Study on germination rate enhancement and seedling raising-method development on Sorbus commixta Hedl. (마가목 종자(種子)의 발아율(發芽率) 향상(向上) 및 육묘기술(育苗技術))

  • This study was conducted to clarify on germination rate enhancement effect and to develope the growing seedling method on Sorbus commixta Hedl. The results were follows; The optimum temperature of stratification and germination of mountain ash was $5^{\circ}C$ the seed germination rate in wet stratification with sand at $5^{\circ}C$ for 90 days was 22.6% and the highest germination of 78.9% was observed when seed was soaked in BA 200ppm for 20 minute after stratification for 90 days at $5^{\circ}C$. The highest germination of large chinese hawthorn was 11.1% when seed was soaked BA 50ppm for 20 minute after stratification 90 days at $5^{\circ}C$. Emergence ratio in box raising seedling and pot seedling showed high, 93 and 95%. Root length and dry root weight in pot seedling for 50 days showed higher than that of direct seedling and box raising seedling, Plant height and dry leaf weight in box raising seedling showed higher than that of pot seedling and direct seedling. The growth increment of seedling was high between 40 days and 50 days after seedling. The excellent seedling of mountain ash was produced in pot seedling for 50 days.

  • This study was carried out to investigate propagation characteristics of seedling and rhizome in Epimedium koreanum NAKAI. Seed germination essentially needed after-ripening duration after harvesting for 250days and more by stratification. At $20^{\circ}C$ seed emergence days and root length were 8 and 0.46cm respectively. The above-ground part of E. koreanum grew until 60days after root cutting and the underground part of it grew after that time. The optimum amount of organic fertilizer was fermented cow-manure with husk 2,000kg/10a that showed $57.5cm^2$ in leaf area per plant, 6.2cm in new rhizome length, and 0.43% in icariin content, an index component of E. koreanum, while inorganic N-fertilizer made E. koreanum worse as it increased. In nutriculture extruded rice hull media was better for growth than perlite media at 1/2 NHRI solution.

  • Korean ginseng(Panax gmseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In environment stresses, soil condition is the most important factor, among which nutrients, especially inorganic materials such as N, P, K, Ca, Mg, Fe, etc., influence greatly on the ginseng growth. However, present ginseng field soils in Korea contain so much amount of such inorganic materials that a variety of remarkable disorders were noted in many ginseng plantations, resulting in decrease of qualitative ginseng production. Therefore, it is required to search for genetic resources and genes tolerant to salt stress for the development of ginseng cultivars. Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. Murashige & Skoog(MS) media with 2.5 folds of $KNO_3,\;NH_4NO_3,\;MgSO_4\;7H_2O,\;KH_2PO_4,\;and\;CaCl_2\;2H_2O$ was established for the selection of ginseng lines tolerant to salt stress under the embryo culture.

Growth Characteristics and Yield of the Three Species of Genus Angelica (당귀의 종별 생육특성 및 생산성 비교)

  • In genus Angelica, three species have been used and cultivated for medicinal material in orient, A. gigas in Korea, acutiloba in Japan and sinensis in China. A. gigas (Chamdangui) is different from A. acutiloba or A. sinensis in the contents chemical components and their efficacious. In Korea, in particular, A. acutiloba has been cultivated in a small acreage from several decades ago, and recently A. sinensis has been trying to cultivate. A trial of adaptation and cultivation of the three species was performed in different altitudinal regions such as Suwon (50 m of sea level), Jinbu (500 m of sea level) and Taebak (700 m of sea level). A. gigas and acutiloba grew and produced roots successfully at all locations, but A. sinensis could grow only at high altitudinal region, Taebaek. A. sinensis survived 70.3% at Taebaek, but only 45.2% at Jinbu and 3.6% at Suwon. The dry root yield of A. sinensis was as low as 1,600 kg/ha at Taebaek and 1,270 kg/ha at Jinbu comparing farmers' average root yield of A. gigas, 3,000 kg/ha however, no root could be harvested at Suwon. The results, showed it was that A. sinensis could not grow at low altitudinal plane area except high altitude area in Korea. Even though it could grow, the root yield was too low to support the income.

  • Fertilization response on production and quality of flue-cured tobacco as to different level of nitrogen, phosphorus, and potassium were studied in a field experiment. The results were summarized as follows: 1. Growth and yield of flue-cured tobacco were significantly better in higher nitrogen fertilization levels, regardless of soil fertility, but the negative correlation was recognized between the quality of leaves and the amounts of nitrogen application. While, both fertilizers of phosphorous and potassium did not have should little effect on the tobacco yield and quality. 2. The optimum ratio of N, P, and K fertilizer applications were decided by the appearance of the proper yield and the best quality of tobacco leaves. The proportion of N:P:K was 2:1:4. 3. The single effect (Complete plot minus Non-fertilized plot) of N, P, and K on yield and quality of cured leaf was greatly affected by nitrogen, but the combined effect (Nutrient deficiency plot minus Non-fertilzier plot) of that were only slightly affected by P and K.

