한국인삼(韓國人蔘)의 Saponin에 관(關)한 연구(硏究)제3보(第三報) -산지별(産地別), 부위별(部位別), 재배기간별(栽培期間別) 인삼(人蔘) 및 가공중(加工中) Saponin함량(含量)에 관(關)하여- (Saponins of Korean Ginseng Panax ginseng C.A. Meyer (Part III) -Saponins of ginseng by the cultivating locations, sampling seasons, plant parts, growing stages and the processings-)

  • 인삼(人蔘)의 유효성분(有效成分)으로 알려진 saponin을 Thin layer chromatography로 전개(展開)하여 Digital Densitometer를 사용(使用)하여 한국인삼(韓國人蔘)의 산지별(産地別), 부위별(部位別), 재배연도별(栽培年度別) saponin함량(含量) 및 그 saponin fraction의 조성비(組成比)를 정량(定量)하고, 홍삼(紅蔘) 및 인삼(人蔘)엑기스 제조중(製造中) 일어나는 saponin함량(含量)의 변화(變化)를 연구(硏究)하여 다음과 같은 결과(結果)를 얻었다. 1. 산지별(産地別) saponin 함량(含量)에는 별로 차이(差異)가 없었으며, panaxadiol을 aglycone으로 하는 saponin군(群)과 panaxatriol을 aglycone으로 하는 sapon군(群)의 조성비(組成比)는 ($1.7{\sim}2.6$) : 1정도였다. 2. 부위별(部位別)로 볼 때, saponin함량(含量)은 미삼(尾蔘)이 12.7%로서 3.3%인 백삼(白蔘)의 4배에 가까운 높은 값을보였다. 그리고 saponin fraction별(別)로 볼 때, panaxadiol을 aglycone으로 하는 saponin fraction도 미삼(尾蔘)이 백삼(白蔘)보다 많으나, panaxatriol을 aglycone으로 하는 saponin fraction은 반대(反對)로 미삼(尾蓼)이 백삼(白蔘)보다 적었다. TLC의 2차원전개결과(次元展開結果), ginsenoside-Rd는 백삼(白蔘)에만 나타나고, 미삼(尾蔘)에서는 나타나지 않는 반면(反面), Rf와 $Rg_1$은 미삼(尾蔘)에서는 분리(分離)되었으나, 백삼(白蔘)에서는 $Rg_1$만이 존재하고, Rf는 분리(分離)되지 않았다 3. 재배연도별(栽培年度別) 근부(根部)의 saponin함량(含量)은 재배기간(栽培其間)에 따라 일정(一定)한 경향(傾向)을 찾아볼 수 없으나, 인삼근당평균(人蔘根當平均) saponin함량(含量)은 2년근(年根)이 90.3mg, 3년근(年根)이 254.2mg. 4년근(年根)이 404.2mg, 5년근(年根)이 996.9mg, 6년근(年根)이 1377.1mg으로 재배기간(栽培其間)이 길어질수록 현저(顯著)하게 높았다. 그리고 인삼지상부(人蔘地上部)의 saponin함량(含量)도 재배기간(栽培其間)에 길어질수록 높아지는 경향(傾向)을 볼 수 있었다. 그러나, saponin fraction별(別)볼 때는 $5{\sim}6$년(年)의 수확기(收穫期)에 가까워질수록 panaxatriol을 aglycone으로 하는 saponin fraction의 함량(含量)이 높았다. 4. 홍삼제조중(紅蔘製造中)의 saponin fraction별(別) panaxadiol을 aglycone으로 하는 saponin fraction의 조성비(組成比)도 건삼(乾蔘)에 비(比)하여 홍삼(紅蔘)이 낮아졌으나, panaxatriol을 aglycone으로 하는 saponin fraction의 조성비(組成比)는 건삼(乾蔘)에 비(比)하여 홍삼(紅蔘)이 오히려 높았다. 홍삼(紅蔘)의 Thin layer chromatogram에는 건삼(乾蔘)의 그것에 나타나지 않았던 수개(數個)의 미확인(未確認) spot를 더 볼 수 있었다. 5. Ethanol과 물로 미삼(尾蔘)을 추출(抽出)하여, 29.9%의 인삼(人蔘)엑기스를 얻었는데, 이 엑기스에는 미삼(尾蔘)으로부터 추출수율(抽出收率)이 94.2%에 상당(相當)하는 saponin이 이행(移行) 함유(含有)되어 있었다.

