더덕 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 항 당뇨효과 (Antidiabetic effect of ethanol extract on Codonopsis lanceolata root in streptozotocin induced diabetic rats)

  • Streptozotocin(STZ)을 45mg/kg.b.w의 용량으로 흰쥐의 미정맥에 투여한 후 유발된 당뇨 흰쥐에게 1일 1회 7일간 1,000mg/kg의 용량으로 투여 후 glucose 함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) 활성과 glycogen 함량, triglyceride(TG), total cholesterol등의 지질대사에 관여하는 물질들을 측정한 결과 더덕 뿌리 에탄올 추출물 투여군이 glucose, TG, total cholesterol등의 함량과 G-6-Pase 활성의 유의적인 감소(p<0.05)를 나타내었으며 glycogen 함량과 G-6-PDH, GK의 활성이 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 더덕뿌리 에탄올 추출물이 항당뇨 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

  • 본 실험은 문헌상으로 혈당강하효과가 알려져 있는 인동초를 에탄올로 추출하여 그 추출물을 streptozotocin(STZ)으로 당뇨를 유발시킨 흰쥐에게 투여 후 혈당강하작용을 확인하였다. 또한, 그 유효 성분의 추적을 위하여 여러 용매(hexane, chloroform, ethylacetate, butanol, water)로 계통분획하여 그 추출물의 항 당뇨효능을 분석한 결과 hexane, water층의 분획물이 당뇨유발 흰쥐에서 glycogen 함량증가와 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dedehydrogenase (G-6-PDH)등의 당대사효소 활성에 효과가 있었다.

  • Streptozotocin(STZ)으로 유도된 당뇨 흰쥐에게 연교의 에탄올 추출물을 1일 1회 7일간 1,000mg/kg.b.w의 용량으로 투여 후 glucose함량과 이에 관여하는 효소(glucokinase(GK), glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH)활성과 glycogen 함량, triglyceride(T.G), total cholesterol등의 지질대사에 관여하는 물질들의 함량을 측정한 결과 연교 에탄올 추출물 투여군이 glucose, T.G, total cholesterol 등의 함량과 G-6-Pase 활성이 유의적인 감소를 나타내었으며 glycogen 함량과 GK의 활성이 유의적인 증가를 나타내었다. 이와 같이 연교 에탄올 추출물을 1,000mg/kg.b.w을 당뇨 흰쥐에게 투여한 결과 혈당저하, 지질대사의 개선 효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

어유(魚油)식이에 의한 흰쥐체내의 생화학적 변화연구(I) -간장조직내 지방산 합성효소의 변화 - (The Study of Biochemical Changes Induced by Fish Oil Diet in Rat(I) - Changes in Hepatic Lipogenic Enzyme Activity -)

  • 어유식이(魚油食餌)가 간장조직내 lipogenic enzyme의 활성도에 미치는 영향을 조사하고자 고등어유를 식이(食餌) 10%(w/w) 되게 먹이를 조제하여 13일동안 사육한 흰쥐의 간장조직의 glucose-6-phosphate dehydrogenase와 malic enzyme의 활성도는 lard를 섭취한 군(群)보다 $3{\sim}6$일의 섭취로 훨씬 저하되었으며 콩기름에 비하여는 같거나 또는 약간 낮은 값을 보였다. 고등어 또는 장어유를 어유(魚油)로 사용하여 먹이의 $0{\sim}10%$로 함량을 변화시키며 총지방량을 10%로 일정히 하고자 콩기름 또는 쇠기름을 배합유로 사용하였을 때 glucose-6-phosphate dehydrogenase의 활성은 어유(魚油)가 2 % 포함하였을 때부터 malic enzyme의 활성은 5% 포함하였을 때, 어유(魚油)를 포함하지 않은 군(群)에 비하여 감소를 보였다.

구척 에탄올 추출물이 Streptozotocin 유발 당뇨 흰쥐에 대한 항당뇨 및 항산화 작용에 미치는 효과 (Antidiabetic and Antioxidative Effects of Cibotium barometz in Streptozotocin-Induced Diabetic Rats)

  • Streptozotocin(STZ)으로 유도된 당뇨 흰쥐에게 구척 에탄올 추출물을 1일 1회 7일간 1,000 mg/kg.b.w의 용량으로 투여 후 glucose함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH)과 glucokinase(GK)활성과 glycogen 함량과 triglyceride(T.G), total cholesterol 등의 지질대사에 관여하는 물질들과 항산화작용에 관여하는 glutathione-s-transferase(GST),Catalase(CAT), glutathione peroxidase(GSH-Px)활성과 glutathione(GSH)함량을 측정한 결과 구척 에탄올 추출물 투여군이 glucose, T.G, total cholesterol 등의 함량과 G-6-Pase 활성의 유의적인 감소(p<0.05)를 나타내었으며 glycogen 함량과 G-6-PDH, GK의 활성이 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 구척 에탄올 추출물이 항당뇨, 항산화작용의 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

