Experimental Studies on the Effect of Houttuyniae herba Aqua-acupuncture on the Growth of Melanoma B16 Tissue and Mononuclear Cell Subsets in Mice (어성초(魚腥草) 약침(藥鍼)이 Melanoma B16 피부암(皮膚癌)의 증식억제효과(增殖抑制效果)와 Mononuclear cell subsets에 미치는 영향(影響))

  • Experimental studies were performed to investigate the effect of Houttuyniae herba Aqua-acupuncture on the growth of Melanoma 816 tissue and Mononuclear cell subsets in mice. The results are as follows : 1. In the study of lymphocytes numbers in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 2. In the study of $CD4^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 3. In the study of $CD8^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, with stastistical significance. 4. in the study of lymphocytes numbers in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 5. In the study of $CD4^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 6. In the study of $CD8^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 7. In the measurement of melanoma tissue weight, Houttuyniae herba Aqua-acupuncture showed increasing effect, but without stastistical significance. According to above results, it is concluded that Houttuyniae herba Aqua-acupuncture is effective upon immune responses in Melanoma 816 inoculated mice.

Inhibitory Effect of 7-O-butyl Naringenin on Growth of Helicobacter pylori ATCC 26695

  • The antimicrobial effect of a novel flavonoid (7-O-butyl naringenin) on Helicobacter pylori ATCC 26695 and its inhibitory effects on the urease activity of the strain were evaluated by comparing with quercetin and naringenin. H. pylori was cultured with brain heart infusion supplemented with 5% horse serum at $37^{\circ}C$ under 10% $CO_2$ atmosphere and the inhibitory effects of flavonoids against the strain were detected using micro-plate methods. During 12 hr of incubation time, the optical densities of phenol red reduced (pink color) in the urea broth by producing ammonia were detected at 560 nm with a spectrophotometer. The results indicated that both quercetin and 7-O-butyl naringenin were effective against the growth of H. pylori. Moreover, inhibitory effect of 7-O-butyl naringenin on the growth of H. pylori was about two-fold higher than quercetin at the same concentration. With regard to H. pylori urease activity, 7-O-butyl naringenin had a greater inhibitory effect than did naringenin or quercetin at the same concentration.

Research to Fungicide Sensitivity of Colletotrichum spp. Isolated from Apple Fruits in Cheongsong, Korea (사과 주 재배지 경북 청송 지역 사과 탄저병원균의 살균제에 대한 감수성 조사)

  • Apple grower in the Cheongsong region, Korea has reported the increased loss of apple yield due to severe bitter rot incidence. We noticed that this indience is because the Colletotrichum population has developed resistance to commonly used fungicides. We isolated 39 Colletotrichum isolates from 13 orchards in Cheongsong, and all the isolated Colletotrichum species were identified as C. siamense or C. fructicola. These 39 strains were tested for mycelial growth and conidial germination against 12 fungicides. trifloxystrobin (30-55% in recommended concentrations) was shown lower inhibitory effect on mycelial growth. However, the inhibition of conidial germination was shown higher than mycelial growth (62-100%). Kresoxim-methyl was shown lower inhibitory effect on mycelial growth (29-55%). conidial germination inhibitory effect was shown 51% to 96%. dithianon was shown diversity response to inhibition of mycelial growth (43-100%). Tebuconazole was shown high inhibitory effect on mycelial growth (84-100%) and conidial germination inhibitory effect was shown to be 64 to 100%. metconazole has been found to display with high inhibitory effect on mycelial growth (79-100%) and conidial germination (70-80%). fluazinam was shown to possess high inhibitory effect on mycelial growth (87-100%) and conidial germination (94-100%). This study provides basic information for the effective management of apple bitter rot.

