It has been investigated what could be the selective marker distinguishing the immature from mature spermatozoa and whether fi -glucuronidase and fi -glucosidase are dependent on androgen in the luminal fluid of the epididymis or not. The contents of hexose, hexosamine and sialic acid in the epithelial cells, luminal fluid and spermatozoa of the epididymis were examined and the patterns of protein bands were compared in each group of the luminal fluid by SDS-PAGE. Lactate dehydrogenase, glucose-6-phosphatase, Na+ -K+ -ATPase and MgNa-ATPase showed higher activities in the cauda than the caput epididymal spermatozoa but only $Mg^2$+-ATPase activity appeared to be changed significantly. When the contents of hexose, hexosamine and sialic acid were analyzed and compared quantitatively, those of hexose were significantly different in the luminal fluid of caput and cauda epididymis, those of hexosamine in the epithelial cells and those of sialic acid in the epithelial cells and luminal fluid. When SDS-PAGE has been performed in each group, the band of MW 33-37 KD which was absent in the luminal fluid of caput epididymis appeared obviously in the luminal fluid of cauda epididymis and ako apeared in the cauda sperm crude membrane fraction. In addition, $\beta$ -glucuronidase and $\beta$ -glucosidase activities and their dependence on androgen were measured and the SDS-PAGE patiems of proteins and/or glycoproteins in the luminal fluid were examined. The activities of these two enzymes in the luminal fluid of the epididymis decreased significantly from the 5th day after castration. When testosterone was injected, the activity of $\beta$ -glucuronidase began to increase significantly from the 5th day following injection and that of $\beta$ -glucosidase from the loth day. On the other hand, the band of about MW 21 KD was newly observed in the lumen of caput epididymis when testosterone was administered.
The effects of high carbohydrate diet on growth, serum physiological response, and hepatic heat shock protein 70 expression in Wuchang bream were determined at $25^{\circ}C$ and $30^{\circ}C$. At each temperature, the fish fed the control diet (31% CHO) had significantly higher weight gain, specific growth rate, protein efficiency ratio and hepatic glucose-6-phosphatase activities, lower feed conversion ratio and hepatosomatic index (HSI), whole crude lipid, serum glucose, hepatic glucokinase (GK) activity than those fed the high-carbohydrate diet (47% CHO) (p<0.05). The fish reared at $25^{\circ}C$ had significantly higher whole body crude protein and ash, serum cholesterol and triglyceride, hepatic G-6-Pase activity, lower glycogen content and relative levels of hepatic growth hormone (GH) gene expression than those reared at $30^{\circ}C$ (p<0.05). Significant interaction between temperature and diet was found for HSI, condition factor, hepatic GK activity and the relative levels of hepatic GH gene expression (p<0.05).
Grape products have been known to exert greater antioxidant and anti-obesity than anti-hyperglycemic effects in animals and humans. Omija is used as an ingredient in traditional medicine, and it is known to have an anti-hyperglycemic effect. We investigated whether the combined extracts of grape pomace and omija fruit (GE+OE) could reduce fat accumulation in adipose and hepatic tissues and provide beneficial effects against hyperglycemia and insulin resistance in type 2 diabetic mice. C57BL/KsJ-db/db mice were fed either a normal control diet or GE+OE (0.5% grape pomace extract and 0.05% omija fruit extract, w/w) for 7 weeks. GE+OE decreased plasma leptin and resistin levels while increasing adiponectin levels and reducing the total white adipose tissue weight. Furthermore, GE+OE lowered plasma free fatty acid (FFA), triglyceride, and total-cholesterol levels as well as hepatic FFA and cholesterol levels. Hepatic fatty acid synthase and glucose 6-phosphate dehydrogenase activities were decreased in the GE+OE group, whereas hepatic ${\beta}$-oxidation activity was increased. Furthermore, GE+OE supplementation not only reduced hyperglycemia and pancreatic ${\beta}$-cell failure but also lowered blood glycosylated hemoglobin and plasma insulin levels. The homeostasis model assessment of insulin resistance levels was also decreased and the decrease seems to be mediated by the lowered activities of hepatic glucose-6-phosphatase and phosphoenolpyruvate carboxykinases. The present data suggest that GE+OE may have the potential to reduce hyperglycemia, insulin resistance, and obesity in patients with type 2 diabetes.
