Growth Inhibition on the Strain Isolated from Spoiled Red Bean Paste

  • Growth inhibitory effects of ethanol extracts of green tea and pine needles on Bacillus stearothermophilus isolated from spoiled red bean paste were detected at concentrations higher than 750 ppm, and antimicrobial activity of pine needle extract was slightly higher than that of green tea exract. Growth inhibitory effect of pine needle extract in nutrient broth adjusted to pH 6.0, water-activity 0.92, and $45\;^{\circ}$Brix was observed at 500 ppm. These results indicated growth of B. stearothermophilus could be inhibited by adding pine needle and green tea extracts.

Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과)

  • Growth and DNA synthesis inhibitory effects of extracts from root bark of Ulmus parvifolia on MG-63 human osteosarcoma cells, HT-29 human colon cancer cells and K-562 leukemia cancer cells were studied. The root bark extract of Ulmus parvifolia was extracted with methanol, hot water and juice. The methanol extract showed the highest inhibitory effect on growth of MG-63, HT-29 and K-562 cancer cells by >85%. The treatment of hot water and juice extracts from root bark of Ulmus parvifolia also inhibited growth of the above cancer cells with increasing concentration. DNA synthesis of MG-63 and HT-29 cancer cells was significantly inhibited by adding methanol, hot water and juice extracts from root bark of Ulmus parvifolia with increasing concentration, showing that the inhibitory effect of growth was more effective on HT-29 cancer cells. These results suggest that the methanol extract from root bark of Ulmus parvifolia may have specific active com-pounds on anticancer effect. The hot water extract also showed a strong inhibitory effect on growth of cancer cells, indicating that the active compounds may be stable to heat.

  • The study was performed to investigate the inhibitory action on the growth rate of Candida albicans under the various immunological conditions, Candida albicans was grown in broth media containing 0.1ml. (in total broth volume 4.0ml.) of normal human serum, diabetic patient serum, albumin solution, artifical bulla content and nutrient solution as control subject. The inhibitory action of Candida albicans was indicated by mesurement of transparency rate with use of the spectrophotometer. The results are as follows: 1. Normal human serum shows inhibitory effect on the growth of the candida distinctly. 2. Albumin solution reveals almost similar to that of the effect of normal human serum. 3. Artificial bulla content which obtained by irradiation of ultraviolot ray after application of 1% 8-Methoxy-psolaren cream and the diabetic patient serum shows reduction of inhibitory effect as compared with that of the normal human serum. 4. It is estimated that the titer of negative effect of diabetic patient serum is not related with the variation of immunoglobulin titer in patient serum.

Inhibitory effects of Euphorbiae lathyridis Semen extract on cell growth in HT-29 human colon cancer cells (속수자 추출물의 HT-29 대장암세포 증식에 대한 억제효과)

  • Objectives. In this study, we investigate that methanol extract of Euphorbiae lathyridis Semen contributes to growth inhibitory effect on the HT-29 human colon cancer cells. Methods. Euphorbiae lathyridis Semen (ELS) was extracted with 80% methanol. HT-29 cells were treated with different concentrations of ELS extract for 24-72 hrs. Growth inhibitory effect was determined by MTT assay. Cell apoptosis was determined by surveying caspases cascades activation using Western blot. Cell cycle arrest was analyzed by flow cytometry with PI staining. Results. Exposure to ELS extract showed in inhibitory effects on HT-29 cell growth as a dose-dependent manner. Cell growth inhibition by ELS extract was related with induction of cell apoptosis with DNA fragmentation through the activation of caspases-3, caspase-9 and PARP cleavage. Conclusion. ELS extract significantly inhibited cell growth and induced cell apoptosis in HT-29 human colon cancer cells, therefore, These results suggest that ELS extract can be used as chemoprevention agent of colon cancers.

Enzymological Evaluation of Bowel Inflammation Inhibitory Activity and Intestinal Microbial Flora Improvement by Enzymatic Hydrolysate of Schizandrae Fructus (오미자 효소가수분해물에 의한 장내 세균총 개선 및 장관 염증 억제 효과의 효소학적 평가)

  • Schizandrae Fruits has been used as a traditional Oriental medicine for treatment of many stress-induced diseases. In the present study, we investigated inhibitory activity of enzymatic hydrolysate of Schizandrae fructus (SC-EX) in growth of tested intestinal microorganism and activity of bowel inflammation related enzyme. SC-EX was added to the proteose peptone-yeast extract-fildes (PYF) media to investigation the effect on the growth of type culture of intestinal microorganism. The growth of lactic acid bacteria such as Bifidobacterium species and Lactobacillus species was accelerated by more than 3% concentration of SC-EX. But, growth of harmfulness bacteria such as E.coli, Clostridium sp. Staphylococcus sp. Streptococcus sp. was inhibited by more than 3% concentration of SC-EX. Also, SC-EX was exhibited a concentration-dependent inhibitory activity of the bowel inflammation related enzymes. The SC-EX was showed 76% and 92% inhibitory activity of 5-lipoxygenase and cyclooygenase at 5% additional concentration respectively. Our results indicated that SC-EX may possess improvement effect on the intestinal flora and Anti-inflammatory effect on the bowel.

