A Convenient HPLC/ELSD Method for the Quantitative Analysis of Betaine in Lycium chinense

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

약침용 홍삼추출액의 급성독성시험에 관한 연구 (Studies on the Safety Assessment of Red-ginseng Radix Extract Solution for Herb-acupuncture)

  • The components of Red-ginseng radix extract solution for herb- acupuncture were analyzed by HPLC. According to the Medical Product Safety Administration Guidelines for safety assessment, mice and rats were used for acute toxicity test. The results were summarized as follows; 1. In the Saponin contents(%) of Red-ginseng radix extract, Ginsenoside $Rb_1$ Saponin was 0.27% in raw material, 1.67% in extract powder and Ginsenoside Rc Saponin was 0.16% in raw material, 1.12% in extract powder and Ginsenoside Rd Saponin was 0.08% in raw material, 0.54% in extract powder. 2. There were no abnormal findings in acute toxicity test treated with Red-ginseng radix extract solution for herb-acupuncture and $LD_{50}$ could not be measured.

  • Morphine produces wide-spread immunesuppression, such as lymphokine production, phagocytic activity, natural killer cell activity, cell proliferation, and cell-mediated immunity. On the other hand, one of the representative pharmacological act ions of Panax ginseng is immune enhancement. In this study, we investigated the effects of ginseng total sponin (GTS), and Adaptagen (trade name of ginseng product) on morphine-induced immunesuppression. Morphine hydrochloride (10 mg/kg, sc), GTS (400 mg/kg, oral), Adaptagen (400 mg/kg, oral ) were administered to mice for 14 consecutive days.

북한의 인삼 산업 현황과 연구 동향 (Current Status and Trends of the Ginseng Industry and Research in North Korea)

  • 인삼은 우리나라를 대표하는 약용작물로 남한뿐만 아니라 북한에서도 매우 중요하게 취급되고 있다. 그러나 북한에 대한 정보 접근이 어렵기 때문에 실제 북한에서 인삼의 재배, 연구와 개발 동향, 관련 산업 현황에 대해서는 잘 알려지지 않고 있다. 본 논문에서는 비록 제한적이긴 하나 구할 수 있는 서지 자료를 통해 북한에서의 인삼 산업 현황과 연구 동향을 파악하고자 하였다. 북한약전에는 인삼이 "고려인삼"으로 수재되어 있으며 '개성지방에서 재배한 6년생 인삼의 뿌리'로 정의하고 있다. 또한 북한약전에는 22종의 인삼제제가 수재되어 있다. 북한에서는 필수약품(2002) 중 건위소화약, 지사약, 보약 등 10개 약품에 인삼이 들어가고, 그 외에도 다양한 건강보조식품과 화장품, 치약 등이 개발되어 유통·판매되고 있다. 2014년 이후 북한에서 인삼 산업과 연구가 전반적으로 활발해지고 있다. 이때 개성의 인삼재배지역을 대폭 확장하고 설비를 보수하였다. 2016년, 개성고려인삼가공공장을 무균화, 현대화, 무진화된 설비로 정비하여 가동 중이라고 한다. 2017년 이후 품질관리 연구에 관심을 돌려 2019년 품질관리규정과 인증제를 도입하기에 이르렀다. 1990년대에 인삼 제품개발 연구가 많았고 2000년부터 인삼의 약리작용 및 치료·임상연구에 대한 연구가 보고되었다. 또한, 수확량을 높이기 위한 인삼재배와 인삼가공업 관련 연구가 중시되고 있다. 인삼은 우리나라를 대표하는 약용작물이라는 점, 남과 북이 모두 중요시하고 있다는 점, 남과 북이 상호 협력하면 큰 시너지 효과를 얻을 수 있다는 점에서 남북 교류협력에 있어서 매우 적합한 소재이다.