The purpose of this study was to determine the effects of taurine supplementation on the hepatic lipid peroxidation, activiteis of defense enzymes and membrane stability during rat hepatocarcinogenesis. Hepatocarcinogenesis was induced by Solt & Farber modification. Lipid peroxide contents of carcinogen treated group which was not supplemented with taurine were lower than those of control group. This might be that peroxide is decreased because of the activation of detoxifing enzyme. Glutathione S-transferase(GST) activites of carcinogen treated groups were significantly (p<0.05) increased compared to those of control groups. The GST activities of group supplemented with taurine before treatment of carcinogen and during the all period of experiment were only less increased. In carcinogen treated groups, glutathione peroxidase(GPx) activites of groups supplemented with taurine were higher than those of non supplemented group. By carcinogen treatemtn, glucose 6-phosphatase(G6Pase) activites, index of membrane stability were decreased, but in carcinogen treated groups supplemented with taurine, they were less decreased. These results suggest that taurine supplementation seems to inhibit lipid peroxidation, to change the activities of defense enzymes and to prevent to membrane disintegration during chemically induced hepatocarcinogenesis.
To evaluate the effect of cyclohexane(CH) treatment on the liver cytochrome P-450 dependent aniline hydroxylase(CYPdAH) activity in alcohol-pretreated animals, CH(1.56 g/kg body weight) was intraperitoneally administered to Sprague-Dawley male rats, which had been drunk 15% alcohol in distilled water for 1,3 and 6 weeks. CH was injected to rats 4 times every other day and the animals were sacrificed at 24 hours after injection of CH. In the alcohol-pretreated rats, liver injuries were not demonstrated on the basis of the liver weight per body weight, the levels of serum alanine aminotransferase and hepatic microsomal glucose-6-phosphatase activities. By the CH treatment, alcohol-pretreated animals showed the significantly increased activity of hepatic microsomal CYPdAH. Concomitantly $V_{max}$ value in CYPdAH was more increased, whereas $K_{M}$ value more decreased in alcohol-pretreated animals by the treatment of CH. In conclusion, the increasing cause of microsomal CYPdAH in CH-treated rats pretreated with alcohol may be due to induction of enzyme protein in rat liver.r.r.
The purpose of this study was to examine the effect of taurine supplementation time on the activity of the enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats, Sprague-Dawley male rats were fed the purified diet for 7 weeks. They were supplemented with or without 1% taurine in drinking water before or after STZ injection or during all experimental procedure. In comparison to diabetic group without taurine, glucose-6-phosphatase(G6Pase) activity was decreased in diabetic group supplemented with taurine before STZ injection, and it was increased in diabetic group supplemented with taurine after STZ injection, but the difference was not significant. Glutathione S-transferase(GST) activity was significantly increased by the injection of STZ. However, the GST activities of diabetic groups exposed to taurine after STZ injection or during all experimental procedure were significantly decreased. Glutathione peroxidase(GPx) activities was significantly decreased by STZ injection. However, only in diabetic group supplemented with taurine before STZ injection, GPx activities was not decreased by the STZ injection. These results suggest that taurine supplementation may change the activities of GSH-related enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats and that may be helpful for the prevention of diabetic complication.

To investigate an effect of the ethanol extract of Lycium chinense(EELC) on the activities of enzymes scavenging oxygen free radicals or detoxicating alcohol. The ground Lycium chinense was extracted with 30% edible ethanol and then diluted with 6% ethanol to contain 2% EELC(w/v). Three different groups of male Sprague-Dawley rats had taken a drink EELC, ethanol(ETH) or water(control), respectively for 2 months. At the end of experimental period, the animals were sacrificed and obtained the following findings. The EELC-treated animals showed the highest activity of hepatic glucose-6-phosphatase among three groups. The activities of xanthine oxidase and cytochrome p-450 from EELC treatment group were lower than those from ETH-treated group. However, the activity of superoxide dismutase was higher in the EELC-treated group than the ETH-treated(p<0.005). Furthermore, hepatic alcohol or aldehyde dehydrogenase activity, and glutathione content and glutathione peroxidase were significantly higher in EELC-treated animals than in ETH-treated those. The activity of glutathione S-transferase in liver was appeared the orderly higher value in EELC, ETH and control-treated group. As the result, EELC may affect the reduction of oxygen free radical production and help the detoxication of ethanol.

