낮은 농도에서부터 인체 골육암 세포의 증식을 억제시켰다. 인체 결장암세포와 백혈암세포에서도 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액은 낮은 농도인 1 mg/ml에서부터 활성을 나타내어 40% 이상으로 암세포 증식 억제효과를 나타내어 앞서의 MG-63 골육암 세포에서 보다 그 증식 억제효과가 높은 것으로 나타났다. 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액을 골육암과 결장암세포에 투여한 2일 후에 세포내의 DNA 합성에 미치는 영향을 측정한 결과, 농도 의존적으로 암세포의 DNA 합성을 저해하는 것을 살펴 볼 수가 있었다. 따라서 느릅나무 근피 추출물은 인체 암세포 증식을 크게 억제하였으며 열탕 추출물에서도 항암활성 효과를 보여 느릅나무 근피 유래 생리활성 물질은 열에 안정한 것으로 여겨진다.

  • Chemokine and Growth Factor are major mediumtors of immuno-inflammatory pathway. The purpose of this study is to investigate whether productions of Chemokine and Growth Factor in lipopolysaccharide (LPS)-induced mouse macrophage RAW 264.7 cells are modulated by Gallic acid (GA), which is easily founded in tannin-containing natural materials such as red wine, green tea, grape juice, and Corni Fructus. Productions of Chemokine and Growth Factor were analyzed by High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on $xMAP^{(R)}$ (multi-analyte profiling beads) technology. At first, cell culture supernatant was obtained after treatment with LPS and GA for 24 hour. Then, the antibody-conjugated beads were added and incubated for 30 minutes. After incubation, detection antibody was added and incubated for 30 minutes. And Strepavidin-conjugated Phycoerythrin (SAPE) was added. After incubation for 30 minutes, the level of SAPE fluorescence was analyzed on Bio-plex Suspension Array System. Based on fluorescence intensity, concentrations of Chemokine and Growth Factor were determined. The results of the experiment are as follows. GA significantly inhibited the production of interferon-inducible protein (IP)-10, keratinocyte-derived chemokine(KC), and vascular endothelial growth factor (VEGF) in LPS-induced RAW 264.7 cells at the concentration of 25, 50, 100, 200 uM (p<0.05). GA significantly inhibited the production of monocyte chemoattractant protein-1(MCP-1) and macrophage-colony stimulating factor(M-CSF) in LPS-induced RAW 264.7 cells at the concentration of 50, 100, 200 uM (p<0.05). GA diminished the production of granulocyte macrophage-colony stimulating factor (GM-CSF) in LPS-induced RAW 264.7 cells. But GA did not show the inhibitory effect on the production of leukemia inhibitory factor (LIP) and macrophage inflammatory protein (MIP)-2 in LPS-induced RAW 264.7 cells. These results suggest that GA has the immuno-modulating activity related with its inhibitory effects on the production of IP-10, KC, MCP-1, VEGF, and M-CSF in LPS-induced macrophages.

  • [6]-Gingerol, a major pungent ingredient of ginger (Zingiber officinale Roscoe, Zingiberaceae) has a wide array of pharmacologic effects. Our previous studies have demonstrated that [6]-gingerol inhibits mouse skin tumor promotion and anchorage-independent growth of cultured mouse epidermal cells stimulated with epidermal growth factor. In this study, we have investigated the molecular mechanisms underlying anti-tumor promoting effects of [6]-gingerol on mouse skin carcinogenesis. (omitted)

한약재 추출물의 항균효과 검색 (Screening of Antimicrobial Activity for Medicinal Herb Extracts)

