Asian Ladybird, Harmonia axyridis, as a Biological Control Agent: Control Effects of Aphid Populations in the Greenhouses at Different Seasons (생물적 방제 인자로서의 무당벌레(Harmonia axyridis): 하우스에서 계절에 따른 진딧물 방제효과)

  • Application of the Asian ladybird (Harmonia axyridis) to control several species of aphids in the plastic green houses in mind, control effects of aphid populations regulated by the Asian ladybird were observed. The green peach aphid, the turnip aphid, and the cotton aphid were present on mustard plants, Angelia utlis, ornamental kales, and egg plants at greenhouses in spring, summer, and winter. Adults and larvae of the Asian ladybird used in experiments were collected from aggregated sites at Taejon in the autumn and reared on the cotton aphid in the laboratory. In winter, more number of adults and larvae of ladybirds than in other seasons were needed to control aphid population in successively double plastic greenhouses with supplied subterranean water for keeping warmth. In spring and summer, it was possible to keep the aphid populations low when necessary by manipulating ladybird populations according to the density of aphids. On the other hand, the innate increasing rate of aphid, the aphid population density at the time of applying ladybird, the predacious ability of ladybird at specific developmental stages, and needed periods should be taken into account to control aphids. In addition, the environmental factors, for example, optimum temperature and humidity should be considered to be biologically effective when ladybirds are released to greenhouses.

Identification of an Entomopathogenic Fungus, Beauveria bassiana SFB-205 toxic to the Green Peach Aphid, Myzus persicae

  • To select entomopathogenic fungi controlling aphids effectively, several isolates were screened against second instars of Myzus persicae nymphs in the glasshouse using conidia suspension at $1.0{\times}10^5$ conidia/ml. Among these isolates, SFB-205 conidia showed the highest insecticidal activity about 32.7% efficacy to M. persicae at 4 days after application in the glasshouse. The attachment of SFB-205 conidia on the surface of M. persicae nymphs, and germination and penetration were observed using scanning electron microscopy. SFB-205 was identified as Beauveria bassiana species through the comparison of 5.8 s rRNA genes. There were 24 polymorphisms between SFB-205 and the previously reported isolate, B. bassiana ATCC74040 using six kinds of primer combinations in amplified fragment length polymorphism (AFLP) analysis. The B. bassiana SFB-205 might be used as a practical biological control agent for the green peach aphid, M. persicae in the field.

Biological Characteristics of the Aphid-eating Gall-midge, Aphidoletes aphidimyza (Diptera: Cecidomyiidae) as a Biological Control Agents of Aphids (진딧물천적인 진디혹파리(Aphidoletes aphidimyza)의 생물학적 특성)

  • The aphid-eating gall-midge, Aphidoletes aphidimyza (Rondani), as predator of aphids was wildly used as one of the biological control agents for control of several kinds of aphids. Their some kinds of biological characteristics were examined. The developmental periods of A. aphidimyza from egg to adult with the green peach aphid as prey were 40 and 12 days at 15$^{\circ}C$ and 30$^{\circ}C$, respectively. Supplying the cotton aphid as prey, their developmental periods were shorter than with the green peach aphid. In case of the preservation of pupa in the cold condition, emergence rates were over 90% for 1 and 2 weeks at 5$^{\circ}C$ and l0$^{\circ}C$, respectively. Adult females of A. aphidimyza began mating and laying eggs at 2-3 days after emergence, and they laid about 200 eggs for lo days of average life span. Most of adults were emerged from pupa at 6 to 8 pm during a day, and they mainly acted in the early night.

