Comparison of the Antioxidative Effects and Content of Anthocyanin and Phenolic Compounds in Different Varieties of Vitis vinifera Ethanol Extract

  • This study was a quantitative HPLC analysis of four anthocyanins and five phenolic compounds contained in the skins, vines and seeds of the Campbell Early, Muscat Bailey A and Neo Muscat grape varieties. In the phenolic analysis, the seeds of the Campbell Early were found to contain 1.9, 1.8 and 1.6 times higher quantities of gallic acid, catechin and epicatechin relative to other grape seeds. Three anthocyanins, cyanidin, peonidin and pelargonidin, were also found to be higher in the skins of the Campbell Early relative to other grape skins. Therefore, the Campbell Early is the most useful grape variety with regard to the extraction of these six compounds from these grape seeds and skins. The free radical scavenging effects of grape seeds were also compared, and the results indicated that the Campbell Early seeds were most effective among them.

Identification and Characterization of New Record of Grape Ripe Rot Disease Caused by Colletotrichum viniferum in Korea

  • In 2016, grape fruits showing ripe rot symptom were found in fields of Korea. The fungus was isolated and identified as Colletotrichum viniferum based on morphological characteristics and nucleotide sequence data of the internal transcribed spacer, glyceraldehyde-3-phosphate dehydrogenase and ${\beta}$-tubulin. To our knowledge, this is the first report of C. viniferum causing grape ripe rot disease of grape fruits in Korea.

포도와 포도 가공품에 함유되어 있는 trans-resveratrol의 함량 분석 (Analysis of trans-Resveratrol Contents of Grape and Grape Products Consumed in Korea)

  • 본 연구는 생물학적, 비생물학적인 스트레스에 대한 방어물질중의 한가지인 trans-resveratrol를 포도와 포도가공품(포도주, 포도쥬스)으로부터 분석하는 방법을 확립하였다. trans-Resveratrol 기기분석 조건은 다음과 같다. 분석 column은 Nucleosil 100-5 C18을 사용하였으며 이동상은 acetonitrile: water(40:60, v/v)로서 UV검출기(306 nm)를 이용하여 0.3 mL/min의 유속으로 측정하였다. 분석한 결과, 재현성과 반복성의 CV%는 10%이하였으며, 회수율은 $90{\sim}110%$이었다. 또한, 포도, 포도주 그리고 포도쥬스의 최소검출한계는 14.5 ng/g, 3.62 ng/mL 그리고 4.02 ng/mL이고, 최소정량한계는 14.8 ng/g, 3.69 ng/mL 그리고 4.10 ng/mL이었다. 표준물질인 trans-resveratrol 농도가 $0.048{\sim}2.4\;{\mu}g/mL$일때 $R^2$ 값은 0.999이었다. 32품종의 포도, 9종류의 포도주 그리고 9종류의 포도쥬스 내의 trans-resveratrol 함량은 각각 $0.76{\sim}207.14\;{\mu}g/100\;g$, $5.41{\sim}275.66\;{\mu}g/L$ 그리고 $63.25{\sim}751.55\;{\mu}g/L$로 분석되었다.

포도주스의 보충섭취가 흡연성인의 혈장 항산화 영양상태 및 DNA 손상 개선에 미치는 영향 (The Effects of Purple Grape Juice Supplementation on Improvement of Antioxidant Status and Lymphocyte DNA Damage in Korean Smokers)

  • The purpose of this project was to evaluate whether daily fruit juice consumption could reduce the DNA damage in healthy subjects. The study was performed using 67 healthy volunteers (29 smokers, 38 nonsmokers) who were supple-mented with 480 m1 of grape juice for 8 weeks. Eight weeks of grape juice consumption did not change any anthropometric parameters. Lymphocyte DNA damage before the study was significantly greater (p<0.05) in smoker than nonsmoker, but, grape juice consumption significantly reduced DNA damage in both smoker (26%) and nonsmoker (I7%) to the level where there was no difference remained between the two groups after the intervention trial. This preventive effect of grape juice against DNA damage was not affected by sex of the subjects in non-smokers. Plasma $\alpha$-carotene, Iyco-pene and ${\gamma}$-totopherol was significantly increased after the trial in smokers, while erythrocyte catalase was significan-tly increased in both smokers and nonsmokers. Total radical-trapping antioxidant potential (TRAP) level in all subjects was significantly reduced after the intervention, while GSH-Px activity was increased only in nonsmokers. These results suggests that daily consumption of grape juice may protect DNA damage in peripheral lymphocytes, and supports the hypothesis that grape juice might exert their effect partially via a decrease in oxidative damage to DNA in humans partly by improving their antioxidative defense system.

