Radiomodifying Potential of Panax ginseng in Liver of Swiss Albino Mice against Gamma Radiation

  • Panax ginseng occupies an important role in the folk medicine of China, Korea and Japan. The present study was undertaken to determine the radioprotective efficacy of ginseng root extract in the liver of Swiss albino mice. The animals were divided into 4 groups. Group I-Only vehicle was administered. Group II-The animals received 10 mg/kg body weight ginseng root extract i.p. for 4 consecutive days. Group III-Animals were irradiated with 8Gy gamma radiation at the dose rate of 1.69 Gy/min at the distance of 80 ems. Group IV-Animals were given by ginseng root extract (10 mg/kg body weight) continuously for 4 days and on 4th day they were irradiated with 8 Gy gamma radiation after 30 min. The animals from above groups were autopsied on 1,3,7,14 and 30 days. Biochemical estimations of phosphatases (acid & alkaline), LDH (lactate dehydrogenase), LPO (lipid peroxidation) and GSH (reduced glutathione) in liver and SGOT (serum glutamate oxaloacetate transaminase), SGPT (serum glutamate pyruvate transaminase) and alkaline phosphatase in serum were done. In ginseng treated group acid phosphatase (ACP), alkaline phosphatase (ALP), LPO and LDH in liver and SGOT, SGPT and alkaline phosphatase in serum did not show any significant alteration. However, a significant increase in GSH content in liver was recorded. In irradiated group there was a significant increase in ACP, ALP and LPO content in liver and SGOT & SGPT in serum was noted. Whereas, a significant decrease was recorded in GSH and LDH activity in liver and alkaline phosphatase activity in serum. Pretreatment of ginseng with radiation significantly alters the biochemical parameters in liver and serum. A significant decline in ACP, ALP activity and LPO content in liver and SGOT and SGPT activity in serum was observed. However, a significant increase in GSH content and LDH activity in liver and ALP activity in serum was estimated. The present study suggests that pretreatment of ginseng before irradiation significantly protects the liver and maintains the enzyme activity.

복분자 추출물이 Lipopolysaccharide로 유도된 간 손상에 대한 항산화 효과 (Anti-Oxidative Effects of Rubus coreanum Miquel Extract on Hepatic Injury Induced by Lipopolysaccharide)

  • The protective effects of Rubus coreanum Miquel (RCM) extract against LPS-induced hepatotoxicity were studied in rats. Squrague-Dawley rats were intraperitoneally administered the RCM at 100 mg/kg per day for three weeks. Then single dose of LPS (5 mg/kg) was injected into rats. Four hours later, they were anesthesized with ether and dissected. We examined the levels of glutamate oxaloacetate transaminase (AST), glutamate pyruvate transaminase (ALT), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) in sera, superoxide dismutase (SOD) in mitochondrial fraction and catalase (CAT), glutathione peroxidase (GPx) in liver homogenate. LPS-treatment markedly increased the levels of AST, ALT, ALP, LDH and significantly decreased those of SOD, CAT and GPx. But RCM-pretreatment decreased the levels of AST, ALT, ALP and LDH by 57.9%, 37.4%, 62% and 69% respectively and increased those of SOD, CAT and GPx by 82.9%, 64.2% and 96.7% respectively. Subsequently, the protective effects of RCM was evaluated through histopathological examination of liver tissue. The LPS treatment increased the state of necrosis and cirrhosis surrounding the central veins (CV) and sinusoid, but RCM-treatment decreased the state of necrosis and cirrhosis in the liver tissue. These results demonstrated that protective effects of RCM against LPS-induced hepatotoxicity.

Sodium Cyanide로 유도된 신경아세포종 세포주에서 오수유의 신경상해 보호효과 (Anti-neuronal Injury Effect of Evodiae Fructus Water Extract in Sodium Cyanide-induced SK-N-SH Cell Lines)

  • Objectives : This study investigated effect of Evodiae fructus water extract (EVOR) on apoptotic cell death induced by NaCN in SK-N-SH neuroblastoma cell lines. NaCN stimulates glutamate release which can activate glutamate receptors to initiate excitotoxic processes. This study examines the role of EVOR in mediating NaCN-induced cytotoxicity. Methods & Results : Cytotoxicity was assessed by measuring lactate dehydrogenase (LDH) in the culture media. NaCN(0.1mM) produced cytotoxicity following 12hrs of incubation. NaCN-induced cytotoxicity was partially blocked by EVOR. The treatment of EVOR in simultaneous exposure of cultures to NaCN provided complete protection against cytotoxicity. NaCN-induced cytotoxicity was found to inhibit DNA fragmentation, repaired by cell cycle and simultaneous exposure to NaCN, regenerated with neurite outgrowh by EVOR. These results indicate thaf damage by NaCN in neumnal cell cultures was repaired by EVOR, whereas NaCN-induced cytotoxicity is blocked Primarily by activation of anti-apoptosis. Conclusions : These results suggest that EVOR may be beneficial for the treatment of dementia and other degenerative problems of the central nervous system.