결명자 에탄올 추출물이 고콜레스테롤 식이를 섭취한 흰쥐의 체내 지질함량에 미치는 영향 (Effect of Cassia tora Ethanol Extract on the Lipid Levels of Serum and Liver in Rats Fed High Cholesterol Diet)

  • 결명자의 에탄올 추출물이 고콜레스테롤 식이를 섭취한 횐쥐의 체내 지질함량에 미치는 영향을 알아보기 위하여 정상식이군, 고콜레스테롤 대조군, 고콜레스테롤 식이에 결명자 에탄올 추출물을 각각 0.25%와 0.5%씩 첨가하여 공급하는 실험군으로 나누어 4주간 사육한 후 혈청 및 간장의 지질 함량, glucose-6-phosphate dehydrogenase와 malic enzyme 활성변화, 혈청 AST, ALT활성 및 지질과산화물의 함량의 변화를 조사하였다. 체중증가량과 식이섭취량은 추출물을 공급한 실험군에서 높았으나, 식이효율은 각 군간 차이가 없었다. 결명자 추출물군의 혈청 총콜레스테롤, 유리콜레스테롤, 중성지방 및 유리지방산 함량은 대조군에 비하여 감소하는 경향이었다. 간장중의 총 콜레스테롤과 중성지방의 함량은 고콜레스테롤 식이에 의하여 유의하게 증가하였으나 결명자 추출물을 첨가함으로서 증가된 간장 지질함량은 유의하게 감소하였다. 간장의 glucose-6-phosphate dehydrogenase와 magic enzyme 활성 을 측정한 결과, 결명자 추출물군에서는 대조군에 비하여 활성이 유의하게 감소하였으며, 추 출물의 첨가농도가 높을수록 현저하였다. ALT, AST 활성 역시 결명자 추출물군은 고콜레스테롤 대조군에 비하여 감소하는 경향이었고, 간장 조직중의 지질과산화물 함량도 결명자 추출물을 첨가함에 따라 통계적 유의차는 없었으나 감소하는 경향이었다.

Subcellular Characterization of Porcine Oocytes with Different Glucose-6-phosphate Dehydrogenase Activities

  • The in vitro maturation (IVM) efficiency of porcine embryos is still low because of poor oocyte quality. Although brilliant cresyl blue positive (BCB+) oocytes with low glucose-6-phosphate dehydrogenase (G6PDH) activity have shown superior quality than BCB negative (-) oocytes with high G6PDH activity, the use of a BCB staining test before IVM is still controversial. This study aimed to shed more light on the subcellular characteristics of porcine oocytes after selection using BCB staining. We assessed germinal vesicle chromatin configuration, cortical granule (CG) migration, mitochondrial distribution, the levels of acetylated lysine 9 of histone H3 (AcH3K9) and nuclear apoptosis features to investigate the correlation between G6PDH activity and these developmentally related features. A pattern of chromatin surrounding the nucleoli was seen in 53.0% of BCB+ oocytes and 77.6% of BCB+ oocytes showed peripherally distributed CGs. After IVM, 48.7% of BCB+ oocytes had a diffused mitochondrial distribution pattern. However, there were no significant differences in the levels of AcH3K9 in the nuclei of blastocysts derived from BCB+ and BCB- oocytes; at the same time, we observed a similar incidence of apoptosis in the BCB+ and control groups. Although this study indicated that G6PDH activity in porcine oocytes was correlated with several subcellular characteristics such as germinal vesicle chromatin configuration, CG migration and mitochondrial distribution, other features such as AcH3K9 level and nuclear apoptotic features were not associated with G6PDH activity and did not validate the BCB staining test. In using this test for selecting porcine oocytes, subcellular characteristics such as the AcH3K9 level and apoptotic nuclear features should also be considered. Adding histone deacetylase inhibitors or apoptosis inhibitors into the culture medium used might improve the efficiency of IVM of BCB+ oocytes.