Induction of Nrf2/ARE-mediated cytoprotective genes by red ginseng oil through ASK1-MKK4/7-JNK and p38 MAPK signaling pathways in HepG2 cells

  • Background: The induction of cellular defensive genes such as phase II detoxifying and antioxidant enzymes is a highly effective strategy for protection against carcinogenesis as well as slowing cancer development. Transcription factor Nrf2 (nuclear factor E2-related factor 2) is responsible for activation of phase II enzymes induced by natural chemopreventive compounds. Methods: Red ginseng oil (RGO) was extracted using a supercritical $CO_2$ extraction system and chemical profile of RGO was investigated by GC/MS. Effects of RGO on regulation of the Nrf2/antioxidant response element (ARE) pathway were determined by ARE-luciferase assay, western blotting, and confocal microscopy. Results: The predominant components of RGO were 9,12-octadecadienoic acid (31.48%), bicyclo[10.1.0] tridec-1-ene (22.54%), and 22,23-dihydrostigmasterol (16.90%). RGO treatment significantly increased nuclear translocation of Nrf2 as well as ARE reporter gene activity, leading to upregulation of heme oxygenase-1 and NAD(P)H:quinone oxidoreductase 1. Phosphorylation of the upstream kinases such as apoptosis signal-regulating kinase (ASK)1, mitogen-activated protein kinase (MAPK) kinase (MKK)4/7, c-Jun N-terminal kinase (JNK), and p38 MAPK were enhanced by treatment with RGO. In addition, RGO-mediated Nrf2 expression and nuclear translocation was attenuated by JNK inhibitor SP600125 and p38 MAPK inhibitor SB202190. Conclusion: RGO could be used as a potential chemopreventive agent, possibly by induction of Nrf2/ARE-mediated phase II enzymes via ASK1-MKK4/7-JNK and p38 MAPK signaling pathways.

전통적 탁주증자법으로 처리한 홍삼의 일부 항산화 및 항암효과 (Antioxidant and Anticancer Activities of Ginseng Treated with Traditional Rice Wine Steam Process Method)

  • 본 연구는 인삼의 가공방법을 개발하고 제품을 다양화하기 위해 우리나라의 전통주인 탁주로 인삼을 증자한 후 홍삼의 항산화성 및 아질산염 소거능, 항암효과를 비교 연구하였다. $A1\~~A9$시료의 $60\%$ 에탄올 추출물에서의 DPPH 라디칼 소거효과는 1,000 ppm에서 인삼의 증자 횟수와 비례하여 증가하였고, 특히 A3에서 $79.5{\pm}3.3\%$로 라디칼 소거율이 뚜렷하게 증가함을 보였다. 아질산염 소거능은 pH 1.2에서 W.G는 $25.9{\pm}4.4\%$, R.G는 $12.9{\pm}1.1\%$, $A1\~A9$$26.2{\pm}0.1\~56.1{\pm}0.5\%$로 증자 횟수가 증가할수록 높은 아질산염 소거능을 나타냈다. 인체유래 간암세포(Hep3B)에서의 항암효과로 실험에서 W.G는 에탄올 추출물 5,000 ppm으로 처리했을 때, $19.6{\pm}4.5\%$로 나타나 증식 억제율이 미미하였으나, R.G는 에탄을 추출물 5,000 ppm으로 처리했을 때 $54.5{\pm}6.1\%$를 나타냈으며, A9에서는 $96.3{\pm}2.4\%$로 높은 항암효과 가 관찰되었다. 이상의 결과로 볼 때 탁주를 이용한 증자홍삼이 일반 홍삼 및 백삼보다 본 실험의 항산화 및 항암 system(in vitro)을 이용하여 실험한 결과 더 높은 항암 및 항산화 효과를 나타내었다.

  • To investigate the possibility of Panax ginseng as a therapeutic agent for the immune suppression, ginseng total saponin (GTS) extracted from korean red ginseng was tested on immune functions from morphine-induced immune suppressed mice. To study how immune functions are affected by morphine and also to test whether GTS can be an useful therapeutic agent for morphine toxicity, several parameters were employed, body weight, immune organ weight, B cell functions, and T cell function. Morphine impaired the development of body weight and immune organ weight such as spleen and thymus. Morphine also depressed a B-cell function, antibody production. T-cell functions studied by type IV hypersensitivity test were most markedly affected by morphine treatment. GTS restored most of morphine-induced immune suppression. GTS restored the morphine-induced decrease in spleen weight to body weight ratio in a dose dependent manner, but not the body weight decrease. Also all of the morphine-induced impairments of B cell functions and cellmediated immunity were fully recovered by GTS. These results suggest that ginseng product could be very helpful for the treatment of immune suppression occurring in morphine abusers.