BACKGROUND/OBJECTIVES: Recently, anthocyanins have been reported to have various biological activities. Furthermore, anthocyanin-rich purple corn extract (PCE) ameliorated insulin resistance and reduced diabetes-associated mesanginal fibrosis and inflammation, suggesting that it may have benefits for the prevention of diabetes and diabetes complications. In this study, we determined the anthocyanins and non-anthocyanin component of PCE by HPLC-ESI-MS and investigated its anti-diabetic activity and mechanisms using C57BL/KsJ db/db mice. MATERIALS/METHODS: The db/db mice were divided into four groups: diabetic control group (DC), 10 or 50 mg/kg PCE (PCE 10 or PCE 50), or 10 mg/kg pinitol (pinitol 10) and treated with drugs once per day for 8 weeks. During the experiment, body weight and blood glucose levels were measured every week. At the end of treatment, we measured several diabetic parameters. RESULTS: Compared to the DC group, Fasting blood glucose levels were 68% lower in PCE 50 group and 51% lower in the pinitol 10 group. Furthermore, the PCE 50 group showed 2-fold increased C-peptide and adiponectin levels and 20% decreased HbA1c levels, than in the DC group. In pancreatic islets morphology, the PCE- or pinitol-treated mice showed significant prevention of pancreatic ${\beta}$-cell damage and higher insulin content. Microarray analyses results indicating that gene and protein expressions associated with glycolysis and fatty acid metabolism in liver and fat tissues. In addition, purple corn extract increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased phosphoenolpyruvate carboxykinase (PEPCK), glucose 6-phosphatase (G6pase) genes in liver, and also increased glucose transporter 4 (GLUT4) expressions in skeletal muscle. CONCLUSIONS: Our results suggested that PCE exerted anti-diabetic effects through protection of pancreatic ${\beta}$-cells, increase of insulin secretion and AMPK activation in the liver of C57BL/KsJ db/db mice.

The aim of this study was to investigate the hypolgycemic activity of water extract of fermented yacon (Smallanthus sonchifolius) leaves tea (Yacon LWE) in high-fat diet (HFD)/streptozotocin (STZ)-induced diabetic mice. Male ICR mice were fed with a HFD (37% calories from fat) for 4 weeks prior to intraperitoneal injection with STZ (100 mg/kg body weight). Diabetic mice were supplemented with two doses of Yacon LWE (0.16% and 0.8%, wt/wt) for 6 weeks. The supplementation of high-dose Yacon LWE significantly lowered blood glucose levels and plasma ALT and AST activities compared with the control group. High-dose Yacon LWE also improved the insulin tolerance without any changes in plasma and pancreatic insulin concentrations in HFD/STZ-induced diabetic mice. Yacon LWE supplementation increased the insulin staining of pancreatic $\beta$-cells in a dose-dependent manner. Both 0.16% and 0.8% of Yacon LWE significantly elevated plasma leptin concentration, hepatic glucokinase activity and glucokinase/glucose-6-phosphatase ratio compared with the control group. However, glycosylated hemoglobin concentration was not different among the groups. These results suggest that high-dose Yacon LWE lowers the blood glucose level partly by enhancing insulin sensitivity and hepatic glucose metabolism in type 2 diabetic mice.
To study the effect of dietary $\omega 6/\omega 3$ fatty acid ratios on the preneoplastic lesions and lipid peroxidation in rat hepatocellular chemical carcinogenesis, placental glutathione S-transferase(GST-P) positive foci area and numbers, glucose 6-phosphatase(G6Pase) activity, thiobarbituric acid reactive substances (TBARS) were measured. Male Sprague-Dawley rats were fed 5 different diets-low $\omega 6/\omega 3$ ratio with fish oil (Low-F), low $\omega 6/\omega 3$ ratio with perilia oil(Low-P), moderate ratio with perilia oil(Moderate), blend of 10 different commercial fats and oils(High-BL) and high $\omega 6/\omega 3$ ratio(High)-for 8 weeks. Hepatocarcinogenesis was induced by modified Ito model. The area of GST-P positive loci was the lowest in Moderate group and in ascending order of Low-F < Low-P < High-BL < High. But statistically, only Moderate and High groups were significantly different. The number of GST-P positive foci showed the same trend as foci area. The activities of G6Pase, membrane stability marker, were increased as $\omega 6/\omega 3$ ratio decreased. Lipid peroxidation values (TBARS) were the lowest in Low-F group and it is significantly different from Moderate, High-BL and High groups. When dietary $\omega 6/\omega 3$ ratio was moderate(4.06), hepatocarcinogenesis was suppressed compared with high or low $\omega 6/\omega 3$ ratios. Blend fat, commonly consumed among Koreans, did not show any suppressive effect on carcinogenesis because of high ratio(6.7). These results suggest that dietary $\omega 6/\omega 3$ ratio influences hepatocellular chemical carcinogenesis. It is recommended that appropriate $\omega 6/\omega 3$ ratio should be around 4.0. and we recommend to use more $\omega 3$ fatty acid in food preparation to reduce the risk of hepatocarcinogenesis.
