A report of 9 unrecorded radiation resistant bacterial species in Korea

  • Five bacterial strains, ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 assigned to the genus Deinococcus were isolated from soil samples collected from Namyangju-si in Gyeonggi-do, Gangnam-gu and Dongdaemun-gu in Seoul, Korea. In addition, four bacterial strains, KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 assigned to the genus Hymenobacter were isolated from soil samples collected from Gangnam-gu and Dongdaemun-gu in Seoul, in South Korea. The five Deinococcus species were Gram-stain positive, pink-pigmented, and short-rod or coccus shaped. The four Hymenobacter species were Gram-stain negative, red-pigmented, and short-rod shaped. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strains ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 were most closely related to Deinococcus citri $NCCP-154^T$ (with 99.8% similarity), Deinococcus grandis DSM $12784^T$ (99.0%), Deinococcus marmoris DSM $12784^T$ (98.8%), Deinococcus claudionis $PO-04-19-125^T$ (98.7%), and Deinococcus radioresistens $8A^T$ (99.8%), respectively. KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 were most closely related to Hymenobacter algoricola $VUG-A23a^T$ (99.1% similarity), Hymenobacter elongatus $VUG-A112^T$ (99.1% similarity), Hymenobacter gelipurpurascens $Txg1^T$ (99.1% similarity), and Hymenobacter psychrotolerans $Tibet-IIU11^T$ (99.3% similarity), respectively. These nine species have never been reported in Korea; thus, five Deinococcus species are reported in the family Deinococcaceae, order Deinococcales, class Deinococci, phylum Deinococcus-Thermus and four Hymenobacter species are reported in the family Cytophagaceae, order Cytophagales, class Cytophagia, phylum Bacteroidetes.

Novel Photocatalytic and Antibacterial Activities of Three-Dimensional (3D) Polycrystalline Anatase TiO2 Structures

  • We report three-dimensional polycrystalline anatase TiO2 structures (3D a-TiO2) for environmental and bio-medical applications. The 3D a-TiO2 was synthesized without thermal treatment by the growth of rod-like polycrystals on Degussa P25 (P25) via low temperature (< $85^{\circ}C$) modified alkali hydrothermal processing. X-ray diffraction and high-resolution transmission electron microscopic results showed that the rod-like polycrystals of 3D a-TiO2 possessed the highly anatase nanostructures. The photocatalytic activity of 3D a-TiO2 was found to be 2.2 times higher than that of P25. The recyclability of the 3D a-TiO2 was found to be high: the decolorization rate was 94.8% of the initial value after fifteen cycles. In addition, 3D a-TiO2 exhibited excellent antibacterial activities for the sterilization of gram-negative Escherichia coli (E. coli) and gram-positive Staphylococcus aureus (S. aureus). Even at the 10th recycled use, more than 98.4% of E. coli and S. aureus can be killed. These results indicated that 3D a-TiO2 might have utility in several promising applications such as photocatalytic water/air purification and bactericidal agents.

Isolation and Characterization of an Agar-degrading Bacterium, Isolated from The Sea Water

  • An agar degrading bacterium, This strain was isolated from sea water in Jeju. The strain is Gram-negative, rod and strictly aerobe for growth. The identification bases on the 16S rRNA gene sequencing showed that the strain was closely related to the genus Agariovarans sp. and named Agariovorans sp. JA1. The strain grew on agar as a sole carbon and energy source and produced an extra cellular agarase. For the increase of agarase productivity, 0.5% agar, yeast extract and $NaNO_3$ were used as carbon, organic and inorganic nitrogen source, respectively. For effective production of agarase and growth, the pH, temperature and NaCl concentration were was pH 7, $25^{\circ}C$ $^{\sim}$ $30^{\circ}C$ and 2%, respectively.

Identification and morphology of scanning electron microscopy(sem) of bacteria isolated from dental biofilm (덴탈 바이오필름에서 분리한 세균의 주사전자현미경적 관찰 및 동정)

  • In this study, specimens such as tongue, supragingival and subgingival biofilm were taken from total 20 scaling subjects who visited the oral prophylaxis practice lab at department of dental hygienics, J Health College in order to observe bacterial distributions and morphology using scanning electron microscopy(sem). as a result, this study came to the following conclusions: 1. According to observation of tongue, supragingival and subgingival biofilm through sem, it is found that there are round colonies of gram-positive cocci and gram-negative bacilli on blood agar medium. 2. The observation of bacterial morphology on dental biofilm through sem, cocci in chain cocci in cluster and bacillus(rod) respectively. 3. For tongue biofilm, it is found that a variety of bacterial species are detected, such as Granulicatolla adiacens(1), Gemella morbillorum(3), Streptococcus mitis(2), Streptococcus sanguinis(1), Aerococcus viridans (2), Streptococcus equinus(1), Leuconostoc spp.(1), Gemella haemolysans (1) and Lactococcus lactis spp.(1) respectively. 4. For supragingival biofilm, it is found that a variety of bacterial species detected, such as Aerococcus viridans(1), Gemella haemolysans(2), Leuconostoc spp.(2), Gemella morbillorum(1) and Pseudomonas fluoescens (1) respectively. 5. For subgingival biofilm, it is found that a variety of bacterial species detected, such as Leuconostoc spp.(1), Staphylococcus lugdunensis(1) and Streptococcus salivarius(1) respectively.