Biotransformation of Ginsenosides by Eoyukjang-derived Lactic Acid Bacteria in Mountain-cultivated Ginseng

  • Biotransformation of ginsenosides by microorganisms alters the absorption and bioavailability of ginseng as a medicinal herb. In this study, we isolated two kinds of fermenting microorganisms from Eoyukjang, which is a traditional Korean fermented food made from soybean. Next, we identified and detected their ability to convert major ginsenosides to compound K. The two microorganisms, referred to as R2-6 and R2-15, had 100% similarity with Lactobacillus plantarum subsp. plantarum ATCC $14917^T$ and Lactobacillus rhamnosus JCM $1136^T$, respectively. The optimal pH and growth temperature of the isolates were determined to be pH 6-7 and $30^{\circ}C$. After fermentation for 30 days, the major ginsenosides in the mountain-cultivated ginseng were transformed to the highly bioactive ginsenoside, compound K, in the final product.

  • Objectives The aim of this experiment is to provide an differentiation of ginseng, red ginseng, cultivated wild ginseng(CWG), and red wild ginseng(RWG) through component analysis using HPLC(High Performance Liquid Chromatography, hereafter HPLC). Methods Comparative analyses of ginsenoside $Rg_3$, ginsenoside $Rh_2$, and ginsenosides $Rb_1$ and $Rg_1$ of various ginsengs were conducted using HPLC. Results 1. CWG was relatively heat-resistant and showed slow change in color during the process of steaming and drying, compared to cultivated ginseng. 2. Ginsenoside $Rg_3$ was not detected in cultivated ginseng and CWG, whereas it was high in red ginseng and RWG. Ginsenoside $Rg_3$ was more generated in red ginseng than in RWG. 3. Ginsenoside $Rh_2$ appreared during steaming and drying of cultivated ginseng, whereas it was more increased during steaming and drying of CWG. 4. Ginsenoside $Rg_1$ content was more increased during steaming and drying of cultivated ginseng, whereas it was more decreased during steaming and drying of CWG. 5. Ginsenoside $Rb_1$ content was increased about 500% during steaming and drying of cultivated ginseng, whereas it was increased about 30% during steaming and drying of CWG, indicating that ginsenoside $Rb_1$ was more generated in red ginseng than in RWG. 6. Ginsenoside $Rg_3$ content was higher, whereas ginsenoside $Rg_1$ content was lower in 11th RWG than in 9th RWG, indicating that ginsenoside $Rg_3$ content was increased and $Rg_1$ content was decreased as steaming and drying continued to proceed. Ginsenoside $Rh_2$ and $Rb_1$ contents began to be increased, followed by decreased after 9th steaming and drying process. Conclusions Above experiment data can be an important indicator for the dentification of ginseng, red ginseng, CWG, and RWG. And the following studies will be need for making good product using CWG.

The Change of Ginsenoside Composition in the Ginseng (Panax ginseng) Flower Buds by the Ultrasonication and Vinegar Process

  • The purpose of this study was to develop a new ginseng (Panax ginseng) flower buds extract with the high concentration of ginsenoside Rg3, Rg5, Rk1, Rh1 and F4, the Red ginseng special component. Chemical transformation from the ginseng saponin glycosides to the prosapogenin was analyzed by the HPLC. The ginseng flower buds were processed at the several treatment conditions of the ultrasonication (Oscillator 600W, Vibrator 600W) and vinegar (about 14% acidity). The result of UVGFB-480 was the butanol fraction of ginseng flower buds that had been processed with ultrasonication and vinegar for 480 minutes gained the highest amount of ginsenoside Rg5 (3.548%), Rh1 (2.037%), Rk1 (1.821%), Rg3 (1.580%) and F4 (1.535%). The ginsenoside Rg5 of UVGFB-480 was found to contain 14.3 times as high as ginseng flower buds extracts (GFB, 0.249%).