A Case of Neonatal Intrahepatic Cholestasis Caused by Citrin Deficiency (NICCD) Confirmed by SLC25A13 Mutation

  • Citrullinemia type 2 (citrin deficiency) is an autosomal recessive inborn error metabolism, caused by the SLC25A13 gene mutation. Citrin deficiency is associated with two clinical phenotype; neonatal-onset type II citrullinemia (CTLN2), also known as neonatal intraphepatic cholestasis caused by citrin deficiency (NICCD) and adult-onset CTLN2. Clinical manifestations of NICCD include poor growth, intrahepatic cholestasis, liver dysfunction and increased plasma citrulline, methionine, threonine, arginine. The molecular diagnosis could be confirmed by SLC25A13 gene mutation analysis. A 3-month-old male infant with persistent jaundice was referred for evaluation. Newborn screening was normal at birth. Mild elevation of serum ammonia and AST/ALT were observed. Plasma amino acid analysis showed significantly elevated citrulline, methionine, threonine. DNA sequence analysis of the SLC25A13 gene revealed two compound heterozygous mutations, c.[852_855del]($p.Met285Profs^*2$) and [1180+1G>A]. We suggest that NICCD should be considered as one of the cause of in infants with cholestatic jaundice, although the newborn screening was normal.

  • Carbamoyl phosphate synthetase 1 deficiency (CPS1D) is a rare autosomal recessive urea cycle disorder characterized by hyperammonemia. CPS1D is caused by mutations in the CPS1 gene on chromosome 2q35. Based on the age of onset, there are two phenotypes: the neonatal type and the delayed-onset type. The severity of clinical manifestation depends on the degree of CPS1 residual enzymatic activity, and can result in hyperammonemia and neurological dysfunction. We report a case of CPS1D in a neonate who developed vomiting, decreased consciousness and hyperammonemia at 25th day after birth. She showed excellent response to treatment including hydration, ammonia-lowering drugs and a low-protein diet without hemodialysis. Her growth, development and neurological outcomes were fair at the last follow-up at 17 months of age.

  • 'Nongjin-go' is a new breed strain of Lentinula edodes, saw-dust bag variety. It is a cross combination of dikaryon Lentinula edodes ASI 3305mut and monokaryon L5-16 of L. edodes ASI 3305(Sanjo701ho). We crossbred them by 2011 and verified productive capacity from 2012 to 2013 in Rural Development Administration. Optimum temperature of mycelial growth is $30^{\circ}C$ and it of fruit-body primodium formation is range from 15 to $23^{\circ}C$. Nongjigo is agricultured at mid-high temperature well. Fruiting body is platy-hemisphere, light brown and centralizing. And bast is formed around edge of pileus. Yield productions per period is regular than 'Sanjo701'. Plastic bag culture medium is 1.5 kilogram and culture periods are 90~100 days. As its browning of pileus in culture is a little slow, Light and ventilation is needed a lot in light-culturing. Humidity is controlled properly for its color in fruit-body growing. Tested culture medium is consisted of 80% Oak-Tree saw-dust and 20% rice-bran.

Stain improvement in the white button mushroom 'Seolgang' and its varietal characteristics in Agaricus bisporus

  • The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, 'Seolgang' was developed by crossing two monokaryons 'CM020913-27' and 'SSU423-31'. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of 'Seolgang' on CDA was better at $20^{\circ}C$ and $25^{\circ}C$ when it was compared with that of '505 Ho'. The mature cap shape of new strain 'Seolgang' is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain '505 Ho', the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of '505 Ho'. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of '505 Ho'. Genetic analysis of the new strain 'Seolgang' showed different profiles compared to '505 Ho', CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.

Medium development of Flammulina velutipes by using herb medicine refuse (폐 한방슬러지를 이용한 팽이버섯의 배지개발)

  • pH of oriental medicine sludge was 5.3, which was similar to 5.2 of the main ingredient, corncob. Its sugar content, however, was 4.8 mg/g, which was 2.5 times higher than concorb's 1.9 mg/g. According to the addition content analysis of oriental medicine sludge by using blood agar plate, the experimental group showed much more robust growth than the control group. 10% of oriental medicine sludge was added to corncob and pine tree sawdust for test-tube culture. Then they were cultivated at $25^{\circ}C$ for 6 days after inoculating Flammulina velutipes liquid spawn. The control group and experimental group showed 2.2~3.4 and 5.8~6.4 cm hypae growths respectively. At the field test for 10% herbal medicine refuse, mushroom yield dropped by 5% compared to the control group. However, it had distinctively lower number of deformity and the 2nd grade products. An economic analysis was conducted based on the cultivation facility that produces 160,000 mushrooms per day. The analysis demonstrated that the facility can save 50,000,000 won in the starting year and 130,000,000 won in the following years from the unit cost of production excluding labour and operation cost.