Effect of Various Biodegradable Chelating Agents on Growth of Plants under Lead stress (생분해되는 다양한 킬레이트들이 납에 노출된 식물의 성장에 미치는 영향)

  • Phytoextraction is a method of phytoremediation using plants to remediate metal-contaminated soils. Recently, various chelating agents were used in this method to increase the bioavailability of metals in soils. Even though phytoextraction is an economic and environment-friendly method, this cannot be applied in highly metal-contaminated areas because plants will not normally grow in such conditions. This research focuses on identifying chelating agents which are biodegradable and applicable to highly metal-contaminated areas. Lead (Pb) as a target metal and cysteine (Cys), histidine (His), citrate, malate, oxalate, succinate, and ethylenediamine (EDA) as biodegradable chelating agents were selected. Ethylenediamine tetraacetic acid (EDTA) was used as a comparative standard. Plants were grown on agar media containing various chelating agents with Pb to analyze the effect on root growth. Cys strongly increased the inhibitory effect of Pb on root growth of plants, while, His did not affect on it significantly. The inhibitory effect of oxalate is weak, and malate, citrate, and succinate did not show significant effects. Both EDTA and EDA diminished the inhibitory effect of Pb on root growth. The effect of EDA is correlated with decreased Pb uptake into the plants. In conclusion, as biodegradable chelating agents, EDA is a good candidate for highly Pb-contaminated area.

Inhibitory Effect of Zostera japonica on Growth of Human Cancer Cells (애기거머리말 추출물의 암세포 성장 억제효과)

  • In this study, crude extracts of the marine eelgrass Zostera japonica and their solvent-partitioned fractions were evaluated for their inhibitory effect against AGS, HT-1080 and MCF-7 human cancer cells using MTT assay. Each of the crude extracts (acetone/methylene, chloride, and methanol) of Z. japonica showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts were partitioned between $CH_2Cl_2$ and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and the aqueous layer was then fractionated into n-BuOH and $H_2O$, successively. Growth inhibition effects of solvent-partitioned fractions from Z. japonica on human cancer cells increased in a dose-dependent manner. Among these tested samples, the 85% aq. MeOH fraction revealed good inhibitory effects on the growth of AGS and HT-1080 human cancer cells, while the n-hexane fraction exhibited good inhibitory effects on the growth of AGS and MCF-7 human cancer cells. In addition, 85% aq. MeOH and n-hexane fractions enhanced mRNA expression of p53 gene. These results suggest that there is further scope for the isolation of active compounds from Z. japonica, which should show much stronger anticancer activity.

Anti-microbial Activities Against Oral Microbes and Growth-Inhibitory Effect on Oral Tumor Cell of Extracts of Perilla and Mugwort (들깨잎과 쑥 추출물의 구강병 원인균에 대한 항균 및 KB 세포 증식 억제효과)

  • Methanol extracts of Perilla and Mugwort were stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activities and inhibitory effect on growth of oral tumor cell of each extract were investigated. Each extracts of Perilla and Mugwort were investigated to anti-microbial effects on oral microbes by means of agar diffusion method and MIC. These results suggest that the hexane extracts of Perilla and Mugwort have antimicrobial activities against S. mutans and potent inhibitory Effect to KB cell growth.

In vitro Screening of UVA Phototoxicity Inhibitors using the Natural Products (In vitro 실험법에 의한 천연물 중의 UVA 광독성 억제제 검색)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.

  • In its study, We investigated the effects of Areca catechu L on the growth and acid production of Streptococcus mutans JC-2 in broth system and the activity of glucosyltransferase. The results were summarized as follow; 1. The growth of Streptococcus mutans JC-2 was suppressed by adding Areca catechu L in broth system. Especially, its inhibitory effect was significant at 2,000ppm of concentration. 2. Areca catechu L decreased the acid production of Streptococcus mutans JC-2. Decrease of pH according to acid production was less in presence of Areca catechu L than in absence. 3. Areca catechu L exerted the inhibitory effect against glucosyltransferase activity form Streptococcus mutans JC-2.
