녹비작물 재배가 유기포도 수량 및 안토시아닌 함량에 미치는 영향 (Effect of Green Manure Cultivations on Yield and Anthocyanin Content in Organic Grapevine)

  • In sustainable viticulture, green manure represents a safe and non-polluting way to bring large quantities of organic matter into the soil. The cultivation of green manure crops plays an important role in soil quality and sustainability of agricultural system. This study was conducted to evaluate the effects of green manure fertilization on yield and quality of the Campbell Early grape variety in the organic vineyard. Hairy vetch was the more productive green manure crop, yielding higher dry materials. Average yields of grape were significantly greater in hairy vetch+rye ($13.02ton\;ha^{-1}$) than nature weed ($11.65ton\;ha^{-1}$), respectively. The concentrations of total as well as individual anthocyanins were consistently higher with hairy vetch treatment compared with rye and nature weed, thus making the green manure cultivation is an environmentally friendly cultivation to increase the yield and anthocyanin contents in organic grape.

Phenolic plant extracts are additive in their effects against in vitro ruminal methane and ammonia formation

  • Objective: The methane mitigating potential of various plant-based polyphenol sources is known, but effects of combinations have rarely been tested. The aim of the present study was to determine whether binary and 3-way combinations of such phenol sources affect ruminal fermentation less, similar or more intensively than separate applications. Methods: The extracts used were from Acacia mearnsii bark (acacia), Vitis vinifera (grape) seed, Camellia sinensis leaves (green tea), Uncaria gambir leaves (gambier), Vaccinium macrocarpon berries (cranberry), Fagopyrum esculentum seed (buckwheat), and Ginkgo biloba leaves (ginkgo). All extracts were tested using the Hohenheim gas test. This was done alone at 5% of dry matter (DM). Acacia was also combined with all other single extracts at 5% of DM each, and with two other phenol sources (all possible combinations) at 2.5%+2.5% of DM. Results: Methane formation was reduced by 7% to 9% by acacia, grape seed and green tea and, in addition, by most extract combinations with acacia. Grape seed and green tea alone and in combination with acacia also reduced methane proportion of total gas to the same degree. The extracts of buckwheat and gingko were poor in phenols and promoted ruminal fermentation. All treatments except green tea alone lowered ammonia concentration by up to 23%, and the binary combinations were more effective as acacia alone. With three extracts, linear effects were found with total gas and methane formation, while with ammonia and other traits linear effects were rare. Conclusion: The study identified methane and ammonia mitigating potential of various phenolic plant extracts and showed a number of additive and some non-linear effects of combinations of extracts. Further studies, especially in live animals, should concentrate on combinations of extracts from grape seed, green tea leaves Land acacia bark and determine the ideal dosages of such combinations for the purpose of methane mitigation.

청포도 즙의 첨가가 곤약젤리의 품질특성에 미치는 영향 (Effects of Adding Green Grape Juice on Quality Characteristics of Konjak Jelly)

  • This study examined the quality characteristics of jelly prepared with green grape juice (GJ). The pH, $^{\circ}Brix$ value, color, texture, and sensory evaluation of the jelly were measured. The pH of the jelly significantly decreased with increasing amount of GJ over the range of 3.25-5.27. The $^{\circ}Brix$ value of the jelly showed a significantly higher result as the amount of GJ increased (p<0.001). Lightness (L) and redness (a) decreased with increasing amount of GJ, and yellowness (b) increased. In the texture measurement, the GJ-100 sample group with a high substitute rate of GJ showed high hardness, gumminess, and chewiness (p<0.001). The results of principal component analysis (PCA) showed that the sample groups with high GJ content were classified as having relatively strong yellowness, sweet aroma, metallic aroma, grassy aroma, sweetness, sourness, green grape skin taste, and astringency. In the acceptance test, the GJ-50 sample group was evaluated to be high in flavor (p<0.001) and overall acceptance (p<0.01). However, sample groups consisting of 50% or more GJ were evaluated to be significantly strong in terms of astringency. Therefore, further study needs to be conducted about improving astringency in the future.

녹차, 메밀, 포도잎 추출물이 고콜레스테롤 급여 흰쥐의 지질대사, TBARS 및 혈액응고에 미치는 영향 (Effects of Green Tea, Buckwheat and Grape Leaves Extracts on Lipid Metabolism, Antioxidative Capacity, and Antithrombotic Activity in Rats Fed High Cholesterol Diets)