Cell Growth Inhibitory Effect of Tissue Cultured Root of Wild Panax ginseng C.A. Mayer Extract on Various Cancer Cell Lines

  • This study was performed to investigate the cell growth inhibitory effect of tissue cultured root of wild Panax ginseng C.A. Mayer (tcwPG). The human stomach carcinoma cell line, MKN 74, was incubated with 70% EtOH extract of tcwPG or Panax ginseng C.A. Mayer (PG) for 24 hrs. tcwPG inhibited cell growth at a concentration of $250{\mu}g/ml$. However, Panax ginseng extract did not inhibit cell growth at the same concentration. We also tested the ethyl acetate and $H_2O$ fractions of tcwPG. The inhibitory effect of the ethyl acetate fraction on cell proliferation in MKN 74 cells was more potent than that of the crude extract, and the inhibitory effect of the $H_2O$ fraction was less than that of the ethyl acetate fraction. When we separated tcwPG into polar and non-polar saponin fractions and then measured cell growth inhibition, the non-polar saponin in tcwPG exhibited cytotoxicity. To compare the effects of tcwPG on various cancer cell lines, we measured cytotoxicity in MKN 74 (stomach cancer cell line), SW 620 (colon cancer cell line) and PC 3 (prostate cancer cell line). All three cell lines showed cell growth inhibition, and the cell growth inhibitory effects were not quite different in the various cell lines. The non-polar saponins of tcwPG arrested PC 3 cells at G1-phase as did Panax ginseng.

Anticancer Effect of Doenjang in in vitro Sulforhodamine B (SRB) Assay (된장의 in vitro Sulforhodamine B (SRB) Assay에 의한 암세포 증식 억제 효과)

  • Growth inhibitory effect of doenjang(Korean soypaste) methanol extracts in SRB assay using AGS human gastric adenocarcinoma cell, Hep 3B human hepatocellular carcinoma cell and HT 29 human colon cancer cell was studied. The treatment of doenjang methanol extracts(2mg/assay) to the AGS, Hep 3B and HT 29 cancer cells inhibited the growth of the cancer cells by 55%, 60%, and 71%, respectively. Doenjang methanol extracts exhibited the highest inhibitory effect among other soybean fermented foods and original materials in the SRB assay. In addition, to separate active compounds of doenjang methanol extracts, we fractionated the doenjang with hexane, methanol, dichloromethane, ethylacetate and butanol. Growth inhibitory effect on the AGS, Hep 3B, HT 29 and MG 63 cancer cells was the highest in the fractions of dichloromethane and ethylacetate among other solvent fractions of the doenjang. These results showed that some compounds contained in the fractions of dichloromethane and ethylacetate might play a role on the anticanceric effect of doenjang.

Allelopathic Effect of Ganghwa mugwort (Artemisia spp.) on Seed Germination and Seedling Growth of Plants (강화약쑥 추출물이 종자발아 및 유식물 생장에 미치는 알레로파시 효과)

  • This study was conducted to identify allelopathic effect of Ganghwa domestic Artemisia spp., named Sajabalssuk and Ssajuarissuk, for various receptor plants including clover (Trifolium repens L.), alfalfa (Medicago sativa L.), lawn grass (Zoysia japonica Steud.), dandelion (Taraxacum platycarpum Dahlst.), and dahurianpatrinia (Patrinia scabiosaefolia Fisch. ex Trevir). Receptor plants were treated with the aqueous and essential oil extract of Artemisia plants. In consequence, their allelopathic effects were evaluated by measuring seed germination rates, seedling growth, and dry weights of the receptor plants. The seed germination and seedling growth of the receptor plants were inhibited by all treatments of both aqueous and essential oil extracts of the Artemisia plants, and, in addition, the inhibitory effects were increased according to the higher concentration. Among the donor plants, A. $sp.^*III$ showed most effective allelopathic effect. Comparing the alleopathic effect among the receptor plants, seed germination was most inhibited in lawn grass while inhibitory effect of seedling growth was comparatively higher in dandelion. Although inhibitory effects were comparatively lower, the allelopathic effects of Artemisia plants were identified in clover and alfalfa since the seedling growth of these plants were inhibited more than 70%. Thus, in result, Ganghwa domestic Artemisia spp. could be possibly used for weed control since natural products of the plants showed inhibitory effects on seed germination and seedling growth of various receptor plants.