Effects of Ginsenoside Total Saponins on Experimental Irritable Bowel Syndrome in Rats

  • In the previous study, we reported that the in viかo inhibitory effect of ginsenosides, active ingredient of Panax ginseng, on $5-HT_{3A}$ receptor channel activity is coupled to in vivo anti-vomiting and anti-nausea effect. In the present study, we further investigated that the inhibitory effect of ginsenosides, active ingredient of Panax ginseng, on 5-HT3A receptor channel activity is also coupled to attenuation of irritable bowel syndrome (IBS), which is induced by colorectal distention (CRD) and $0.6\%$ acetic acid treatment. The CRD-induced visceral pains induced by CRD and acetic acid treatment are measured by frequency of contractions of the external oblique muscle in conscious rats. Treatment of GTS significantly inhibited CRD-induced visceral pain with dose-dependent manner. The $EC_{50}$ was $5.5{\pm}4.7$ mg/kg ($95\%$ confidence intervals: 1.2-15.7) and the antinociceptive effect of GTS on visceral pain was persistent for 4 h. We also compared the effects of protopanaxadiol (PD) ginsenosides and protopanaxatriol (PT) ginsenosides with saline on acetic acid-and CRD-induced visceral pain, and found that protopanaxatriol (PT) ginsenosides was much more potent than PD ginsenosides in attenuating CRD-induced visceral pain. These results indicate that U ginsenosides of Panax ginseng are components far attenuation of experimentally CRD-induced visceral pains.

정상 및 기억손상 유도 동물의 수동회피반응에 대한 홍삼 사포닌의 효과 (Effects of Red Ginseng Saponin on Normal and Scopolamine-induced Memory Impairment of Mice in Passive Avoidance Task)

  • This study was performed to examine the effect of red ginseng total saponin and extract on memory in mice using one trial step-down type passive avoidance method. Red ginseng total saponins (No. 1: PD/PT ratio=1.24, No. 2: PD/PT ratio=1.47) were prepared with the different mixing ratio by using the parts of red ginseng. In single administration of total saponin No. 1 (100 mg/ kg, bw) or No. 2 (50 mg/kg, bw) increased the latency time as compared with control group but was not statistically significant. Treatment of total saponin No. 1 (50 mg/kg, bw) for 10 days produced an increase of latency time but was not statistically significant. In scopolamine-induced memory deficient mice total saponin No. 1 (50 mg/kg, bw) and No. 2 (100 mg/kg, bw) significantly improved the latency time. These results show that red ginseng total saponin may improve the memory of sco-polamine-induced memory deficient mice and have nootropic activity.

생쥐에서 디젤배기가스 입자에 의한 기도염증과 기도 과민성에 미치는 홍삼 조사포닌의 영향 (The Effect of Crude Saponins of Korean Red Ginseng against Airway Inflammation and Airway Hyperresponsiveness Induced by Diesel Exhaust Particles in Mice)

  • The objective of this study was to investigate the effect of crude ginseng total saponins (CGS) against airway inflammation (AI) and airway hyperresponsiveness (AH) induced by diesel exhaust particles (DEP) in mice. AI and AH were induced by the intratracheal instillation with 0.1 $mg/m{\ell}$ of DEP suspension once a week for 10 weeks combined with ovalbumin (OVA) sensitization. Mice were also treated orally with 75 $mg/m{\ell}$ of CGS, 5 days a week for 10 weeks. Oral CGS treatment decreased in the level of serum immunoglobulin (IgE) and histamine increased by DEP and OVA, and declined respiratory resistance. It also dropped an enhanced infiltration of eosinophils in the bronchoalveolar lavage fluid (BALF) of mice, and an increased T helper type 2 cell derived cytokine levels such as of interleukin (IL)-4, IL-13 and IL-5 in the BALF. However, it did not influence T helper type 1 cytokine such as interferon-gamma in the BALF. These results indicate that CGS may alleviate allergen-related AI and AH in mice and may play an important role in the modulation of asthmatic inflammation.

랫트의 학습능력에 대한 홍삼 사포닌의 효과 (Effect of Red Ginseng Saponins on Learning Behavior of Rats in the Water Maze)

  • This study was performed to investigate the effect of ginseng saponin from Korean red ginseng on the learning and memory. Total (50, 100 mg/kg, bw) and panaxadiol saponin (15, 30 mg/kg, bw) treated groups did not show the difference of the time score and the number of error in comparison with control group. Panaxatriol saponin (15, 30 mg/kg, bw) significantly decreased both the time score and the number of error in water maze test. These results indicate that panaxatriol saponin from Korean red ginseng may improve the learning ability of rat in water multiple T-maze.

  • Effects of total ginseng saponin, 20-S-protopanaxadiol saponin, 20-S-protopanaxatriol saponin and playcodon saponin on the osmotic behavior of liposomes were investigated by optical measurement. These saponins showed different activities on liposomal membrane, and cholesterol in liposomes was an important factor to this variation of saponin activities.