Characterization of a New Acidophilic Acetobacter sp. Strain HA Isolated from Korean Traditional Fermented Vinegar

  • A new strain of acidophilic, acetogenic bacterium, Acetobacter sp. strain HA was isolated by selective enrichment from the traditionally fermented rice wine vinegar in Korea. It was a gram-negative, non-motile short rod and oxidized acetate and lactate. The optimal temperature and pH for growth were $28^{\circ}C$ and 4.0, respectively. The strain HA differed from other Acetobacter species by growing well on methanol, xylitol, inositol, dulcitol, D-xylose, L-arabinose, and D-mannose as sole sources of carbon and energy. The isolated strain HA did not produce $\gamma$-pyrones from glucose and did not produce ketone bodies from glycerol. The quinone system used in this study was an ubiquinone-9 isoprene unit. The guanine-plus-cytosine content of the DNA was 50.7 mol%, and the major cellular fatty acids were $C_{18:1} and C_{16:0}$.

  • An organic solvent-tolerant Pseudomonas savastanoi strain BCNU 106 was isolated.. This bacterium was gram negative rod, and showed oxidase positive, catalase positive, and decaboxylase positive reactions. The Pseudomonas strain oxidized cholesterol on the medium supplemented with an organic solvent. The strain BCNU 106 is able to grow in presence of organic solvents of which log $P_{ow}$ is between 1.5 to 7.0. The strain was well grown in a medium supplemented with a 10% volume of a organic solvent (toluene, p-xylene, cyclohexane, mesitlylene) containing $cholesterol(20mg/m{\ell)$. Major conversion compound were 7-dehydrocholesteryl dimethylphosphate, cholesta-4,7-dien-3-one, and cholesta-3,5-dieone.

Isolation and Identification of Bacterium Producing Extracellular Cytosine deaminase (세포외 Cytosine Deaminase 생산균의 분리 및 동정)

  • A bacterium, strain YK 391 producing extracellular cytosine deaminase, has been isolated from soil sample collected near Taegu City and identified. The strain YK 391 was observed to be a motile Gram-negative rod, and did not produced capsule nor spore. The bacterium produced acid from glucose and trehalose, not from arabinose. Esculine was nto hydrolyzed. The isolate could grow anaerobically at 37$\circ $C, but not at 4$\circ $C. Palmitoleic and palmitic acids comprised over 80% of the fatty acid composition of the strain. The strain. The strain YK 391 was identified as Chromobacterium violaceum YK 391 based on its morphological and physiolohical characteristics, and on the fatty acid composition. The extracellular cytosine deaminase produced by Chromobacterium violaceum YK 391 is believed to be unique because it was active not only on cytosine and 5-fluorocytosine but also on cytidine.

  • Organic solvent tolerant bacterium, designated as strain BCNU 106 is a gram negative, rod-shaped aerobe and grows on benzene, toluene, ethylbenzene, and xylenes (BTEX) as a sole carbon source. According to 16S rDNA analysis and fatty acid analysis, strain BCNU 106 showed highest similarity to Pseudomonas syringae var. savastanoi (Pseudomonas savastanoi). Strain BCNU 106 was able to utilize toluene, ethylbenzene, both o-, m-, p-xylene , m-cresol and o-cresol. The degradation of o-, m-, p-xylene by strain BCNU 106 is particularly important, since o-xylene is a compound of considerable environmental interest, owing to its recalcitrance; and very few microorganism have been reported to utilize both o-, m-, p-xylene as a sole carbon source.

Isolation, Identification, and Culture Conditions of the Strain Producing Eicosapentaenoic Acid

  • The bacterium producing EPA was isolated from the intestines of the marine fishes. The strains were studied for their identification and their culture conditions. The selected strain was gram negative, rod(0.7 $\times$ 2.4 $\mu m$ in size) and motile with a single polar flagellum. This strain was identified as a Pseudomonas sp. on the basis of its morphological, cultural and physiological characteristics. The strain showed a maximum productivity of phospholipid at $20^{\circ}C$ after 48 hours of culture time with an initia1 pH of 7.0 in the PYM-glucose medium. Under these culture conditions, the production of phospholipid was about 0.3 mg/ml and 0.06 mg/mg dry cell weight.

A study on the prevalence of Riemerella anatipestifer from domestic ducks in Cheonan-Asan city (충남 천안아산지역에서 사육중인 육용오리의 리메렐라 감염율 조사)

  • This survey was carried out to investigate the infection rate of Riemerella anatipestifer from domestic ducks by isolation. A total of 145 samples (nasal swabs and carcasses) were collected from farms to examine the biochemical properties and antimicrobial susceptibility test from November 2006 to February 2008. Riemerella anatipestifer was isolated from 13 of 145 sample and isolation rate was higher in the ducks below 25 days (76%, 10/13 isolates) than those over 26 days (23%, 3/13). However, there was no significant difference in isolation rate between region. The biochemical properties of isolate were Gram negative (-), non-fermentative rod that grows fastidiously on routine media. In the test of antimicrobial drug susceptibility the isolates were susceptible to tetracycline(100%), enrofloxacin (77%), ampicillin (70%).
