  • An Extremely halophilic bacterium was isolated from solar salts. The isolated strain was Gram-negative, facultatively anaerobic, and motile bacterium. The colony was circular, smooth, and red-orange color. The strain showed pleomorphism depending on magnesium ion concentrations. The range of temperature and pH for growth of the isolate were 35 -45$\circ$C and 7.0 - 9.0. NaCl concentration for growth of it was 4.3 - 5.0 M. The isolate was catalase and oxidase positive, and sensitive to bacitracin. It showed starch hydrolyzing and acid forming characteristics. DNA G+C content was 62.7 mol%. The morphological, physiological and biochemical characteristics of the isolate resembled those of the Haloarcula vallismortis, therefore it was identified as Haloarcula sp. EH-1.

  • A halophilic bacterium was isolated from domestic marine. The bacterium was gram negative and motile. Transmission electron micrograph after cultivating for 6-, 20-, 72-, and 144 hrs showed that it was bacilli and contained intracellular granules, which were pleomorphic, larger in density by the time and considered to be PHB since they were positive on the sudan black B staining. The presence of sodium chloride was critical, because the isolated marine bacterium could not multiply and even produce any immunostimulant in the deficiency of sodium chloride. The strain produced an immunostimulant, which was investigated for the biological characteristics. The optimal conditions for the production of the immunostimulant were 1 % dextrose and 1 % yeast extract in artificial sea water for carbon and nitrogen sources, respectively. The initial pH and growth temperature for the production were 8.0 and $30^{\circ}C$ under the presence of oxigen, respectively.

  • The increasing spread of antibiotic-resistant pathogens has raised the interest in alternative antimicrobial treatments. In our study, the functionally active gram-negative bacterium bacteriophage CP933 endolysin was produced in Nicotiana benthamiana plants by a combination of transient expression and vacuole targeting strategies, and its antimicrobial activity was investigated. Expression of the cp933 gene in E. coli led to growth inhibition and lysis of the host cells or production of trace amounts of CP933. Cytoplasmic expression of the cp933 gene in plants using Potato virus X-based transient expression vectors (pP2C2S and pGR107) resulted in death of the apical portion of experimental plants. To protect plants against the toxic effects of the CP933 protein, the cp933 coding region was fused at its Nterminus to an N-terminal signal peptide from the potato proteinase inhibitor I to direct CP933 to the delta-type vacuoles. Plants producing the CP933 fusion protein did not exhibit the severe toxic effects seen with the unfused protein and the level of expression was 0.16 mg/g of plant tissue. Antimicrobial assays revealed that, in contrast to gram-negative bacterium E. coli (BL21(DE3)), the gram-positive plant pathogenic bacterium Clavibacter michiganensis was more susceptible to the plant-produced CP933, showing 18% growth inhibition. The results of our experiments demonstrate that the combination of transient expression and protein targeting to the delta vacuoles is a promising approach to produce functionally active proteins that exhibit toxicity when expressed in plant cells.

  • Studies on the adsorption of four kinds of bacteria, Staphylococcus aureus, Sarcina lutea, Escherichia coli and Serratia marcescens by activated Korean Kaoline have been carried out to innovate utilization as adsosrbent preparations. In connection with particle size and size fraction, the adsorption was examined by colony counting and spectroscopy. Korean kaolin was purified from Hadong white species of preminum grade and three size fractions were derived from passage through BS # 100, #200 and # 325 mesh sieves, respectively. These were activated at $105^{\circ}$or $280^{\circ}$ for three hours and at $550^{\circ}C$ for one hours. The results indicated that the adsorbing power of Korean kaolin was superior for S. aureus and S. lutea, but E. coli and S. marcescens were not adsorbed by clays. The smaller the particle size, the greater was adsorbing power for Gram-positive bacteria. Threre appears to be justification for its investigations as an ingredient in intestinal adsorbent preparations.

  • A heterotrophic nitrifying bacterium was isolated from the compost and analyzed for its characteristics. This bacterium was found to be a Gram positive rod, catalase positive, and motile. Nitrite production was detected on the ammonium acetate medium through the violet color formation. BBL test showed that this strain has high homology with Bacillus strains. Phylogenetic analysis using 16S rDNA revealed that the bacterium has 94% of similarity with Mycobacterium smegmatis strain.