  • 천연식품보존료 개발 목적의 일환으로 항균효력이 있을 것으로 추정되는 한약재 중에서 목단피, 황백, 연교, 오미자, 자초, 길경 등 20종을 대상으로 물과 95% 에탄올로 항균성물질을 추출하여 각종 미생물에 대한 항균력을 검토하였다. 물 추출물의 경우, 모든 세균에 대하여 증식억제 효과가 나타난 것은 치자, 구기자, 오미자 등의 3종이었으며, 그중에서 오미자가 증식 억제력이 제일 강하였다. 에탄올 추출물의 경우, 대부분의 세균에 대하여 증식억제 효과가 나타난 것은 치자, 오미자, 자초 등의 3종이었으며, 목단피, 황백, 금은화 등의 3종은 그람양성균에만 효과가 나타났는데, 특히 황백의 경우, S. aureus에 대하여는 0.01%의 첨가로 균의 증식이 억제되었다. 한편 이들 한약재들의 물 추출물은 진균류에 대해서는 증식억제 효과가 좋지 않았으나, 에탄올 추출물의 경우, 목단피, 황백, 치자, 자초 등의 4종이 증식억제 효과가 우수하였고, 그중 황백이 균증식 억제력이 제일 강하였다. 전 시료를 통하여 에탄올 추출물이 물 추출물 보다 $2{\sim}100$배나 항균력이 높았으며, 특히 자초의 에탄올 추출물은 맛과 향 등의 관능검사결과 이미 이취가 적을 뿐만 아니라 항균력도 우수하여 천연식품 보존료로서 이용 가능성이 높았다.

한국산 현미 및 율무 추출물에 의한 돌연변이 및 인체 암세포주 증식 억제 효과 (Inhibitory Effects of Methanol Extracts from Korean Orysa sartiva and Coix lachryma-jobi var. ma-yuen on Mutagenicity and Growth of Human Cancer Cells)

  • 본 연구에서는 비교적 양질의 영양소를 가진 현미 및 율무 메탄올 추출물의 항돌연변이 및 인체 암세포 증식억제 효과에 대하여 알아보고자 하였다. Ames test를 이용한 돌연변이 억제 실험에서 $AFB_1$ (0.6 mg/plate)에 대해 현미 메탄올 추출물은 농도의 증가와 더불어 돌연변이 억제 효과가 증가하여 첨가농도 2.5 및 5 mg/plate일 때 각각 70% 및 76%의 억제 효과를 나타내었다. 율무 메탄올 추출물의 경우도 첨가농도 2.5 및 5 mg/plate일 때 각각 60% 및 76%의 항돌연변이 효과를 관찰 할 수가 있었다. 직접돌연변이원인 MNNG에 대해서도 현미 및 율무 메탄올 추출물들은 첨가농도 5 mg/plate에서 각각 79% 및 69%의 항돌연변이 효과를 나타내었다. 인체 위암세포(AGS)에 현미 메탄올 추출물을 5, 10, 20 mg/ml의 농도별로 처리했을 때 농도 의존적으로 암세포 증식 억제 효과가 증가하였다. 율무 메탄올 추출물의 경우도 현미 메탄올 추출물과 유사하게 농도 의존적으로 암세포의 증식을 억제시켰으며 첨가농도 20 mg/ml일 때 77%의 증식 억제효과를 살펴 볼 수가 있었다. 인체 결장암세포(HT-29)의 경우에서도 이들 현미 및 율무추출물은 낮은 농도에서부터 그 억제효과가 나타나 농도 의존적으로 암세포의 증식을 억제시켰다.

인진쑥 추출물의 병원성 세균에 대한 생체내 증식 억제 효과 (Inhibitory Effects of Artemisia capillaris Extract on the Pathogenic Bacteria in Mice)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate the inhibitory effects of orally administrated Artemisia capillaris extracts on the pathogenic bacteria in 200 ICR mice. The experimental groups showed inhibitory effects on the bacteria in $1{\sim}3$ days after inoculation. After 21 days of inoculation, no viable bacterial cells appeared in the feces of both experimental groups while they did appear in the control group. The results of these studies indicate Artemisia capillaris extract exhibited excellent antimicrobial and inhibitory effects on the food poisoning pathogenic bacteria; S. enteritidis, E. coli O157:H7, L. monocytogenes and S. aureus.