The Antioxidant Activity of Various Cultivars of Grape Skin Extract

  • The aim of this study was to analyze the antioxidant properties of different cultivars of grape skin extract in an in vitro system. The extracts were prepared from eight grape cultivars: 'Campbell Early' (CE), 'Kyoho' (K), 'New Kyoho' (NK), 'Muscat of Alexandria' (MOA), 'Seibel' (S), 'Morgen Schow' (MS), 'Gold Finger' (GF), and 'Meru' (M). The total phenolic acid contents were highest in MS and K. Resveratrol content was high in NK (50.88 mg/l g of coat), and quercetin content was significantly higher in K (0.68 mg/l g of coat) than in the other grape species (0.21-0.44 mg/l g of coat). The K and MS grape species, in which total phenol content was comparatively high (K: $24.15\;{\mu}g/mL$, MS: $25.52\;{\mu}g/mL$), also showed a high level of electron donating activity (K, 53%; MS, 59%). The hydrogen radical scavenging activity of M (50.36%) was significantly higher than the other grape species, including the S (50.21%), MS (49.43%), and K (49.06%) cultivars. Antioxidant activity varied depending on grape species, but overall it was highest in the MS and K cultivars.

Stimulating Effects of Far-infrared Ray Radiation on the Release of Antioxidative Phenolics in Grape Berries

  • This research was conducted to determine the effect of far-infrared ray (FIR) irradiation of grape berries as a potential application for manufacturing grape products with a high amount of antioxidant chemicals. Two grape cultivars, the red grape cv. Campbell Early and the white grape cv. Thompson Seedless, produced increased amounts of crude extracts, in the FIR treatments compared to a non-FIR treatment control with same temperature. However, total phenolic concentrations and antioxidant activity in a 'Campbell Early' increased in the extracts following FIR treatment, whereas those of 'Thompson Seedless' did not increase significantly. High performance liquid chromatography (HPLC) analysis indicated that functional components affecting antioxidant activity were significantly increased in the extract of 'Campbell Early' following FIR treatment. Our results indicate that application of FIR treatment in heat process of grapes increases levels of antioxidative phenolic chemicals and it may help to enhance the availability of antioxidative compounds in various grape food products.

포도과피 첨가가 분쇄돈육의 이화학적·관능적 특성에 미치는 영향 (Effect of Grape Skin on Physicochemical and Sensory Characteristics of Ground Pork Meat)

  • Purpose: This study aimed to investigate the effect of grape skin on the physicochemical properties and sensory score of ground pork meat. Methods: Four types of ground pork were evaluated: T0 without grape skin, T1 with 0.3% grape skin, T2 with 0.7% grape skin, and T3 with 1.0% grape skin. Results: There was no significant group wise difference in VBN content, L-value, b-value, chemical composition of raw and cooked meat, cooking yield, water holding capacity, moisture retention, fat retention, hardness, springiness, cohesiveness, gumminess, chewiness, taste, texture, juiciness, or palatability. Total polyphenol content was highest in T3, and DPPH radical scavenging activity was highest in T2 and T3 (p<0.001). The pH was highest in T0, and was lowest in T3 (p<0.001). The a-value of T2 and T3 were significantly higher than that of T0 (p<0.05). Flavor was highest in T2 among samples (p<0.01). Conclusion: The study results suggest that grape skin may be a useful ingredient in ground pork meat in terms of antioxidant potential, color and flavor.

