Study on the Characteristics of Growth, Yield, and Pharmacological Composition of Licorice (Glycyrrhiza uralensis Fisch.) in a Temperature Gradient Tunnel (온도구배터널에서 기온상승에 따른 만주감초의 생육, 수량, 약리성분 특성에 관한 연구)

  • Background: Studies have suggested that the northern provinces of Gangwon-do are good sites for licorice (Glycyrrhiza uralensis Fisch.) cultivation in Korea, as they have similar temperatures to its original locations in northern China. However, poor growth and freezing injury are often reported in Korea. Therefore, it is necessary to reassess the domestic cultivation site of licorice. Methods and Results: To determine the optimum temperature for cultivating licorice, the growth, yield, and pharmacological characteristics of G. uralensis were assessed in a temperature gradient tunnel at Eumseong, Chungcheongbuk-do, Korea in 2017. Plant height increased until the temperature rose to $5.9^{\circ}C$ above the local external temperature. Yield (㎏/10a) increased by 46.9% when the growing temperature was $1.5^{\circ}C$ to $3.0^{\circ}C$ (T2) above the external temperature and by 72.6% when the growing temperature was $3.0^{\circ}C-4.5^{\circ}C$ (T3) above the external temperature. However, a difference of $4.5^{\circ}C-5.9^{\circ}C$ (T4) above the external temperature, decreased the yield by 9.8% compared to that at T2. The glycyrrhizin content of G. uralensis roots in each temperature band was 0.72%, 0.53%, 0.91%, and 0.84% (T1, T2, T3, T4), these differences appear to result form individual plant variation rather than growth temperature. Conclusions: Based on the results of this study, we estimate that the temperature-based optimum cultivation site for G. uralensis in Korea is the south central region, rather than the northern province of Gangwon-do. Improvement in growth and yield maybe observed if the plantations in the central Jecheon (Chungcheongbuk-do, Korea) are expanded into the south central region.

Biological Activities in roots of Glycyrrhiza uralensis Fisch (감초 세근의 생리활성 탐색)

  • The biological activities of ethanol, ethanol: water(1 : 1v/v) and water extracts from Glycyrrhiza uralensis Fisch, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin were compared. About 50% of the growth of MCF7, A549, Hep3B and AGS cells were inhibited in adding 1.0 g/L of the crude extracts, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin. For example, the ethanol extract inhibited 76%, 66% in MCF7 and Hep3B cells by adding 1.0 g/L. For cytotoxicity on human normal liver cell(WRL-68), the crude extracts were scored as above 26%. For the result of antimutagenecity using CHO V79 cell, the crude extracts proved more effective than other samples. The growth of human immune B and T cells were enhanced up to $1.2{\sim}1.3$ times by adding the crude extracts. In inhibitory effect of ${\alpha}-glucosidase$ activity was showed that the ethanol extract, water extract and ethanol: water (1 : 1v/v) extract were appeared 65%, 68%, 62% in adding 1.0 g/L. The higher enhancement of glutathione -S-transferase activity was observed in the ethanol extract as 257% compared to the control in adding 1.0 g/L. From the results, the biological activities of the crude extracts were equivalent or higher than glycyrrhizin and enzymatically hydrolyzed glycyrrhizin.

Biotransformation of Liquiritin in Glycyrrhiza uralensis Fisch Extract into Liquiritigenin by Plant Crude Enzymes (식물 유래 조효소에 의한 감소 Liquiritin의 Liquiritigenin으로의 변환)

  • Liquiritin in licorice (Glycyrrhiza uralensis Fisch) extract was treated with three different plant crude enzymes (Prunus dulcis enzyme; PDE, P. armeniaca enzyme; PAE and P. persica enzyme; PPE) for biotransformation. The resulting product of liquiritin was analyzed by TLC and HPLC. The ${\beta}glucosidase$ activities of crude enzymes were 259.6 U/g (PDE), 407.6 U/g (PAE) and 445.8 U/g (PPE), respectively. The liquiritin was converted to liquiritigenin after 12 hours of incubation with the crude enzymes. Liquiritigenin content reached its maximum level after the treatment with PPE at $37^{\circ}C$.

Nematicidal activity of Glycyrrhiza uralensis Fisch. root extracts on Meloidogyne incognita eggs and juveniles

  • This study evaluated the in vitro nematicidal activity of Glycyrrhiza uralensis root extracts (GuRE) on Meloidogyne incognita eggs and juveniles. The results showed that treatment of M. incognita eggs with 2.0 mg/mL GuRE for 5 and 10 d resulted in 64.0 and 68.1% hatch inhibition, respectively. Furthermore, the relative mortality of J2 was 96.2% after treatment with 2.0 mg/mL GuRE for 48 h. Changes in the shape of the eggs and juveniles were determined after incubation with 2.0 mg/mL GuRE for 5 d and 48 h, respectively. These preliminary results suggest that GuRE can be used as an environment-friendly bio-nematicide to control root-knot nematodes. In the future, in vivo assays should be conducted using GuRE to ascertain its potential for widespread application as a nematicide.