인산염이 Listeria monocytogenes Scott A 성장에 미치는 영향 (Effect of Polyphosphates on the Growthof Listeria monocytogenes Scott A)

  • To investigate the antimicrobial effect of polyphosphates as a food additive, the growth and structural change of Listeria monocytogentes Scott A were examined in relation to polyphosphates concentration and incubation temperature. Up to 10,000 ppm of polyphosphates, the growth rate of strain was gradually inhibited with increasing polyphosphates concentration and decreasting the incubation temperature. Minimal inhibitory concentration of polyphosphates to the growth of strain was about 12,000 ppm. It was observed , using both scanning electron microscopy(SEM) and transmission electron microscopy(TEM), that 0.9% polyphosphates treatment was resulted in the destruction of cell wall and outflow of cell ingredients. The antimicrobial effects of polyphosphates were more effective than those of dehydroacetate and potassium sorbate at 13$^{\circ}C$ and 4$^{\circ}C$. The growth rate the strain in beef was significantly inhibited by the treatment of 0.9% polyphosphates and storaged at cooling temperature.

Inhibitory effects of Coptis chinensis extract on the growth and biofilm formation of Streptococcus mutans and Streptococcus sobrinus

  • Streptococcus mutans and Streptococcus sobrinus play important roles in dental caries. Coptis chinensis is a natural product with antimicrobial activity against enterobacteria; however, its effects on oral streptococci are still unknown. Therefore, the effects of C. chinensis on the growth and biofilm formation of the representative cariogenic bacteria S. mutans and S. sobrinus were investigated for the possible use of C. chinensis as an anticaries agent. The C. chinensis extract was diluted with sterile distilled water, and 0.1-2.5% of the extract was used in the experiment. The effects of the C. chinensis extract on the growth and glucan formation of S. mutans and S. sobrinus were measured by viable cell counting and spectrophotometry at 650 nm absorbance, respectively. Crystal violet staining was also carried out to confirm the C. chinensis extract's inhibitory effect on biofilm formation. The C. chinensis extract significantly inhibited the growth of S. mutans and S. sobrinus at concentrations of ≥ 0.3% as compared with the control group. The viable cell count of colonies decreased by 1.7-fold and 1.2-fold at 2.5% and 1.25%, respectively, compared with the control group. The biofilm formation of S. mutans and S. sobrinus was inhibited by > 20-fold at C. chinensis extract concentrations of ≥ 1.25% as compared with the control group. In summary, the C. chinensis extract inhibited the growth and biofilm and glucan formation of S. mutans and S. sobrinus. Therefore, C. chinensis might be a potential candidate for controlling dental caries.

적미 추출물과 분획물의 항산화 활성 및 암세포 성장억제효과 (Effect of Methanol Extracts of Red Colored Rices on Antioxidant Activity and Growth Inhibitory Activities of Cancer Cells)

  • 자광도와 홍향미, 동진현미를 물과 methanol로 추출하여 과산화지질생성 억제능 및 4종류의 인체유래의 암세포 즉, 간암세포 HepG2, 위 암세포 SNU-1, 유방암세포 MCF-7 및 대장암세포 SNU-C4에 대한 성장억제효과를 검토하였다. 물 추출물보다는 methanol 추출물에서, 동진현미보다는 자광도와 홍향미에서 높은 과산화지질생성 억제능과 암세포의 성장억제효과가 나타났다. 과산화지질생성 억제능은 홍향미 methanol 추출물(1 mg/assay)에서 80%, 자광도 methanol 추출물(1 mg/assay)에서 68%였다. 암세포 성장억제효능은 MCF-7에 자광도 methanol 추출물 첨가시 57%로 가장 낮았다. 또한 methanol 추출물을 극성이 서로 다른 4개의 유기용매로 분획하여 얻은 획분으로 과산화지질생성 억제능과 인체유래 암세포의 성장억제효과를 확인하였다. 자광도와 홍향미 methanol 추출물 분획물의 암세포에 대한 성장억제효과는 세포주에 따라 다소 차이는 있으나 chloroform, hexane 및 ethyl acetate 획분에서 성장억제효과가 큰 것으로 나타났다.