  • 본 연구에서는 Sprague Dawley 종 수컷 흰쥐에게 콜레스테롤이 1% 함유된 사료를 급여하여 고지혈증을 유발시키면서 녹차추출물, 메밀추출물, 포도잎추출물 추출물을 500 mg/kg B.W.의 농도로 4주간 매일 같은 시각에 경구투여하여 흰쥐의 혈액과 간장 내의 지질조성과 혈액응고기전 및 체내 지질과산화에 미치는 영향을 관찰하여 다음과 같은 결과를 얻었다. 전군에서 식이섭취량과 체중증가량 및 식이효율은 유의한 차이가 없어서 실험식이가 정상적인 성장에 영향을 미치지 않았다. 녹차추출물투여군은 고콜레스테롤 급여 대조군에 비해서 흰쥐의 꼬리에서의 출혈시간을 증가시켜서 항혈전효과가 있었다. 고콜레스테롤 급여로 인해 흰쥐에게 고지혈증이 유발되었으며 실험 시료를 경구투여하는데 따른 혈액중의 지질대사 개선효과는 없었다. 고콜레스테롤 급여로 흰쥐의 간장은 비대해지고 간장내 중성지방과 총콜레스테롤 함량도 현저히 증가하였으며 녹차추출물, 메밀추출물, 포도잎추출물은 간장 내 총콜레스테롤 함량을 감소시켜 간장 내 지질대사의 개선효과가 있었다. 고콜레스테롤의 급여는 혈장 내의 지질과산화물의 함량을 증가시키며 포도잎추출물은 체내 지질과산화를 억제시키는 효과가 있었으며 간에서는 메밀과 포도잎추출물이 지질과산화를 억제하였다.

포도 저장을 위한 아황산가스 발생제의 제조 및 처리 방법 (Preparation and Treatment of Sulfur Dioxide Gas Generating Agent for Storage of Grape Fruits)

  • 아황산가스 발생제를 자가 제조하여 포도의 저장 중 병해발생 방지에 이용하는 방법을 확립하기 위하여 sulfite 화합물의 아황산가스 발생 정도와 지속 기간을 측정하고 아황산가스 발생 조절 및 간이 검출 방법을 개발하였다. 포도의 저장 중 잿빛곰팡이병을 일으키는 B. cinerea의 포자 발아와 균사생장은 각각 400 ppm과 3200 ppm의 아황산가스 농도 조건에서 억제되었다. 5종류의 sulfite 및 bisulfite 화합물 중 sodium hydrosulfite는 아황산가스 발생량이 가장 높았으며 이를 비닐 포장하여 포장의 pinhole수를 조절하거나 산 또는 알카리의 pH 조절제를 혼합하면 가스 발생량과 발생 지속 기간을 조절할 수 있었다. Malachite green 용액은 아황산가스에 의해 정량적으로 탈색되어 아황산가스 검출에 이용할 수 있었으며 여과지에 흡수하여 건조할 경우 아황산가스 검출지로 활용할 수 있었다. MBA 포도를 속효성과 지효성의 아황산가스 발생제를 동시에 처리하여 저온 저장할 경우 병해 발생을 감소시킬 수 있었다.

Effective extraction of antioxidantive oligomeric proanthocyanidins from mountain grape seeds

  • The interest of oligomeric proanthocyanidins(OPCs) as therapeutic agents against diseases involving radical damage is growing. Proanthocyanidins are a class of polyphenolic compounds in several plant species and are oligomers of flavan-3-ol monomer units. Polyphenols in green and black tea, grape seeds, grapes and wine have raised much attention but mountain grape seed has not been investigated intensively up to now This study investigated the total OPCs contents and the total antioxidant activity of mountain grape seeds. Total antioxidant activity using DPPH method was employed and OPCs contents were determined by means of the UV-VIS spectrophotometer. The total OPCs yield of mountain grape seeds was about 1.45 % and total antioxidant activity was 15.8 ${\mu}g/m{\ell}$.

Effective Extraction of Oligomeric Proanthocyanidin (OPC) from Wild Grape Seeds

  • The Oligomeric proanthocyanidin (OPC) in green and black tea, grape seeds, grapes and wine has raised much attention but that OPC in wild grape seed remains to be intensively investigated. This study investigated the total OPC contents and total antioxidant activity of wild grape seeds and developed an efficient extraction process with various temperatures, solvent compositions and times. Also, a chromatography column packed with the Dia-ion HP-20 resin was used for further purification of the ope. The total OPC contents were determined with the Folin-Ciocalteu reagent, and the antioxidant activity using total antioxidant potential (TAP) and 1,1-dipheny|-2picrylhydrazy| (DPPH). The yield of final purified OPC was 1.78 (+)-catechin equivalent (CE) g/100 g, with $IC_{50}$ activities of TAP and DPPH of 31.60 and $15.70\;{\mu}\;g/mL$. These activi­ties of the final purified OPC were about two times higher than that of the BHA used as a refer­ence sample.

