Primed human pluripotent stem cells (hPSCs) are highly dependent on glycolysis rather than oxidative phosphorylation, which is similar to the metabolic switch that occurs in cancer cells. However, the molecular mechanisms that underlie this metabolic reprogramming in hPSCs and its relevance to pluripotency remain unclear. Cha et al. (2017) recently revealed that downregulation of SIRT2 by miR-200c enhances acetylation of glycolytic enzymes and glycolysis, which in turn facilitates cellular reprogramming, suggesting that SIRT2 is a key enzyme linking the metabolic switch and pluripotency in hPSCs.
Cancer cells are known to drastically alter cellular energy metabolism. The Warburg effect has been known for over 80 years as pertaining cancer-specific aerobic glycolysis. As underlying molecular mechanisms are elucidated so that cancer cells alter the cellular energy metabolism for their advantage, the significance of the modulation of metabolic profiles is gaining attention. Now, metabolic reprogramming is becoming an emerging hallmark of cancer. Therapeutic agents that target cancer energy metabolism are under intensive investigation, but these investigations are mostly focused on the cytosolic glycolytic processes. Although mitochondrial oxidative phosphorylation is an integral part of cellular energy metabolism, until recently, it has been regarded as an auxiliary to cytosolic glycolytic processes in cancer energy metabolism. In this review, we will discuss the importance of mitochondrial respiration in the metabolic reprogramming of cancer, in addition to discussing the justification for using mitochondrial DNA somatic mutation as metabolic determinants for cancer sensitivity in glucose limitation.

Objective: This study was performed to understand transcriptional changes in the genes involved in gluconeogenesis and glycolysis pathways following castration of bulls. Methods: Twenty Korean bulls were weaned at average 3 months of age, and castrated at 6 months. Liver tissues were collected from bulls (n = 10) and steers (n = 10) of Korean cattle, and hepatic gene expression levels were measured using quantitative real-time polymerase chain reaction. We examined hepatic transcription levels of genes encoding enzymes for irreversible reactions in both gluconeogenesis and glycolysis as well as genes encoding enzymes for the utilization of several glucogenic substrates. Correlations between hepatic gene expression and carcass characteristics were performed to understand their associations. Results: Castration increased the mRNA (3.6 fold; p<0.01) and protein levels (1.4 fold; p<0.05) of pyruvate carboxylase and mitochondrial phosphoenolpyruvate carboxykinase genes (1.7 fold; p<0.05). Hepatic mRNA levels of genes encoding the glycolysis enzymes were not changed by castration. Castration increased mRNA levels of both lactate dehydrogenase A (1.5 fold; p<0.05) and lactate dehydrogenase B (2.2 fold; p<0.01) genes for lactate utilization. Castration increased mRNA levels of glycerol kinase (2.7 fold; p<0.05) and glycerol-3-phosphate dehydrogenase 1 (1.5 fold; p<0.05) genes for glycerol utilization. Castration also increased mRNA levels of propionyl-CoA carboxylase beta (mitochondrial) (3.5 fold; p<0.01) and acyl-CoA synthetase short chain family member 3 (1.3 fold; p = 0.06) genes for propionate incorporation. Conclusion: Castration increases transcription levels of critical genes coding for enzymes involved in irreversible gluconeogenesis reactions from pyruvate to glucose and enzymes responsible for incorporation of glucogenic substrates including lactate, glycerol, and propionate. Hepatic gluconeogenic gene expression levels were associated with intramuscular fat deposition.

A method for the depolymerization of polycarbonate waste by glycolysis using ethylene glycol without catalyst was explored in order to get the monomer bisphenol A (BPA). The effect of operation variables such as reaction time, reaction temperature, EG/PC weight ratio and the kinetic of glycolysis were studied. It was found that the polymerization reaction has two different activation energies depending on the reaction temperature. A drop in activation energy with temperature indicates that the reaction mechanism has shifted from one of a succession of elementary steps to another in the series. The maximum yield of BPA of 95.6% was achieved at reaction temperature $220^{\circ}C$ for 85min with EG/PC weight ratio 4.
The tazarotene-induced gene 1 (TIG1) protein is a retinoidinducible growth regulator and is considered a tumor suppressor. Here, we show that DnaJ heat shock protein family member C8 (DNAJC8) is a TIG1 target that regulates glycolysis. Ectopic DNAJC8 expression induced the translocation of pyruvate kinase M2 (PKM2) into the nucleus, subsequently inducing glucose transporter 1 (GLUT1) expression to promote glucose uptake. Silencing either DNAJC8 or PKM2 alleviated the upregulation of GLUT1 expression and glucose uptake induced by ectopic DNAJC8 expression. TIG1 interacted with DNAJC8 in the cytosol, and this interaction completely blocked DNAJC8-mediated PKM2 translocation and inhibited glucose uptake. Furthermore, increased glycose uptake was observed in cells in which TIG1 was silenced. In conclusion, TIG1 acts as a pivotal repressor of DNAJC8 to enhance glucose uptake by partially regulating PKM2 translocation.