Effects of Dietary Supplementation of Glycyrrhiza uralensis Fisch and Solanum nigrum L. Mixture for Alternate the Antibiotics on Productivity and Blood Composition in Broiler Chickens (항생제 대체 감초와 용규 혼합 급여가 육계의 생산성과 체액 성상에 미치는 영향)

  • The objective of this study was to evaluate the effects of dietary Solanum nigrum L. (SNL) and Glycyrrhiza uralensis Fisch (GUF) mixture powder on performance and blood compositions of broilers. Seven-day old Ross broiler male chickens were randomly given into one of the 5 dietary treatments for 5 weeks: 1) no SNL-GUF and antibiotics (control); 2) 5 ppm avilamycin (T1); 3) 0.5% SNL and GUF (T2); 4) 1.0% SNL and GUF (T3); and 5) 2.0% SNL and GUF (T4). Each dietary group (n = 10) had three replicates. Body weight gain, feed intake and feed conversion ratio of broiler chickens were not significantly different among treatments. Carcass weight was highest at both 4 (P<0.05) and 6 weeks (P>0.05) in the treatment with 1.0% SNL and GUF. Liver, but not Spleen, weights at both 4 and 6 weeks were significantly higher in the treatments with 0.5% SNL and GUF treatment (P<0.05). The contents of protein, albumin, bilirubin, glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), glucose, triglyceride (TG) and cholesterol in blood serum at the 4weeks and 6 weeks were not significantly influenced. These results suggest that addition of SNL and GUF could be use a substitutes for antibiotics since they did not influence to the productivity and the blood composition of the broilers.

Protective Effect of Glycyrrhiza New Domestic Variety on the Acute Pancreatitis (국내 감초 신품종의 급성 췌장염 보호 효과)

  • Glycyrrhiza korshinskyi Grig. (Gk), which is a new variety of Glycyrrhiza, was developed by the National Institute of Horticultural and Herbal Science in Korea Rural Development Administration. It is improved the limitations of traditional Glycrrhiza species such as G. uralensis Fisch., G. glabra L., and G. inflata Batal. and has been reported various pharmacological effects. The aim of this study is to investigate the protective effect of Gk on the acute pancreatitis, because which has not been revealed yet. AP was induced via intraperitoneal injection of cerulein (50 ㎍/kg) hourly for 7 times in C57BL/6 mice. Gk water extract (20 or 50 mg/kg) or saline was administrated via oral gavage 1 h before the first injection of cerulein. The mice were sacrificed at 5 h after the final injection of cerulein. The inflammatory cell infiltration in pancreatic tissue was decreased by the administration of Gk water extract compared to the AP group. Also, serum amylase and lipase levels were decreased by the administration of Gk water extract compared to the AP group. These results suggest that Gk have protective effect on the acute pancreatitis.

Inhibitory Effects of Oriental Medicinal Herbs on Enzymatic Activity of Sialyltransferases (ST3Gal I and ST6Gal I) Associated with Cancer

  • In many cases of human cancer, the appearance of hypersialylated glycan structures is related to a precise stage of the disease ; this may depend on the elebated sialyltransferase activity during carcinogenesis. The aim of this study was to investigate the inhibitory effects of Oriental medicinal herbs on enzymatic activities of two kinds ofsialyltransferase, Gal $\beta$ 1,3GalNAc$\alpha$2,3-sialyltransferase(ST3Gal I) and Gal $\beta$ 1,4GlcNAc $\alpha$2,6-sialyltransterases(ST6Gal I), which are well known as glycosyltransterases associated with cancer. The aqueous extracts of Scutellaria Baicalensis Georgi, Coptidis Rhizoma, Glycyrrhiza urlensis Fisch, Bupleuri Radix and Platycodi Radix were prepared and tested, respectively. At concentration of 100$\mu$g, Glycyrrhiza uralensis Fisch showed the highest inhibitory effects(about 42% and 57%, respectively) on ST3Gal Iand ST6Gal Iactivities. ST3GAl I was inhibited about 23% by Scutellaria baicalensi G댁햐, but not by the other samples, whereas ST6Gal I was inhibited about 20% and 40%,respectively, by Scutellaria baicalensis Georgi and Bupleuri Radix. All inhibitory effects were obtained in a concentration-dependent manner.

The Effects of Gamcho on Histamine and Cytokine Production In Murine Asthma Model (감초가 천식모델 생쥐의 plasma내 histamine과 폐조직내 cytokien 생성에 미치는 효과)

  • This experiment was designed to investigate the effect of Gamcho(Glycyrrhiza uralensis Fisch, GLU) in asthma. We measured histamine, IL-1β, IL-4, 1L-5, IL-6, IL-10, IL-13, in plasma of ovalbumin induced asthmatic mouse. The results were obtained as follows: GLU decreased the proliferation of histamine, IL-1β, IL-4, IL-5, IL-6, IL-13 significantly. GLU increased the proliferation of IL-10 significantly. According to the above results, it is suggested that GLU extract might be useful applied for prevention and treatment of allergic asthma.
