그래핀 옥사이드가 탄소나노튜브기반 바이오센서 전극의 포도당 산화효소 담지능및 민감도에 미치는 영향 (Effects of the Graphene Oxide on Glucose Oxidase Immobilization Capabilities and Sensitivities of Carbon Nanotube-based Glucose Biosensor Electrodes)

  • 본 연구에서는 다중벽탄소나노튜브(multi-walled carbon nanotube, MWCNTs) 기반 바이오센서 전극의 포도당 산화효소(glucose oxidase, GOD) 담지능을 높여 그 민감도를 개선하고자 그래핀 옥사이드(graphene oxide, GO)를 첨가하여 전극을 제조하였다. GO 첨가로 인하여 MWCNTs의 분산뿐만 아니라 전극의 친수성 및 표면에너지가 증가하였다. 또한, MWNCTs 0.05 g에 GO 0.05 g를 첨가하였을 때 $K_m$ (Michaelis-Menten constant)이 0.105로 가장 높은 값을 나타냈음에도 불구하고 GOD 담지능이 높아졌으며, 민감도가 $121{\mu}A\;mM^{-1}$까지 향상됨을 알 수 있었다. 이러한 실험 결과는 GO첨가에 의한 MWCNTs의 분산 안정성 향상, MWCNTs 전극 표면에서 친수성으로 개질 및 표면 자유에너지 증가가 GOD 담지능에 영향을 미친 것으로 사료된다.

효소연료전지의 Anode 제조조건이 성능에 미치는 영향 (Effect of Fabrication Method of Anode on Performance in Enzyme Fuel Cells)

  • Anode는 효소를 이용한 효소전극과 cathode는 PEMFC용 전극을 이용해 효소연료전지를 구동하였다. 효소 anode는 graphite 분말과 효소로서 글루코스 산화제, 전자매개체로 ferrocene을 혼합해 압축해서 만들고 Nafion 이오노머로 코팅하였다. Anode 제조조건을 변화시키며 성능을 측정해 효소 anode 제조 최적조건을 찾았다. 효소 anode 압축 시 최적 압력은 8.89 MPa이고, 효소 anode의 graphite 성분비가 60%일 때 최고의 출력밀도를 나타냈다. Anode 기질 용액의 최적 glucose 농도는 1.7mol/l이었다. 효소 anode는 Nafion 용액에 1초, 2회 침지에 의해 안정화되었다.

불가사리(Asterias amurensis) 장내에서 분리된 종속영양세균의 탄소원 이용 특성 (Characteristics of Carbon Source Utilization by Heterotrophic Bacteria Isolated from Internal Organs of Starfish (Asterias amurensis))

  • 전라남도 장흥군 인근 해안에서 채집한 불가사리(Asterias amurensis)를 대상으로 불가사리의 장내에 분포하는 종속영양세균의 균체수를 측정하였고, 분리동정된 균주들을 대상으로 탄소원 이용 특성을 조사하였다. 불가사리의 장내에 분포하는 종속영양세균의 균체수는8.65$\pm$0.65 $\times$$10^3$ cfu . g$^{-1}$이 였으며, 분리 동정된 24균주(그람음성균 11균주, 그람양성균이 13균주)중에서 Vibrio속,Staphylococcus속, Corynebacterium속이 높게 나타났다. 불가사리 장내에서 분리 동정된 세균들은 그람양성의 간균이 높은 비율을차지하였고, catalase와 oxidase의 활성은 각각 54.2%,20.8%로 나타났다. 그람음성균의 경우, 탄소원으로 glucose의 이용률이 가장 높았고, 그 다음으로 sucrose, mannose, maltose,그리고 trehalose의 순으로 높은 이용률을 나타냈으며, adipate, phenol acetate는 이용하지 못하였다. 그람양성균의 경우, glucose가 가장 높은 이용률을 나타냈고, 그 다음은 fructose, trehalose, maltose 순으로 높은 이용률을 보였다.

토양 미생물로부터 생산된 Extracellular Cholesterol Oxidase의 특성 (Characterization of Extracellular Cholesterol Oxidase Produced from Soil Microorganism)

  • 본 연구에서는 산업적으로 사용될 수 있는 안정하고 활성이 높은 cholesterol oxidase를 생산하는 균주를 얻기 위해 토양 미생물로부터 균주를 선별하였다. 선별된 균주에 대하여 세포외 효소 활성을 측정한 결과 BEN 115로 명명한 균주가 가장 높은 효소 활성도를 나타내었다. 이 균주는 형태학적, 생리학적 특성과 배양형태 및 G+C 함량을 분석한 결과 Nocardia속으로 확인되었다. 최적 효소 생산 조건을 조사한 결과 기존 yeast malt extract broth 조성인 0.4% yeast extract, 0.4% glucose, 1% malt extract에서 가장 높은 활성을 나타냈다. 본 균주에서 생산된 extracellular cholesterol oxidase는 SDS-PAGE와 Western blot 결과 분자량이 55, 57 kDa인 두 종류의 효소가 존재하는 것으로 나타났다. BEN 115에 대한 효소학적 특성을 연구한 결과 온도 안정성은 $55^{\circ}C$까지 효소 활성이 유지되었고, pH 안정성은 pH $3.5{\sim}9.5$의 범위까지 안정한 것으로 나타났으며 최적 온도와 최적 pH는 각각 35oC와 pH 5.5인 것으로 나타났다. 또한 detergent(Triton X-100, Triton X-114 그리고 Tween 80) 첨가 시 효소 활성이 첨가하지 않은 대조군보다 약 $1.6{\sim}2.0$배 증가하는 것으로 나타났다. Campesterol, sitosterol 그리고 stigmasterol에 대한 기질 특이성은 cholesterol(100%)과 상대적으로 비교 시 각각 50%, 50% 그리고 27%의 기질 특이성을 나타내었다.