  • Prevotella intermedia has been known as one of the important bacterial species involved in the endodontic infections and various periodontal diseases. Polyphosphate has been widely used to prevent decomposition of food and known to have an inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of poly phosphate on the growth of Prevotella intermedia, a gram negative bacterium. Prevotella intermedia G8GK3(ATCC 49046) was grown in the presence of polyphosphates with different chain lengths. Inhibitory effect of each polyphosphate, which was added at the beginning or at the early exponential growth phase of Prevotella intermedia, was determined by measuring optical density of the bacterial cells at 540nm, viable cells and lysis of Prevotella intermedia. The results from this study were as follows : 1. Poly phosphate inhibited the growth of Prevotella intermedia. 2. The minimum inhibitory concentration(MIC) of poly phosphate appeared to be 0.05%. 3. Polyphosphates with chain lengths of 5 and 65 demonstrated the greatest inhibitory effect on the growth of Prevotella intermedia. 4. Polyphosphate was bactericidal to Prevotella intermedia, demonstrating the growth inhibition of the bacterium. 5. Polyphosphate induced lysis of Prevotella intermedia. The overall results suggest that polyphosphate has a bactericidal effect on Prevotella intermedia, causing the lysis of the bacterium.

  • The treatment of Patients with bacteraemia and septicemia requires accurate and rapid identification of the pathogen so that the physician can be guided regarding the selection of the proper antimicrobial therapy. The usual procedure is to withdraw an aliquot of the positive blood culture sample for gram staining and subculturing on the media for the growth and subsequent identification, and susceptibility determinations. It was noticed that during the process some microbiologists would sniff the effluent gases that are products of metabolism and in some cases guess the identity of the bacterium. That Prompted us to engage in systematic investigation of two gram positive and two gram negative bacteria using an electronic nose that had been proven successful in distinguishing the aroma of coffee beans from different sources. The investigation was successful in illustrating the efficacy of such a device in this clinical setting to distinguish Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Enterococcus faecalis. Use of Clostridium septicum Alpha Toxins for Isolation of Various Glycosylphosphatidylinositol-Deficient Cells

  • In eukaryotic cells, various proteins are anchored to the plasma membrane through glycosylphosphatidylinositol (GPI). To study the biosynthetic pathways and modifications of GPI, various mutant cells have been isolated from the cells of Chinese hamster ovaries (CHO) supplemented with several exogenous genes involved in GPI biosynthesis using aerolysin, a toxin secreted from gram-negative bacterium Aeromonas hydrophila. Alpha toxin from Gram-positive bacterium Clostridium septicum is homologous to large lobes (LL) of aerolysin, binds GPI-anchored proteins and possesses a cell-destroying mechanism similar to aerolysin. Here, to determine whether alpha toxins can be used as an isolation tool of GPI-mutants, like aerolysin, CHO cells stably transfected with several exogenous genes involved in GPI biosynthesis were chemically mutagenized and cultured in a medium containing alpha toxins. We isolated six mutants highly resistant to alpha toxins and deficient in GPI biosynthesis. By genetic complementation, we determined that one mutant cell was defective of the second subunit of dolichol phosphate mannose synthase (DPM2) and other five cells were of a putative catalytic subunit of inositol acyltransferase (PIG-W). Therefore, C. septicum alpha toxins are a useful screening probe for the isolation of various GPI-mutant cells.

  • Recently, we investigated characteristics of a bacterium which has never been reported as an inulin hydrolyzing microorganism. Several properties of the isolated microorganism were aerobic, rod typed, spore forming and Gram positive. According to the Bergey's manual, this bacterium tentatively appeared to be Becillus subtilis. This bacterium produced inulase constitutively in the media containing giucose, sucrose or cellulose without inulin as a sole carbon source. Also, inulase activities of the bacterium per unit culture volume showed 1.1 unit/ml comparable to 0.9 unit/ml of Kluyveromyces fragilis with relatively short culture time.

  • A new rod-shaped endospore-forming bacterium isolated from the hindgut flora of the termite, Reticulitermes speratus kyushuenesis Morimoto is described. The isolate stained Gram positive, but the KOH test and the test for L-alanine aminopeptidase were negative. The length of a single cell varies from 2.5-9.0 $\mu $m, and the cell is about 0.5-0.7$\mu $m thick. The isolate had a high cellulolytic ability and was identified as Bacillus amyloliquefaciens.
