Background: Some plants have harmful effects on fungi and bacteria as well as other plants. Incorporating such plant into soil as green manure is effective in reducing population densities of soil pathogens. Methods and Results: Twenty-three species of green manure crops were cultivated after the harvest of 6-year-old ginseng and then incorporated into the soil at the flowering stage. The following year, the root rot ratio of 2-year-old ginseng and soil chemical properties were investigated. In the absence of green manure addition, the $NO_3$ content, electric conductivity (EC), and K content decreased by 95%, 79% and 65%, respectively. In the presence of green manure addition, $P_2O_5$ and $NO_3$ contents reduced by 41% and 25%, respectively. The "survived root ratio" of 2-year-old ginseng significantly increased by 56.2%, 47.5%, and 47.3%, in the Sorghum sudanense, Ricinus communis and Helianthus tuberosus treatment, respectively. In addition, there was a significant increase in the "survived root ratio" in the Secale cereale, Chrysanthemum morifolium, Atractylodes macrocephala, and Smallanthus sonchifolius treatments. The "survived root ratio" of ginseng showed a significant positive correlation with the soil pH and a negative correlation with the $NO_3$ contents, and EC. Conclusions: Cultivation of plant form the Chrysanthemum family as green manure, using mainly the rhizomes was effective for the control of root rot disease of ginseng.
Panax ginseng is an important medicinal plant that has been used worldwide for geriatric, tonic, stomachic, and aphrodisiac treatments. Ginsenosides contained in the ginseng root are the main substances having active functions for human body. The price of ginseng is very expensive due to a complex process of cultivation, and the yield of ginseng is limited, which cannot meet the demand of the increasing market. Researchers have applied plant biotechnology to solve the problems but there are still things to be determined towards ginsenoside production by large-scale adventitious root culture. In this experiment, 5 to 20 liter bioreactors were employed to determine optimal conditions for adventitious root culture and ginsenoside production of Panax gineng. Callus was induced from the ginseng root on MS agar medium containing 1.0 mg. $L^{-1}$ 2,4-D and 0.1 mg. $L^{-1}$ kinetin. Then the callus was cultured on MS agar medium supplemented with 2.0 mg. $L^{-1}$ IBA, 0.1 mg. $L^{-1}$ kinetin, and 30 g. $L^{-1}$ to induce adventitious roots. The maximum root growth and ginsenoside production were obtained in 1/2 MS medium. 2.0 mg. $L^{-1}$ naphthalene acetic acid resulted in greater root growth than 2.0 mg $L^{-1}$ indole-3-butyric acid. Ginsenoside content increased with 2.0 mg. $L^{-1}$ benzyl adenin or kinetin. High concentrations of benzyl adenin (above 3.0 mg. $L^{-1}$ ) decreased the adventitious root growth and ginsenoside productivity. N $H_{4}$$^{+}$ inhibited the ginsenoside accumulation, while high concentrations of $K^{+}$, $Mg_{2}$$^{+}$, and $Ca_{2}$$^{+}$ increased it. N $H_{4}$$^{+}$ at 0.5 and 1.0 times of the normal amount in 3/4 SH medium resulted in the greatest biomass increase, but the highest ginsenoside productivity was obtained when N $O_{3}$$^{-}$ was used as the sole nitrogen source in the medium. Most microelements at high concentrations in the medium inhibited the root growth, but high concentrations of MnS $O_4$enhanced the root growth. Root dry weight increased with increasing sucrose concentrations up to 50 g. $L^{-1}$ , but decreased from 70 g $L^{-1}$ Ginsenoside productivity was maximized at the range of 20 to 30 g. $L^{-1}$ sucrose. In the experiment on bioreactor types, cone and balloon types were determined to be favorable for both adventitious root growth and ginsenoside production. Jasmonic acid was effective for increasing ginsenoside contents and Rb group ginsenosides mainly increased. These results could be employed in commercial scale bioreactor cultures of Panax ginseng.x ginseng.