  • Purpose: The purpose of this study was to investigate the adherence to diabetes control recommendations (blood glucose testing, medication, diet, exercise) in patients with type 2 diabetes at home and to analyse the correlation between adherence and blood glucose level. Method: Participants, numbering 214, were recruited from the endocrinology outpatient department of a tertiary care hospital in an urban city. The data were collected by a self report adherence questionnaire. Glycosylated haemoglobin (HbA1c) was determined by the high-performance liquid chromatography technique and fasting blood glucose was analyzed by the glucose oxidase method. Result: Medication adherence was higher than diet or exercise adherence. The frequency of blood glucose testing was lower for middle school graduates than college graduates. Diet adherence was significantly lower for participants who were obese, who did not have a spouse, and who had hyperglycemia. Medication and diet adherence were negatively correlated with HbA1c. Conclusion: A diet education program should be developed for patients with type 2 diabetes who are obese, who do not have a spouse, and who have hyperglycemia.

개인용 혈당측정기의 신뢰성 보증시험 개발 (Development of Reliability Demonstration Test for Personal Blood Glucose Meters)

  • The value of blood sugar is measured by a personal blood glucose meter which consists of two parts: a glucose strip and a blood glucose meter. A personal blood glucose meter makes use of electric media which are composed of glucose oxidase and electron. This study is to propose a method of reliability demonstration test derived from field data analysis and FMEA(Failure Mode and Effect Analysis). Detailed Conditions for reliability demonstration test are selected from the comparison of various failure mechanisms. The most dominant failure mechanism is wear-out which is caused by strip insertion/extraction. The testing device that can reproduce the failure mechanism of strip insertion/extraction is made to conduct reliability demonstration test. Using the testing device, it is confirmed that target lifetime of selected devices is more than 2 years.

Blood glucose levels, insulin concentrations, and insulin resistance in healthy women and women with premenstrual syndrome: a comparative study

  • Objective: To compare the blood glucose levels, insulin concentrations, and insulin resistance during the two phases of the menstrual cycle between healthy women and patients with premenstrual syndrome (PMS). Methods: From January of 2011 to the August of 2012, a descriptive cross-sectional study was performed among students in the School of Medicine of Jahrom University of Medical Sciences. We included 30 students with the most severe symptoms of PMS and 30 age frequency-matched healthy controls. We analyzed the serum concentrations of glucose, insulin, and insulin resistance by using the glucose oxidase method, radioimmunometric assay, and homeostasis model assessment of insulin resistance equation, respectively. Results: No significant differences between the demographic data of the control and PMS groups were observed. The mean concentrations of glucose of the two study groups were significantly different during the follicular and luteal phases (p=0.011 vs. p<0.0001, respectively). The amounts of homeostasis model assessment of insulin resistance of the two study groups were significantly different in the luteal phase (p=0.0005). Conclusion: The level of blood glucose and insulin resistance was lower during the two phases of the menstrual cycle of the PMS group than that of the controls.

Immobilization of Penicillium citrinum by Entrapping Cells in Calcium Alginate for the Production of Neo-Fructooligosaccharides

  • This work describes neo-fructooligosaccharides (neo-FOSs) production using the immobilized mycelia of Penicillium citrinum. Some critical factors were evaluated to optimize maximal production of neo-FOS. Optimal alginate and cell concentrations were determined to be $1.96\%$ alginate and $7.17\%$ cell, respectively, by statistical analysis. The optimal concentration of $CaCl_{2}$, which is related to bead stability, was determined to be 2 M. It was possible to increase the neo-FOS production by adding 15 units of glucose oxidase to the batch reaction. By co-immobilizing cells and glucose oxidase, neoFOS productivity increased $123\%$ compared with the whole-cell immobilization process. Based on the results above, a co-immobilization technique was developed and it can be utilized for large-scale production.

호도약침액(胡桃藥鍼液)의 유리기(遊離基) 소거(消去)와 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究) (Radical Scavenging and Antioxidant Effects of Juglandis Semen Extract(JSE))

  • This study was performed to determine if Juglandis semen extract(JSE) has free radical scavenging and antioxidant activities. Superoxide anion generation by xanthine oxidase/xanthine and in neutrophils activated by phorbol-12, 13-dibutyrate was inhibited by JSE and its effect was dose-dependent. JSE also inhibited generation of $H_2O_2$ induced by glucose oxidase/glucose and in opossum kidney cells treated with antimycin A. JSE exerted a direct $H_2O_2$ scavenging effect. Exposure of opossum kidney cells to 1mM tBHP caused a significant increase in lipid peroxidation, which was prevented by JSE. JSE also prevented tBHP-induced LDH release. These data suggest that JSE has free radical scavenging and antioxidant activities. However, further studies should be carried out to find the active ingredient(s) of JSE that exerts radical scavenging action.