Biological Control of Aphid Using Fungal Culture and Culture Filtrates of Beauveria bassiana

  • Aphids are one of the most destructive pests in crop production such as pepper, cucumber, and eggplants. The importance of entomopathogenic fungi as alternative pest control agents is increasing. Conidia of entomopathogenic fungi are influenced by environmental conditions, such as temperature and relative humidity, and cause slow and fluctuating mortality. These factors have prevented wider application and use of biocontrol agents. For investigation of means of mitigation of such problems, we conducted bioassays with 47 fungal culture filtrates in order to evaluate the potential of secondary metabolites produced by entomopathogenic fungi for use in aphid control. Among 47 culture filtrates cultured potato dextrose broth, filtrate of Beauveria bassiana Bb08 showed the highest mortality (78%) against green peach aphid three days after treatments. Filtrate of Bb08 cultured in Adamek's medium showed higher toxicity as 100% to third instar nymphs of the aphid compared with seven other filtrates cultured in different broths amended with colloidal chitin or oil. The culture filtrates and fungal cultures from media amended with colloidal chitin or oil had lower control efficacies than filtrates without these additives in three different media. These results indicate that the fungal culture fluid or culture filtrate of B. bassiana Bb08 cultured in Adamek's medium has potential for development as a mycopesticide for aphid control.

Resistance development and cross-resistance of green peach aphid, Myzus persicae (Homoptera : Aphididae), to imidacloprid (Imidacloprid에 대한 복숭아혹진딧물의 저항성 발달 및 교차저항성)

  • Studies on the resistance monitoring of green peach ahpid, Myzus persicae, its development pattern by artificial selection with imidacloprid and cross-resistance were carried out to develope resistance management strategy. Resistance ratios of M. persicae collected at Hwachon and Dunnae among 5 locations in alpine cultivation area appeared to be high as 37.2 and 16.5, respectively. Resistance of aphid to imidacloprid developed slowly up to 20 time selection, and after that it grew quickly. Imidacloprid-resistant aphid strain showed low cross-resistance ratios(<10) to most of organophosphates, carbamates, and mixed insecticides except pirimicarb(487.8), but high ratios to acetamiprid(143.0) which is one of the neonicotinoids like imidacloprid, and pyrethroids such as deltamethrin(14.9), flucythrinate(12.9) and halothrin(15.9).

Effect of Temperature at Different Performance Stages of Aphelinus varipes (Hym. Aphelinidae) Parasitizing the Green Peach Aphid, Myzus persicae (복숭아혹진딧물에 기생하는 목화면충좀벌의 발육단계별 온도영향)

  • Developmental period, mummification, pupal mortality, and sex ratio of Korean strain of Aphelinus varipes F$\ddot{o}$rster parasitizing the green peach aphid, Myzus persicae were studied at 20, 25, 30, and $35^{\circ}C$ in controlled climate cabinets. The plastic container with a leaf disc of sweet pepper (Capsicum annuum L.) was used as experimental units. Green peach aphids at different developmental stages were reared in four different temperatures and presented to A. varipes for 12 hours. A female adult of A. varipes was allowed to parasitize for 12 hours in each plastic petri dish. The periods from egg to adult of A. varipes were 18.3 days, 14.7 days, 10.4 days, and 9.3 days at $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$, respectively. The pupal mortalities of A. varipes were 0%, 1.5%, 2.8%, and 10.2% at $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$, respectively. The mummifications of A. varipes were significantly different in all developmental stages of the aphid. The mummification of A. varipes parasitizing green peach aphid was the highest at 1st and 2nd in $20^{\circ}C$ and $25^{\circ}C$, but the highest at 3rd (84.4-85.0%) in $30^{\circ}C$ and $35^{\circ}C$. The host feedings of A. varipes were $20^{\circ}C$ (12.5%), $25^{\circ}C$ (17.3%), $30^{\circ}C$ (16%), and $35^{\circ}C$ (10.9%), respectively. The sex ratios of A. varipes female were $20^{\circ}C$ (49%), $25^{\circ}C$ (46%), $30^{\circ}C$ (69%), and $35^{\circ}C$ (60%), respectively.