  • 인삼의 부위별(部位別) saponin조성(組成)에 대하여 검토하였으며 아울러 시판인삼차(市販人蔘茶)을 분석하여 saponin의 함량(含量) 및 분포형태를 검토하므로써 이들 제품의 품질을 추정하고자 하였다. (1) 백삼에는 타부위(他部位)보다 $2.6{\sim}6.6$배의 Ra(O)를 함유하였다. (2) 미삼(尾蔘), 피삼, 뇌두에는 $Rb_1$, $Rb_2$, Rc가 많았고 total saponin의 약 50%에 달하였다. (3) Rbc와 Rg(f)의 비(比)는 인삼잎이 0.64 : 1, 백삼이 2 : 1, 뇌두가 3 : 1, 인삼꽃이 3.2 : 1, 피삼이 5.8 : 1, 미삼이 7 : 1 정도였고 백삼은 미삼에 비하여 Rg(f)의 상대함량(相對含量)이 약 3배정도 많았다. (4) 13종(種)의 시판(市販)인삼차를 분석한 결과 panaxadiol계(系) saponin과 panaxatriol계(系) saponin의 비(比)는 $0.8{\sim}8\;:\;1$로 많은 차이가 있었다.

Ginseng Total Saponin Attenuate Cardiac Hypertrophy Induced by Homocysteine in Rats

  • Recent studies have shown that Panax ginseng has a variety of beneficial effects on the cardiovascular system. Homocysteine (Hcy), which is derived from methionine, has been closely associated with the increased risk of cardiovascular diseases. In the present study, whether the in-vivo long-term co-administration of ginseng total saponins (GTS), active ingredients of Panax ginseng, with L-methionine (Met) inhibits methionine-induced hyperhomocysteine (HHcy) and H-Hcy-induced cardiovascular dysfunctions was investigated, and it was found that the plasma Hcy level, which was measured after 30 and 60 days, in the GTS+Met co-administration group was more significantly reduced than in the Metalone-treatment group. The left-ventricle (LV) wall thickness of the heart was likewise examined in each treatment group, and it was found that the co-administration of GTS with Met significantly reduced the Met-induced LV wall thickness. The results of the study indicate that the in-vivo long-term co-administration of GTS with Met not only inhibited H-Hcy induced by long-term Met-alone administration but also attenuated the H-Hcy-induced cardiovascular dysfunctions in rats.

홍삼 활성 성분이 생쥐 간 조직에서 Glutathione 및 지질과산화에 미치는 항산화 효과 (Effects of Red Ginseng Component Administration on Glutathione and Lipid Peroxidation Levels in Mice Liver)

  • 홍삼의 물 추출물, 알코올 추출물, 지용성 추출물, 총 사포닌, PD 및 PT 각 홍삼 추출물을 생리식염수에 녹여 50mg/kg/0.1ml용량으로 4주령 수컷 생쥐에게 15일간 경구투여 하여 생쥐 간으로부터 항산화 효소(GPX)의 활성도 변화와 GSH, GSSG 및 MDA의 함량 변화에 미치는 영향을 조사하였다. GPX의 활성도는 PD, PTTS순으로 활성도가 높은 것으로 조사되었으며, 항산화 물질인 환원형 GSH 또한 같은 순으로 증가하는 것으로 사되었다. 한편 GSSG 함량은 지용성 추출물, PD, 알코올 추출물 순으로 감소하는 것으로 조사되었다. 동물조직세포내의 산화환원반응과 해독작용상태의 평가에 중요한 GSSG/total GSH비율은 지용성 추출물, PD, 총 사포닌 순으로 감소하는 것으로 조사되었다. 활성 산소에 의해 생성된 지질과산화의 최종산물인 MDA의 함량은 GSSG/total GSH 비율의 조사 결과와 비슷한 경향을 보였다 이와 같은 결과를 종합하여 볼 때 항산화 효소 활성과 항산화 물질 활성 증대를 강화시키는 홍삼 성분으로는 지용성 추출물, PD 및 총 사포닌인 것으로 조사되었으며, 이러한 성분들이 산화적 손상에 대한 방어기전 향상에 좋은 것으로 생각된다.

Change in Ginsenosides and Maltol in Dried Raw Ginseng during Extrusion Process

  • Although widely applied in the food industry, extrusion cooking has not been applied to the traditional red ginseng process for steaming and drying ginseng. We therefore investigated the change in the effective components in red ginseng (total saponins, ginsenosides and maltol) from extruded raw ginseng. The variables were the drying temperature of the sliced raw ginseng (80 and $90^{\circ}C$) before the extrusion process and the moisture content (15 and 22%, w.b.) during the extrusion process. Ginsenosides Rg1 and Rg2 were detected in dried ginseng at $80^{\circ}C$, but ginsenoside Rg3, which was contained in red ginseng, was not detected. On the other hand, ginsenosides Rg1, Rg2 and Rg3 were detected in extruded ginseng at moisture contents of 15 and 22%. Total ginsenosides were highest at $90^{\circ}C$ drying temperature and 22% moisture content for the extrusion process.