The study was conducted to investigate the tissue and chemical characteristics of rusty root epidermal cells. In histological study, the rusty symptoms were frequently observed in the epidermis of ginseng root and to be yellow under microscopic observation. Disks of the epidermal cell tissue of the rusty root were usually 2 and 3 times greater in the number of cell layer and thickness of cell wall than the healthy root, respectively. The color degree of methanol extracts from the rusty root epidermis was 5.5 times higher than that of the healthy root. And the extracts of rust matter in the root epidermis were easily dissolved in polar solvents compared to nonpolar solvents. UV-absorption spectra of methanol extracts in various fractions of phenolics showed a maximum peak between 275∼280 nm. The crude lipids and phenolic compounds such as acid insoluble bound phenolics, acid insoluble esterified phenolics, acid insoluble condensed phenolics, insoluble bound phenolics and free phenolics were also more in the rusty root epidermis than in the healthy one. Fe content in the rusty root epidermis was 2.7 times higher than that of healthy one. It was presumed that the phenolic compounds(precursor of the rusty) in association with lipid and iron in the root epidermis might defence the root when ginseng root was depressed by the unfavorable conditions in soil and/or portions of a root system were subjected to anoxic conditions.

Background: Ginseng root rot disease, caused by Cylindrocarpon destructans and Fusarium solani is a major cause of replant failure in continuous cropping ginseng. Methods and Results: To control replant injury in soil infected with C. destructans and F. solani, biosolarization was performed by covering the plot with transparent polyethylene film after adding green manure of maize and sunflower for the summer season. Per 10 a, fresh and dry weight of maize was 10.1 and 2.5 tons, respectively, and that of sunflower was 8.1 tons and 1.2 tons, respectively. Mean maximum temperature at 20 cm depth was $33.2^{\circ}C$, $41.5^{\circ}C$ and $41.8^{\circ}C$ in the control, maize-incorporated and sunflower-incorporated plots, respectively. The elapsed time over $40^{\circ}C$ was 36.4 h in the maize-incorporated plot and 77.3 h in the sunflower-incorporated plot. Biosolarization increased $NO_3$ content in soil, while content of organic matter, Ca, and Mg was decreased. Electrical conductivity, $NO_3$ and $P_2O_5$ in soil significantly increased after two years of biosolarization. The number of spores of C. destructans in soil was significantly decreased by biosolarization, and sunflower treatment was more effective than maize treatment in decreasing the number of spores. Root yield of 3-year-old ginseng was significantly increased by biosolarization, however, there was no significant difference between maize and sunflower treatments. Rate of root rot in 3-year-old ginseng decreased to 16.5% with the incorporation maize and 5.0% with the incorporation of sunflower, while that in control 25.6%. Conclusions: Biosolarization was effective in inhibiting ginseng root rot by decreasing the density of root rot disease and improving soil chemical properties.
Root discs of 4-year-old ginseng, Panax ginseng C. A. Meyer, were inoculated with the higher($10^8$ colonyforming units(CFU)/ml) and lower($10^6\;or\;10^5$ CFU/ml) initial inoculum levels of a plant-growth promoting rhizobacterium(PGPR), Paenibacillus polymyxa GBR-1 to examine rot symptom development and bacterial population changes on the root discs. At the higher inoculum level, brown rot symptoms developed and expanded on the whole root discs in which the bacterial population increased continuously up to 4 days after inoculation. In light and electron microscopy, ginseng root cells on the inoculation sites were extensively decayed, which were characterized by dissolved cell walls and destructed cytoplasmic contents. However, no rot symptoms were developed and the bacterial population increased only during the initial two days of inoculation at the lower inoculum level($10^6$ CFU/ml) of P. polymyxa GBR-1. At the lower inoculum level($10^5$ CFU/ml), boundary layers with parallel periclinal cell divisions, structurally similar to wound periderm, were formed internal to the inoculation sites, beneath which the cells were intact containing numerous normal-looking starch granules and no disorganized cell organelles, suggesting that these structural features may be related to the suppression of symptom development, a histological defense mechanism.
This study was divised to observe an inhibitory toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng(NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L Is known to be a lipolytic factor that was partially purified from the ascites fluid of sarcoma 180-hearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol precipitate component of large root NPG(mean 46.8%) was higher (mean 1.8 times) than that of water extract component in final reaction concentration ,5001g1m1, on the other side inhibitory effect of water extract component in small root NPG(mean 43.9%) was higher(mean 1. 2 times) than that of ethano1 precipitate component, respectively. In a way inhibitory effect of ethanol precipitate component in large root NPG(47.6%), when final reaction concentration of sample were 1,000 U g/ml, was about 4095 lower than that of Korean red ginseng, respectively.