In this work, ultrathin and two-dimensional (2D) cobalt hydroxide [Co(OH)2] nanosheets were synthesized by a sonication assisted liquid-phase exfoliation of bulk Co(OH)2. The resulting exfoliated Co(OH)2 is a hexagonal mono-layered nanosheet with a high specific surface area of 27.5 ㎡ g-1. The depolymerization of polyethylene terephthalate (PET) based on glycolysis reaction was also performed using an exfoliated Co(OH)2 catalyst. Excellent catalytic reaction performances were demonstrated; a high PET conversion and bis(2-hydroxyethyl) terephthalate (BHET) yield of both 100% using the nanosheet catalyst were achieved within a reaction time and temperature of 30 min and 200 ℃, respectively. The long-term stability of exfoliated Co(OH)2 catalysts was also demonstrated by recyclability tests of the catalyzed glycolysis reaction of PET over four cycles, showing both 100% of high PET conversion and BHET yield.
Hepatocellular carcinoma (HCC) is an aggressive and incurable cancer. Although understanding of the molecular pathogenesis of HCC has greatly advanced, therapeutic options for the disease remain limited. In this study, we demonstrated that SETD5 expression is positively associated with poor prognosis of HCC and that SETD5 depletion decreased HCC cell proliferation and invasion while inducing cell death. Transcriptome analysis revealed that SETD5 loss downregulated the interferon-mediated inflammatory response in HCC cells. In addition, SETD5 depletion downregulated the expression of a critical glycolysis gene, PKM (pyruvate kinase M1/2), and decreased glycolysis activity in HCC cells. Finally, SETD5 knockdown inhibited tumor growth in xenograft mouse models. These results collectively suggest that SETD5 is involved in the tumorigenic features of HCC cells and that targeting SETD5 may suppress HCC progression.

Barley growing in paddy fields often suffers from wet-injury due to oxygen deficiency in rhizospere caused by excessive water in the soil. This study was conducted to investigate responsiveness of growth, development and anaerobic glycolysis enzymes to acute hypoxia in barley seedlings. Barley seedlings at the third leaf stage were subjected to hypoxia (1 ppm dissolved oxygen) by sparging the culture solution with nitrogen gas for up to seven days. Length and fresh weights of the shoot and root were affected little by hypoxia for up to 5 days. But root dry weight was slightly decreased by hypoxia for 7 days. In the root, alcohol dehydrogenase and lactate dehydrogenase activities increased drastically under hypoxia, reaching at their maximum levels in 3 to 5 days of hypoxia and decreasing slightly thereafter. However, the activities of both enzymes changed little in the shoot. Increases of their activities in the root were contributed by all the isozymes found in barley. These results suggest that barley seedlings first adapt to hypoxia by rapidly activating fermentative glycolysis to stabilize cellular pH and to increase energy production for the following morphological adaptative changes.
A new route for PBT (Poly butylene terephthalate) production from recycled PET (Poly ethylene terephthalate) has been explored. The route consists of glycolysis of PET (Poly ethylene terephthalate) wastes using 1,4-butandiol into BHBT oligomers and polycondensation of the oligomers into PBT oligomer. This process uses post-consumer or post-industrial recycled PET and converts it into high-end PBT type engineering thermoplastic via a chemical recycling process. Zink acetate was used as a catalyst for both glycolysis and polycondensation. Two types of reactor for the glycolysis, batch and semi-batch reactor, were investigated and their performances were compared. Semi-batch reactor removes ethylene glycol (EG) and THF (tetrahydrofuran) during the reaction. Amounts of EG and THF generated during the glycolysis reaction were measured and used as criteria for the reactor performance. Performance of semi-batch reactor was shown to be better than that of batch reactor. Optimum reaction condition for the semi-batch reactor was BD/PET ratio of 4, and reaction temperature of $220^{\circ}C$, giving high EG yield (max 91%) and low production of THF. In addition, it was confirmed that the molecular weight of PBT oligomer increases in accordance with the progress of the polycondensation reaction.

The catalytic glycolysis process is the method of chemical recycling where the polyol and carbamate compounds recovered by transesterification reaction are reused to produce new polyurethane foams. In this work, ethylene glycol, diethylene glycol, and 1,4-butanediol were used to decompose polyurethane foams and various metallic acetates were provided as catalysts. The catalytic glycolsis of polyurethane foams was taken place in the reaction temperature of $180{\sim}200^{\circ}C$. The reaction rates of catalytic glycolysis reaction were indicated by the viscosity of the reaction products at different reaction times. IR and GPC analysis showed the types and the molecular weight distributions of the products. The catalytic glycolysis was profitable for using ethyleneglycol at high temperature. The activities of the catalysts are suitable for K, Na, Tl acetate, and the products are composed of comparatively high-contained amine compounds and carbamate compounds. In the case of Sr acetate and Quinoline, the reaction rate was somewhat low. However, the content of polyol was high and the content of the side-products was low. The foams which were prepared by blending up to 20wt% of recovered polyol with virgin polyols showed better physical properties in tensile strength, hardness, tear strength, and compressive strength compared to those of polyurethane foams from virgin polyol.