Reuse of Yuza Seed By-product for Production of Aphicidal Rhamnolipid by Pseudomonas sp. EP-3 (Pseudomonas sp. EP-3 rhamnolipid 에 의한 진딧물 살충성 생산을 위한 유자씨앗 부산물의 재활용)

  • BACKGROUND: Yuza seed by-product has been produced in a large amount from the agricultural farms in the southern area of Korea. It has been mostly abandoned after commercial process for the production of juice, jam and tea. The study on the reuse of the yuza seed by-product has received much attention as a bio-resource material for the production of active compound in agriculture. METHODS AND RESULTS: Insecticidal rhamnolipid-producing Pseudomonas sp. EP-3 was grown in mineral salt media with the yuza seed by-product at 2, 20, 50 and 100 g/L. The growth of EP-3 was accompanied by a increase in insecticidal activity against green peach aphid. The highest insecticidal activity was observed when EP-3 was grown in the medium containing 50 g/L of the seed sample, producing approximately 996 mg/L of rhamnolipid at 96 h. Palmitic acid, stearic acid, oleic acid and linoleic acid were determined as the major fatty acids of the seed sample. The EP-3 cultures grown on the fatty acid mixture extracted from the seed sample showed a aphid mortality similar to that of cultures grown on the seed sample. The EP-3 cultures grown on 50 g/L of the seed sample showed aphid mortality more than 90% under greenhouse conditions. CONCLUSION: This study suggested that the yuza seed by-product may be used as a renewable material for microbial production of rhamnolipid against green peach aphid.

Relation of Aphicidal Activity with Cuticular Degradation by Beauveria bassiana SFB-205 Supernatant Incorporated with Polyoxyethylene-(3)-Isotridecyl Ether

  • The application of Beauveria bassiana SFB-205 supernatant incorporated with polyoxyethylene-(3)-isotridecyl ether (TDE-3) significantly reduced the population of two species of aphids including cotton aphid, Aphis gossypii, and green peach aphid, Myzus persicae, much higher in cotton aphid, compared with supernatant incorporated with Tween 80, which allows the relationship of aphicidal activity with the degradation of aphid cuticles to be determined. Overall, the degradation of the cuticles induced by the supernatant was more remarkable in conjunction with TDE-3 than Tween 80, and this phenomenon was more observable in cotton aphid through SDS-polyacrylamide gel electrophoresis, revealing high correlation with their aphicidal activities.

Effects of Polyethylene Net Barrier on the Control of Aphid - borne Potato Virus Y in Tobacco (폴리에칠렌 차단망 설치에 의한 연초의 진딧물 전염 PVY 방제효과)

  • Potato virus Y (PVY) is an important viral disease in burley tobacco in Korea and is mostly transmitted by green peach aphid, Myzus penicae, in nature. Effects of barrier nets on the immigration of the aphid population into tobacco fields and on the control of PVY were investigated in 3 tobacco fields with 1.8 m - height polyethylene (PE) nets on their outer - sides in Iseo - Myeon, Wanju - Kun, Cheonbuk. Immigration of aphids to the tobacco yields started at late April and reached maximum at early June. The immigrating aphid population was apparently blocked by the barrier nets to be reduced by 54-73%. PVY severity was also reduced by the barrier nets. However, the control value was variable, ranging 24-67%, which suggests that effect of the PE net barrier on the prevention of aphid-borne virus might be variable depending on the location and slope of the fields.

First Record of the Entomopathogenic Fungus Zoophthora radicans on the Green Peach Aphid Myzus persicae in Korea (복숭아 혹진딧물에서 발견된 국내 미기록 곤층병원 사상균 Zoophthora radicans에 관한 보고)

  • An entomopathogenic fungus, Zoophthora radicans, was found causing epizootics in populations of the green peach aphid, Myzus persicae, on March 27, 1998 at Seongju Gun, Kyungbuk Province in Korea. Anamorhic characteristics of the fungus were described using light and scanning electron microscopy. Distinguishing characteristics of Z. radicans is formation of capilliconidia on a single slender secondary conidiophore.