This study was carried out to have the basic and applied information to develop the cultivation methods and models and to increase the productivity of high-quality ginseng. Plant height was dramatically increased from one year old to four years old, and then it was grown up very slowly. Stem length was shown similar tendency as the plant height did. Its range were from 9.4 cm in one year old to about 42.4 cm in four year-old ginseng. The leaf area was clearly increased until four years old, $10.1cm^2$ in one year old to $204.9cm^2$ in four years old, while it was slightly increased after four years old. Root length was continuously increased from one year old until four years old, and then it was grown up very slowly. Tap root length which was measured was appeared about 8 cm from three years old to six years old, showing not much different among the year-olds. The root diameter was continuously increased from one year old to six year-old ginseng, about 2.9 mm and about 19 mm, respectively. The moisture content was higher on the shoot than that on the root. It was slightly decreased as they were getting old. In shoot part, the rate of leaves and stems based on dry weight was about 1 : 1. In the root, the dry weight of tap root was heavier than that of lateral root. The dry weight of shoot was decreased after four years old, while it of the root was continuously increased until six years old, resulting the production of ginseng root. Based on the results of this study, growth characteristics of shoot and root of ginseng were dramatically increased from one year old to four years old and then their growths were appeared different between shoot and root, showing not much growing in shoot and keep growing in root.
This study was carried out to investigate the correlation of agronomic characters, their path coefficients in 2, 3 and 4-year old ginseng plants, and to provide a useful information for ginseng breeding. Correlation coefficients between stem fen변h, number of leaves and number of Iraflets in 2-year age, and stem diameter and leaf length in 3-year age showed highly significant correlation with number of fruits and root weight in 4-year age. The path coefficient analysis indicated that stem length and number of leaflets might give indirect effects on root weight regardless of plant age. On the other hand, stem length and number of leaflets in 2-year age and, stem diameter and leaf length in 3-year age showed direct effects on root weight in 4-year old ginseng. These results may be used for selection of high-yielding ginseng plants. Key words Selection information, correlation and path coefficient analysis.
This study was conducted to know the respiration rate in different organ of 4 years old ginseng plant on June 14 and in different root conditions of 6 years old on October 1. Respiration rate of each organ was the increasing order of young berry, leaf, peduncle, stem and root at all temperature(15$^{\circ}C$ to 30$^{\circ}C$). Temperature coefficients of respiration rate from 15$^{\circ}C$ to 30$^{\circ}C$ of each organ were 2.39 in young berry, 1.75 in leaf, 1.57 in root, 1.17 in stem and 1.16 in peduncle. There was no difference between respiration rate of large size root (117${\pm}$8.8g) and that of small size (54${\pm}$4.0g) in 6 years old ginseng. Respiration rate was decreased with the decrease in the water content in root, especially in small size root. And respiration rate of red skin root was higher than that of healthy root.

Panax ginseng Meyer (Korean ginseng) is in the spotlight of Oriental medicine and is proclaimed as the king of medicinal plants owing to its adaptogenic characteristics. Ginseng root rot is a devastating disease caused by the fungus, Ilyonectria mors-panacis that generally attacks younger roots (~2 years), leading to defects in root quality, ginsenoside accumulation and also life cycle of the plant. Hence, there is an indispensable need to develop strategies resulting in tolerance against ginseng root rot. In the present study, we evaluated the effect of silica nanoparticles(N-SiO2) in Panax ginseng during I. mors-panacis infection. Long term analysis (30 dpi) revealed a striking 50% reduction in disease severity index upon 1mM and 2mM treatment of N-SiO2. However, N-SiO2 did not have any direct antifungal activity against I. mors-panacis. Membrane bound sugar efflux transporter, SWEET (Sugars Will Eventually be Exported Transporters) was identified in ginseng and as expected, its expression was suppressed upon N-SiO2 treatment in the root rot pathosystem. Furthermore, the total and reducing sugars in the apoplastic fluid clearly revealed that N-SiO2 regulates sugar efflux into apoplast. In a nut shell, N-SiO2 administration induces transcriptional reprogramming in ginseng roots, leading to regulated sugar efflux into apoplast resulting in enhanced tolerance against I. mors-panacis.