배양된 흰쥐 대뇌 피질 astrocytes의 세포기능에 대한 화학적 무산소증 유도물의 효과 (Effects of Chemical Anoxia Inducers on Cellular Functions of Cultured Rat Cortical Astrocytes)

  • The effects of antimycin A(AA), dodium azide ($NaN_3$) and 2,4-dinitrophenol (DNP), which inhibit mitochondrial ATP production, on cellular functions of cultured astrocytes were studied. High concentrations of AA $(50{\;}\mu\textrm{g}/ml),{\;}NaN_3$ (100mM) and DNP (20mM) significantly decreased 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) reduction, which was known to be related to mitochondrial function and then cel viability. AA ($50{\;}\mu\textrm{g}/ml$) increased lactate dehydrogenase (LDH) release and decreased [$^3H$] glutamate uptake, suggesting severe damage of cellular function by the concentrations of the compounds. Meanwhile, low concentrations of AA $(\leq{;\}10{\;}\mu\textrm{g}/ml),{\;}NaN_3{;\}(\leq{\;}50mM)$ and DNP ($\leq{\;}5mM$) significantly increased MTT reduction, the effect of which was specific to astrocytes. AA (5 and $10{\;}\mu\textrm{g}/ml$) did not affect LDH release and [$^3H$] glutamate uptake, indicating that these compounds increased MTT reduction at the low concentrations without cellular membrane damage. However, the low concentrations of AA produced significant decrease of MTT reduction in a glucose-free medium. Low concentrations of AA (1 and $5{\;}\mu\textrm{g}/ml$) did not change ATP production of astrocytes in the medium containing 10 mM glucose, but completely inhibited in a glucose-free medium, suggesting marked increase of cytosolic ATP production by the blockade of mitochondrial ATP production with low concentrations of AA. These results suggest that astrocytes have ability to enhance neuronal function or survival under conditions of incomplete ischemia or early by enhancement of glycolysis, and that cellular reduction of MTT occurs not only mitochondrially but also extramitchondrially.

환경 독성 억제효과를 가진 커큐민 나노스피어가 마우스의 사료 효율 및 악취저감에 미치는 영향 (Effects of Anti-ecotoxicological Curcumin Nanospheres on Feed Efficiency and Fecal Odor in Mice)

  • Curcumin ($C_{21}H_{20}O_6$) is a hydrophobic polyphenol found in turmeric. Although curcumin has been used as a natural medicine, its major limitation is related to poor absorption from the gut. Therefore, we developed a method for preparation of Curcumin Nanospheres (CN) to improve the aqueous-phase solubility of curcumin and investigate the functional role of CN in promoting feed efficiency and odor reduction in mice. CN showed inhibitory effects on actate dehydrogenase (LDH) cytotoxicity induced by ecotoxic substance toluene in gut epithelial HCT116 cells. In addition, the weights of internal organs (liver, heart, kidneys, and spleen) and the levels of serum Glutamate Oxaloacetate Transaminase (GOT), Glutamate Pyruvate Transaminase (GPT), and LDH did not show significant differences between mice administered oral CN for two weeks and compared to the control group. Interestingly, CN not only reduced hydrogen sulfide ($H_2S$) and ammonia ($NH_3$) levels and fecal odor, but also improved feed efficiency in mice. These results demonstrate that oral nano-delivery of anti-ecotoxicological CN is a functional system to deliver curcumin to the gut to improve feed efficiency and reduce fecal odor in mice.

