Optimization of Regeneration Condition Under Agrobacterium-mediated Transformation in in vitro Cultured Korean Soybean

  • Soybean is the important crop in Asian countries as protein source, oil production and animal feed. Improving soybean using genetic transformation is the principal tool in nowadays. Developing herbicide resistant transgenic soybean plants through Agrobacterium-mediated transformation has been worked in many previous studied. However, the transformation efficiency is still low. Many attempts try to find the optimum media condition for plant regeneration after infection. After transformation, the plant regeneration is very important condition to promote growth of transgenic plant. In this study, we optimized a regeneration condition for two Korean soybean cultivar, Dawonkong and Pungsannamulkong using cotyledon, cotyledonary nodes and hypocotyl as explant. The results showed that shoot regeneration of cotyledonary nodes on B5 medium containing 2 mg/L 6-benzylaminopurine showed the highest percentage of regeneration in Dawonkong (75.8%) while Pungsannamulkong presented high number of shoots 2.12 shoots per explant. For transformation condition, co-cultivation in 7 days showed a high number of GUS positive expression. Most of explants can survived under media including 5 mg/L of glufocinate which refers phosphinotricin for 2-week selection. Washing with 400 mg/L of cefotaxime in several times and selection in plant regeneration media with 400 mg/L of cefotaxime can prevent bacteria growth, effectively.

Comparison of Gold Biosensor Combined with Light Microscope Imaging System with ELISA for Detecting Salmonella in Chicken after Exposure to Simulated Chilling Condition

  • In this study, the performance of a gold biosensor combined with light microscope imaging system (GB-LMIS) was comparatively evaluated against enzyme-linked immunosorbent assay (ELISA) for detecting Salmonella under simulated chilling condition. The optimum concentration of antiSalmonella polyclonal antibodies (pAbs) was determined to be 12.5 and 100 ㎍/ml for ELISA and GBLMIS, respectively. GB-LMIS exhibited a sufficient and competitive specificity toward three tested Salmonella among only. To mimic a real-world situation, chicken was inoculated with Salmonella cocktail and stored under chilling condition for 48 h. The overall growth of Salmonella under chilling condition was significantly lower than that under non-exposure to the chilling condition (p < 0.05). No significant differences in bacterial growth were observed between brain heart infusion and brilliant green broth during the enrichment period (p > 0.05). Finally, both GB-LMIS and ELISA were employed to detect Salmonella at every 2-h interval. GB-LMIS detected Salmonella with a competitive specificity by the direct observation of bacteria on the sensor using a charge-coupled device camera within a detection time of ~2.5 h. GB-LMIS is a feasible, novel, and rapid method for detecting Salmonella in poultry facilities.

  • Czochralski법에 의한 ZnWO₄단결정을 [100], [101], [001] 방향으로 성공적으로 성장시켰다. 각 축 방향에 따른 성장조건이 rotation speed, pulling rate, 성장된 결정의 직경 등의 변수를 가지고 조사되어졌다. 성장된 결정의 냉각시 발생되는 균열을 annealing 효과에 의하여 방지할 수 있었다. 성장된 결정의 방위는 Laue back reflection으로 결정하였다. 각 축 방향으로 성장된 결정의 미세구조적 특징이 논하여졌으며, 경도, 열팽창계수 및 유전상수의 물리적 특성이 평가되어졌다.

  • In this paper, we show the existence of solution of the neutral stochastic functional differential equations under non-Lipschitz condition, a weakened linear growth condition and a contractive condition. Furthermore, in order to obtain the existence of solution to the equation we used the Picard sequence.

Distribution and Habitat Condition of Crypsinus hastatus in Buyeogun (부여군 고란초의 생육지 환경조건과 분포)

  • Background : This text was conducted to evaluate the distribution, shape characteristics and habitat condition of Crypsinus hastatus in Buyeo Korea. Methods and Results : In Buyeo, Crypsinus hastatus was distributed at an altitude of 31.2 m, a slope of 82.5% and a distance from river water of 27.2 m. The leaf blade was oblong, 2.7 cm long, 1.2 cm wide with a petiole length of 2.3 cm. The total population comprised 2,100 individuals, with the highest proportion being individuals with a leaf blade of length 0.5 - 1 cm. The temperature and light intensity of the growth point were lower, but the humidity higher, than that of growth around. The chemical properties of the soil were as follows: pH 4.42, organic matter 77.99 g/kg, electrical conductivity 0.50 dS/m, nitrate nitrogen 20.50 mg/kg and available phosphate 165.7 mg/kg. Exchangeable cation content of potassium, magnesium, sodium and calcium was 0.35, 0.70, 0.09 and $2.04cmol^+/kg$, respectively. Conclusions : In Buyeo, Crypsinus hastatus was distributed in $5.3m^2$ area of Nakhwaam, Naeseongri and Jeongamri. However, the population of the medicinal resources seemed to decrease owing to the poor environmental conditions of the habitat.