유기 반추동물 전용 수입산 유기사료 및 국내산 유기 부산물의 사료적 가치 평가 (Nutritional Evaluation of Imported Organic Feeds and Locally Produced Agricultural By-products for Organic Ruminant Farming)

  • This study was conducted to evaluate the nutritional value of locally produced organic agricultural by-products to substitute imported organic feeds for organic ruminant farming. Imported organic feeds (corn grain, soybean meal, soybean seed, oat grain, barley grain, wheat grain, buckwheat, sunflower seed meal) and byproducts (rice bran, grape seed meal, rice straw, soybean hull, soybean curd, rice hull, green kernel rice, and crushed rice grain) were analyzed for chemical composition and NDF, ADF, mineral, and amino acid contents and anti-nutritional factors. Dry matter, NDF and ADF contents in organic feeds were higher than those in conventional feeds. Especially, the 9.65% fat content of organic soybean meal was 6 times higher than the 1.95% fat content of conventional soybean meal. Fat contents of rice bran, grape seed meal, green kernel rice, and crushed rice grain were 25.66, 6.09, 3.57 and 1.59%, respectively. Protein contents of soybean hull and soybean curd were 14.68 and 19.87%, respectively, which are highest among organic by-products. Levels of aflatoxin in all feeds were below the safety level. Therefore, organic rice bran, green kernel rice and crushed rice as energy source, and soybean hull and soybean curd as protein source could partial replace imported feeds for organic ruminant farming.

Comet Assay를 이용한 Flavonoids와 항산화 비타민의 인체임파구 세포 DNA 손상 보호 효과 (Protective Effect of Flavonoids on Lymphocyte DNA Damage Using Comet Assay)

  • The present study was attempted to investigate and compare the antioxidant potency of several well-know flavonoids, antioxidant vitamin and commercially available popular beverages. The antioxidant potency was assessed by the effect on reducing oxidative DNA damage of human lymphocytes. Cellular oxidative DNA damage was measured by SCGE (single-cell gel electrophoresis), also known as comet assay. Lymphocytes were pre-treated for 30 minutes with wide ranges of doses of apigenin, kaempferol, luteolin, myricetin, rutin, quercetin, $\alpha$-tocopherol (10,25,50,100,200,500,1000 $\mu$M) ,green tea extract or grape juice (10,50,100,250,500,1000 $\mu$g/mL) followed by a $H_2O$$_2$(100 $\mu$M) treatment for 5 min as an oxidative stimulus. The physiological function of each antioxidant substance on oxidative DNA damage was analyzed as tail moment (tail length $\times$ percentage migrated DNA in tail) and expressed as relative DNA damage score after adjusting by the level of control treatment. Cells treated with $H_2O$$_2$alone (positive control) had an extensive DNA damage compared with cells treated with phosphate buffered saline (PBS, negative control) or pre-treated with all the tested samples. Of all the six flavonoids, quercetin was the most potent antioxidant showing the lowest $ED_{50}$/ of 8.5 $\mu$g/mL (concentration to produce 50% protection of relative DNA damage). The antoxidant potency of individual flavonoids were ranked as follows in a decreasing order; luteolin (18.4 $\mu$g/mL), myricetin (19.0 $\mu$g/mL) , rutin (22.2 $\mu$g/mL) , apigenin (24,3 $\mu$g/mL) , kaempferol (25.5 $\mu$g/mL). The protective effect of $\alpha$-tocopherol was substantially lower (highest $ED_{50}$value of 55.0 $\mu$g/mL) than all the other flavonoids, while the protective effect was highest in green tea and grape juice with low ED5O value of 7.6 and 5.3, respectively. These results suggest that flavonoids, especially quercetin, and natural compounds from food product, green tea and grape juice, produced powerful anti-oxidative activities, even stronger than $\alpha$-tocopherol. Taken together, supplementation of antioxidants to lymphocytes followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species.

  • 7주령의 F1B golden Syrian 햄스터에게 동맥경화 유발식이에 녹차추출물, 메밀추출물 및 포도잎 추출물을 함유한 타블렛 제품을 각각 5%와 10%로 첨가하여 7주간 사육하였을 때 햄스터 체내의 지질성분과 대동맥 내에서의 지방의 축적 정도에 미치는 영향을 조사하였다. 실험 결과 녹차, 메밀, 포도잎 추출물을 함유한 타블렛 제품은 동맥경화유발식이를 섭취하는 햄스터의 혈중 중성지방과 총콜레스테롤치를 감소시켰고, 대동맥궁내에서의 지방의 축적을 예방하였고, 간장내 중성지방과 총콜레스테롤치를 감소시키는 것으로 나타났다. 특히 5%를 첨가한 군보다 10%를 첨가한 군에서 더 높은 효과를 나타내어 농도 의존적인 경향을 보였다. 이러한 결과를 통해 녹차, 메밀 및 포도잎 추출물 분말을 이용하여 식이 보조제의 형태로 개발한 타블렛 제품이 동맥경화를 예방하는 식품으로써 효과가 있음을 확인할 수 있었다.