Serum Mineral and Haematobiochemical Profile of Microfilariae Infected Cattle in India: Its Effects on Production and Therapy

  • A survey was under taken of six district of Northern India viz. Bareilly, Pilibhit, Udham Singh Nagar, Nainital, Almora and Rampur. The age, breed, sex and physiological status recorded. A total number of 854 cattle examined out of which lactating (274 cases), non lactating (302 cases) heifers (128 cases), calves (82 cases) and adult male (68 cases) were examined. An incidence of 4.92 percent (42) of microfilarisis was recorded. The highest prevalence was observed in Rudrapur District of Udham Singh Nagar (33.33%, 4/12), followed by Lalkaun in Nanital District (21.74%, 10/46), Rampur (12.50%, 2/16), Bareilly (8.16%, 8/98) and Pilibhit (1.22%, 1/82). No infection was observed in Almora region. Amongst 854 cattle of different group incidence was highest in adult male (12.20%, 10/82), followed by non lactating (3.82%, 12/314) and lactating (2.70%, 2/74), (7.64%, 12/157) was found in Heifers. For haemeto-biochemical, serum minerals estimations and therapeutic study 32 animals suffering from filariasis and 18 healthy animals were taken. 16 animals were treated with ivermectin $@200{\mu}g/kg $ body weight. Effect of this disease on production has also been estimated for which body weight and milk production was observed. The main clinical manifestations observed were anaemia, loss of appetite, debility, oedematous swelling especially in the abdominal region, increased heart rate, and respiration rate. Haematological changes indicated decrease in hemoglobin, total erythrocyte count, packed cell volume, erthrocyte fragility and neutrophil, whereas there was significant increase in erythrocytes sedimentation rate (ESR), total leukocyte count (TLC), lymphocyte and eosinophils. Biochemical changes showed significant reduction in the values of serum albumin, A : G ratio, where as there was significant increase in blood glucose, blood urea nitrogen (BUN), globulin, total lipid, total cholesterol, phospholipids, serum bilirubin. Serum mineral profile also altered markedly, which indicate a significant decrease in Ca, Cu, Fe, Zn, and Mn with increase value of Na and Cl. There was no significant change in P and K values. Enzyme pattern in micro filaria infected animal indicated increased level of AST, ALT, alkaline phosphatase, ornithine carbamyl transferase, sorbitol dehydrogenase, glutamate dehydrogenase, isocitric dehydrogenase and lactate dehydrogenase. In blood gas values and acid/base balance, there was an increase in $PVCo_2$ and $PVo_2$. It has been observed that microfilaria infected cattle showed decrease in body weight and milk production. Animal treated with ivermectin showed the return of these above values toward normalcy.

헛개 열매, 줄기, 가지 간장의 항산화 활성 및 관능적 품질 특성 (Antioxidant Activity and Sensory Evaluation in Soy Sauce with Fruit, Stem, or Twig of Hovenia dulcis Thunb)

  • Home-made soy sauces with or without Hovenia dulcis Thunb (Hutgae) originated from different parts such as fruits, stems, and twigs were prepared according to the Korean traditional procedure. Soy sauces supplemented with Hutgae were evaluated for their activities of 2,2-diphenyl-1-picrylhydrazyl radical scavenging (DPPH) and alcohol dehydrogenase (ADH), free amino acid profiles, and sensory quality. All soy sauce types containing Hutgae had a strong DPPH activity as compared to the general type of soy sauce without Hutgae (GSC). Among Hutgae groups, DPPH activities of soy sauce supplemented with Hutgae stems was higher than that of soy sauces with either Hutgae fruits or twigs. ADH activities of soy sauces with Hutgae ranged from 14% to 55%, thus indicating that the functional activity of Hutgae was not altered during soy sauce preparations. Total free amino acid content of GSC was 295.5 mg%, and that of soy sauce with Hutgae fruits (346.8 mg%) was the highest when compared to Hutgae stems (272.3 mg%) and Hutgae twigs (225.6 mg%). In amino acid profiles, aspartate, arginine, histidine, and lysine levels were higher in soy sauces with Hutgae compared to GSC, whereas isoleucine, leucine, and phenylalanine levels were lower. Particularly, high levels of aspartate, glutamate, threonine, and lysine were presented in Hutgae twigs, whereas for Hutgae fruits and Hutgae stems, the levels of serine, glycine and arginine, and proline and methionine were high, respectively. According to sensory evaluations, Hutgae stems were preferred than GSC, due to the lower offensive smell and higher umami tastes. These findings demonstrate that soy sauce with Hutgae stems has potential protective effects against hangovers, improves the taste, and implies a possible functional ingredient.