STudies on the Microbial Pigment(I) (미생물의 색소에 관한 연구. 제1보)

  • The bacteria of red colonies isolated from soil were identified as Serratia marcescens. The best solvent for pigment extraction was n-buthanol and the pigment was identified as prodigiosene. The extracted pigment was stable on temperature and light but not on acidity. The redpigment color changed into red in alkaline solution. The maximum absorbancy of pigment was 466 nm in alkaline condition and 540 nm in acid condition. And the pigment formed single spot on the TLC(starch). By the result of infra red spectrum, the red pigment has the same absorption pattern comparing with, the prodigisin produced by S. marcescens strain Nima. It was confirmed that the pigment was secondary metabolite and that the maximal peak of production appeared at 30 hrs after the inoculation, when the bacterial growth was in statinary state. Referring to the effect of temperature, the pigment was not formed at $36^{\circ}C$ and the optimal temperature for both of bactrial growth and pigmentation was $30^{\circ}C$. The optimal range of pH for pigmentation was 5.0 and under the condition the bacterial growth was not affected at all. Examining the effects of light, the bacterial pigment ation was more increased in darkness than in visible light.

  • The effect of solidification condition and vibration on structure refinement was investigatedfor unidirectionally solidified $AI-CuAI_{2}$ eutectic composites. Eutectic composites were unidirectionally solidifiedunder vibration with variation of growth rates (R) and thermal gradient((G), $32^{\circ}C$/cm, and 35$^{\circ}C$/cm. A Study on the Mixed-Seeding Rates of Exotic Grasses for Slope Revegetation Measures (사면(斜面) 녹화용(綠化用) 외래초종(外來草種)의 혼파조합(混播組合)에 관(關)한 연구(硏究))

  • This study was conducted to find out the growth characteristic of mixed-seeding in the grasses for slope revegetation measures. Seeding treatment with six combination types of five grasses used for this experiment such as orchardgrass(Dactylis glomerata 'Potomac'), perennial ryegrass(Lolium perenne 'Manhattan'), tall fescue(Festuca arundinacea 'Jaguar'), kentucky bluegrass(Poa pratensis 'Majestic') and weeping lovegrass(Eragrostis curvula). The growth of orchardgrass, perennial ryegrass and tall fescue were good on mixed-seeding condition in case of number of individuals, heights and coverages at this experiment. These plants will useful for revegetation measures mixed-seeding, but weeping love grass and kentucky bluegrass did not show a desirable growth in mixed-seeding condition, and these plant prefered mixed seeding to single seeding. Regeneration percents of orchardgrass, perennial ryegrass and tall fescue were higher than that of weeping love grass and kentucky bluegrass, and coverage kept on good condition through one year after seeding, therefore, these plants will effective for initial stage revegetation measures.

Condition of the most suitable inoculation and manufacture of spawn of ear mushroom (목이재배를 위한 배지선발 및 최적 접종조건)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of ear mushroom(Auricularia auricula). Sawdust spawn of media composition for optimal growth were found to be quercus sawdust combination of 20% rice bran and poplar sawdust combination of 10% wheat bran were the best of the optimal combination. The optimal condition of the most suitable inoculation amount showed that quercus spawn and poplar spawn were 15g, 25g, respectively.

  • This review provides an overview of dietary nucleotides as an alternative to in-feed antibiotics for weaning pigs. Dietary nucleotides are composed of DNA or RNA molecules and are normally contained in protein-rich feed ingredient, brewer's yeast, yeast extract, and milk. Weaning pigs are suffering from several stresses, such as environmental challenges (i.e. crowding, transportation, and feeding). Such stressors can damage the intestinal epithelium and cause an invasion by Escherichia coli, secondary inflammatory responses, and post weaning diarrhea. To overcome weaning disorder, people often use antibiotics which reduce symptoms and boost growth performance. However, since antibiotics were banned due to concerns of antibiotic resistant bacteria, researchers are studying alternative materials to antibiotics. Dietary nucleotides are one of the alternative materials for replacing antibiotics and can be used in abnormal conditions, such as weaning diarrhea, low digestibility, and disease condition. Nucleotides have substances that have important roles in cell division and cell growth, affecting growth performance, intestinal condition, and immunological effect at the weaning stage. However, nucleotides' composition is very different between sources and this aspect makes it difficult to utilize nucleotides at the weaning stage. Therefore, this review paper focuses on i) the characteristics and functions of dietary nucleotides and ii) the effect of dietary nucleotides on the growth performance and immune system of pigs.