Comparative and Interactive Biochemical Effects of Sub-Lethal Concentrations of Cadmium and Lead on Some Tissues of the African Catfish (Clarias gariepinus)

  • Cadmium is a strong toxic heavy metal which presents in paints and liquid wastes and causes oxidative stress in fish. On the other hand, lead is widely used for different purposes, e.g. lead pipes, it targets vital organs such as liver and kidney causing biochemical alterations. The present study evaluates the effects of 60 days exposure to Cd and Pb either single or combined together in African catfish. Sixty-four fishes were divided into 3 groups and exposed to $CdCl_2$ (7.02 mg/L) or $PbCl_2$ (69.3 mg/L) or a combination of them along with control group. Activities of acid phosphatase (ACP), lactate dehydrogenase (LDH) and glucose-6-phosphate dehydrogenase (G-6-PDH) were estimated. Moreover, gill, liver and kidney were assayed for activities of superoxide dismutase (SOD), catalase (CAT) and levels of glutathione (GSH) and malondialdehyde (MDA). Individual exposure showed that both Cd and Pb significantly decreased LDH activity and SOD activity in the kidney. Pb significantly increased G-6-PDH activity and decreased GSH level in the gill. CAT activity in liver and kidney elevated significantly on Cd exposure while lead caused a significant depletion in the liver and significant elevation in the kidney. Both Cd and Pb significantly increased MDA levels in liver and kidney while Pb increased its level in gills. The combined exposure resulted in normalization of LDH, G-6-PDH activity, and CAT activity in liver and kidney as well as GSH level in both tissues and MDA in gill and kidney. The combination increased SOD activity and MDA level in liver and decreased SOD activity in kidney and GSH level in gills. In conclusion, the antioxidant system of African catfish was adversely affected by prolonged exposure to Cd and Pb. The combined exposure caused less damage than individual exposure and returned most parameters to those of controls.

Anti-obesity Effects of Mulberry Leaf and Yacon Extract in Obese Rats

  • We evaluated the effect of anti-obesitic activity of MYE (mulberry leaf + Yacon tuber) extracted from Morus alba as muberry leaf and Smallanthus sonchifolia as yacon. 1%, 3%, or 5% of MYE were treated to Sprague-Dawley rats exposed to a high-fat diet. MYE treated rats were suppressed weight during four weeks, and they lost weight significantly after six weeks. Common blood chemistry panels related to liver function revealed significant improvement in the MYE-treated groups. The expression of leptin as indicators for obesity was decreased in perirenal fat. Such results indicate that MYE could be a promising candidate for the improvement of obesity. In addition, MYE effected on deceased glucose metabolism, reducing the activities of glucose-6-phosphate dehydrogenase (G-6-PDH) and glucokinase related to glycogen synthesis. The fatty liver was observed in high-fat diet-treated rats, resulting from increased number of adipose cells and Ito cells. However, this pathologic change was significantly improved by administration of MYE. MYE have significant effects on antioxdative function and glycometabolism against high fat diet. Thereby, it seems that MYE prevent fatty liver by high-fat diet. Thus it is suggested that MYE would be worth being developed as an biofunctional food to prevent undesirable effects caused by obesity.

Dexamethasone enhances glucose uptake by SGLT1 and GLUT1 and boosts ATP generation through the PPP-TCA cycle in bovine neutrophils

  • Background: Clinical dexamethasone (DEX) treatment or stress in bovines results in extensive physiological changes with prominent hyperglycemia and neutrophils dysfunction. Objectives: To elucidate the effects of DEX treatment in vivo on cellular energy status and the underlying mechanism in circulating neutrophils. Methods: We selected eight-month-old male bovines and injected DEX for 3 consecutive days (1 time/d). The levels of glucose, total protein (TP), total cholesterol (TC), and the proinflammatory cytokines interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α in blood were examined, and we then detected glycogen and adenosine triphosphate (ATP) content, phosphofructosekinase-1 (PFK1) and glucose-6-phosphate dehydrogenase (G6PDH) activity, glucose transporter (GLUT)1, GLUT4, sodium/glucose cotransporter (SGLT)1 and citrate synthase (CS) protein expression and autophagy levels in circulating neutrophils. Results: DEX injection markedly increased blood glucose, TP and TC levels, the Ca2+/P5+ ratio and the neutrophil/lymphocyte ratio and significantly decreased blood IL-1β, IL-6 and TNF-α levels. Particularly in neutrophils, DEX injection inhibited p65-NFκB activation and elevated glycogen and ATP contents and SGLT1, GLUT1 and GR expression while inhibiting PFK1 activity, enhancing G6PDH activity and CS expression and lowering cell autophagy levels. Conclusions: DEX induced neutrophils glucose uptake by enhancing SGLT1 and GLUT1 expression and the transformation of energy metabolism from glycolysis to pentose phosphate pathway (PPP)-tricarboxylic acid (TCA) cycle. This finding gives us a new perspective on deeper understanding of clinical anti-inflammatory effects of DEX on bovine.