EFFECT OF POLYPHOSPHATE ON THE GROWTH OF ORAL BACTERIUM, PREVOTELLA INTERMEDIA (구강세균 Prevotella intermedia의 성장에 따른 polyphosphate의 영향에 관한 연구)

  • Prevotella intermedia has been known as one of the important bacterial species involved in the endodontic infections and various periodontal diseases. Polyphosphate has been widely used to prevent decomposition of food and known to have an inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of poly phosphate on the growth of Prevotella intermedia, a gram negative bacterium. Prevotella intermedia G8GK3(ATCC 49046) was grown in the presence of polyphosphates with different chain lengths. Inhibitory effect of each polyphosphate, which was added at the beginning or at the early exponential growth phase of Prevotella intermedia, was determined by measuring optical density of the bacterial cells at 540nm, viable cells and lysis of Prevotella intermedia. The results from this study were as follows : 1. Poly phosphate inhibited the growth of Prevotella intermedia. 2. The minimum inhibitory concentration(MIC) of poly phosphate appeared to be 0.05%. 3. Polyphosphates with chain lengths of 5 and 65 demonstrated the greatest inhibitory effect on the growth of Prevotella intermedia. 4. Polyphosphate was bactericidal to Prevotella intermedia, demonstrating the growth inhibition of the bacterium. 5. Polyphosphate induced lysis of Prevotella intermedia. The overall results suggest that polyphosphate has a bactericidal effect on Prevotella intermedia, causing the lysis of the bacterium.

  • Polyphosphate has been used to prevent decomposition of foods and has been shown to have inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of polyphosphate on the growth of Porphyromonas endodontalis, a gram negative endodontopathic bacterium. Porphyromonas endodontalis ATCC 35406 was grown in the presence of polyphosphates with different chain lengths. Inhibitory effect of each polyphosphate which was added at the beginning or during the culture, was determined by measuring the optical density of the bacterial cell at 540nm and by viable cell count. The results from this study were as follows : 1. Polyphosphates were shown the growth inhibition of the Porphyromonas endodontalis. 2. The minimal inhibitory concentration(MIC) of polyphosphate was observed to be 0.04%. 3. Polyphosphates with chain lengths of 25 and 75 demonstrated the greatest inhibitory effect on the growth of Porphyromonas endodontalis. 4. Polyphosphates are bactericidal to Porphyromonas endodontalis, demonstrating the growth inhibition of the bacterium. The overall results suggest that use of polyphosphate may affect the growth of Porphyromonas endodontalis. Further studies will be needed to confirm the effect of, polyphosphate.

Growth Inhibitory Effects of Chloride Salts and Organic Acid Salts Against Food-Borne Microorganisms (Chloride염 및 유기산 칼슘염의 식중독 미생물에 대한 증식 억제 효과)

  • The growth inhibitory effects of chloride salts and organic acid salts against six food-borne microorganisms (Bacillus cereus ATCC 11778, Escherichia coli O157:H7 ATCC 43894, Listeria monocytogenes ATCC 19111, Salmonella Typhimurium ATCC 14028, Staphylococcus aureus ATCC 25923, Vibrio parahaemolyticus ATCC 17802) were determined using Bioscreen C in broth medium. The growth inhibitory concentrations of sodium chloride and potassium chloride on B. cereus were 7 and 9%, respectively. E. coli O157:H7 and S. aureus were inhibited by treatment of 3% calcium chloride. Magnesium chloride showed growth inhibitory effect on B. cereus, S. Typhimurium, and S. aureus at 5%. The order of growth inhibition effects by organic acid salts was calcium propionate>calcium acetate>calcium lactate. Calcium chloride (3%) with 0.01% lactic acid showed strong inhibition on the growth of S. Typhimurium and exhibited stronger growth inhibition than calcium chloride alone (5%). We concluded that calcium chloride and calcium propionate had strong growth inhibitory activities and that calcium chloride and sodium chloride in combination with lactic acid had stronger inhibitory activities than that of chloride salts alone.

Chemical Components and Biological Activities of Red Onion Powder (붉은 양파 분말의 화학성분 및 생리활성)

  • We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.