포도 식사가 일부 과체중 여대생들의 체중조절과 혈액성상에 미치는 영향 (The Effect of Grape Diet on Weight Control Serum Components in Korean Overweight Female College Students)

  • This study was to investigate the effect on weight control and changes in serum glucose, total protein , GPT, GOT , cholesterol, triglyceride, calcium, and phosphorus contents of seventeen overweight female college students during experimental periods [before grape diet(0 day-, after grape diet (3 day), after control diet(3 day), after normal diet(30 day)]. The mean weight, height , BMI, and Rohrer index of the subjects at the onset of the study was 58.91$\pm$5.22kg, 159.94$\pm$6.01cm, 22.96$\pm$1.17, and 20.06$\pm$1.48 respectively. During the study subjects, conducted grape diets, control diets, and normal diets to control their weight. They analyzed their changes in anthrophometric variables. The mean weight was significantly decreased from 58.29$\pm$7.85kg to 57.2$\pm$8.23kg after normal diet(p<0.05). Therefore BMI (body mass index) and Rohrer index was also significantly decreased after experimental periods. The analysis of dietary intake showed that the grape diet induced unbalanced low nutrient intake, but the intake of total calories and other nutrients were almost recovered their onset of the study during control and normal diets. The concentration of serum glucose, cholesterol, triglyceride GOT, and GTP were significantly(p<0.05) decreased and significantly(p<0.05) increased again during experimental periods. Calcium concentrations were not affected by grape diet, but phosphorus concentration was significantly reduced after grape and control diet.

Impact of a reduction in the quality of Shine Muscat on the grape variety market using the Armington model

  • We devised a grape variety model to estimate the impact of lowering the Shine Muscat quality level on the grape market. Shine Muscat has become a popular grape variety in Korea. Accordingly, the area devoted to the harvesting of Shine Muscat has increased dramatically since 2016. Our study examines how a reduction in the quality of Shine Muscat affects other grapes such as Campbell Early, giant peak, and Muscat Bailey A (MBA). The Armington model was used to impose consumer preferences and product differentiation assumptions. We found that a decrease in the consumer preference for Shine Muscat realized by lowering the quality of Shine Muscat largely reduces the price of this variety. Also, the prices of other grape varieties fell via a substitute effect. Moreover, if grape varieties were more differentiated, the reduction in the price of Shine Muscat would be greater, while the decreases in the prices of other grape varieties would be smaller. These results imply that farmers of Shine Muscat must continue with quality management efforts to avoid the negative effect of changing consumer behavior with regard to Shine Muscat against a reduction in its quality. Our model introduces a product differentiation model for the fruit market and helps policymakers and farmers understand the impact of changing market conditions in the fruit market.

고콜레스테롤혈증 유발식이를 급여한 흰쥐에서 머루씨 분말첨가가 항산화활성과 혈액지질조성에 미치는 효과 (Antioxidative Activity and Serum Lipid Composition of Wild Grape Seed Powder in Rats fed Hypercholesterolemic Diet)

  • This study is to find out the antioxidative effect and serum lipid composition of wild grape seed powder in vivo. 20 white Sprague Dawley rats of six weeks old were divided into 2 groups and AIN-93 basic diet, high fat and cholesterol were provided. And they were examined to know how wild grape seed powder worked for antioxidative effect and serum lipid composition. For the comparing group, wild grape seed powder consisting 5% of the diet weight was provided and the quantity of protein, fat, carbohydrate, and cellulose was controlled following the analysis of the ingredients. The rats were fed for four weeks with experimental diet. Serum lipid and the antioxidant enzyme activity in blood and liver microsome were measured after 4 weeks of experiment. The results are as follows; There was no difference between the experimental groups in the initial body weight, final body weight, weight gain and FER. Food intake was higher in the group wild grape seed powder was provided than in the control group(p<0.05). Serum total cholesterol in the control group was significantly higher than that in the group wild grape seed powder was provided.(p<0.05). There was no difference serum HDL cholesterol and LDL cholesterol between the groups. Serum triglyceride showed no significant difference between the groups. In blood, glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group. The glutathione reductase activity of blood showed no difference between the groups. In liver, the glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group(p<0.05). Glutathione reductase activity in liver showed no difference in accordance with the supplementation of wild grape seed powder.