Biochemical, Haematological and Thyroid Hormone Profile in Healthy Indian Kathiawari Horses

  • Normal haematological and biochemical indices along with thyroid hormone status were studied in healthy Kathiawari horses of different age groups (yearling, young stock, adults and old stock) belonging to either sex. Effect of both age and sex was observed on thyroid hormone levels, haematological and biochemical indices. In females, hemoglobin levels was significantly lower in yearlings than adult animals while total leukocyte counts were higher in yearlings than equids of other age groups. Sex had effect only on total erythrocyte counts, mean corpuscular hemoglobin concentration and mean corpuscular hemoglobin in horses of 1-3 years age group (young stock) and on packed cell volume in adult female and male equids. Among biochemical indices, activities of enzymes were observed to be influenced both by age and sex. Creatine kinase, gamma glutamyl transferase, glutamate pyruvate transaminase, glutamic oxaloacetate transaminase and lactate dehydrogenase activities were significantly higher in young and adult equids than animals of other age groups in Kathiawari horses while activity of alkaline phosphatase was significantly higher in yearlings than equids belonging to other age groups in both male and females. However, activity of sorbitol dehydrogenase was unaltered due to both sex and age factor. Albumin, bilirubin direct, bilirubin total, cholesterol, creatinine, protein, triglyceride and uric acid were statistically different in various age and sex groups of horses. Calcium, magnesium and chloride contents were almost same in various age groups of male horses. Significantly higher levels of $T_3$ and $T_4$ were observed in both male and female yearlings as compared to equids of other age groups in both the sexes.

High-Molecular-Weight Poly-Gamma-Glutamate Protects Against Hypertriglyceridemic Effects of a High-Fructose Diet in Rat

  • We studied the effects of 2 different dosages of high-molecular-weight poly-${\gamma}$-glutamic acid (hm ${\gamma}$-PGA) derived from Bacillus subtilis chungkookjang on lipid metabolism in a high-fructose diet-induced hypertriglyceridemic animal model. For 4 weeks, rats were fed either AIN-93 diet (normal control, NC; n = 10) or modified AIN-93 diet in which cornstarch was substituted with 63% fructose (n = 30) to induce hypertriglyceridemia. After 4 weeks, the hypertriglyceridemic rats were treated with daily oral doses of 0 mg (hypertriglyceridemic control, HC), 2.5 mg (hypertriglyceridemic, low hm ${\gamma}$-PGA, HL), or 5 $mg{\cdot}kg{\cdot}bw^{-1}{\cdot}d^{-1}$ (hypertriglyceridemic, high hm ${\gamma}$-PGA, HH) hm ${\gamma}$-PGA for 4 weeks. The HL and HH groups exhibited significantly lower levels of serum triglyceride, total cholesterol, LDL cholesterol, and free fatty acids than the HC group. The administration of hm ${\gamma}$-PGA reduced serum ALT and AST levels. The activities of lipogenic enzymes such as hepatic malic enzyme and glucose-6-phosphate dehydrogenase as well as glucose-6-phosphate dehydrogenase mRNA expression were significantly decreased by hm ${\gamma}$-PGA administration (p < 0.05). These results indicate that hm ${\gamma}$-PGA has an anti-hypertriglyceridemic effect in high-fructose diet-induced hypertriglyceridemic rats.

Pathogenicity of Korean isolates of Acanthamoeba by observing the experimental infection and zymodemes of five isoenzymes

  • To determine the pathogenicity of Acanthamoeba spp. isolated in Korea and to develop a isoenzymatic maker, the mortality rate of infected mice, in vitro cytotoxicity against target cells and isoenzyme band pattern were observed. Five isolates of Acanthamoeba spp. (YM-2, YM-3, YM-4, YM-5 and YM-7) were used in this study as well as three reference Acanthamoeba spp. (A. culbertsoni, A. hatchetti, and A. royreba). According to the mortality rate of infected mice, Korean isolated could be categorized into three groups: high virulent (YM-4), low virulent (YM-2, YM-5, YM-7) and the nonpathogenic group (YM-4), In addition, the virulence of Acanthamoeba spp. was enhanced by brain passage in mice. In the cytotoxicity assay against chinese hamster ovary cells, especially, the cytotoxicity of brain-passaged amoebae was relatively higher than the long-term cultivated ones. The zymodeme patterns of glucose-6-phosphate dehydrogenase (G6PD), malate dehydrogenase (MDH), hexokinase (HK), glutamate oxaloacetate transaminase (GOT) and malic enzyme (ME)of Acanthamoeba spp. were different among each isolate, and also between long-term cultrued amoebae and brain passaged ones. In spites of the polymorphic zymodemes, a slow band of G6PD and K, and an intermediate band of MDH were only observed in pathogenic Acanthamoeba spp., which should be used as isoenzymatic makers.
